One-pot synthesis of novel ferrocene grafted N-methyl-spiropyrrolidines has been accomplished in good yields through a facile 1,3-dipolar cycloaddition reaction of various azomethine ylide derived from ninhydrin and sarcosine with various ferrocene derivatives as dipolarophile. The regiochemical and stereochemical outcome of the cycloaddition reaction is ascertained by X-ray crystallographic studies of one of the cycloadducts.  相似文献

A facile and rapid one-pot synthesis of ferrocene bearing novel spiropyrrolizidines has been accomplished by [3+2]-cycloaddition reaction of various azomethine ylides derived from isatin/5,7-dibromoisatin and l-proline with various ferrocene derivatives as dipolarophile in good yield. A Lewis acid-catalyzed [3+1+1] cycloaddition reaction between aliphatic isocyanides and azomethine ylides generated in situ from aziridines, leading to pyrrolidine derivatives, has been developed. This reaction proceeds smoothly under mild conditions and can also be modified by employing aromatic isocyanides to generate four-membered heterocycles, azetidines, through a [3+1] cycloaddition reaction.  相似文献

Highly regio- and stereoselective synthesis of glucosylspiro-oxindole/acenaphthenone derivatives was accomplished through 1,3-dipolar cycloaddition (1,3-DC) reaction. The glucosyl α,β-unsaturated-7,3-lactone (a sugar lactone) derived from d-glucose was used as a chiral dipolarophile in 1,3-DC reaction for the first time. The azomethine ylides generated from sarcosine/l-proline/thiazolidine-4-carboxylic acid/piperidine-2-carboxylic acid with isatin and acenaphthoquinone were trapped by the sugar lactone to give a series of glucosylspiro-pyrrolidine derivatives in good yields.  相似文献

Synthesis of pyrrolidinyl-spirooxindoles fused to sugar lactone has been achieved by a one pot three component 1,3-dipolar cycloaddition (1,3-DC) reaction. A unique dipolarophile (α,β-unsaturated lactone) derived from d-glucose/d-galactose reacted with azomethine ylide generated in situ from isatin/N-substituted isatin and secondary amino acids (sarcosine/proline/piperidine-2-carboxylic acid) to give the corresponding cycloadducts in good yield. A chiral thiourea catalyst possessing an amine function catalyzes an asymmetric [3+2] cycloaddition of azomethine ylides to nitroolefins to provide highly functionalized pyrrolidines with high diastereo- and enantioselectivities (up to 98:1:1 dr, 92% ee). The reaction proceeds in a stepwise manner consisting of Michael addition and subsequent intramolecular aza-Henry reaction. Both reactions are promoted by the thiourea catalyst, and the reaction rate of the latter step is efficiently enhanced by the addition of 2,2,2-trifluoroethanol.  相似文献

An efficient synthesis of dispiro-oxindolopyrrolizidine and dispirooxindolothienopyrrole derivatives has been expediently accomplished via a one-pot, three-component 1,3-dipolar cycloaddition reaction. High regioselectivity was achieved on ultrasonication in the presence of silica as a catalyst. The regio- and stereochemical polar [3+2] cycloaddition of the azomethine ylides, which were generated in situ by the reaction of isatin derivatives and proline, with trans-β-nitrostyrene and (E)-1-phenyl-2-nitropropene were studied using experimental and theoretical methods. In comparison with trans-β-nitrostyrene, when the reactions were performed with (E)-1-phenyl-2-nitropropene, a remarkable inversion in the regioselectively was observed. The regioselectivity of the reactions was investigated using global and local reactivity indices and frontier molecular orbital (FMO) analysis at the B3LYP/6-31G(d,p) level of theory. The effects of the electronic and steric factors on the regioselectivity of the reactions were discussed. The inspection of geometries and energetics of transition states revealed the importance of weak interactions in regioselectivity of the cycloaddition reactions.  相似文献

2-Arylidene-1,2,3,4-tetrahydro-naphthalen-1-ones undergo regioselective 1,3-dipolar cycloaddition reaction with the azomethine ylide derived from isatin and sarcosine by decarboxylative route affording a series of 1-N-methyl-spiro[2.31]oxindole-spiro[3.211]111, 211,311,411, -tetrahydro-naphthalen-111-one-4-aryl-pyrrolidines. 2-Arylidene-1,2,3,4-tetrahydro-naphthalen-1-ones also undergo cycloaddition reaction with azomethine ylide derived from paraformaldehyde and sarcosine affording a series of 1-N-methyl-spiro[3.21]11,21,31,41-tetrahydro-naphthalen-11-one-4-aryl-pyrrolidines. Expeditious syntheses of cis-1-methyl-2,3,3a,4,5,9b-hexahydro-1H-pyrrolo-[3,2h]isoquinoline/[2,3-f]quinoline have been developed. The syntheses started with commercially available materials and afforded excellent overall yields in straightforward steps. Intramolecular azomethine ylide-alkene [3+2] cycloaddition is the key step in the construction of these pyrroloisoquinoline and pyrroloquinoline scaffolds. This route is much more atom-economic than those reported in the literature and is appropriate for scale-up synthesis.  相似文献

Efficient synthesis of tricyclic indole derivatives bearing a substituent at the 3-position of the indole nucleus was achieved by the [3+2] cycloaddition reaction of transition metal-containing azomethine ylides derived from N-(o-alkynylphenyl)imines with vinyl ethers. Third-row transition metal complexes, especially PtCl2, turned out to be highly efficient for the reaction of internal alkynes and imidate substrates with wide generality. Moreover, as strong support for the reaction mechanism, the intermediate Pt-carbene complex was found to undergo intramolecular C-H bond insertion reaction to give tetracyclic indoline derivatives when benzyl vinyl ether was employed as a dipolarophile. This protocol provided a facile synthesis of highly functionalized tricyclic indole derivatives found as the basic skeleton of the mitosene family, such as mitomycin C.  相似文献

AgOAc/ThioClickFerrophos complex-catalyzed 1,3-dipolar cycloaddition of glycine imino ester, the precursor of azomethine ylide, with aryl- and alkylidene malonates afforded the corresponding exo-cycloadducts, that is, proline ester derivatives in high yields with high enantiomeric excess (up to 99% ee).
