Cyclic RGD peptides are potent antagonists for the αvβ3 integrin receptor. In this Letter, microwave-assisted solid-phase synthesis of cyclic RGD peptides is described. In a coupling reaction between Fmoc-Arg(Pbf)-OH and high-loading H-Gly-Trt(2-Cl) resin, multiple coupling reactions were required for completion under the conventional HBTU activation. We found that the use of COMU, a new coupling reagent, under microwave heating to 50 °C accelerated the reaction even inside the resin. This method was applicable to the synthesis of linear pentapeptides, H-Asp(OtBu)-Xxx-Yyy-Arg(Pbf)-Gly-OH (Xxx = d-Phe(p-Br) or d-Tyr, Yyy = Lys(Boc) or MeVal). Cyclization of these peptides followed by deprotection gave the desired cyclic RGD peptides with high purity.  相似文献

Cu2+ complexes with peptides containing three histidine residues have very specific metal binding abilities and can mimic the structures of various multi-histidine metal binding sites in proteins. The main goal of the work concerns the investigations of coordination abilities of the group of N-terminally protected Ac-His-Arg-His-Gly-His-Gly, Ac-His-Gly-His-Arg-His-Gly, Ac-Gly-His-His-Arg-His-Gly and Ac-His-His-Gly-His-Arg-Gly, and their unprotected analogs His-Arg-His-Gly-His-Gly, His-Gly-His-Arg-His-Gly, Gly-His-His-Arg-His-Gly and His-His-Gly-His-Arg-Gly towards Cu2+ ions. Detailed spectroscopic (UV/Vis, CD and EPR) and potentiometric studies have been made. The stoichiometry and binding mode for each ligand–Cu2+ system were determined.  相似文献

Linear and cyclic olefin peptides containing the substrate sequence for human T-cell leukemia virus type-1 (HTLV-1) were efficiently synthesized on a solid support using the Horner-Emmons reaction. The precursor peptide aldehyde was prepared by oxidation of the corresponding peptide alcohol with Dess-Martin periodinane. The oxidation reaction proceeded quantitatively on a cross-linked ethoxylate acrylate resin (CLEAR) support instead of a polystyrene-based support. Cyclization on the solid support was achieved via an amide bond formation mediated by EDC/HOAt to yield a single major product. The linear olefin peptide was cleaved by HTLV-1 protease at the scissile site, whereas the cyclic olefin peptide functions as a competitive inhibitor rather than a substrate.  相似文献

An aminooxy-containing peptide, the nucleophile partner for oxime ligations, is usually grafted on a NH2-peptide resin by activating a protected aminooxyacetic acid as an active ester. Here, we have shown that its subsequent coupling to NH2-peptide resin competes with the overacylation of the -NH-O- nitrogen and that the overacylation level increases with the basicity of the reaction mixture. Moreover, we found that overacylation is prevented when the COOH of the Aoa-derivatives is engaged in an amide bond.  相似文献

In the course of the chemical synthesis of human protein mitogaligin, we present here a simple method to prepare peptide thioesters using Fmoc chemistry. The hydroxyl side chain of serine was reacted with a trichloroacetimidate Wang resin to anchor it on solid phase. After peptide elongation and orthogonal unmasking of the C-terminus, the amino thioester was introduced under optimized conditions to avoid epimerization.  相似文献

Chain-growth condensation polymerization of 3-(4-octyloxybenzylamino)benzoic acid esters bearing an alkoxy group on the benzene ring was investigated for the synthesis of polyamides having a specific conformation owing to intramolecular hydrogen bonding of CONH?OR. The 4-octyloxybenzyl group on the nitrogen of the amide linkage was easily removed with trifluoroacetic acid after polymerization to afford the desired polyamides, which had lower solubility than expected. Furthermore, we found that a cyclic triamide was selectively obtained by slow addition of the base to a solution of the monomer.  相似文献

Sodium dithionite initiated reactions of 1-bromo-1-chloro-2,2,2-trifluoroethane (1) with methyl and trimethylsilyl ethers of cyclopentanone and cyclohexanone enols (2a-d) in a MeCN/H2O system were investigated. 2-(2,2,2-Trifluoroethylidene)cyclopentanone (4a) and 2-(2,2,2-trifluoroethylidene)-cyclohexanone (4b), respectively, were obtained as the main products and isolated in reasonable yields. The reaction with a 1:1 mixture of 5- and 3-methyl substituted 1-methoxycyclohexenes, 2e and 2f, revealed strong influence of steric hindrance on the reaction rate; a mixture of 2-(2,2,2-trifluoroethylidene)-5-methylcyclohexanone (6) and 2-(2,2,2-trifluoroethylidene)-3-methylcyclohexanone (7) in a 9:1 ratio was formed. Ketones 4a and 4b were reduced to the corresponding alcohols 8 and 9 and the reaction of 4b with hydrazine gave an indazole derivative 10.  相似文献

The axially chiral monophosphine 2-(diphenylphosphino)-2′-methoxy-1,1′-binaphthyl (MeO-MOP) is a versatile ligand. We report a shorter (four steps from chiral BINOL), more atom-economical synthetic route to MeO-MOP. (R)-BINOL is transformed into its monomethyl ether by the Mitsunobu reaction, and the latter is reacted with triflic anhydride to give its triflate. The C-P coupling of the triflate and diphenylphosphine oxide catalyzed by palladium give a phosphine oxide, the precursor of (R)-MeO-MOP, which is reduced with trichlorosilane to furnish (R)-MeO-MOP.   相似文献

以(1S,5S)-(-)-α-蒎烯为原料合成了系列新型(1S,2S,3S,5R)-N-烷基-3-蒎胺类化合物. (-)-α-蒎烯经硼氢化氧化、重铬酸吡啶盐(PDC)氧化得到(1S,2S,5R)-(-)-3-蒎酮; 在BF3•(C2H5)2O催化下(1S,2S,5R)-(-)-3-蒎酮与伯胺化合物反应生成Schiff碱, 再经KBH4或NaBH4还原得到(1S,2S,3S,5R)-N-烷基-3-蒎胺类化合物. 采用FT-IR, 1H NMR, 13C NMR和GC-MS等分析手段对合成所得(1S,2S,5R)-N-烷基-3-蒎烷亚胺和(1S,2S,3S,5R)-N-烷基-3-蒎胺类化合物的结构进行了表征. 考察了(1S,2S,3S,5R)-N-烷基-3-蒎胺类化合物对大肠杆菌(E. coli)、金黄色葡萄球菌(S. aureus)、枯草芽胞杆菌(B. subtilis)、荧光假单胞菌(P. fluorescens)、白色念珠菌(C. albicans)、黑曲霉(A. niger)和米根霉(R. oryzae)等细菌和真菌的抑菌和杀菌活性. 结果表明(1S,2S,3S,5R)-N-正庚基-3-蒎胺对真菌和细菌均表现出良好的杀菌和抑菌活性.  相似文献

Synthesis of bis-ornithine, a new Cα,α-disubstituted α-amino acid bearing orthogonally protected α and δ amine groups is reported. Bis-ornithine (bisOrn) and dipeptides containing bis-ornithine have been synthesized in solution up to multigram scale. As a first example, one of these compounds Boc-βAla-bisOrn(Alloc)2-OH has been attached to a solid support and used as template for the synthesis of a symmetrical assemblage of peptides.  相似文献

The guanidinylation of a peptide chain on a polymeric support under microwave conditions using derivatives of thioureas—S-alkylisothioureas, pyrazole-carboxamidine, and guanidine as guanidinylating reagents is described. The best results are obtained with N,N′-di-Z-S-methylisothiourea and N,N′-di-Z(2-Cl)-S-methylisothiourea. It is found that guanidinylation with reagents containing Boc groups is accompanied by side reactions.  相似文献

本文通过2-溴-N-(2-硝基苯)乙酰胺和取代的喹唑啉酮在NaH催化下发生亲核取代反应,再经过氢化、酸化合成了4个新型常山碱类抗球虫药物-N-(2-氨基苯基)-2-喹唑啉酮乙酰胺盐酸盐(1∶1)。其中,2-溴-N-(2-硝基苯)乙酰胺通过溴乙酰氯与邻硝基苯胺反应制备,取代的喹唑啉酮使用取代的邻氨基苯甲酸与甲酰胺反应合成。所有目标化合物的结构均经1H NMR,IR和HRMS等方法确证。  相似文献

N,N′-1,2-phenylenebis(2-thienylideneimine) (PTI) can be obtained via the condensation of o-phenylenediamine and thiophene-2-carboxaldhyde in cyclohexane. It isomerized to its cyclic form 1-(2-thienylmethyl)-2-(2-thienyl)benzimidazole (TMTBI) readily in the presence of Fe2(CO)9. Coordination reactions of PTI with cuprous iodide and silver nitrate in acetonitrile resulted in the formation of [Cu(TMTBI)I]2 and [Ag(TMTBI)2]NO3, respectively, in which the PTI ligand had been isomerized to TMTBI. Both PTI, TMTBI, and complexes were characterized by NMR, IR, and Mass spectroscopies as well as elemental analysis. The crystal structures of TMTBI and [Cu(TMTBI)I]2 were determined by single-crystal X-ray diffraction. Crystallographic details for X-ray structures are as follows. TMTBI is crystallized in the monoclinic space group P21/n with a = 8.953(3), b = 9.150(1), c = 17.436(2) Å, β = 93.73(2)°, and Z = 4. The final R factor was 0.050 (Rw = 0.066) for 1337 observed reflections. Complex [Cu(TMTBI)I]2 is crystallized in the triclinic space group $ {\rm P}\bar 1 $ with a = 9.961 (2), b = 10.825(2), c = 9.184(2) Å, α = 112.98(2), β = 110.78(2), γ = 88.71 (2)°, and Z = 2. The final R factor was 0.056 (Rw = 0.057) for 2363 observed reflections.  相似文献

本文采用配体1, 2-二(邻氨基苯氧基)乙烷(L)分别与AgPF6, AgCF3SO3, AgNO3和AgSbF6 进行配位反应,依次得到了四个配合物1 [Ag2(L)2(PF6)]、2 [Ag2(L)2(CF3SO3)2]、3 [Ag(L)NO3]n 和4 [Ag(L)2SbF6]n,并通过FTIR、元素分析、以及X射线单晶衍射等对配合物的结构与组成进行了表征。单晶衍射结果表明,配合物1和2为双核银(I)配合物,3和4为银(I)的配位聚合物。配合物1具有穴状结构,2经Ag?Ag键桥连两个配体形成扭曲的非平面结构。聚合物3的结构为一维(1D)“之”字链,4具有三维(3D)多孔的结构框架。在四个配合物结构中,相应的抗衡阴离子均未参与Ag(I)进行配位作用。  相似文献

A method for the solid phase chemical synthesis of AMPylated Ser/Thr containing peptides is described. Peptides were phosphonylated using the H-phosphonate method, then the adenosine moiety was introduced using 2′,3′-isopropylidene adenosine and the condensing reagent PyBOP. Oxidation with iodine yielded the desired AMPylated product in good yield following acidolytic cleavage from the resin support.  相似文献

An efficient, inexpensive, and heterogeneous catalyst, [Al(H2PO4)3], was applied in a three-component, one-pot cyclocondensation reaction of isatoic anhydride with primary amines (or ammonium salts) and aldehydes to afford the corresponding quinazolinone derivatives in excellent yields. Reactions occurred under thermal solvent-free conditions. It was found that this solid acidic catalyst could be easily recovered and reused for at least three cycles without any loss of activity.  相似文献

用三苯基氢化锡,三对甲苯基氢化锡作为锡氢化试剂与9-乙炔基-9-芴醇进行反应,合成了2个有机锡化合物:[Z]-2-(三苯基锡基)-1-(9-芴醇)乙烯(1)和[Z]-2-(三对甲苯基锡基)-1-(9-芴醇)乙烯(2)。化合物1和2分别与ICl,Br~2,I~2反应,得到6个有机锡一卤化物,6个有机锡二卤化物和2个有机锡混合卤化物(3-16)。有机锡一碘化物7,13和有机锡二碘化物8,14与KOH乙醇溶液反应,分别得到相应的有机锡氢氧化物17,18和有机锡氧化物19,20。有机锡二碘化物8,14分别与含氮双齿配体1,10-邻菲罗啉(Phen),2,2'-联吡啶(Bipy),8-羟基喹啉(Oxin)反应,得到6个相应的配合物21-26。26个新化合物通过元素分析,锡含量测定,IR,^1HNMR测定对其结构进行了表征。同时测定了化合物2的晶体结构,晶体属单斜晶系,空间群P2~1/c。化合物2是以Sn原子为中心扭曲的四面体构型。  相似文献
