This paper carries the results of an evaluation of various materials, which may be used to aid in the release of a fused bead from its mould during a wavelength‐dispersive x‐ray fluorescence (WD‐XRF) measurement. The following bead‐releasing agents were studied: NaI, LiBr, NH4I, and LiI. Each was incorporated in different quantities, as a solid and/or in an aqueous solution, together with a flux, into samples of ceramic raw materials. Release agent interference in the WD‐XRF measurement was analysed, and the optimum quantity of release agent needed to obtain suitable beads for WD‐XRF measurement was determined. The best results were obtained for LiI, which yielded reproducible beads without significant interference in the WD‐XRF measurement when a relatively small quantity (0.11 LiI g/bead) was used. The encapsulation-dehydration cryopreservation protocol is critically dependent upon the evaporative desiccation step, which must optimise survival with the retention of glass stability on sample cooling and rewarming. Desiccation is usually achieved evaporatively by drying in a sterile airflow. However, chemical desiccation using silica gel has advantages for laboratories that do not have environmental control and/or which are exposed to high relative humidities and risks of microbial contamination. This study characterised thermal profiles of silica gel-desiccated encapsulated shoot-tips of two Ribes species using Differential Scanning Calorimetry. For both species silica gel-desiccation at 16 degrees C for 5 h decreased bead water content from ca. 75 to 28% fresh weight (3.8 to 0.4 g x g(-1) dry weight); further desiccation (for 6 and 7 h) reduced the bead water content to 21% (0.3 g x g(-1) dry weight). These changes in water status altered the thermal properties of beads for both species. After 7 h desiccation over silica gel stable glass transitions were observed on both cooling and rewarming of beads containing meristems. Tg mid-point temperatures ranged from -78 to -51 degrees C (cooling) and from -88 to -54 degrees C (warming) [at cooling and warming rates of 10 and 5 degrees C min(-1), respectively] after 5 to 7 h silica gel-desiccation. Post-cryopreservation viability of both species was ca. 63%. Thermal analysis studies revealed that an encapsulation/dehydration protocol using silica gel as a desiccant should comprise a minimum 5 h drying (at 16 degrees C). This reduces bead moisture content to a critical point (ca. 0.4 g x g(-1) dry weight) at which stable glasses are formed on cooling and rewarming. It is concluded that silica gel has advantages for use as a desiccant for alginate-encapsulated plant meristems by promoting stable vitrification and is useful in laboratories and/or geographical locations where environmental conditions are not under stringent control.  相似文献

We study the jamming of bead assemblies placed in a cylindrical container whose bottom is pierced with a circular hole. Their jamming behavior is quantified here by the median jamming diameter, that is the diameter of the hole for which the jamming probability is 0.5. Median jamming diameters of monodisperse assemblies are obtained numerically using the Distinct Element Method and experimentally with steel beads. We obtain good agreement between numerical and experimental results. The influence of friction is then investigated. In particular, the formation of concentric bead rings is observed for low frictions. We identify this phenomenon as a boundary effect and study its influence on jamming. Relying on measures obtained from simulation runs, the median jamming diameter of bidisperse bead assemblies is finally found to depend only on the volume-average diameter of their constituting beads. We formulate this as a tentative law and validate it using bidisperse assemblies of steel beads.  相似文献

We present a new method to calibrate a quadrant photodiode used as position detector to monitor latex beads trapped in optical tweezers. The method combines the dragging Stoke’s force with the thermal noise analysis (power spectral density (PSD)) of the Brownian motion of the trapped bead. Position detector calibrations used in other similar methods normally utilise a bead attached to the coverslip: the voltage-position calibration factor is found by translating the bead across the waist of a laser beam. The so determined calibration factor is then assumed to be the same when beads are investigated in the optical trap. This procedure presents some drawbacks since attached beads can be affected by proximity effects due to the coverslip glass surface which alter the position sensor response itself. On the contrary, our method is able to provide, simultaneously, the calibration factor, the trap stiffness, and the local viscosity of the medium making use of a single trapped bead.  相似文献

Colloidal particles coated by polyelectrolyte multilayers of alternatingly positive and negative charge are shown to interact strongly with lipid vesicles. We have studied two cases: (i) the interaction between beads and small unilamellar vesicles (vesicles diameter smaller than the particles one), where we found evidence for coating of the beads with lipid bi- or multilayers in the form of an increase in bead diameter and changes in the beads surface potential; (ii) the interaction of beads with giant vesicles (vesicles larger than the particles), where we observed by fluorescence microscopy the spreading of the vesicle on the bead manipulated with an optical tweezer. Giant fluctuations of the vesicles are suppressed due to the adhesion of the vesicle to the bead and direct observation of the coating process shows that lipid coverage is not limited to the direct vesicle-bead contact area, but is rather extended to the entire bead. A set of tank experiments was performed to investigate acoustic transmission across a roughened fluid-fluid interface with the intention to test heuristic Bragg scattering predictions used to explain observations of anomalous transmission in field experiments. In the tank experiments, two immiscible fluids (vegetable oil floating on glycerin) formed the layers. Small polystyrene beads were floated at the interface to simulate roughness. An array of hydrophones placed in the bottom layer (glycerin) was used to measure the acoustic levels transmitted across the interface. This array was also employed as a beamformer to determine the apparent angle and sound speed of the scattered signals. Data were acquired at subcritical grazing angles in the frequency range of 100-200 kHz for three different bead diameters and for various configurations in which the locations of the beads floating on the interface were varied. Results of these measurements demonstrated that a significant amount of acoustic energy can be scattered into the bottom layer by beads floating at the interface. The scattered levels increased with increasing bead diameter. However, discrepancies occurred between observed propagation properties and the Bragg predictions. By comparing the processed tank data to a computer simulation of the same it was determined that these discrepancies are a consequence of near-field reception of the scattering by the bead array and ignoring the directionality of the scattering by the beads. Consequences to observations made in field experiments are discussed.  相似文献

We investigate the physical meaning of coarse-grained beads generated by coarse graining of nonbonded particles such as solvent molecules in a solution. Starting from the partition function, we analytically coarse grain an N-particle fluid to a system containing N-2 of the original particles plus a bead representing the two remaining particles. As a direct consequence of the lack of bonding interactions, the resulting effective potential becomes independent of the bead coordinates, i.e., ideal-gas-like, in the thermodynamic limit. Thus, there are no conservative forces between coarse-grained beads representing assemblies of nonbonded molecules nor between these beads and any other species in the system.  相似文献

We report an experimental study of the statistical properties of vibrated granular rings. In this system, a linked rod and bead metallic chain in the form of a ring is collisionally excited by a vertically oscillating plate. The dynamics are driven primarily by inelastic bead-plate collisions and are simultaneously constrained by the rings' physical connectedness. By imaging many instances of the ring configurations, we measure the ensemble averages and distributions of several physical characteristics on the scale of individual beads and composite ring. We study local properties such as inter-bead separation and inter-bonds angles, and global properties such as the radius of gyration and center-of-mass motion. We characterize scaling with respect to the size of the chain.  相似文献

Optical tweezers are very often used for measurement of piconewton range forces. Depending on the displacement of the trapped bead, the trap may become stiffer which causes considerable underestimation of the measured force. We have shown, both by theory and experiment, that such a stiffening occurs for beads larger than 0.5 μm in radius. For the first time, we have shown that the displacement at which the stiffening starts is size dependent and that the stiffening starts at higher forces for larger beads. We have shown that for the applications, which simultaneous force measurement and position sensing are on demand (such as biopolymer stretching), mid-range sized (~1.5 μm in radius) beads could be the best choice.  相似文献

We report an experimental study of the heat transport along a chain of macroscopic beads: at one end, one bead is periodically heated and we record the resulting temperature variations of another bead as a function of position and time. The experimental results show that the chain behaves like a high-order low-pass filter. The measurement of the associated cut-off frequency makes it possible to determine accurately the resistance of the bead-bead contact: we make use of the experimental setup for studying the effects of liquid bridges decorating the bead-bead contacts, which provides important clues for understanding the thermal properties of partially wet granular matter.  相似文献

Locomotion of bacteria by actin polymerization and in vitro motion of spherical beads coated with a protein catalyzing polymerization are examples of active motility. Starting from a simple model of forces locally normal to the surface of a bead, we construct a phenomenological equation for its motion. The singularities at a continuous transition between moving and stationary beads are shown to be related to the symmetries of its shape. Universal features of the phase behavior are calculated analytically and confirmed by simulations. Fluctuations in velocity are shown to be generically non-Maxwellian and correlated to the shape of the bead.  相似文献

We have determined the fluorescence yield of stained micro beads, used for calibration purposes in flow cytometry, as function of the irradiance of the exciting laser beam. A rate equation model has been applied to derive the number of fluorescence molecules carried by each micro bead. To derive in situ photo-physical properties of the specific dye, required for the rate equation model, we discuss an approach based on flow cytometric sorting of micro beads, which have passed two laser beams with properly chosen different irradiances, and subsequent observation of single molecule bleaching employing high sensitivity microscopy. The feasibility of our approach is demonstrated presenting first results concerning saturation of fluorescence of beads in flow and single molecule bleaching by high sensitivity microscopy.  相似文献

We have studied the effect of small amounts of added liquid on the dynamic behavior of a granular system consisting of a mixture of glass beads of two different sizes. Segregation of the large beads to the top of the sample is found to depend in a nontrivial way on the liquid content. A transition to viscoplastic behavior occurs at a critical liquid content, which depends upon the bead size. We show that this transition can be interpreted as a condensation due to the hysteretic liquid bridge forces connecting the beads, and we provide the corresponding phase diagram.  相似文献

Arabidopsis thaliana shoot tips were successfully cryopreserved using encapsulation-dehydration cryopreservation methods. Between one and seven shoot tips were encapsulated within 4 mm calcium-alginate beads. Beads were formed in the presence of 2 M glycerol + 0.4 M sucrose. The time required to make 10 beads, each containing five shoot tips (4 min), was less than the time required to make 50 beads containing one shoot tip (12 min). Shoot tip regrowth after cryoexposure was between 60 and 68%, with one to seven shoot tips per bead. Using five Arabidopsis shoot tips per bead, alginate beads were formed either in the presence of 2 M glycerol + 0.4 M sucrose or 0.5 M sucrose. Beads formed in the presence of glycerol were immediately air-dried to moisture contents between 0.21 to 0.26 g H2O/g FW (0.27 to 0.38 g H2O/g DW). Alginate beads formed in 0.5 M sucrose were incubated in solutions of 0.5, 0.75, and 1 M sucrose for one day each prior to air-dehydration, achieving moisture contents of 0.19 to 0.21 g H2O/g FW (0.23 to 0.27 g H2O/g DW). Shoot tip regrowth after cryoexposure was between 42 and 65%, with no significant differences among treatments. We successfully reduced the amount of time needed for shoot tip processing for Arabidopsis by encapsulating five shoot tips per alginate bead and by using a glycerol-encapsulation method, without lowering shoot tip regrowth levels after cryopreservation.  相似文献

The splash and spill effects caused by the impact of a ball dropped from a height into a granular medium held in a small open container are examined. Different granular media, namely rice, mustard seeds, cream of wheat and plastic beads are used. The quantity of spilled-over granular matter (W, grams) is measured as a function of the ball-drop height and compared for different cases. Digital pictures of the splash process are also recorded. The quantity W is seen to vary approximately linearly with the energy of impact. Interestingly, a distinct upward jump is seen in the spilled quantity at specific impact energy in the case of mustard seeds, which have spherical shape and also exhibit some charging effects. Similar jump was also confirmed for the case of plastic beads with broadly similar properties. Although the parameters such as mass per grain and packing density for the case of mustard seeds are intermediate between those for rice and cream of wheat, the spill quantity for comparable impact energy is considerably higher in the former case. The possible reasons for this non-monotonicity of behavior are discussed in terms of the differences in grain shapes and properties. Experiments are also performed using plastic beads of the same type but with four different sizes to explore the dependence of spilled quantity on bead size. The container size dependence is also examined for various bead types. Interesting systematics are seen, which are discussed qualitatively.  相似文献

A comparative study on the energy dispersive X‐ray fluorescence analytical results of uranium determinations, in uranium and uranium–thorium mixed oxides, using specimens in the form of fused beads and pressed pellets, has been made. It was observed that in case of fusion bead specimens, the intensity of the analyte lines was approximately 1.6 times of that observed in pellet specimens under identical instrumental conditions. In case of uranium oxide samples, the analytical results with bead specimens were slightly better compared with the pellet specimens. However, in case of the uranium–thorium oxide mixtures, the average precision obtained with bead specimen was significantly better (1%, 1 s) in comparison with that achieved using pellet specimens (7%, 1 s). This difference may be due to the hardness of thorium oxide compared with uranium oxide, which affects the homogeneity of the pellet specimens prepared. In fusion bead method of sample preparation, even highly refractory material like ThO2 forms uniform glass beads. Addition of internal standard further improves the analytical results, with reduction in the percent deviation of energy dispersive X‐ray fluorescence results from the expected values to 3% from 7% compared with that obtained using without internal standard. The fusion bead method of sample preparation will be very useful for characterization of sintered (U,Th)O2 pellets, which are highly refractory and difficult to dissolve. We experimentally investigate the segregation of a binary mixture of spherical beads confined between two horizontal vertically vibrating plates. The two kinds of beads are of equal diameter and mass but have different restitution coefficients. Segregation occurs in particular ranges of vibration amplitude and frequency. We find that the collisions between beads at an angle to the horizontal plane induce an effective horizontal repulsive force. When one or both bead types bounce up and down in synchronization, the effective repulsive force between the two types of beads is likely to be larger than that found within a single bead type, resulting in the mixture segregating. Non-horizontal collisions also play a role in stabilizing the segregation state by transferring the horizontal kinetic energy back into vertical motion.  相似文献

We present the apparent mass measurements at the bottom of granular packings for different bead and silo sizes. The redirection parameter K in Janssen theory is found to increase with the ratios of the diameter of the silo to the bead. We attribute this feature to the friction between the beads and the confining wall of silo; it is the role of friction that leads to variations in the shielding of vertical stresses as well as pressure screening.  相似文献
