Starting from 8-oxabicyclo[3.2.1]oct-6-en-3-one and racemic 2,2-dimethyl-8-oxabicyclo[3.2.1]oct-6-en-3-one, the C1-C16 segment of the bryostatins has been synthesized in 30 steps and 9% overall yield (17 steps longest linear sequence). Fragment coupling by dithiane strategy and protecting group manipulations provided an advanced chemodifferentiated northern half segment.  相似文献

Both enantiomers of 8-oxabicyclo[3.2.1]oct-3-en-2-one (6) have been synthesized from 4-hydroxycyclohept-2-enone (3) on the basis of a novel oxidative cyclo-etherification using PhI(OH)OTs (Koser's reagent). (-)-(1S,5R)-8-Oxabicyclo[3.2.1]oct-3-en-2-one [(-)-6, 95% ee] was expeditiously transformed to (+)-sundiversifolide (1).  相似文献

Photocycloadducts from dl-piperitone and 1,1-dimethoxyethylene afforded bicyclo[3.2.1]oct-6-en-8-one derivatives under glc conditions in excellent yields. The synthesis of the title sesquiterpenes was accomplished using the bridged bicyclic compounds as intermediates.  相似文献

β-(2-Oxocyclopentyl)ethylaryl ketones are transformed under acid conditions via a 2-aryl-bicyclo[3.2.1]oct-2-en-8-one intermediate into a mixture of arylcycloheptene carboxylic acids. The scope and the mechanism of the reaction are both discussed.  相似文献

The bicyclic alkenylidenes 9 (bicyclo[3.2.1]oct-2-en-8-ylidene) and 17 (bicyclo[3.3.1]non-2-en-9-ylidene) were claimed to be stabilized foiled carbenes. Our B3LYP and MP2 computations confirm previous experimental data. Moreover, they show that these carbenes are very reactive and rearrange rapidly, mainly through a 1,2-vinyl shift by overcoming a low barrier (1.2 to 5.4 kcal/mol). This is in contrast to the high barriers (up to 30 kcal/mol) predicted for the same type of rearrangements in norborn-2-en-7-ylidene derivatives. In 17 and bicyclo[4.1.1]oct-2-en-7-ylidene (23), the divalent carbon atom is even bent away from the double bond!  相似文献

The lithium aluminium hydride reduction of 2,2,4,4-tetrachloro-8-oxabicyclo[3.2.1]oct-6-en-3-one (8) was reinvestigated. In contrast to most halogeno-substituted oxabicyclic ketones, which give predominantly the corresponding endo alcohols, the expected (3endo)-2,2,4,4-tetrachloro-8-oxabicyclo[3.2.1]oct-6-en-3-ol (9n) is formed in a minute proportion. An X-ray structure analysis of the dominating product gave proof of the exo-alcohol, i.e., (3exo)-2,2,4,4-tetrachloro-8-oxabicyclo[3.2.1]oct-6-en-3-ol (9x). On the other hand, reduction of trichloroketone 11, 2,2,endo-4-trichloro-8-oxabicyclo[3.2.1]oct-6-en-3-one, and the methoxy-substituted chloroketones 13 and 14 provided the corresponding endo alcohols (12 and 15).  相似文献

[reaction: see text] The novel [RhCl(CO)(2)](2)-catalyzed [2 + 2 + 1] cycloaddition of allenenes leading to the bicyclo[4.3.0]non-1(9)-en-8-one as well as the bicyclo[5.3.0]dec-1(10)-en-9-one skeletons has been developed. This method also provides a new procedure for the construction of the bicyclo[4.3.0]non-1(9)-en-8-one skeleton having an alkyl appendage at the ring juncture, which was hardly attained in a satisfactory yield by the Pauson-Khand reaction of the corresponding enynes.  相似文献

Antiaromatic compounds with a closed loop of 4n p-electrons are relatively unstable and often difficult to study. We report in this article the synthesis of alcohols 2-(4'-fluorophenyl)bicyclo[3.2.1]octan-2-ol 11, 2-(4'-fluorophenyl)bicyclo[3.2.1]oct-3-en-2-ol 12, and 2-(4'-fluorophenyl)bicyclo[3.2.1]octa-3,6-dien-2-ol 13 and their transformations into corresponding carbocations 14-16, respectively, in a superacidic medium (FSO3H/SO2ClF) at -120 degrees C. Cations 14-16 are characterized by NMR analysis (1H, 13C, 19F), and 15 and 16 are further characterized by quenching in NaOCH3/H3COH at -120 degrees C. The relative stabilities of 14-16 are determined experimentally by 19F NMR spectroscopy. Cation 16 is found to be experimentally less stable than cation 15 by 3.7 kcal/mol. DFT calculations (structure and energy: B3LYP/6-31G(d); NMR: B3LYP/6-311+G(2d,p)) are performed for alcohols 11-13 and bicyclo[3.2.1]octyl cations 6, 7, 9, 14-16, 26, 28, and 30. In the case of 11-16, data from DFT calculations is in good agreement with experimental data. Because 6,7-dimethylenebicyclo[3.2.1]oct-3-en-2-yl cation 26 is more stable than cation 7 by 1.69 kcal/mol, the inductive effect of sp(2)-hybridized carbon atoms C6 and C7 in carbocations 6 and 16 cannot be the reason for the destabilization of 6 relative to 7 and 16 relative to 15. Destabilization of 6 relative to 7 and 16 relative to 15 and the calculated NICS of 6 (+4.17 ppm) and 16(+3.3 ppm) document that 6 and 16 are bishomoantiaromates.  相似文献

Twelve novel constituents isolated from Burley tobacco condensate by semi-preparative GLC. have been identified as (E)-3,4-epoxy-5-isopropyl-nonane-2,8-dione ( A ), exo-(1-methyl-4-isopropyl-7,8-dioxabicyclo[3.2.1]oct-6-yl)methyl ketone ( B ), exo-1-(1-methyl-4-isopropyl-7,8-dioxabicyclo[3.2.1]oct-6-yl)-ethanol ( C ), (E)-5-isopropyl-8-hydroxy-8-methyl-non-6-en-2-one ( D ), (E)-5-isopropyl-6,7-epoxy-8-hydroxy-8-methyl-nonan-2-one ( E ), endo-2-(1-methyl-4-isopropyl-7,8-dioxabicyclo[3.2.1]oct-6-yl)-propan-2-ol ( F ), 3,3,5-trimethyl-8-isopropyl-4,9-dioxabicyclo[3.3.1]nonan-2-ol ( G ), (E)-5-isopropyl-non-3-ene-2,8-diol ( H ), 5-isopropyl-nonane-2,8-diol ( I ), (E)-5-isopropyl-8-hydroxy-non-6-en-2-one ( J ), 5-isopropyl-8-hydroxy-nonan-2-one ( K ), and (E)-3-isopropyl-6-methyl-hepta-4,6-dien-1-ol ( L ). Compounds A–K were synthesized from norsolanadione ( 2 ), and compound L from 2-isopropyl-5-oxo-hexanal ( 15 ). The relative configuration of the bicyclic internal acetals B, C, F, G and their δ-keto-epoxide precursors A and E is discussed. All these Burley tobacco flavour components belong to a growing family of metabolites structurally related to solanone ( 1 ). They are believed to arise from the breakdown of cembrene-type precursors.  相似文献

The homo-l, 4 adduct obtained from difluorocarbene and bicyclo [2.2.1]hepta-2, 5-diene ( 1 ) was treated successively with HCl, FSO3H and SbF5 in SO2ClF at low temperature. The protic acids underwent electrophilic addition to the cyclopropane part of 1 , giving the corresponding derivatives. However, in FSO3H at ? 50°, protonation of the gem-difluoro grouping also occurred to give the 2-fluoro-6-fluorosulfonylbicyclo [3.2.1]oct-2-en-3-yl cation. The reaction of 1 with SbF5 at ?78° led initially to the formation of the 2-fluorobicyclo [3.2.1 ]octa-2, 6-dien-4-yl cation, which rearranged to 4-fluorotricyclo [5.1.0.05,8]oct-3-en-2-yl cation at ?40°. These rearrangements are discussed in the light of those expected for C8H8F square pyramidal cations.  相似文献

The addition of dichlorocarbene (generated by the interaction of sodium methoxide and ethyl trichloroacetate) to bicyclo[3.2.1]oct-2-ene, its 3-chloro and exo-3,4-dichloro derivatives gives the exo 1 : 1 adducts in yields of 94, 89 and 48%. By suitable chemical reactions of these adducts, convenient syntheses of bicyclo[4.2.1]nona-2,4-diene and bicyclo[4.2.1]non-3-ene, together with their monochloro, dichloro and trichloro derivatives are obtained. Bicyclo[4.2.1]-nonan-3-one is also obtained from bicyclo[4.2.1]non-3-ene in a synthesis starting from the readily available 5-hydroxymethylnorborn-2-ene in an overall yield of 20%.  相似文献

Application of the polybromo ketone-iron carbonyl reaction to natural product synthesis is summarized. The general synthesis of tropane alkaloids has been achieved via the reductive [3+4] cyclocoupling of sym-tetrabromoacetone with N-methoxycarbonylpyrrole as the key step. Ready availability of 8-oxabicyclo[3.2.1]oct-6-en-3-one from the tetrabromoacetone and furan has opened a new, efficient entry to natural C-nucleosides including pscudouridine, pseudocytidinc, and showdomycin. The artificial analogues such as 2-thiopscudouridine, 6-azapseudouridine, pseudoisocytidine, etc., are also obtainable. The oxabicyclic ketones bearing an isopropyl substituent at the appropriate position serve as intermediates for the synthesis of naturally occurring troponoids, nezukone, α-thujaplicin, and hinokitiol (β-thujaplicin). Carbocamphenilone and camphenic acid have been prepared through the [3 + 4] reaction of 1,1,3-tribromo- 3-methylbutan-2-one and cyclopentadiene. The [3+2] cyclocondensation of an α,?a-dibromo ketone and a styrene derivative leads to the single-step synthesis of α-cuparenone. 1,3-Dibromo-3,7-dimethyloct-6-en-2- one derived from nerol (or geraniol) undergoes the biogenctic-type double cyclization. The iron carbonylassisted intramolecular [3 + 2] cyclocoupling gives camphor accompanied by other monoterpenic ketones. A mixture of campherenone and epicampherenone has been obtained from related dibromo ketone prepared from farnesols. The hetero [3+2] reaction by use of dibromo ketones and N,N-dimethylcarboxamides, forming 3 (2H)-furanones, is employable for the preparation of muscarine alkaloid derivatives.  相似文献

The Rh(I)-catalyzed PKR of 1-phenylsulfonyl-1,2-octadien-7-ynes and their aza derivatives exclusively produced the corresponding 9-phenylsulfonylbicyclo[4.3.0]nona-1,6-dien-8-ones and no 4-(phenylsulfonylmethylidene)bicyclo[3.3.0]oct-1-en-3-ones could be detected. Thus, the ring-closing pattern was found to be the same as those of the previously reported 3-phenylsulfonyl-1,2-octadien-7-yne derivatives. However, the formation of 4-(phenylsulfonylmethylidene)-7-oxabicyclo[3.3.0]oct-1-en-3-ones was observed as a minor product when the 5-oxa congeners were used. In addition, a larger ring-sized product, 10-phenylsulfonyl-5-azabicyclo[5.3.0]deca-1,7-dien-9-one derivative, was obtained from the 6-aza derivative of 1-phenylsulfonyl-1,2-nonadien-8-yne.  相似文献

Condensations of amines with 1-chloro cycloheptene in the presence of complex base (NaNH2-t-BuONa) lead to mixtures of enamines and 1,2-cycloheptadiene dimer. Under the same conditions, 2-chloro and 3-chloro bicyclo [3.2.1] oct-2 enes yield only enamines via bicyclo [3.2.1] oct-2 yne. The mechanism of these reactions depends not only upon the halogeno derivatives but also upon “reaction media.” The general problem of the interpretation of elimination-addition reactions is discussed.  相似文献

The proposed total synthesis of hapalindole Q based on bicyclo[2.2. 2]oct-2-ene construction by Diels-Alder reaction and subsequent olefin cleavage is outlined. The β, γ-unsaturated ketones bicyclo[2.2.1]hept-2-en-7-one (10) and 7,7-dimethoxybicyclo[2.2.1]hept-2-en-5-one (15) have been condensed with 1-metalated trans-1-methoxybutadienes (7a or 7b) and 2-isopropenylcyclopentenes (8b or 8c). Oxyanion formation within the resulting alcohols is followed by skeletal rearrangement at room temperature. Careful product analysis has revealed the [3,3] sigmatropic reaction manifold to be followed almost exclusively. Only in the case of 13 is a modest amount (4%) of formal antarafacialretention [1,3] sigmatropic bridgehead carbon migration in evidence. Consequently, the structural features inherent to these alcohols are not conducive to redirecting electronic reorganization to an alternative isomerization process.  相似文献

Racemic 1,1′-methylene[(1RS,1′RS,3RS,3′RS,5RS,5′RS)-8-oxabicyclo[3.2.1]oct-6-en-3-ol] ((±)-6) derived from 2,2′-methylenedifuran has been resolved kinetically with Candida cyclindracea lipase-catalysed transesterification giving 1,1′-methylenedi[(1R,1′R,3R,3′R,5R,5′R)-8-oxabicyclo[3.2.1]oct-6-en-3-ol] (−)-6 (30% yield, 98% ee) and 1,1′-methylenedi[(1S,1′S,3S,3′S,5S,5′S)-8-oxabicyclo[3.2.1]oct-6-en-3-yl] diacetate (+)-8, (40% yield, 98% ee). These compounds have been converted into 1,1′-methylenedi[(4S,4′S,6S,6′S)- and (4R,4′R,6R,6′R)-cyclohept-1-en-4,6-diyl] derivatives.  相似文献
