Abstract

A new nanocomposite was developed by combination of prussian blue (PB) nanoparticles and multiwalled carbon nanotubes (MWNTs) in the matrix of biopolymer chitosan (CHIT). The PB and MWNTs had a synergistic electrocatalytic effect toward the reduction of hydrogen peroxide. The CHIT/MWNTs/PB nanocomposite‐modified glassy carbon (GC) electrode could amplify the reduction current of hydrogen peroxide by ~35 times compared with that of CHIT/MWNTs/GC electrode and reduce the response time from ~60 s for CHIT/PB/GC to 3 s. Besides, the CHIT/MWNTs/PB nanocomposite‐modified GC electrode could reduce hydrogen peroxide at a much lower applied potential and inhibit the responses of interferents such as ascorbic acid (AA) uric acid (UA) and acetaminophen (AC). With glucose oxidase (GOx) as an enzyme model, a new glucose biosensor was fabricated. The biosensor exhibited excellent sensitivity (the detection limit is down to 2.5 µM), fast response time (less than 5 s), wide linear range (from 4 µM to 2 mM), and good selection.  相似文献

In the present study, a novel enzymatic glucose biosensor using glucose oxidase (GOx) immobilized into (3‐aminopropyl) triethoxysilane (APTES) functionalized reduced graphene oxide (rGO‐APTES) and hydrogen peroxide sensor based on rGO‐APTES modified glassy carbon (GC) electrode were fabricated. Nafion (Nf) was used as a protective membrane. For the characterization of the composites, Fourier transform infrared spectroscopy (FTIR), X‐ray powder diffractometer (XRD), and transmission electron microscopy (TEM) were used. The electrochemical properties of the modified electrodes were investigated using electrochemical impedance spectroscopy, cyclic voltammetry, and amperometry. The resulting Nf/rGO‐APTES/GOx/GC and Nf/rGO‐APTES/GC composites showed good electrocatalytical activity toward glucose and H2O2, respectively. The Nf/rGO‐APTES/GC electrode exhibited a linear range of H2O2 concentration from 0.05 to 15.25 mM with a detection limit (LOD) of 0.017 mM and sensitivity of 124.87 μA mM−1 cm−2. The Nf/rGO‐APTES/GOx/GC electrode showed a linear range of glucose from 0.02 to 4.340 mM with a LOD of 9 μM and sensitivity of 75.26 μA mM−1 cm−2. Also, the sensor and biosensor had notable selectivity, repeatability, reproducibility, and storage stability.  相似文献

This work reports on the performance of new glucose biosensors based on the combination of the electrocatalytic properties of metals and carbon nanotubes towards the reduction of hydrogen peroxide with the biocatalytic activity of glucose oxidase (GOx). The bioelectrodes were obtained by dispersing the metal particles, enzyme and multi-wall carbon nanotubes within a mineral oil binder. The strong electrocatalytic activity of copper and iridium towards the reduction of hydrogen peroxide has made possible an important improvement in the sensitivity for the determination of glucose compared to the carbon nanotube composite without metals. A highly sensitive and selective amperometric detection of glucose becomes possible at very low potentials (−0.100 V). The presence of the protein enables a better dispersion of the metals within the composite matrix, thus allowing an additional enhancement in the response to hydrogen peroxide. The influence of the amount of copper in the composite on the analytical performance of the bioelectrode is discussed. A biosensor containing 0.77% w/w Cu and 10.0% w/w GOx gave a fast response (10.0 s), a linear relationship between current and glucose concentration up to 1.20 × 10−2 M, and a detection limit of 2.0 × 10−5 M. A similar behavior was found for a carbon nanotube-composite electrode containing iridium.  相似文献

A simple procedure was developed to prepare a glassy carbon electrode modified with multi walled carbon nanotubes (MWCNTs) and Celestin blue. Cyclic voltammograms of the modified electrode show stable and a well defined redox couple with surface confined characteristic at wide pH range (2–12). The formal potential of redox couple (E′) shifts linearly toward the negative direction with increasing solution pH. The surface coverage of Celestine blue immobilized on CNTs glassy carbon electrode was approximately 1.95×10?10 mol cm?2. The charge transfer coefficient (α) and heterogeneous electron transfer rate constants (ks) for GC/MWCNTs/Celestine blue were 0.43 and 1.26 s?1, respectively. The modified electrode show strong catalytic effect for reduction of hydrogen peroxide and oxygen at reduced overpotential. The glucose biosensor was fabricated by covering a thin film of sol‐gel composite containing glucose oxides (GOx) on the surface of Celestine blue /MWCNTs modified GC electrode. The biosensor can be used successfully for selective detection of glucose based on the decreasing of cathodic peak current of oxygen. The detection limit, sensitivity and liner calibration rang were 0.3 μM, 18.3 μA/mM and 10 μM–6.0 mM, respectively. The accuracy of the biosensor for glucose detection was evaluated by detection of glucose in a serum sample, using standard addition protocol. In addition biosensor can reach 90% of steady currents in about 3.0 sec and interference effect of the electroactive existing species (ascorbic acid–uric acid and acetaminophen) was eliminated. Furthermore, the apparent Michaelis–Menten constant 2.4 mM, of GOx on the nano composite exhibits excellent bioelectrocatalytic activity of immobilized enzyme toward glucose oxidation. Excellent electrochemical reversibility of redox couple, high stability, technically simple and possibility of preparation at short period of time are of great advantages of this procedure for modification of glucose biosensor.  相似文献

A new H2O2 enzymeless sensor has been fabricated by incorporation of thionin onto multiwall carbon nanotubes (MWCNTs) modified glassy carbon electrode. First 50 μL of acetone solution containing dispersed MWCNTs was pipetted onto the surface of GC electrode, then, after solvent evaporations, the MWCNTs modified GC electrode was immersed into an aqueous solution of thionin (electroless deposition) for a short period of time <5–50 s. The adsorbed thin film of thionin was found to facilitate the reduction of hydrogen peroxide in the absence of peroxidase enzyme. Also the modified electrode shows excellent catalytic activity for oxygen reduction at reduced overpotential. The rotating modified electrode shows excellent analytical performance for amperometric determination of hydrogen peroxide, at reduced overpotentials. Typical calibration at ?0.3 V vs. reference electrode, Ag/AgCl/3 M KCl, shows a detection limit of 0.38 μM, a sensitivity of 11.5 nA/μM and a liner range from 20 μM to 3.0 mM of hydrogen peroxide. The glucose biosensor was fabricated by covering a thin film of sol–gel composite containing glucose oxides on the surface of thionin/MWCNTs modified GC electrode. The biosensor can be used successfully for selective detection of glucose based on the decreasing of cathodic peak current of oxygen. The detection limit, sensitivity and liner calibration rang were 1 μM, 18.3 μA/mM and 10 μM–6.0 mM, respectively. In addition biosensor can reach 90% of steady currents in about 3.0 s and interference effect of the electroactive existing species (ascorbic acid–uric acid and acetaminophen) is eliminated. The usefulness of biosensor for direct glucose quantification in human blood serum matrix is also discussed. A magnetic mesoporous carbon material (i.e., mesoporous iron oxide/C, mesoFe/C) is synthesized for protein immobilization, using glucose oxidase (GOx) as model. Transmission electron microscopy images show that mesoFe/C has highly ordered porous structure with uniform pore size, and iron oxide nanoparticles are dispersed along the wall of carbon. After adsorption of GOx, the GOx-mesoFe/C composite is separated with magnet. The immobilized GOx remains its natural structure according to the reflection–absorption infrared spectra. When the GOx-mesoFe/C composite is coated on a Pt electrode surface, the GOx gives a couple of quasireversible voltammetric peaks at −0.5 V (vs. saturated calomel electrode) due to the redox of FAD/FADH2. The electron-transfer rate constant (k s) is ca. 0.49 s−1. The modified electrode presents remarkably amperometric response to glucose at 0.6 V. The response time (t 95%) is less than 6 s; the response current is linear to glucose concentration in the range of 0.2–10 mM with a sensitivity of 27 μA mM−1 cm−2. The detection limit is 0.08 mM (S/N = 3). The apparent Michaelis–Menten constant (K mapp) of the enzyme reaction is ca. 6.6 mM, indicating that the GOx immobilized with mesoFe/C has high affinity to the substrate.  相似文献

Colloidal carbon microspheres (CMS) are dispersed in chitosan (CHIT) solution to form an organic-inorganic hybrid with excellent micro-environment for the immobilization of biomolecules. A novel amperometric biosensor for the determination of hydrogen peroxide (H(2)O(2)) has been constructed by entrapping horseradish peroxidase (HRP) in as-synthesized CMS/CHIT hybrid. The modification of glassy carbon electrode is made by a simple solution-evaporation method. The electrochemical properties of the biosensor are characterized in electrochemical methods. The proposed biosensor shows high sensitive determination and fast response to H(2)O(2) at -0.15 V. The constructed HRP/CHIT/CMS/GC electrode also exhibits a fine linear correlation with H(2)O(2) concentration. The calculated value of the apparent Michaelis-Menten constant, 2.33 mM, suggests that the HRP in CMS/CHIT hybrid keeps its native bioactivity and has high affinity for H(2)O(2).  相似文献

Abstract

Platinum nanowires (PtNW) were prepared by an electrodeposition strategy using nanopore alumina template. The nanowires prepared were dispersed in chitosan (CHIT) solution and stably immobilized onto the surface of glassy carbon electrode (GCE). The electrochemical behavior of PtNW‐modified electrode and its application to the electrocatalytic reduction of hydrogen peroxide (H2O2) are investigated. The modified electrode allows low potential detection of hydrogen peroxide with high sensitivity and fast response time. As an application example, the glucose oxidase was immobilized onto the surface of PtNW‐modified electrode through cross‐linking by glutaric dialdehyde. The detection of glucose was performed in phosphate buffer at –0.2 V. The resulting glucose biosensor exhibited a short response time (<8 s), with a linear range of 10?5?10?2 M and detection limit of 5×10?6 M.  相似文献

The preparation of an enzyme-dispersed carbon-nanotube (CNT) electrode, based on mixing glucose oxidase (GOx) within CNT, is described. The new binderless biocomposite was packed within a 21-gauge needle and used for amperometric monitoring of glucose. The resulting microsensor offers a low-potential highly selective and sensitive detection of glucose. The high sensitivity and selectivity are coupled to a wide linear range, prolonged lifetime and oxygen independence. About 80% of the GOx activity is retained during a 24 h thermal stress at 90 degrees C, reflecting the enzyme-stabilization action of CNT. The marked electrocatalytic action towards hydrogen peroxide allows highly selective detection of the glucose substrate at -0.1 V (vs. Ag/AgCl) with no interferences from coexisting ascorbic acid, acetaminophen or uric acid. Linearity prevails up to 40 mM glucose (with analytically useful signals observed up to 0.1 M). Factors affecting the performance of the CNT-based glucose biosensor were assessed and optimized. The attractive performance of the new needle electrode offers great promise for continuous monitoring of glucose in connection to the management of diabetes, and for the biosensing of other metabolites.  相似文献

Three different kinds of glassy carbon (GC‐R, GC‐K, GC‐G) were equally pretreated, further modified with electrochemically deposited Prussian Blue and used as sensors for hydrogen peroxide at an applied potential of ?50 mV (vs. Ag|AgCl). Their performance was evaluated with respect to the following parameters: the coverage and electrochemistry of the electrodeposited Prussian Blue, the sensitivity and the lower limit of detection for hydrogen peroxide, and the operational stability of the sensors. GC‐R showed the best behavior concerning the surface coverage and the operational stability of the electrodeposited Prussian Blue. For this electrode the sensitivity for hydrogen peroxide (10 μM) was 0.25 A/M cm2 and the detection limit was 0.1 μM. Scanning electron microscopy was used to study the surfaces of the three electrodes before and after the electrodeposition of Prussian Blue and to search for the reason for the three different behaviors between the different glassy carbon materials. The Prussian Blue modified GC‐R was also used for the construction of a glucose biosensor based on immobilizing glucose oxidase in Nafion membranes on top of electrodeposited Prussian Blue layer. The operational stability of the glucose biosensors was studied in the flow injection mode at an applied potential of ?50 mV (vs. Ag|AgCl) and alternatively injecting standard solutions of hydrogen peroxide (10 μM) and glucose (1 mM) for 3 h. For the GC‐R based biosensor a 2.8% decrease of the initial glucose response was observed.  相似文献

The incorporation of carbon nanotubes to a Nafion/tetraruthenated cobalt porphyrin/ glassy carbon electrode (GC/Nf/CoTRP vs GC/Nf/CNTCoTRP) enhanced the amperometric determination of hydrogen peroxide. Both electrodes produced a decrease in the overpotential required for the hydrogen peroxide oxidation in about 100 mV compared to glassy carbon under the same experimental conditions. Nevertheless, for GC/Nf/CNT/CoTRP, the increase in the current is remarkable. The GC/Nf/CoTRP modified electrode gave no significant analitycal signal for hydrogen peroxide reduction. Moreover, a great increase in current is observed with GC/Nf/CNT/CoTRP at ?150mV which suggests a significant increase in the sensitivity of the modified electrode. Scanning electrochemical microscopy (SECM) revealed an enhancement in the electroactivity of the GC/Nf/CNT/CoTRP modified electrode. This fact has been explained in terms of enhanced homogeneity of the electrodic surface as a consecuence of better dispersibility of CNT‐CoTRP produced by a Nafion polyelectrolyte.  相似文献

A novel hydrogen peroxide (H2O2) biosensor was developed by immobilizing hemoglobin on the gold colloid modified electrochemical pretreated glassy carbon electrode (PGCE) via the bridging of an ethylenediamine monolayer. This biosensor was characterized by UV-vis reflection spectroscopy (UV-vis), electrochemical impendence spectroscopy (EIS) and cyclic voltammetry (CV). The immobilized Hb exhibited excellent electrocatalytic activity for hydrogen peroxide. The Michaelis–Menten constant (K m) was 3.6 mM. The currents were proportional to the H2O2 concentration from 2.6 × 10−7 to 7.0 × 10−3 M, and the detection limit was as low as 1.0 × 10−7 M (S/N = 3).  相似文献

In this paper, a novel biosensor was prepared by immobilizing glucose oxidase (GOx) on carbon nanotube-gold-titania nanocomposites (CNT/Au/TiO2) modified glassy carbon electrode (GCE). SEM was initially used to investigate the surface morphology of CNT/Au/TiO2 nanocomposites modified GCE, indicating the formation of the nano-porous structure which could readily facilitate the attachment of GOx on the electrode surface. Cyclic voltammogram (CV) and electrochemical impedance spectrum (EIS) were further utilized to explore relevant electrochemical activity on CNT]Au/TiO2 nanocomposites modified GCE. The observations demonstrated that the immobilized GOx could efficiently execute its bioelectrocatalytic activity for the oxidation of glucose. The biosensor exhibited a wider linearity range from 0.1 mmol L-1 to 8 mmol L^-1 glucose with a detection limit of 0.077 mmol L^- 1.  相似文献

Glucose oxidase showed direct electrochemical transfer at glassy carbon electrodes immobilized with carbon nanotube‐gold colloid (CNT‐Au) composites with poly(diallydimethylammonium chloride) (PDDA) coatings. The modified electrode (GC/CNT/Au/PDDA‐GOD) was employed for the amperometric determination of glucose. Under optimal conditions, the biosensor displayed linear response to glucose from 0.5 to 5 mM with a sensitivity of 2.50 mA M?1 at an applied potential of ?0.3 V (vs. Ag|AgCl reference).  相似文献

Multilayer films of multiwalled carbon nanotubes (MWNTs) were homogeneously and stably assembled on a glassy carbon (GC) electrode using the layer-by-layer (LBL) method based on electrostatic interaction between MWNTs (negatively charged) and a biopolymer chitosan (CHIT) (positively charged). Scanning electron microscopy (SEM) image of the resulting {CHIT/MWNTs}9 film indicated that the substrate was mostly covered with MWNTs in the form of small bundles or single nanotubes. The multilayer film was used to study the electrocatalytic oxidation of NADH. The assembled {CHIT/MWNTs}9/GC electrode could decrease the oxidation overpotential of NADH by more than 350 mV. The {CHIT/MWNTs}9/GC electrode exhibited a wide linear response range of 8 × 10−7 to 1.6 × 10−3 mol · L−1 with a correlation coefficient of 0.997 for the detection of NADH. The response time and detection limit (S/N = 3) were determined to be 3 s and 0.3 × 10−6 mol · L−1, respectively. Another attractive characteristic was that the method was simple and the assembled {CHIT/MWNTs}9/GC electrode was highly stable.  相似文献

Horseradish peroxidase (HRP) was immobilized onto a polyion complex membrane containing positively charged silver nanoparticles (nanosilver), double stranded DNA and poly(thionine) to fabricate highly sensitive and selective electrochemical hydrogen peroxide (H2O2) biosensor on a glassy carbon electrode. The presence of nanosilver provided a biocompatible microenvironment for enzyme molecules, greatly amplified the surface coverage of HRP on the electrode surface, and most importantly could act as a charge carrier. The process of the biosensor construction was characterized by scanning electron microscopy. Voltammetric and time-based amperometric techniques were employed to characterize the properties of the derived biosensor. Under optimal conditions, the biosensor has an electrocatalytic behavior towards the H2O2 reduction, and exhibits a linear range from 1.1 μM to 5.2 mM, with a lower detection limit of 0.2 μM. The apparent Michaelis–Menten constant of the biosensor to H2O2 was estimated to be 1.02 mM. Furthermore, the biosensor exhibited high sensitivity, good reproducibility, and acceptable stability. Importantly, the properties of composite film, together with the bioelectrochemical catalytic activity, could make them useful in the development of bioelectronic devices and investigation of protein electrochemistry at functional interface. A new amperometric biosensor based on adsorption of glucose oxidase (GOx) at the CuGeO3 nanowires (NW) modified glassy carbon electrode (GCE) is reported in this article. The properties of the biosensor were characterized by Fourier transform infrared spectroscopy (FTIR), electrochemical impedance spectroscopy (EIS) and electrochemical techniques. The GCE modified with the CuGeO3 NW/GOD showed excellent electrocatalytical response to the oxidation of glucose. Different parameters including GOx concentration, working potential and pH of supporting electrolyte that governed the analytical performance of the biosensor have been studied in detail and optimized. The biosensor was applied to detect glucose with a linear range of 0.5 to 7 mM. The biosensor exhibited excellent reproducibility and stability.  相似文献

A new amperometric biosensor for glucose was developed based on adsorption of glucose oxidase (GOx) at the gold and platinum nanoparticles-modified carbon nanotube (CNT) electrode. CNTs were covalently immobilized on gold electrode via carbodiimide chemistry by forming amide linkages between carboxylic acid groups on the CNTs and amine residues of cysteamine self-assembled monolayer (SAM). The fabricated GOx/Aunano/Ptnano/CNT electrode was covered with a thin layer of Nafion to avoid the loss of GOx in determination and to improve the anti-interferent ability. The immobilization of CNTs on the gold electrode was characterized by quartz crystal microbalance technique. The morphologies of the CNT/gold and Ptnano/CNT/gold electrodes have been investigated by scanning electron microscopy (SEM), and the electrochemical performance of the gold, CNT/gold, Ptnano/gold and Ptnano/CNT/gold electrodes has also been studied by amperometric method. In addition, effects of electrodeposition time of Pt nanoparticles, pH value, applied potential and electroactive interferents on the amperometric response of the sensor were discussed.

The enzyme electrode exhibited excellent electrocatalytic activity and rapid response for glucose in the absence of a mediator. The linear range was from 0.5 to 17.5 mM with correction coefficient of 0.996. The biosensor had good reproducibility and stability for the determination of glucose.  相似文献
