The association of 10 different tannins and related polyphenols with gramicidin S, a cyclic peptide having a rigid beta-turn structure, has been examined using 1H-NMR spectroscopy. In the presence of pentagalloylglucose and epigallocatechin-3-O-gallate, the proton signals due to proline and the adjacent phenylalanine moieties selectively shifted to up field, suggesting a regioselective association with the beta-turn structure. The association was also supported by the observation of intermolecular nuclear Overhauser effects between epigallocatechin-3-O-gallate and the peptide. In contrast, ellagitannins, biogenetically derived from pentagalloylglucose, showed small and non-selective chemical shift changes, suggesting that interaction with these tannins is relatively weak. The hydrophobicity of the tannin molecules and the steric hindrance of the interaction site are thought to be important in the association. 相似文献

Combining laser desorption with a supersonic expansion together with the selectivity of IR/UV double resonance spectroscopy makes it possible to isolate and characterise the gas phase of remarkable backbone conformations of short peptide chains mimicking protein segments. A systematic bottom-up approach involving a conformer-specific IR study of peptide sequences of increasing sizes has enabled us to map the spectral signatures of the intramolecular interactions, which shape the peptide backbone, in particular H-bonds. The precise data collected are directly comparable to the most sophisticated quantum chemistry calculations of these species and therefore constitute a stringent test for the theoretical methods used. One-residue chains reveal the local conformational preference of the backbone and its dependence upon the nature of the residue. The investigation of longer chains provides evidence for a competition between simple successions of local conformational preferences along the chain and more folded structures, in which a new H-bonding network, involving distant H-bonding sites along the backbone, takes place. From three residues, the issue of helical folding can also be addressed. The present review of the gas phase literature data emphasizes the observation of remarkable secondary structures of biology, including short segments of beta-strands, gamma- and beta-turns, combinations of turns, including a 3(10) helix. It also provides evidence for the flexibility of the peptide chains, i.e., a critical influence of rather minor interactions (like side-chain/backbone interactions) on the conformational stability. Ion mobility spectrometry, with subsequent mass spectrometric detection, has been employed to study the stability of compact
protein conformations of FK-binding protein, hen egg-white lysozyme, and horse heart myoglobin in the presence and absence
of bound ligands. Protein ions, generated by electrospray ionization from ammonium acetate buffer, were activated by collision
with argon gas to induce unfolding of their compact structures. The collisional cross sections (Ω) of folded and unfolded
conformations were measured in the T-Wave mobility cell of a Waters Synapt HDMS (Waters, Altrincham, UK) employing a calibration
against literature values for a range of protein standards. In the absence of activation, collisional cross section measurements
were found to be consistent with those predicted for folded protein structures. Under conditions of defined collisional activation
energies partially unfolded conformations were produced. The degree of unfolding and dissociation induced by these defined
collision energies are related to the stability of noncovalent intra- and intermolecular interactions within protein complexes.
These findings highlight the additional conformational stability of protein ions in the gas phase resulting from ligand binding. 相似文献

The structural and energetic properties of a group of selected amides, of well-known importance for the design of efficient clathrate inhibitors, are calculated with Hartree-Fock and density functional theory, B3LYP, theoretical levels, and a 6-311++g** basis set in the gas phase and a water solution. The conformational behavior of the molecules is studied through the scanning of the torsional potential energy surfaces and by the analysis of the differences in the energetic and structural properties between the isomers. The properties of the amides in water solution are determined within a self-consistent reaction field approach with a polarizable continuum model that allows the calculation of the different contributions to the free energy of solvation. The calculated barriers to rotation are in good agreement with the available experimental data and the comparison of the gas and water results shows the strong effect of the solute polarization. The properties of different amide-water complexes are calculated and compared with available experimental information. 相似文献

A rapid method, based on sonication-assisted extraction in small columns (SAESC) and subsequent quantification and identification by gas chromatography-mass spectrometry (GC-MS), was developed for the determination of cyclic and linear siloxanes in soil. In the experiments with spiked samples (10-50 ng g(-1)), the recovery of cyclic and linear siloxanes ranged from 87.7 to 108.0% and from 84.9 to 107.6%, respectively. The validated method was used to determine the levels of these compounds in various types of soil samples collected from different locations in Spain. The cyclic siloxanes, decamethylcyclopentasiloxane (D5) and dodecamethylcyclohexasiloxane (D6) were detected in all the soil samples analyzed at concentrations from 9.2 to 56.9 ng g(-1) for D5 and from 5.8 to 27.1 ng g(-1) for D6 in agricultural soils and from 22 to 184 ng g(-1) for D5 and from 28 to 483 ng g(-1) for D6 in industrial soils. The total linear siloxanes concentrations (L5-L14) (sum of the 10 congeners) ranged from 191 to 292 ng g(-1) in agricultural soils and from 1411 to 8532 ng g(-1) in industrial soils. 相似文献

Ion mobility-mass spectrometry is starting to be considered as a useful tool in the deconvolution of complex oil and petroleum samples. While ultrahigh resolution mass spectrometry is the incumbent technology in this field, ion mobility offers complementary information related to species size and shape, and also the ability to resolve structural isomers. In this work, a sample of the resins portion of the Saturates, Aromatics, Resins, and Asphaltenes (SARA) fractions of crude oil was analysed using an orthogonal acceleration quadrupole time-of-flight mass spectrometer (oa-QToF MS) that incorporates a travelling wave ion mobility spectrometry (TWIMS) region. The ion mobility data were compared with previously acquired ultrahigh resolution Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS) data and various nitrogen containing families were identified. Ion mobility data were processed in the typical way for the oil and petroleum industry; and the use of high resolution exact mass coupled with mobility data to provide enhanced species resolution was examined. Double bond equivalence (DBE) and carbon number groups were identified using patterns in the ion mobility data, which demonstrated the utility of ion mobility for discovering species relationships within the crude oil sample. The ability to calibrate the ion mobility cell and generate sizes for the detected ions was also recognised as potentially having particular value for the implementation of conversion or hydrotreatment processes in the oil industry. 相似文献

Fast-atom bombardment (FAB) mass spectrometry was used to investigate the interaction of proton and alkali metal ions with dinucleotide analogs such as T-n-T (T = thymine moiety, n = polyether chain, e.g., triethylene, tetraethylene, pentaethylene, and hexaethylene ether 1–4), A-n-T (A = adenine unit 5–8), and T-n-OMe (9–12) in 3-nitrobenzyl alcohol matrix. The [M + H]+ ion is the most abundant ion for the A-n-T series, whereas in 1–4 and 9–12 the (TC2H4)+ ion is the most abundant. Formation of [M + H -C2H4O]+ ions, a characteristic fragmentation of crown ethers under electron ionization, is observed for compounds 1–12 and is more pronounced in 6 and 7. An abundant [M ? H]? ion is observed for all the compounds studied under negative ion FAB due to the presence of the (-CO-NH-CO-) group of thymine, an indication of existence of intramolecular H bonding. The FAB mass spectra of 1–12 with alkali metal ions (Li+, Na+, K+, Rb+, and Cs+) showed formation of abundant metal-coordinated ions ([M + Met]+ and [TC2H4 + Met]+). Compounds 3, 4, 6, 7, and 10–12 showed ions due to the substitution of the thymine moiety by a hydroxyl group ([M + Met ? 108]+, Met = metal ion). For compound 3 alone, substitution of two thymine groups ([M + Met - 216]+) was observed. Metastable ion studies were used to elucidate the structures of these potentially significant ions, and the ion formule were confirmed with high resolution measurements. Selectivity toward metal complexation with ligand size was seen in the T-n-T and A-n-T series and was even more pronounced in A-n-T series. These dinucleotide analogs fall in the following order of chelation of alkali metal ions, acyclic glymes < dinucleotide analogs (acyclic glymes substituted with nitrogen bases) < crown ethers, which places them in perspective as receptor models. 相似文献

Protein identification is routinely accomplished by peptide sequencing using mass spectrometry (MS) after enzymatic digestion.
Site-specific chemical modification may improve peptide ionization efficiency or sequence coverage in mass spectrometry. We
report herein that amino group of lysine residue in peptides can be selectively modified by reaction with a peroxycarbonate
and the resulting lysine peroxycarbamates undergo homolytic fragmentation under conditions of low-energy collision-induced
dissociation (CID) in electrospray ionization (ESI) and matrix-assisted laser desorption and ionization (MALDI) MS. Selective
modification of lysine residue in peptides by our strategy can induce specific peptide cleavage at or near the lysine site.
Studies using deuterated analogues of modified lysine indicate that fragmentation of the modified peptides involves apparent
free-radical processes that lead to peptide chain fragmentation and side-chain loss. The formation of a-, c-, or z-types of
ions in MS is reminiscent of the proposed free-radical mechanisms in low-energy electron capture dissociation (ECD) processes
that may have better sequence coverage than that of the conventional CID method. This site-specific cleavage of peptides by
free radical- promoted processes is feasible and such strategies may aid the protein sequencing analysis and have potential

Environmental analytical chemistry has recently changed focus from analysis of non-polar, persistent contaminants (e.g. polychlorinated biphenyls and dioxins) to more polar and labile compounds that interfere with biological processes. For example, natural and synthetic estrogens and their metabolites have been detected in sewage treatment plant effluents at nanogram/liter concentrations that are similar to those at which both total sex reversal and intersex (containing both testes and ova) is induced in fish exposed to these compounds in laboratory experiments. The development of techniques for the analysis of natural and synthetic estrogens in biological fluids (i.e. serum and urine) has been a priority in the biomedical field. However, the recent recognition that estrogen hormones are contaminants in the environment that may contribute to endocrine disruption has focused attention on the need for highly sensitive and specific techniques that are applicable for trace analysis in complex environmental matrices. Three optimized mass spectrometric protocols have been developed for the determination and quantitation of steroid hormones in environmental matrices using gas chromatography/tandem mass spectrometry (GC/MS/MS), liquid chromatography/mass spectrometry selected ion monitoring, (LC/MS - SIM) and liquid chromatography/tandem mass spectrometry (LC/MS/MS). [structures: see text] A simple and highly efficient Fmoc solid-phase protocol for synthesizing the antimicrobial decapeptide gramicidin S and various labeled analogues is presented. When preparing the linear precursor peptides (1a-e), a systematic permutation of the starting amino acid within the cyclic sequence gave different yields between 51% and 93%. Also the subsequent step of cyclization gave widely diverging yields between 26% and 74%, depending again on the starting amino acid. The ease of cyclization was found to correlate with the tendency of the respective linear precursor peptide to assume a preorganized conformation, as observed by circular dichroism. The overall yield is thus critically dependent on the starting amino acid and can be raised from 20% to 70% using (D)Phe. The choice of coupling agent and its counterion was found to play only a marginal role. Irrespective of being able to assume a preorganized conformation, none of the linear precursor peptides exhibited any antimicrobial or hemolytic activity. Using the optimized protocol, which involves only simple Fmoc-couplings and requires no intermittent purification steps, several gramicidin S analogues (3-8) containing 19F-labeled phenylglycine derivatives and/or 15N-labeled amino acids were synthesized for solid-state NMR structure analysis. 相似文献

The conformational preferences of morphine and heroin were studied in gas phase and with inclusion of solvent effects. At 298.15 K, three conformers are significant for isolated morphine, all of them displaying antiperiplanar arrangement for the C2? C3? O? H unit, and there is only one significantly populated conformer for heroin. Photoexcitation (using 157 nm vacuum ultraviolet radiation) of proton-bound peptide complexes leads to water elimination and the formation of longer amino acid chains. Thus, it appears that proton-bound dimers are long-lived intermediates along the pathway to peptide formation. Product specificity can be controlled by selection of specific complexes and the incorporation of blocking groups at the N- or C-termini. Infrared multiple photon dissociation spectroscopy and hydrogen/deuterium exchange methods are used to confirm the macrocylic structure of a b(6) peptide fragment by direct comparison with a synthetically made cyclic peptide. The acetylation of the peptide N-terminus results in the inhibition of the macrocyclic formation, supporting the "head-to-tail" cyclization mechanism. Differences in hydrogen/deuterium exchange rates for macrocyclic and oxazalone structure peptide fragments are interpreted to be a result of the complex interplay of multiple basic sites in the peptide fragment, supporting the relay mechanism for deuterium exchange with CH(3)OD. 相似文献

Factors governing hydration, regioselectivity and conformational choice in hydrated carbohydrates have been examined by determining and reviewing the structures of a systematically varied set of singly and multiply hydrated monosaccharide complexes in the gas phase. This has been achieved through a combination of experiments, including infrared ion-depletion spectroscopy conducted in a supersonic jet expansion, and computation through molecular mechanics, density functional theory (DFT) and ab initio calculations. New spectroscopic and/or computational results obtained for the singly hydrated complexes of phenyl beta-D-mannopyranoside (beta-D-PhMan), methyl alpha-D-gluco- and alpha-D-galactopyranoside (alpha-D-MeGlc and alpha-D-MeGal), when coupled with those reported earlier for the singly hydrated complexes of alpha-D-PhMan, beta-D-PhGlc and beta-D-PhGal, have created a comprehensive data set, which reveals a systematic pattern of conformational preference and binding site selectivity, driven by the provision of optimal, co-operative hydrogen-bonded networks in the hydrated sugars. Their control of conformational choice and structure has been further revealed through spectroscopic and/or computational investigations of a series of multiply hydrated complexes; they include beta-D-PhMan.(H2O)2,3, which has an exocyclic hydroxymethyl group, and the doubly hydrated complex of phenyl alpha-L-fucopyranoside, alpha-L-PhFuc.(H2O)2, which does not. Despite the very large number of potential structures and binding sites, the choice is highly selective with binding invariably "focussed" around the hydroxymethyl group (when present). In beta-D-PhMan.(H2O)2,3, the bound water molecules are located exclusively on its polar face and their orientation is dictated by the (perturbed) conformation of the carbohydrate to which they are attached. The possible operation of similar rules governing the structures of hydrogen-bonded protein-carbohydrate complexes is proposed. 相似文献
