Synthesis of (2S,3S)-[3-(2)H1]-4-methyleneglutamic acid and (2S,3R)-[2,3-(2)H2]-4-methyleneglutamic acid

(2S,3S)-[3-(2)H1]-4-Methyleneglutamic acid 1a and (2S,3R)-[2,3-(2)H2]-4-methyleneglutamic acid 1b have been synthesised for use in biosynthetic and metabolic studies.

Chiral fluoroacetic acid: synthesis of (R)- and (S)-[2H1]-fluoroacetate in high enantiopurity

A two-step synthesis of (R)- and (S)-[²H₁]-fluoroacetate (sodium salts) in high enantioselectivity is reported. The synthesis is the development of a previous one in which the enantioselectivity has been increased from ～38% ee to >95% ee. The improvement in enantioselectivity applied Bio’s methodology, which involved a deoxyfluorination reaction with DAST on either enantiomer of [²H₁]-benzyl alcohol, adding TMS-morpholine to the reaction. The additive promotes an S_N2 inversion process, and suppresses a competing non-stereospecific S_N1 reaction course, and as a result significantly improves the stereointegrity of the C–F bond formation. The intermediate [²H₁]-benzyl alcohols, [²H₁]-benzyl fluorides and the product [²H₁]-fluoroacetates as their hexyl esters were separately assayed for their stereochemical integrity, using the Courtieu method. This method involved measuring their ²H NMR spectra in a chiral matrix of poly-γ-benzyl l-glutamate. The chiral assay demonstrated that there was no significant loss in stereointegrity during the deoxyfluorination reaction and showed that the enantiomers of [²H₁]-fluoroacetate were generated with high enantiomeric purity (95% ee).     

Solution properties of poly[1-(9-carbazyl)-2,3-(diethoxycarbonyl)-butane-1,4-diyl]

Poly[1-(9-carbazyl)-2,3-(diethoxycarbonyl)-butane-1,4-diyl] (I), obtained by radical copolymerization of 9-vinylcarbazole with diethylfumarate, has been characterized by viscometry and light scattering. The Huggins constant in benzene at 25 is k_H = 0.33. The Mark-Houwink relationship suggests that benzene is a good solvent for the alternating copolymer I (a = 0.67; K = 1.46·10^?2 ml/g). The unperturbed dimension of the chain is given by value (R₀²/M)^0.5 = 5.73·10^?9 cm·mol^0.5 g^?0.5. The steric factor σ = 2.51 indicates a higher flexibility for this polymer than for poly[1-(9-carbazyl)ethylene].     

Synthesis of (−)-(4R,5R)- and (+)-(4S,5S)-1-alkyl-4-(4′-nitrophenyl)-1-azonium-3,7-dioxabicyclo[3.3.0]octane halides

Synthesis and Crystal Structure of Spiro[1-bromo(S)-4-(R)-hydroxy- 5-oxa-6-oxo-bicyclo[3.1.0]hexane-2,2''-(3''-diethyl-(-(S)-4''-Cl-benzyloxyphosphonyl-4''-(1R,2S,5R)-menthyloxybutyrolactone)]

The title compound, spiro[1-bromo(S)-4-(R)-hydroxy-5-oxa-6-oxo-bicyclo[3.1.0]-hxane-2,2'-(3'-diethyl-α-(S)-4'-Cl-benzyloxyphosphonyl-4'-(1R,2S,5R)-menthyloxybutyrolactone)] has been synthesized via the tandem asymmetric reaction and it crystallizes in a monoclinic system, space group P21 with a = 11.067(3), b = 12.484(2), c = 12.356(2)(A), β = 101.95°,C29H39BrC1O10P, Mr = 693.93, V= 1670.2(6) (A)3, Z= 2, Dc= 1.380 g/cm3, λ(MoKα) = 0.071073nm, μ = 1.410 mm-1, F(000) = 720, the final R = 0.0570 and wR = 0.0758 for 6190 observed reflections with I ＞ 2σ(I). The structure is characterized by the special combination of biologic phosphonyl group and one cyclopropane as well as two butyrolactones. The intermolecular hydrogen bond between O(3)-H(3A)…O(10) in the crystal lattice has been observed.
