Microchip was coupled with MS through a stable, sensitive, and controllable sheath-flow nanoelectrospray (nES) interface for glycoprotein and glycopeptide analysis. The nano-ESI (nESI) was made with a delivery capillary, a commercial nES capillary, and a stainless steel (SS) tube which were connected together through a tee unit. High voltage for nES was applied on the SS tube and the commercial nES capillary was used as nES emitter. The delivery capillary was attached to the microchannel for delivering liquid from microchip to the nESI source. The flow rate of sheath liquid was optimized to be 100-200 nL/min which largely reduced the sample dilution. The detection limit of peptides on this microchip/MS platform was at femtomole level. Glycoprotein and glycopeptides were also successfully analyzed on the platform. All the glycoforms and glycopeptides of ribonuclease B (RNase B) were identified with this method. Some structures of the glycopeptides from RNase B were further characterized with MS/MS on the microchip, coupled with a quadrupole IT-MS. 相似文献

A simple and cost-effective laboratory-made liquid junction interface was used for coupling of CE with MS. In this device the capillary column and the spray tip were positioned in the electrode vessel containing appropriate spray liquid. The electrospray potential was applied on the electrode inside the liquid junction. A stable electrospray was produced at nanoliter per minute flow rates generated in the emitter tip without using an external pump. This arrangement provided high durability of the spray tip and independent optimization of the CE separation (use of coated capillaries) and ESI conditions. CE-MS analysis of mixtures of drugs, peptides, tryptic digests of proteins and biological fluids was optimized with respect to the effects of the distance between the separation capillary and electrospray tip and pressure applied on the liquid junction. The sensitivity of the system, in terms of the LOD (base peak monitoring) was below 10 ng/mL for the beta-blocker drugs and below 200 ng/mL for peptide analysis. 相似文献

A new method for the simultaneous separation of cocaine and four metabolites in urine by CE-ESI-MS via a pressurized nanoliquid junction interface was developed. The resolution of cocaine, cocaethylene, benzoylecgonine, norcocaine, and ecgonine methyl ester was achieved in a polyvinyl-alcohol-coated capillary with 75 μm id × 50 cm total length, using a 15 mM ammonium formate electrolyte solution (pH 9.5) in less than 15 min. In addition, to enhance sensitivity, a field-amplified sample injection (FASI) was evaluated in terms of injection time and sample solvent composition. The limits of detection achieved with the FASI method ranged from 1.5 to 10 ng/mL for all the compounds. The detection of the studied compounds was performed using an ion-trap mass spectrometer in a positive ionization mode. A mixture of methanol:water (80:20 v/v) containing 0.1% v/v of formic acid was employed as spray liquid and delivered at ~200 nL/min. Under optimal CE-MS conditions, linearity was assessed in the concentration range of interest for all analytes with correlation coefficients r2 ≥ 0.9913. Intra- and inter-day precision provided a relative standard deviation lower than 1.54% for migration times and lower than 12.15% for peak areas. Finally, urine samples, spiked with the standard mixture, were extracted using a solid-phase extraction procedure and injected under FASI conditions, providing recoveries from 80% to 94% for all analytes. 相似文献

Electrospray ionization (ESI) is the most widely used ionization method in on-line coupling of capillary electrophoresis-mass spectrometry (CE-MS). The conventional coaxial sheath flow electrospray interface is currently being replaced by the more sensitive nanoelectrospray technique. The usual limitation of nanoelectrospray CE-MS interface has been its short lifetime caused by deterioration of the metal coating on the CE capillary terminus. This article describes an easy way to construct a more durable and sensitive nanospray interface for nonaqueous CE-MS. In this approach a very thin glass spray capillary (ca. 30 microm outer diameter) is partly inserted inside the CE capillary, the junction being surrounded by the electrolyte medium, which is in contact with the platinum electrode. The interface was tested with five pharmaceuticals: methadone, pentazocine, levorphanol, dihydrocodeine, and morphine. Detection limits ranged from 12 to 540 fmol. Separation efficiency and reproducibility were also studied. The CE current was found to be stable and the migration times were highly reproducible. All the CE separations were carried out in a nonaqueous background electrolyte solution. 相似文献

We report a new method for fabricating nanospray ionization tips for MS, formed from glass substrates and the inert polymer, parylene-C. Using a single photolithography step, the emitters are formed contiguously with microchannels, such that no dead volumes are observed. In addition, because the devices are very thin (approximately 0.3 mm) and the tips are formed at rectangular corners, the Taylor cone volumes are small, which makes the method attractive for future integration with microfluidic separations. Device performance was demonstrated by evaluating diverse analytes, ranging from synthetic polymers, to peptides, to nucleic acids. For all analytes, performance was similar to that of conventional emitters (pulled-glass capillaries and the Agilent HPLC Chip) with the advantage of rapid, batch fabrication of identical devices. 相似文献

The influence of packing process parameters (packing pressure, application of ultrasound) and the stationary phase particle size (3.5 and 5 μm) on the chromatographic performance of HPLC/MS chips was systematically investigated for proteomic samples. First, reproducibility and detection limits of the separation were evaluated with a low‐complexity sample of tryptic BSA peptides. The influence of adsorbent packing quality on protein identification was then tested with a typical proteomics sample of high complexity, a human plasma protein fraction (Cohn fraction IV‐4). All HPLC/MS chips provided highly reproducible separations of these proteomic samples, but improved packing conditions and smaller particle sizes resulted in chromatograms with narrower peaks and correspondingly higher signal intensities. Improved separation performance increased the peak capacity, the number of identified peptides, and thus the sequence coverage in the proteomic samples, particularly for low sample amounts. 相似文献

An analytical method of CE-MS and CE with an online preconcentration technique induced by a dynamic pH junction, addition of organic solvent and large volume injection was developed for sensitive determination of peptides in biological samples. Leucine enkephalin, methionine enkephalin, dynorphin A, β-endorphin and angiotensin II were used as model peptides. The optimal online preconcentration conditions were obtained at a sample matrix consisting of 100?mM borate buffer (pH 10.0) with 50% v/v acetonitrile and a BGE containing 1?M formic acid at pH 2.0, along with a 25-cm injection length. Under the optimized conditions, a 4.0×10(3)-1.1×10(4)-fold increase in peak intensity was achieved without degrading the peak shape. This online preconcentration method was applied to analyze the intracellular angiotensin II within the peptides extracted from HL1 cells and approximately increase of 1×10(4)-fold sensitivity was achieved compared to normal condition. Thus, the developed method could be applied to the analysis of various peptides for peptidomics study in biological samples. 相似文献

We have developed a method involving extraction with mixtures of solvents under pressure (pressurized liquid extraction (PLE)) for the determination of triazine herbicides in a series of samples from the food industry. The organic extracts obtained were subjected to a clean-up step with SPE, using Oasis MCX sorbents, after which they were analyzed by NACE. Potato was chosen as a representative matrix of horticultural products since it has a high water content. Spiked potato samples were used to optimize extraction conditions. In order to compare the results obtained with NACE, different studies were also conducted using HPLC. The detection limits in NACE were similar to those found with HPLC and were of the order of 10-15 microg/kg, depending on the analyte. Satisfactory results were obtained on applying the method proposed for the potato matrix (PLE with separation by electrophoresis) to other food matrices such as other tubercles, fruits, vegetables and cereals. 相似文献

Piperaceae family is original from tropical regions and it shows more than 700 species around the world. Piper gaudichaudianum Kunth is the specie more abundant in Brazil, occurring from Northeast to South Brazil. In this paper, it was investigated the influence of some experimental parameters on the pressurized liquid extraction (PLE) of P. gaudichaudianum Kunth leaves, using petroleum ether as extractor solvent. The optimization of the main variables involved in the PLE process (extraction temperature and time) has been done by response surface methodology (RSM) using, as responses, the extraction yield and the chromatographic profile (GC/MS) of the extracts. The optimized procedure employed 3 g of ground leaves, 10 min of extraction and one cycle of extraction at 85 degrees C. The major compounds present in the petroleum ether extracts were: palmitic acid, stearic acid and nerolidol. The results presented in this work show the possibility of using a fast and easy process to recover compounds from P. gaudichaudianum Kunth. 相似文献

We have developed a mid‐infrared laser ablation sampling technique for nano‐flow liquid chromatography coupled with tandem mass spectrometry proteomic profiling of discrete regions from biological samples. Laser ablation performed in transmission geometry was used to transfer material from 50‐µm thick tissue sections mounted on a glass microscope slide to a capturing solvent. Captured samples were processed using filter‐aided sample preparation and enzymatically digested to produce tryptic peptides for data‐dependent analysis with an ion trap mass spectrometer. Comparison with ultraviolet laser capture microdissection from neighboring regions on the same tissue section revealed that infrared laser ablation transfer has higher reproducibility between samples from different consecutive sections. Both techniques allowed for proteomics investigation of different organelles without the addition of surfactants. Copyright © 2016 John Wiley & Sons, Ltd. 相似文献

A method based on pressurized liquid extraction and LC‐MS/MS has been developed for determining nine benzoylureas (BUs) in fruit, vegetable, cereals, and animal products. Samples (5 g) were homogenized with diatomaceous earth and extracted in a 22 mL cell with 22 mL of ethyl acetate at 80°C and 1500 psi. After solvent concentration and exchange to methanol, BUs were analyzed by LC‐MS/MS using an IT mass analyzer, which achieved several transitions of precursor ions that increase selectivity providing identification. LOQs were between 0.002 and 0.01 mg/kg, which are equal or lower than maximum residue limits established by the Codex Alimentarius. Excellent linearity was achieved over a range of concentrations from 0.01 to 1 mg/kg with correlation coefficients 0.995–0.999 (n=7). Validation of the total method was performed by analyzing in quintuplicate seven different commodities (milk, eggs, meat, rice, lettuce, avocado, and lemon) at three concentration levels (0.01, 0.1, and 1 mg/kg). The recoveries ranged from 58 to 97% and the RSDs from 5 to 19% depending on the compound and the commodity. The combination of pressurized liquid extraction with LC‐MS/MS provides a sensitive and selective method for the determination of BUs in food. 相似文献

A technique has been developed to enhance analyte focusing for CE for the analysis of physiological samples. High-ionic-strength samples are titrated to low-ionic-strength on-line using pH-mediated sample stacking in conjunction with a dynamic pH junction. This method concentrates analytes by reducing their electrophoretic mobility during field-amplification. Parameters responsible for enhanced focusing were investigated, and an enhanced pH-mediated stacking method was optimized for anionic nucleosides. The process results in ultra-narrow peak widths, for example, 0.28 s for thymidine with a 10 min analysis time. Peak width and resolution with the enhanced stacking method were also compared to normal base stacking and electrokinetic injection. With this technique, mass-loading capacity can be increased without degradation in peak shape and resolution is dramatically improved. 相似文献

This article reports the applicability of online gel permeation chromatography (GPC)-GC/MS for the determination of seven predominant polybrominated diphenyl ethers (PBDEs) in eggs to effectively eliminate matrix interference. Selective pressurized liquid extraction using acidic alumina as a fat retainer was used for cleanup of the PBDEs in eggs. It was selected because of its advantages: simpler operation, minimum time spent on sample handing to get fat-free extracts, and low volume of solvent consumption. After concentration, the extract was directly injected for online GC/MS operated in the negative ion chemical ionization mode with a 15 m capillary column. Recoveries of spiked samples were between 75.1 and 102.0%, with RSDs (n=3) ranging from 3.69-11.47% when spiked at levels of 2 and 20 ng/g, dry mass. The LOD varied from 0.25-34 ng/g, dry mass. The proposed method was proven to be rapid, efficient, and reliable for the trace determination of PBDEs in eggs. 相似文献

A selective pressurized liquid extraction (SPLE) procedure capable of performing simultaneous extraction and clean-up has been demonstrated for multi-residue analysis of organochlorine pesticides (OCPs) in soil. The final method was performed at 100 degrees C for 3 x 10 min using acetone/n-heptane (1:1, v/v). Florisil was placed inside the extraction cell downstream the sample to remove interfering compounds. Extraction of two soil samples by SPLE gave a recovery which was over 80% for beta-endosulfan, endosulfan sulfate, p,p'-DDT and p,p'-DDE compared to PLE with off-line clean-up. The same trend was observed when applying the SPLE method to a certified reference soil sample (CRM 811-050) containing 13 OCPs, where the SPLE method gave 80-90% recovery vis-à-vis the PLE method with off-line clean-up. Feasibility of the SPLE method was further demonstrated by applying it to five real soil samples collected in Ethiopia. 相似文献

A method was developed for high-throughput speciation analysis of chromium by on-line coupling of short-column capillary electrophoresis (SC-CE) and inductively coupled plasma mass spectrometry (ICP-MS). Baseline separation of Cr(III) and Cr(VI) was achieved within 1 min by SC-CE in a 15 cm x 75 microm id fused-silica capillary at 6 kV using 15 mM HNO(3) as running electrolyte. The precisions (RSD, n = 5) of migration time and peak area for Cr(III) and Cr(VI) were in the range of 1.8-2.4% and 2.2-5.7%, respectively. The limits of detection (3sigma) of Cr(III) and Cr(VI) were 1.8 and 1.9 microg/L, respectively. The synthesized samples containing Cr(III) and Cr(VI) species were determined by the developed SC-CE-ICP-MS hyphenated technique, and the recoveries of Cr(III) and Cr(VI) in the synthesized samples were in the range of 103-110% and 90-108%, respectively. 相似文献

A new method based on pressurized liquid extraction followed by LC‐MS/MS analysis has been developed for the identification and quantification of three capsaicinoids (capsaicin, dihydrocapsaicin, and nordihydrocapsaicin) in extracts of Capsicum annuum. For the recovery of three capsaicinoids, the efficiency levels of ultrasonic‐assisted extraction, microwave‐assisted extraction, Soxhlet extraction, and pressurized liquid extraction were compared under different conditions. Pressurized liquid extraction resulted in higher yields. Pressurized liquid extractions were performed using methanol; temperature was set at 100°C and pressure at 1500 psi. LC analysis was performed on a Waters XBridge? C18 column (150 × 2.1 mm, id 3.5 μm) eluted by a mobile phase of 0.1% formic acid and ACN. Data acquisition was carried out in multiple reaction monitoring transitions mode, monitoring two‐reaction monitoring transitions to ensure an accurate identification of target compounds in the samples. The proposed method is rapid, simple, and could be utilized for the routine analysis of three capsaicinoids in C. annuum samples. 相似文献

Due to the wide range of pesticides that can be used in agriculture, the development of fast multiresidue methods that simultaneously determine polar and non-polar pesticides is greatly demanded. This study shows the development and validation of a multiresidue method for the analysis of 98 non-polar pesticides and 28 polar pesticides in soil. A simultaneous extraction step by pressurized liquid extraction was utilized. The optimum results were obtained using ethyl acetate-methanol (3:1, v/v) with 2 min of preheat time and 85 degrees C as the extraction temperature. The final determination of non-polar pesticides was performed by GC, whereas polar pesticides were determined by ultra-performance liquid chromatography (UPLC). Both GC and UPLC were coupled to triple-quadrupole analyzers operating in tandem MS. The optimized extraction procedure was validated. The average extraction recoveries were in the range 72-108% (10 microg/kg) and 71-106% (50 microg/kg), with RSD values < or = 26%. The matrix effect was also evaluated, and matrix-matched standard calibration was finally applied for quantification. The suitability of the method was also checked by the analysis of a certified reference material. Furthermore, 26 real soil samples were analyzed by the proposed methods in order to assess their applicability. Several pesticides (e.g., bifenthrin, triadimefon, or endosulfan) were found in the samples. 相似文献

A new type of monolithic trapping columns with high mechanical strength was prepared by thin-layer sol–gel coating method and applied to trapping intact proteins for on-line capillary liquid chromatography. Monolithic trapping columns were fabricated by entrapping C8 reversed-phase particles into the capillary columns through a sol–gel network, which was formed by hydrolysis and polycondensation of methyltriethoxysilane. Hundreds times of trapping/untrapping for intact proteins were carried out. The trapping columns showed long-term stability up to 300 bar. Recovery, loading capacity and reproducibility of trapping columns were evaluated using four proteins. The recovery of four protein mixtures for the C8 monolithic trapping columns was 99.3% on average. The loading capacity of 5 mm × 320 μm i.d. C8 trapping columns for the protein mixtures was 30 μg. Day-to-day relative standard deviation (RSD) values for recoveries of protein mixtures on the same C8 trapping column ranged from 2.34 to 5.87%, column-to-column RSD values were from 3.01 to 6.81%. The C8 trapping columns were used to trap normal mouse liver intact proteins in a capillary liquid chromatography system. Results demonstrated high efficiency of the monolithic trapping columns for trapping intact proteins for proteomic analysis in on-line capillary liquid chromatography system. 相似文献

A multiresidue method for the analysis, in egg matrices, of residues of nine quinolones used in veterinary medicine, has been developed and validated according to the provisions of Council Decision 2002/657. Compounds were extracted by a pressurized liquid extraction (PLE) technique using a 1:1 mixture of acetonitrile and a phosphoric acid buffer (pH 3.0) at 70 °C. The obtained extract was clear enough, so that no further clean-up was necessary. Analytes were determined by liquid chromatography (LC) with fluorescence detection (FL). Two chromatographic columns were compared: a high-purity silica Inertsil C8 column and a newly developed Kinetex C18 core-shell technology column. Validation was carried out at four concentration levels, using both chromatographic columns. Precision in terms of reproducibility standard deviation was between 7% and 23%, and good recoveries were obtained. Decision limit (CCα) and detection capability (CCβ) values obtained with the Inertsil and Kinetex columns were in the 0.2-19.8 μg kg−1 and 0.4-33.5 μg kg−1 concentration ranges, respectively. The proposed method allows residues of quinolones banned for use with laying hens to be detected and quantified in eggs. About twenty-four samples per day can be processed. 相似文献
