An electrochemiluminescence (ECL) sensor with good long-term stability and fast response time has been developed. The sensor was based on the immobilization of tris(2,2′-bipyridyl)ruthenium(II) (Ru(bpy)32+) into the Eastman-AQ55D–silica composite thin films on a glassy carbon electrode. The ECL and electrochemistry of Ru(bpy)32+ immobilized in the composite thin films have been investigated, and the modified electrode was used for the ECL detection of oxalate, tripropylamine (TPA) and chlorpromazine (CPZ) in a flow injection analysis system and showed high sensitivity. Because of the strong electrostatic interaction and low hydrophobicity of Eastman-AQ55D, the sensor showed no loss of response over 2 months of dry storage. In use, the electrode showed only a 5% decrease in response over 100 potential cycles. The detection limit was 1 μmol l−1 for oxalate and 0.1 μmol l−1 for both TPA and CPZ (S/N=3), respectively. The linear range extended from 50 μmol l−1 to 5 mmol l−1 for oxalate, from 20 μmol l−1 to 1 mmol l−1 for TPA, and from 1 μmol l−1 to 200 μmol l−1 for CPZ.  相似文献

A highly sensitive cathodic stripping voltammetric method for the determination of naringin is presented. It is based on the formation and accumulation of two naringin–mercury complexes at the electrode surface, followed by reduction of the surface species during a differential pulse voltammetric scan. The cathodic stripping responses at −0.25 V and −0.42 V, are evaluated with respect to various experimental conditions, such as composition and pH of the supporting electrolyte, naringin concentration, accumulation potential and preconcentration time. The new method is suitable for the determination of naringin concentrations between 0.1 mg l−1 (1.72×10−7 mol l−1) and 40 mg l−1 (6.88×10−5 mol l−1). A 3σ limit of detection of 32 μg l−1 (55 nmol l−1) can be reached. The relative standard deviation (r.s.d.) is <1.5%. Recovery experiments yielded a mean recovery of 97% (r.s.d.=4.1%). The application of the procedure to the selective determination of naringin in grapefruit juice is demonstrated.  相似文献

Horseradish peroxidase (HRP) was immobilised on silica gel modified with titanium oxide. This material was employed to prepare modified carbon paste electrode. The direct electron transfer of the hydrogen peroxide reduction by HRP was blocked when immobilised on silica–titanium. This biosensor presented a very sensitive response for phenol (1 μmol l−1) at an applied potential of 0 mV vs SCE. The best condition was achieved in phosphate buffer pH 6.8, ratio of hydrogen peroxide/phenol higher than 0.35. The biosensor showed a linear response range between 10 and 50 μmol l−1 of phenol, adjusted by the equation j=−32.8+16.3 [phenol], for n=5 with a correlation coefficient of 0.9995. The response time of the biosensor was about 3 s.  相似文献

The importance of atmospheric hydrogen peroxide (H2O2) in the oxidation of SO2 and other compounds has been well established. A spectrophotometric method for the determination of hydrogen peroxide in rainwater is proposed. This method is based on selective oxidation of hydrogen peroxide using an on-line tubular reactor containing peroxidase immobilized on Amberlite IRA-743 resin. The hydrogen peroxide in the presence of phenol, 4-aminoantipyrine and peroxidase, produces a red compound (λ = 505 nm). Beer's law is obeyed in a concentration range of 1–100 μmol l−1 hydrogen peroxide with an excellent correlation coefficient (r = 0.9991), at pH 7.0, with a relative standard deviation (R.S.D.) <2%. The detection limit of the method is 0.7 μmol l−1 (4.8 ng of H2O2 in a 200 μl sample). Measurements of hydrogen peroxide in rain samples were carried out over the period from November 2003 to January 2005, in the central area of the Juiz de Fora city, Brazil. The concentration of H2O2 varied from values lower than the detection limit to 92.5 μmol l−1. The effects of the presence of nonseasalt (NSS) SO42−, NO3 and H+ in the concentration of hydrogen peroxide in the rainwater had been evaluated. The average concentrations of H2O2, NO3, NSS SO42− and SO42− are 23.4, 18.9, 7.9 and 10.3 μmol l−1, respectively. The pH values for 82% of the collected samples are greater than 5.0. The spectrophotometeric method developed in this work that uses enzyme immobilized on the resin ion-exchange compared with the amperometric method did not present any significant difference in the results.  相似文献

A stannum film electrode has been developed for the simultaneous determination of trace levels of chromium(III) and cadmium(II) by differential pulse anodic stripping voltammetry (DPASV). The stannum film electrode was generated in situ by depositing simultaneously the stannum film and the metals obtained by reduction of Cd(II) and Cr(III) at −1.4 V on a glassy carbon electrode. Then, the reduced products were oxidized by scanning the potential of the electrode from −1.4 to −0.4 V using DPASV. The electrode exhibited well-defined and separated stripping signals for both metals accompanied with a low background contribution. The possible mechanism of this design was proposed. Under the optimized working conditions, the detection limit was 2.0 and 1.1 μg l−1 for Cr(III) and Cd(II) at a deposition time of 3 min. Finally, the stannum film electrode was successfully applied to the determination of Cd(II) in tap water with satisfactory results.  相似文献

The aerobic bacterial respiration rate is an indicator of microbial growth and metabolism, essential for monitoring the oxidation process and organic load content of samples in a diverse field of application from influent streams in wastewater treatment facilities to industrial fermentations. This paper looks at the influence of parameters, such as culture concentration and volume, sample surface area/volume ratio and headspace volume to achieve optimisation of respirometry measurement and thus design a bench-top respirometric device, based on the monitoring of the pressure changes in a closed chamber where a bacterial culture is allowed to respire in contact with a sample. Contrary to traditional respirometry, the goal is detection of bacterial respiration within 5 min in a minimal sample volume. Both qualitative and quantitative data could be derived using a simple equation and fine-tuning of the micro-manometric parameters of the device, with a most important finding being that minimal headspace volume in combination with elevated bacterial populations maximised absolute pressure change response and favoured high sensitivity at short response time, even though the conditions indicated oxygen-limitation. Furthermore, in comparison with a commercially available respirometer the typical respiration rate of stationary phase P. putida M10 gave oxygen uptake rate (OUR) and specific oxygen uptake rate (SOUR) of 38 μmol l−1 min−1 and 5 μmol g−1 min−1, respectively with the ‘classical’ system, while the μ-Warburg device designed here showed a typical response, for the culture with the same dry cell concentration, of 66 μmol l−1 min−1 for the OUR and 9 μmol g−1 min−1 for the SOUR. The remarkable outcome from this data, therefore, is that it appears that the high surface area/volume geometry of the μ-Warburg device design has achieved less respiration limitation, even though the sample is unstirred. This presents important insight regarding future respirometer design.  相似文献

Fenoterol and salbutamol were determined by electrogenerated chemiluminescence (ECL) coupled with flow injection analysis (FIA), using Ru(bpy)32+ as the luminescent substance. Fenoterol and salbutamol oxidize together with the ruthenium 2,2-bipyridyl at a platinum electrode, which leads to an increase in the luminescent intensity, and this increase is proportional to the analyte concentration. For fenoterol a linear calibration curve within the range from 1.0 × 10−5 to 1.0 × 10−4 mol l−1 was obtained with a correlation coefficient of 0.998 (n = 5) and for salbutamol the linear analytical curve was also obtained in this range with a correlation coefficient of 0.995 (n = 5). The relative standard deviation was estimated as ≤2.5% for 3 × 10−5 mol l−1 for fenoterol solution and as ≤1.3% for 5.0 × 10−5 mol l−1 salbutamol solution for 15 successive injections. The limit of detection for fenoterol was 2.4 × 10−7 mol l−1 and for salbutamol was 4.0 × 10−7 mol l−1. Fenoterol and salbutamol were successfully determined in drug tablets and the soluble components of the matrix did not interfere in the luminescent emission. The results obtained using the luminescent methodology were not statistically different from those obtained by UV-spectrophotometry at 95% confidence level.  相似文献

Bismuth as BiCl4 and BH4 ware successively retained in a column (150 mm × 4 mm, length × i.d.) packed with Amberlite IRA-410 (strong anion-exchange resin). This was followed by passage of an injected slug of hydrochloric acid resulting in bismuthine generation (BiH3). BiH3 was stripped from the eluent solution by the addition of a nitrogen flow and the bulk phases were separated in a gas–liquid separator. Finally, bismutine was atomized in a quartz tube for the subsequent detection of bismuth by atomic absorption spectrometry. Different halide complexes of bismuth (namely, BiBr4, BiI4 and BiCl4) were tested for its pre-concentration, being the chloride complexes which produced the best results. Therefore, a concentration of 0.3 mol l−1 of HCl was added to the samples and calibration solutions. A linear response was obtained between the detection limit (3σ) of 0.225 and 80 μg l−1. The R.S.D.% (n = 10) for a solution containing 50 μg l−1 of Bi was 0.85%. The tolerance of the system to interferences was evaluated by investigating the effect of the following ions: Cu2+, Co2+, Ni2+, Fe3+, Cd2+, Pb2+, Hg2+, Zn2+, and Mg2+. The most severe depression was caused by Hg2+, which at 60 mg l−1 caused a 5% depression on the signal. For the other cations, concentrations between 1000 and 10,000 mg l−1 could be tolerated. The system was applied to the determination of Bi in urine of patients under therapy with bismuth subcitrate. The recovery of spikes of 5 and 50 μg l−1 of Bi added to the samples prior to digestion with HNO3 and H2O2 was in satisfactory ranges from 95.0 to 101.0%. The concentrations of bismuth found in six selected samples using this procedure were in good agreement with those obtained by an alternative technique (ETAAS). Finally, the concentration of Bi determined in urine before and after 3 days of treatment were 1.94 ± 1.26 and 9.02 ± 5.82 μg l−1, respectively.  相似文献

A method for the determination of chloramine using a flow injection peroxidase reactor based on the inhibition reaction of the enzyme is developed. The immobilisation of the horseradish peroxidase is performed on the commercial polymer carrier VA Epoxy Biosynth. The peroxidase activity is detected photometrically based on the dehydration of the dye 2,2′-azino-di-[3-ethylenebenzthiazoline-6-sulphonate] (ABTS). The calibration of the method gives a linear concentration range from 0.026 to 1.04 mmol l−1 (SDn=3=below 5%). The detection limit was calculated to 26 μmol l−1. A mixture out of competitive and non-competitive inhibition was analysed based on the Lineweaver–Burk plots.  相似文献

A new method is developed for the determination of perfluorooctanesulfonate (PFOS) and perfluorooctanoic acid (PFOA) in sewage sludge samples. The analytes in sewage sludge samples are extracted by methanol and formic acid, cleaned by C18 solid-phase extraction, then separated, identified and quantitated by liquid chromatography/quadrupole time-of-flight mass spectrometry (LC–QTOF-MS). A C18 column (150 mm × 2.1 mm, 3.5 μm) with gradient elution of MeOH–H2O (60:40) containing 5 mmol/L ammonium acetate and MeOH–H2O (80:20) is used for the chromatographic separation. [M−K] ions at m/z 498.93 for PFOS and [M−COOH] ion at m/z 368.97 for PFOA are selected for QTOF-MS in the negative electrospray ionization mode. The detection limits for PFOS and PFOA in sewage sludge samples are 0.5 and 0.8 ng/g, respectively. The spiked recoveries are in the range of 85–114 and 71–98% for PFOS and PFOA, respectively. The proposed method is successfully applied to the analysis of PFOS and PFOA in 16 sewage sludge samples from China. PFOS and PFOA are detected in most sewage sludge samples and the concentrations of PFOS and PFOA are up to 5383 and 4780 ng/g (oven dry weight), respectively.  相似文献

An atomic absorption spectrophotometric method for the determination of trace copper after adsorption of its 1-nitroso-2-naphthol-3,6-disulfonic acid chelate on Ambersorb 572 has been developed. This chelate is adsorbed on the adsorbent in the pH range 1–8. The copper chelate is eluted with 5 ml of 0.1 mol l−1 potassium cyanide and determined by flame atomic absorption spectrometry (FAAS). The selectivity of the proposed procedure was also evaluated. Results show that iron(III), zinc(II), manganese(II) and cobalt(II) at the 50 μg l−1 level and sodium(I), potassium(I), magnesium(II), calcium(II) and aluminium(III) at the 1000 μg l−1 level did not interfere. A high enrichment factor, 200, was obtained. The detection limit (3σ) of copper was 0.34 μg l−1. The precision of the method, evaluated by seven replicate analyses of solutions containing 5 μg of copper was satisfactory and the relative standard deviation was 1.7%. The adsorption of copper onto Ambersorb 572 can formally be described by a Langmuir equation with a maximum adsorption capacity of 14.3 mg g−1 and a binding constant of 0.00444 l mg−1. The accuracy of the method is confirmed by analysing tomatoes leaves (NIST 1573a) and lead base alloy (NBS 53e). The results demonstrated good agreement with the certified values. This procedure was applied to the determination of copper in waters (tap, river and thermal waters), aluminium foil and tea samples.  相似文献

A solid phase extraction procedure based on biosorption of copper(II), lead(II), zinc(II), iron(III), nickel(II) and cobalt(II) ions on Aspergillus fumigatus immobilized Diaion HP-2MG has been investigated. The analytical conditions including amounts of A. fumigatus, eluent type, flow rates of sample and eluent solutions were examined. Good recoveries were obtained to the spiked natural waters. The influences of the concomitant ions on the retentions of the analytes were also examined. The detection limits (3sigma, N = 11) were 0.30 μg l−1 for copper, 0.32 μg l−1 for iron, 0.41 μg l−1 for zinc, 0.52 μg l−1 for lead, 0.59 μg l−1 for nickel and 0.72 μg l−1 for cobalt. The relative standard deviations of the procedure were below 7%. The validation of the presented procedure is performed by the analysis of three standard reference materials (NRCC-SLRS 4 Riverine Water, SRM 1515 Apple leaves and GBW 07605 Tea). The procedure was successfully applied for the determination of analyte ions in natural waters microwave digested samples including street dust, tomato paste, black tea, etc.  相似文献

An analytical method for analysing acrylamide in coffee was validated. The analysis of prepared coffee includes a comprehensive clean-up using multimode solid-phase extraction (SPE) by automatic SPE equipment and detection by liquid chromatography tandem mass spectrometry using electrospray in the positive mode. The recoveries of acrylamide in ready-to-drink coffee spiked with 5 and 10 μg l−1 were 96±14% and 100±8%, respectively. Within laboratory reproducibility for the same spiking levels were 14% and 9%, respectively. Coffee samples (n = 25) prepared twice by coffee machines and twice by a French Press Cafetière coffee maker contained 8±3 μg l−1 and 9±3 μg l−1 acrylamide. Five ready-to-drink instant coffee prepared twice contained 8±2 μg l−1. Hence, the results do not show significant differences in the acrylamide contents in ready-to-drink coffee prepared by coffee machine, French Press or from instant coffee. Medium roasted coffee contained more acrylamide (10 μg l−1) than dark roasted coffee (5 μg l−1). Males aged 35–45 years, drinking on average 1.1 l coffee per day are exposed to the highest doses of acrylamide from coffee. The dietary intake of acrylamide from coffee comprises, on an average, 10 μg day−1 for males and 9 μg day−1 for females aged 35–45 years. Probabilistic modelling of the exposure of Danish consumers (all adults) to acrylamide from coffee shows a mean exposure of 6.5 μg day−1 and a 95 percentile of 18 μg day−1.  相似文献

Homocystine was for the first time, chemically linked to a highly cross-linked agarose support (Novarose) to be employed as a chelating adsorbent for preconcentration and AAS determination of nickel in table salt and baking soda. Nickel is quantitatively adsorbed on a small column packed with 0.25 ml of the adsorbent, in a pH range of 5.5–6.5 and simply eluted with 5 ml of a 1 mol l−1 hydrochloric acid solution.

A factorial design was used for optimization of the effects of five different variables on the recovery of nickel. The results indicated that the factors of flow rate and column length, and the interactions between pH and sample volume are significant.

In the optimized conditions, the column could tolerate salt concentrations up to 0.5 mol l−1 and sample volumes beyond 500 ml. Matrix ions of Mg2+ and Ca2+, with a concentration of 200 mg l−1, and potentially interfering ions of Cd2+, Cu2+, Zn2+ and Mn2+, with a concentration of 10 mg l−1, did not have significant effect on the analyte's signal. Preconcentration factors up to 100 and a detection limit of 0.49 μg l−1, corresponding to an enrichment volume of 500 ml, were obtained for the determination of the analyte by flame AAS. Application of the method to the determination of natural and spiked nickel in table salt and baking soda solutions resulted in quantitative recoveries. Direct ETAAS determination of nickel in the same samples was not possible because of a high background observed.  相似文献

Ascorbic acid (AA) could be determined in large quantities of a co-existing oxidant. The incorporation of an on-line reagent regeneration step based on redox reaction eliminates the baseline drift in the procedure. This makes it possible to adopt a circulatory flow injection method (cyclic FIA) and to determine AA repetitively. The method is based on the reduction of iron(III) to iron(II) by the analyte, the reaction of the produced iron(II) with 1,10-phenanthroline (phen) in a weak acidic medium to form a colored complex, and the subsequent oxidation reaction of iron(II) to iron(III) by the co-existing peroxodisulfate. A solution (50 ml) of 3.0×10−4 mol l−1 ferric chloride, 9.0×10−4 mol l−1 phen and 5.0×10−2 mol l−1 ammonium peroxodisulfate in acetate buffer (0.2 mol l−1, pH 4.5) is continuously circulated at a constant flow rate of 1.0 ml min−1. Into this stream, an aliquot (20 μl) of the sample solution containing AA is quickly injected by means of a six-way valve. The complex formed is monitored spectrophotometrically (at 510 nm) in the flow system. The stream then returns to the reservoir after passing through a time-delay coil (50 m). The iron(II)–(phen)3 complex is oxidized to iron(III)–(phen)3 complex by peroxodisulfate which exists excessively in the circulating reagent solution. The proposed method allows as many as 300 repetitive determinations of 15 mg l−1 AA with only 50 ml reservoir solution. The contents of AA in commercial pharmaceutical products were analyzed to demonstrate the capability of the developed system.  相似文献

A simple and rapid flow injection (FI) method is reported for the determination of phosphate (as molybdate reactive P) in freshwaters based on luminol chemiluminescence (CL) detection. The molybdophosphoric heteropoly acid formed by phosphate and ammonium molybdate in acidic conditions generated chemiluminescence emission via the oxidation of luminol. The detection limit (3× standard deviation of blank) was 0.03 μg P l−1 (1.0 nM), with a sample throughput of 180 h−1. The calibration graph was linear over the range 0.032–3.26 μg P l−1 (r2=0.9880) with relative standard deviations (n=4) in the range 1.2–4.7%. Interfering cations (Ca(II), Mg(II), Ni(II), Zn(II), Cu(II), Co(II), Fe(II) and Fe(III)) were removed by passing the sample through an in-line iminodiacetate chelating column. Silicate interference (at 5 mg Si l−1) was effectively masked by the addition of tartaric acid and other common anions (Cl, SO42−, HCO3, NO3 and NO2) did not interfere at their maximum admissible concentrations in freshwaters. The method was applied to freshwater samples and the results (26.1±1.1–62.0±0.4 μg P l−1) were not significantly different (P=0.05) from results obtained using a segmented flow analyser method with spectrophotometric detection (24.4±4.45–84.0±16.0 μg P l−1).  相似文献

In this contribution, a plasmon resonance light scattering (PRLS) detection method of ferulic acid (FA) is proposed based on the formation of silver nanoparticles (NPs). It was found that, FA acted as a reducing agent in alkaline medium and could be oxidized by AgNO3, resulting in the formation of silver NPs. The formed silver NPs, which were identified by measuring the plasmon resonance absorption spectra, PRLS spectra and transmission electron microscopy (TEM) image, display characteristic plasmon resonance optical absorption and PRLS band in the visible region. It was found that the PRLS intensity, which could be easily measured using a common spectrofluorometer, was in proportion to the concentration of FA over the range from 0.2 to 2.0 μmol l−1 with the corresponding limits of determination (3σ) of 15.2 nmol l−1. With that, ferulate sodium injection samples have been detected with R.S.D. lower than 3.0% and recoveries over the range of 101.2–104.5%. On the other hand, the present reaction maybe provides the basis of an environmentally friendly approach for the synthesization of silver NPs.  相似文献

A simple GF-AAS method for speciation analysis of chromium in mineral waters and salinas was developed. Cr(VI) species were separated from Cr(III) by solid-phase extraction with APDC (ammonium pyrrolidinedithiocarbamate). The APDC complexes were formed in the sample solution under proper conditions, adsorbed on Diaion HP-2MG resin and the resin was separated from the sample. After elution with concentrated nitric acid Cr(VI) was determined by GF-AAS. Total chromium was determined by GF-AAS directly in the sample and Cr(III) concentration was calculated as the difference between those results.

The detection limit of the method defined as 3 s of background variation was 0.03 μg l−1 for Cr(VI) and 0.3 μg l−1 for total chromium. RSD for Cr(VI) determination at the concentration of 0.14 μg l−1 was 9%, and for total chromium at the concentration of 5.6 μg l−1 was 5%. The recovery of Cr(VI) was in the range of 94–100%, dependently on type of the sample.

The investigation of recovery of the spiked Cr(VI) showed that at concentration levels near 1 μg l−1 and lower recovery may be reduced significantly even by pure reagents that seem to be free from any reductants.  相似文献

A continuous flow system for the determination of lead in home made spirituous beverages was developed. The determination was based on the formation of a neutral chelate of the element with ammonium pyrrolidine dithiocarbamate, its adsorption onto a minicolumn packed with sodium faujasite type Y synthetic zeolite, followed by elution with methyl isobutyl ketone and determination by flame atomic absorption spectrometry. Ethanol and copper interfere strongly in the determination and therefore, must be separated prior to the analysis. Copper is removed by precipitation with rubeanic acid, while ethanol is eliminated by rotaevaporation. Sample solutions containing Pb2+ in the concentration range from 5 to 120 μg l−1 at pH 2.5 could be analyzed, by using a preconcentration time of 3 min. Preconcentration factors from 80 to 140 were achieved for a sample volume of 6 ml and the detection limit varied from 1.4 to 3.5 μg l−1, depending on the matrix composition. The relative standard deviations for 60 μg l−1 Pb was 3.2% (n = 10) and the recovery of spikes (20, 40, 60 and 80 μg l−1) added to the samples was estimated within 92–105% range, suggesting that lead can be quantitatively determined in such samples. Determining lead in several samples by an alternative technique further checked the accuracy. Finally, the concentrations of Pb2+ determined in 28 samples of Venezuelan spirituous beverages were in 12.6–370.0 μg l−1 range, depending on the fermenting material based on different mixtures of agave, raw sugar cane and white sugar.  相似文献

A novel electrochemical DNA biosensor based on methylene blue (MB) and zirconia (ZrO2) thin films modified gold electrode for DNA hybridization detection is presented. Zirconia thin films were electrodynamically deposited onto the bare gold electrode in an aqueous electrolyte of ZrOCl2 and KCl by cycling the potential between −1.1 and +0.7 V (versus Ag/AgCl) at a scan rate of 20 mV s−1. Oligonucleotide probes with phosphate group at the 5′ end were attached onto the zirconia thin films because zirconia is affinity for phosphoric group. The surface density of the immobilized DNA molecules at the zirconia interface was investigated by fluorescence spectroscopy method. Hybridization was induced by exposure of the ssDNA-containing Au electrode to complementary ssDNA in solution. The decreases in the peak currents of MB, an electroactive label, were observed upon hybridization of probe with the target. The cathodic peak current (ip) of MB after hybridization with the target DNA was linearly related to the logarithmic value of the target DNA concentration ranging from 2.25×10−10 to 2.25×10−8 mol l−1. A detection limit of 1.0×10−10 mol l−1 of oligonucleotides can be estimated.  相似文献
