An efficient enantioselective method for the determination of etoxazole in orange pulp, peel, and whole orange was developed using liquid chromatography with tandem mass spectrometry. The enantioseparation was performed on a Chiralpak AD‐3R column at 30ºC using acetonitrile with 0.1% formic acid solution (80:20, v/v) as the mobile phase in less than 5 min. Quantification was achieved using matrix‐matched standard calibration curves. The overall mean recoveries for two enantiomers from orange pulp and whole orange were 91.0–99.6% and the orange peel was 92.6–103.1%, with relative standard deviations of 0.8–5.4% intraday and 2.0–4.8% interday at 1, 10, and 100 μg/kg levels, and 1.3–5.2% intraday and 3.5–4.3% interday at 5, 50, and 500 μg/kg levels, respectively. The limits of quantification for all enantiomers in three matrices did not exceed 5 μg/kg. Moreover, the absolute configuration of etoxazole enantiomers had been determined by the combination of experimental and predicted electronic circular dichroism spectra, and the first eluted enantiomer was confirmed as (S)‐etoxazole on a Chiralpak AD‐3R column while (R)‐etoxazole was first on three cellulose chiral columns. The application of the proposed method to real sample analysis suggests its potential use in enantioselective determination of etoxazole enantiomers in citrus.  相似文献

A novel and sensitive method was developed for the determination of tebuconazole enantioselectively using reversed‐phase LC‐MS/MS. The separation and determination were performed using on an amylose‐based chiral stationary phase, a Lux 3u Amylose‐2 column (150 mm×2.0 mm), under isocratic conditions at 0.3 mL/min flow rate. A series of chiral stationary phases were investigated and the effect of mobile phase composition on the enantioseparation was discussed. Parameters including the matrix effect, linearity, precision, accuracy and stability were evaluated. Under optimal conditions, the overall mean recoveries for two enantiomers from the soil, tomato, cucumber, pear and apple samples were 79.3–101.1% with 2.8–11.5% intra‐day relative standard deviations (RSDs) and 4.1–8.6% inter‐day RSDs at 5, 25 and 50 μg/kg levels; the mean enantiomer recoveries from the water samples were 89.6–101.9% with 3.3–10.2% intra‐day RSDs and 5.1–7.7% inter‐day RSDs at 0.25, 0.5 and 2.5 μg/kg levels. The limits of detection (LODs) for all enantiomers in tomato, cucumber, pear, apple, soil and water were less than 0.6 μg/kg, whereas the limit of quantification (LOQ) did not exceed 2.0 μg/kg. The results indicate that this proposed method is convenient and reliable for the enantioselective determination of tebuconazole enantiomers in foods and environment samples.  相似文献

A novel and sensitive dispersive liquid–liquid microextraction method based on the solidification of the floating organic drop combined with high‐performance liquid chromatography and ultraviolet detection was used for the determination of atorvastatine in blood serum samples. The chromatographic separation of atorvastatin was carried out using methanol as the mobile phase organic modifier. Various parameters affecting the extraction efficiency were optimized, such as the kind and volume of extraction solvent (1‐undecanol) and disperser solvent (acetonitrile), pH, and the extraction time. The calibration curve was linear in the range of 0.2–6000 μg/L of atorvastatin (r2 = 0.995) with a limit of detection of 0.07 μg/L. The relative standard deviation for 100 μg/L of atorvastatin in human plasma was 8.4% (n = 4). The recoveries of plasma samples spiked with atorvastatin were in the range of 98.8–113.8%. The obtained results showed that the proposed method is fast, simple, and reliable for the determination of very low concentrations of atorvastatin in human plasma samples.  相似文献

Dispersive liquid–liquid microextraction method was developed for the determination of the amount of phthalate esters in bottled drinking water samples and dispersive liquid–liquid microextraction samples were analyzed by GC–MS. Various experimental conditions influencing the extraction were optimized. Under the optimized conditions, very good linearity was observed for all analytes in a range between 0.05 and 150 μg/L with coefficient of determination (R2) between 0.995 and 0.999. The LODs based on S/N = 3 were 0.005–0.22 μg/L. The reproducibility of dispersive liquid–liquid microextraction was evaluated. The RSDs were 1.3–5.2% (n = 3). The concentrations of phthalates were determined in bottled samples available in half shell. To understand the leaching profile of these phthalates from bottled water, bottles were exposed to direct sunlight during summer (temperature from 34–57°C) and sampled at different intervals. Result showed that the proposed dispersive liquid–liquid microextraction is suitable for rapid determination of phthalates in bottled water and di‐n‐butyl, butyl benzyl, and bis‐2‐ethylhexyl phthalate compounds leaching from bottles up to 36 h. Thereafter, degradation of phthalates was observed.  相似文献

A quick, easy, cheap, effective, rugged, and safe (QuEChERS) sample pretreatment method coupled with LC–MS was developed for the determination of 11 pesticides in tobacco. Sample pretreatment parameters and instrumental parameters of LC–MS were investigated, and the optimal conditions were selected. Under the optimized conditions, the 11 pesticides were detected simultaneously with a good linear relationship (r2 = 0.9993–0.9999) and high precisions (less than 5% of the RSD of peak areas). The LODs were in the range of 0.1–5.0 μg/L. Compared with SPE clean‐up, QuEChERS greatly simplified the sample pretreatment with simple solvent extraction system. After QuEChERS pretreatment, no serious matrix effects were observed. Used for the analysis of real samples, metalaxyl was found in cigarette and tobacco samples at 63.47 and 132.27 ng/g, respectively. The recoveries for 11 pesticides were in the range of 70.03–118.69%, and RSDs were less than 10%. The proposed method is simple, low cost, and has good reproducibility.  相似文献

A simple, efficient, solvent‐free, and commercial readily available approach for determination of five volatile chlorinated hydrocarbons in water samples using the static headspace sampling and gas chromatography with electron capture detection has been described. The proposed static headspace sampling method was initially optimized and the optimum experimental conditions found were 10 mL water sample containing 20% w/v sodium chloride placed in a 20 mL vial and stirred at 50ºC for 20 min. The linearity of the method was in the range of 1.2–240 μg/L for dichloromethane, 0.2–40 μg/L for trichloromethane, 0.005–1 μg/L for perchloromethane, 0.025–5 μg/L for trichloroethylene, and 0.01–2 μg/L for perchloroethylene, with coefficients of determination ranging between 0.9979 and 0.9990. The limits of detection were in the low μg/L level, ranging between 0.001 and 0.3 μg/L. The relative recoveries of spiked five volatile chlorinated hydrocarbons with external calibration method at different concentration levels in pure, tap, sea water of Jiaojiang Estuary, and sea water of waters of Xiaomendao were in the range of 91–116, 96–105, 86–112, and 80–111%, respectively, and with relative standard deviations of 1.9–3.6, 2.3–3.5, 1.5–2.7, and 2.3–3.7% (n = 5), respectively. The performance of the proposed method was compared with traditional liquid–liquid extraction on the real water samples (i.e., pure, tap, and sea water, etc.) and comparable efficiencies were obtained. It is concluded that this method can be successfully applied for the determination of volatile chlorinated hydrocarbons in different water samples.  相似文献

An electrothermal atomic absorption method (ETAAS) for the direct determination of trace elements (Cd, Cr, Cu, Mn, Se) both in blood fractions (erythrocytes, plasma and lymphocytes) and whole blood was developed. Zeeman background correction and graphite tubes with L’vov platforms were used. Samples were ¶diluted with HNO3/Triton X-100 and pipetted directly ¶into the graphite tube. Ashing, pretreatment and atomization steps were optimized carefully for the different fractions and elements applying different matrix modifiers ¶for each element. For the lymphocyte fraction a multi-fold injection technique was applied. Low detection limits ¶of the ETAAS method (Cd 0.13 μg/L, Cr 0.11 μg/L, ¶Cu 0.52 μg/L, Mn 0.13 μg/L, Se 0.7 μg/L of whole blood) combined with small quantities of sample necessary for analysis allow determination of trace elements in this matrix. Verification of possible differences in the trace element status of humans was performed with statistical significance (P < 0.05). In addition, a contribution to the determination of normal values of essential elements was achieved. The method was applied for determination of trace elements in human blood and blood fractions of two groups (n = 50) different in health status.  相似文献

A novel and sensitive method was developed for the simultaneous determination of fenbuconazole and its main metabolites enantioselectively using chiral liquid chromatography coupled with tandem mass spectrometry. The separation and determination were performed using reversed-phase chromatography on a cellulose chiral stationary phase, a Chiralcel OD-RH (150 mm×4.6 mm) column, under isocratic conditions at 0.5 mL/min flow rate. The effects of three cellulose-based columns and three amylose-based columns on the separation were also investigated. The elution orders of the eluting enantiomers were identified by an optical rotation detector. The QuEChERS (acronym for Quick, Easy, Cheap, Effective, Rugged and Safe) method and solid-phase extraction (SPE) were used for the extraction and clean-up of the soil and water samples, respectively. Parameters including the matrix effect, linearity, precision, accuracy and stability were evaluated. Under optimal conditions, the mean recoveries for all enantiomers from the soil samples were 82.5-104.1% with 2.7-9.5% intra-day relative standard deviations (RSD) and 5.7-11.2% inter-day RSD at 5, 25 and 50 μg/kg levels; the mean enantiomer recoveries from the water samples were 81.8-104.6% with 2.6-11.4% intra-day RSD and 5.3-10.4% inter-day RSD at 0.25, 0.5 and 2.5 μg/L levels. Coefficients of determination R2≥0.9991 were achieved for each enantiomer in the soil and water matrix calibration curves within the range of 1.0-125 μg/L. The limits of detection (LOD) for all enantiomers in the soil and water were less than 0.8 μg/kg, whereas the limit of quantification (LOQ) did not exceed 2.5 μg/kg. The results of the method validation confirm that this proposed method is convenient and reliable for the enantioselective determination of the enantiomers of fenbuconazole and its main metabolites in soil and water.  相似文献

We developed and validated a simple, sensitive, selective and reliable LC–ESI‐MS/MS method for direct quantitation of dropropizine enantiomers namely levodropropizine (LDP) and dextrodropropizine (DDP) in rat plasma without the need for derivatization as per regulatory guidelines. Dropropizine enantiomers and carbamazepine (internal standard) were extracted from 50 μL rat plasma using ethyl acetate. LDP and DDP resolved with good baseline separation (Rs = 4.45) on a Chiralpak IG‐3 column. The mobile phase consisted of methanol with 0.05% diethylamine pumped at a flow rate of 0.5 mL/min. Detection and quantitation were done in multiple reaction monitoring mode following the transitions m/z 237 → 160 and 237 → 194 for dropropizine enantiomers and the internal standard, respectively, in the positive ionization mode. The proposed method provided accurate and reproducible results over the linearity range of 3.23–2022 ng/mL for each enantiomer. The intra‐ and inter‐day precisions were in the ranges of 3.38–13.6 and 5.11–13.8 for LDP and 4.19–11.8 and 8.89–10.1 for DDP. Both LDP and DDP were found to be stable under different stability conditions. The method was successfully used in a stereoselective pharmacokinetic study of dropropizine enantiomers in rats following oral administration of racemate dropropizine at 100 mg/kg. The pharmacokinetic results indicate that the disposition of dropropizine enantiomers is not stereoselective and chiral inversion does not occur in rats.  相似文献

A rapid dispersive micro‐solid phase extraction (D‐μ‐SPE) combined with LC/MS/MS method was developed and validated for the determination of ketoconazole and voriconazole in human urine and plasma samples. Synthesized mesoporous silica MCM‐41 was used as sorbent in d ‐μ‐SPE of the azole compounds from biological fluids. Important D‐μ‐SPE parameters, namely type desorption solvent, extraction time, sample pH, salt addition, desorption time, amount of sorbent and sample volume were optimized. Liquid chromatographic separations were carried out on a Zorbax SB‐C18 column (2.1 × 100 mm, 3.5 μm), using a mobile phase of acetonitrile–0.05% formic acid in 5 mm ammonium acetate buffer (70:30, v /v). A triple quadrupole mass spectrometer with positive ionization mode was used for the determination of target analytes. Under the optimized conditions, the calibration curves showed good linearity in the range of 0.1–10,000 μg/L with satisfactory limit of detection (≤0.06 μg/L) and limit of quantitation (≤0.3 μg/L). The proposed method also showed acceptable intra‐ and inter‐day precisions for ketoconazole and voriconazole from urine and human plasma with RSD ≤16.5% and good relative recoveries in the range 84.3–114.8%. The MCM‐41‐D‐μ‐SPE method proved to be rapid and simple and requires a small volume of organic solvent (200 μL); thus it is advantageous for routine drug analysis.  相似文献

A novel chiral method was developed and validated to determine N‐acetyl‐glutamine (NAG) enantiomers by liquid chromatography–tandem mass spectrometry (LC–MS/MS). Enantioseparation was achieved on a Chiralpak QD‐AX column (150 × 4.6 mm i.d., 5 μm) using methanol–water (50 mm ammonium formate, pH 4.3; 70:30, v/v) at a flow rate of 500 μL/min. The detection was operated with an electrospray ionization source interface in positive mode. The ion transition for NAG enantiomers was m/z 189.0 → 130.0. The retention time of N‐acetyl‐l ‐glutamine and N‐acetyl‐d ‐glutamine were 15.2 and 17.0 min, respectively. Calibration curves were linear over the range of 0.02–20 μg/mL with r > 0.99. The deviation of accuracy and the coefficient of variation of within‐run and between‐run precision were within 10% for both enantiomers, except for the lower limit of quantification (20 ng/mL), where they deviated <15%. The recovery was >88% and no obvious matrix effect was observed. This method was successfully applied to investigate the plasma protein binding of NAG enantiomers in rats. The results showed that the plasma protein binding of NAG enantiomers was stereoselective. The assay method also exhibited good application prospects for the clinical monitoring of free drugs in plasma.  相似文献

A porous and highly efficient polyaniline‐based solid‐phase microextraction (SPME) coating was successfully prepared by the electrochemical deposition method. A method based on headspace SPME followed by HPLC was established to rapidly determine trace chlorophenols in water samples. Influential parameters for the SPME, including extraction mode, extraction temperature and time, pH and ionic strength procedures, were investigated intensively. Under the optimized conditions, the proposed method was linear in the range of 0.5–200 μg/L for 4‐chlorophenol and 2,4,6‐trichlorophenol, 0.2–200 μg/L for 2,4‐dichlorophenol and 2–200 μg/L for 2,3,4,6‐tetrachlorophenol and pentachlorophenol, with satisfactory correlation coefficients (>0.99). RSDs were <15% (n = 5) and LODs were relatively low (0.10–0.50 μg/L). Compared to commercial 85 μm polyacrylate and 60 μm polydimethylsiloxane/divinylbenzene fibers, the homemade polyaniline fiber showed a higher extraction efficiency. The proposed method has been successfully applied to the determination of chlorophenols in water samples with satisfactory recoveries.  相似文献

For the first time, the low‐density solvent‐based vortex‐assisted surfactant‐enhanced emulsification liquid–liquid microextraction, followed by GC‐flame photometric detection has been developed for the determination of eight organophosphorus pesticides in aqueous samples. A small volume of organic extraction solvent (toluene) was dispersed into the aqueous samples by the assistance of surfactant and vortex agitator. The extraction was performed in a special disposable polyethylene pipette, allowing using the reagents with lower density than water as extraction solvents. The influence parameters were systemically investigated and optimized: toluene (30 μL) and Triton X‐100 (0.2 mmol/L) were used as the extraction solvent and the surfactant, respectively, and the extraction was performed for 1 min under room temperature without adding sodium chloride. Under the optimum conditions, the validation parameters such as the RSD (n = 6; 2.1–11.3%), LOD (0.005 and 0.05 μg/L), and linear range (0.1–50.0 μg/L with correlation coefficients (0.9958–0.9992) showed the method was satisfying. The proposed method has been successfully applied to the determination of the organophosphorus pesticides in real samples with recoveries between 82.8 and 100.2%.  相似文献

This article described a new method for the sensitive determination of organophosphorus pesticides in water samples using SPE in combination with GC‐flame photometric detection. In the procedure of method development, TiO2 nanotubes were used as SPE adsorbents for the enrichment of organophosphorus pesticides from water samples. Several factors, such as eluent and its volume, sample pH, sample volume, sample flow rate, and concentration of humic acid, were optimized. Under the optimal conditions, the proposed method had good linear ranges as 0.1–40 μg/L for each of them, LOD of 0.11, 0.014, and 0.0025 μg/L, and LOQs of 0.37, 0.047, and 0.0083 μg/L for chlorpyrifos, phorate, and methyl parathion, respectively. The proposed method was validated with real environmental water samples and the spiked recoveries were over the range of 86.5–115.1%. All these results indicated that TiO2 nanotubes, as a new SPE adsorbent, would be used widespread for the preconcentraiton and determination of environmental pollutants in the future.  相似文献

A novel dispersive liquid‐phase microextraction method without dispersive solvents has been developed for the enrichment and sensitive determination of triclosan and triclocarban in environmental water samples prior to HPLC‐ESI‐MS/MS. This method used only green solvent 1‐hexyl‐3‐methylimidazolium hexafluorophosphate as extraction solvent and overcame the demerits of the use of toxic solvents and the instability of the suspending drop in single drop liquid‐phase microextraction. Important factors that may influence the enrichment efficiencies, such as volume of ionic liquid, pH of solutions, extraction time, centrifuging time and temperature, were systematically investigated and optimized. Under optimum conditions, linearity of the method was observed in the range of 0.1–20 μg/L for triclocarban and 0.5–100 μg/L for triclosan, respectively, with adequate correlation coefficients (R>0.9990). The proposed method has been found to have excellent detection sensitivity with LODs of 0.04 and 0.3 μg/L, and precisions of 4.7 and 6.0% (RSDs, n=5) for triclocarban and triclosan, respectively. This method has been successfully applied to analyze real water samples and satisfactory results were achieved.  相似文献

A LC‐MS/MS method was developed and validated for the determination of cyclosporine A (CsA) and its three phase 1 metabolites AM1, AM9, and AM4N in whole blood and lymphocytes isolated on the Histopaque gradient. 200 μL of whole blood was precipitated with 10 mol/L zinc sulfate in acetonitrile/methanol (40:60, v/v) and lymphocytes isolated from 1.5 mL blood were extracted with acetonitrile/methanol (40:60, v/v). The analytes and internal standard cyclosporine D were separated on RP column BEH C18, 2.1×50 mm, 1.7 μm using gradient LC‐MS/MS analysis in positive electrospray mode. Time of analysis was 5 min. Linearity in blood was 5–2000 μg/L for CsA, AM1, and AM9; 2–500 μg/L for AM4N; and 2–500 μg/L for all substances in lymphocytes. Coefficient of variations was 1.8–9.8% and recovery was 92.0–110.0%. The method was used in early and chronic renal transplant patients for therapeutic drug monitoring of CsA to compare either its share in lymphocytes as target organ or binding to one lymphocyte. The same parameters were calculated for all metabolites tested.  相似文献

A method for the rapid pretreatment and determination of bisphenol A in water samples based on vortex‐assisted liquid–liquid microextraction followed by high‐performance liquid chromatography with fluorescence detection was proposed in this paper. A simple apparatus consisting of a test tube and a cut‐glass dropper was designed and applied to collect the floating extraction drop in liquid–liquid microextraction when low‐density organic solvent was used as the extraction solvent. Solidification and melting steps that were tedious but necessary once the low‐density organic solvent used as extraction solvent could be avoided by using this apparatus. Bisphenol A was selected as model pollutant and vortex‐assisted liquid–liquid microextraction was employed to investigate the usefulness of the apparatus. High‐performance liquid chromatography with fluorescence detection was selected as the analytical tool for the detection of bisphenol A. The linear dynamic range was from 0.10 to 100 μg/L for bisphenol A, with good squared regression coefficient (r2 = 0.9990). The relative standard deviation (n = 7) was 4.7% and the limit of detection was 0.02 μg/L. The proposed method had been applied to the determination of bisphenol A in natural water samples and was shown to be economical, fast, and convenient.  相似文献

A simple homogenous liquid‐phase microextraction methodology applying octanoic acid as the extraction phase was introduced for determination of chlorobenzenes. In this approach, phase separation phenomenon occurred by changing pH of the solution. The extraction phase was isolated based on filtration of emulsion instead of centrifugation and it was introduced to the high‐performance liquid chromatography instrument as an on‐line procedure. This method was used for extraction and determination of five chlorobenzenes in different samples. Experimental design and response surface methodology were used for the optimization of various parameters influencing the extraction efficiency of the method. Under optimal conditions, chlorobenzenes were effectively extracted, and preconcentration factors of 255–294 were obtained. The calibration curves were investigated in the concentration range of 1–200 μg/L and good linearity was achieved with coefficient of determinations better than 0.997. Limits of detection of 0.1 and 0.3 μg/L and suitable precision with relative standard deviations better than 5.1% (n = 5) were attained. Finally, the proposed method was applied to determine the concentration of chlorobenzenes in different samples and acceptable recoveries were gained.  相似文献
