A novel o‐phenanthroline‐immobilized ionic‐liquid‐modified hybrid monolith for capillary electrochromatography was synthesized based on chloropropyl‐silica, which was prepared by the in situ polymerization of tetramethoxysilane and 3‐chloropropyltrimethoxysilane via a sol–gel process. The morphology of the hybrid monolith was characterized by scanning electron microscopy, and relatively stable anodic electroosmotic flow was observed under a broad pH ranged from pH 3.0 to 9.0. The separation mechanism was investigated by separating four neutral molecules (toluene, dimethylformamide, formamide, and thiourea). The obtained hybrid monolith possessed an obviously reversed‐phase retention mechanism, but when the acetonitrile content in the mobile phase was >90% v/v, a weak hydrophilic mechanism was observed on the resultant o‐phenanthroline‐modified chloropropyl‐silica hybrid monolith. The reproducibility of the column was also investigated by measuring relative standard deviations of the migration time for four neutral molecules. Relative standard deviations of run to run (n = 3), day to day (n = 3), and column to column (n = 3) were in the range of 0.4–0.7, 0.9–2.1, and 1.4–3.3%, respectively. Basic separations of various polar analytes including phenols and aromatic amines were successfully achieved.  相似文献

Pepsin‐modified affinity monolithic capillary electrochromatography, a novel microanalysis system, was developed by the covalent bonding of pepsin on silica monolith. The column was successfully applied in the chiral separation of (±)‐nefopam. Furthermore, the electrochromatographic performance of the pepsin‐functionalized monolith for enantiomeric analysis was evaluated in terms of protein content, pH of running buffer, sample volume, buffer concentration, applied voltage, and capillary temperature. The relative standard deviation (%RSD) values of retention time (intraday <0.53, n = 10; interday <0.53, n = 10; column‐to‐column <0.70, n = 20; and batch‐to‐batch <0.80, n = 20) indicated satisfactory stability of these columns. No appreciable change was observed in retention and resolution for chiral recognition of (±)‐nefopam in 50 days with 100 injections. The proteolytic activity of this stationary phase was further characterized with bovine serum albumin as substrate for online protein digestion. As for monolithic immobilized enzyme reactor, successive protein injections confirmed both the operational stability and ability to reuse the bioreactor for at least 20 digestions. It implied that the affinity monolith used in this research opens a new path of exploring particularly versatile class of enzymes to develop enzyme‐modified affinity capillary monolith for enantioseparation.  相似文献

A novel sulfoalkylbetaine‐based zwitterionic organic‐silica hybrid monolith was synthesized by using 3‐dimethyl‐(3‐(N‐methacrylamido) propyl) ammonium propane sulfonate (DMMPPS, neutral sulfoalkyl‐betaine monomer). The added amount of zwitterionic monomer was significantly increased when DMMPPS was used instead of the conventionally used acidic sulfoalkyl‐betaine monomer, that is, the N,N‐dimethyl‐N‐ methacryloxyethyl‐N‐(3‐sulfopropyl) ammonium betaine, and this led to a significantly improved hydrophilicity of the monolith. The DMMPPS‐based organic‐silica hybrid monolith exhibited good mechanical stability and excellent separation performance. About ~20 μm plate height (corresponding to column efficiency of ~50 000 plates/m) was obtained for nucleoside at the linear velocity of 1 mm/s. The proposed monolithic column was successfully applied to separate purines/pyrimidines, nucleotides, and tryptic digest of bovine hemoglobin in a nano‐HILIC mode, and the results demonstrated that such monolith has the potential for separation of a variety of hydrophilic substances.  相似文献

A simple “one‐pot” approach for the preparation of a new vinyl‐functionalized organic–inorganic hybrid monolithic column is described. In this improved method, the hydrolyzed alkoxysilanes of tetramethoxysilane and triethoxyvinylsilane were used as precursors for the synthesis of a silica‐based monolith, while 1‐hexadecene and sodium ethylenesulfonate were used as vinyl functional monomers along with azobisisobutyronitrile as an initiator. The effects of reaction temperature, urea content, and composition of organic monomers on the column properties (e.g. morphology, mechanical stability, and chromatographic performance) were investigated. The monolithic column was used for the separation of neutral solutes by reversed‐phase pressurized capillary. Furthermore, the monolith can separate various aromatic amines, which indicated its excellent cation‐exchange capability and hydrophobic interactions. The baseline separation of the aromatic amines was obtained with a column efficiency of up to 78 000 plates/m.  相似文献

A zirconium terephthalate metal‐organic framework‐incorporated poly(N‐vinylcarbazole‐co‐divinylbenzene) monolith was fabricated in a capillary by a thermal polymerization method. The optimized monolith had a homogeneous structure, good permeability, and stability. The monolith could be used for the effective enrichment of fungicides through π‐π interactions, electrostatic forces, and hydrogen bonds. The potential factors that affect the extraction efficiency, including ionic strength, solution pH, sample volume, and eluent volume, were investigated in detail. The monolith‐based in‐tube solid‐phase microextraction coupled with ultra‐high‐performance liquid chromatography and high‐resolution Orbitrap mass spectrometry was performed for the analysis of five fungicides (pyrimethanil, tebuconazole, hexaconazole, diniconazole, and flutriafol) in environmental samples. Under the optimized conditions, the linear ranges were 0.005–5 ng/mL for pyrimethanil, 0.01–5 ng/mL for flutriafol, and 0.05–5 ng/mL for other fungicides, respectively, with coefficients of determination ≥0.9911. The limits of detection were 1.34–14.8 ng/L. The columns showed good repeatability (relative standard deviations ≤9.3%, n = 5) and desirable column‐to‐column reproducibility (relative standard deviations 5.3–9.4%, n = 5). The proposed method was successfully applied for the simultaneous detection of five fungicides in water and soil samples, with recoveries of 90.4–97.5 and 84.0–95.3%, respectively.  相似文献

A new multiple monolithic fiber solid‐phase microextraction using a polydopamine‐based monolith as the extraction medium is proposed. The monolith was synthesized by facile in situ copolymerization of N‐methacryldopamine and dual cross‐linkers (divinylbenzene/ethylenedimethacrylate) in the presence of N ,N‐dimethylformamide. The effect of the contents of N‐methacryldopamine and porogen in the polymerization mixture on the extraction performance was investigated thoroughly. A series of characterization studies was performed to validate the structure and properties of the monolith. The prepared multiple monolithic fibers were used for the extraction of triazine herbicides in environmental water samples. After the optimization of the extraction parameters, a convenient, sensitive, cost‐effective, and environmentally friendly method for the determination of trace triazine herbicides in water samples was developed by coupling multiple monolithic fibers solid‐phase microextraction with high‐performance liquid chromatography and diode array detection. The results indicated that the limits of detection and quantification for the target compounds were 0.031–0.14 and 0.10–0.45 μg/L, respectively. Good precision and reproducibility were obtained with the relative standard deviations below 10%. The developed method was applied to the analysis of the triazine herbicides in different water samples (lake, river, and farmland waters). The recoveries of the method were in the range between 79.6 and 117%.  相似文献

A porous crosslinked organic polymer based on N‐acryloxysuccinimide (NAS) and ethylene dimethacrylate (EDMA) was prepared inside 75 µm i.d. fused silica capillary as functionalizable monolithic stationary phase for electrochromatographic applications. Succinimide groups on the monolith surface provide reactive sites able to react readily through standard electrophile‐nucleophile chemistry. Propargylamine was used to prepare alkyne functionalized poly(NAS‐co‐EDMA). Onto this thiol‐reactive polymer surface was grafted adamantane units via a photochemically‐driven addition reaction. Chemical characterization was performed in situ after each synthetic step by means of Raman spectroscopy and grafting kinetics was investigated to ensure quantitative grafting of 1‐adamantanethiol. The as‐designed monolithic stationary phase exhibited typical reversed‐phase separation mechanism as evidenced by the linear increase of the logarithm of retention factor of neutral aromatic solutes with the increase of the aqueous buffer content in the mobile phase.  相似文献

A monolithic capillary column with a mixed‐mode stationary phase of reversed‐phase/hydrophilic interaction chromatography was prepared for capillary liquid chromatography. The monolith was created by an in‐situ copolymerization of a homemade monomer N,N‐dimethyl‐N‐acryloxyundecyl‐N‐(3‐sulfopropyl) ammonium betaine and a crosslinker pentaerythritol triacrylate in a binary porogen agent consisting of methanol and isopropanol. The functional monomer was designed to have a highly polar zwitterionic sulfobetaine terminal group and a hydrophobic long alkyl chain moiety. The composition of the polymerization solution was systematically optimized to permit the best column performance. The columns were evaluated by using acidic, basic, polar neutral analytes, as well as a set of alkylbenzenes and Triton X100. Very good separations were obtained on the column with the mixed‐mode stationary phase. It was demonstrated that the mixed‐mode stationary phase displayed typic dual retention mechanisms of reversed‐phase/hydrophilic interaction liquid chromatography depending on the content of acetonitrile in the mobile phase. The method for column preparation is reproducible.  相似文献

A novel mercaptotetrazole‐silica hybrid monolithic column was prepared for capillary liquid chromatography, in which the thiol‐end mercaptotetrazole was mixed with hydrolyzed γ‐methacryloxypropyltrimethoxysilane and tetramethyloxysilane for the co‐polycondensation and thiol‐ene click reaction in a one‐pot process. The effects of the molar ratio of silanes, the amount of mercaptotetrazole, and the volume of porogen on the morphology, permeability and pore properties of the as‐prepared mercaptotetrazole‐silica hybrid monoliths were investigated in detail. A series of test compounds including alkylbenzenes, amides and anilines were employed for evaluating the retention behaviors of the mercaptotetrazole‐silica hybrid monolithic columns. The results demonstrated that the mercaptotetrazole‐silica hybrid monoliths exhibited hydrophobic, hydrophilic as well as ion‐exchange interaction. The run‐to‐run, column‐to‐column and batch‐to‐batch reproducibilities of the mercaptotetrazole‐silica hybrid monoliths were satisfactory with the relative standard deviations less than 1.4 (= 5), 3.9 (= 3) and 4.0% (= 5), respectively. In addition, the mercaptotetrazole‐silica hybrid monolith was further applied to the separation of sulfonamides, nucleobases and protein tryptic digests. These successful applications confirmed the promising potential of the mercaptotetrazole‐silica hybrid monolith in the separation of complex samples.  相似文献

A chloropropyl-functionalized silica (CP-silica) hybrid monolithic column was synthesized within the confines of a capillary via the sol–gel process using tetramethoxysilane (TMOS) and (3-chloropropyl)-trimethoxysilane (CPTMS) as the precursors. The resulting CP-silica hybrid monolith inside the capillary showed homogeneous macroporous morphology and was well attached to the inner wall of the capillary. The obtained CP-silica hybrid monolithic capillary column demonstrated the inherent hydrophobic property and could be applied as a reversed-phase stationary phase in CEC directly. Due to the great chemical reactivity of the incorporated chloro groups on the hybrid silica monolithic matrix, the chloropropyl moieties on the surface of the hybrid silica monolith matrix could be conveniently further modified by a tertiary amine of N,N-dimethyl-N-dodecylamine (DMDA) via the nucleophilic substitution reaction at 70 °C to introduce a dodecyl groups (C12) onto the CP-silica hybrid monolithic matrix. The resulting C12-silica hybrid monolithic column not only demonstrated the significantly enhanced hydrophobic property in the separation of alkylbenzenes in reversed-phase capillary electrochromatography (RP-CEC), but also the strong electroosmotic flow (EOF) in a wide pH range. Five alkylbenzenes could be baseline separated in 3 min with column efficiency ranging from 189 700 to 221 000 N/m with a 70% ACN running buffer in CEC.  相似文献

A range of porous carbon‐based monolithic (PCM) rods with flow‐through pore sizes of 1, 2, 5 and 10 μm, were produced using a silica particle template method. The rods were characterised using SEM and energy‐dispersive X‐ray spectroscopy, BET surface area and porous structure analysis, dilatometry and thermal gravimetry. SEM evaluation of the carbon monolithic structures revealed an interconnected rigid bimodal porous structure and energy‐dispersive X‐ray spectroscopy analysis verified the quantitative removal of the embedded silica beads. The specific surface areas of the 1, 2, 5 and 10 μm rods were 178, 154, 84 and 125 m2/g after pyrolysis and silica removal, respectively. Shrinkage of the monolithic rods during pyrolysis is proportional to the particle size of the silica used and ranged from 9 to 12%. Mercury porosimetry showed a narrow distribution of pore sizes, with an average of ~700 nm for the 1 μm carbon monolith. The suitability of bare and surface oxidised PCM rods for the use as a stationary phase for reversed and normal phase LC was explored. The additional modification of PCM rods with gold micro‐particles followed by 6‐mercaptohexanoic acid was performed and ion‐exchange properties were evaluated.  相似文献

A novel type of macroporous molecularly imprinted hybrid silica monolithic column was first developed for recognition of proteins. The macroporous silica-based monolithic skeleton was synthesized in a 4.6 mm i.d. stainless steel column by a mild sol–gel process with methyltrimethoxysilane (MTMS) as a sole precursor, and then vinyl groups were introduced onto the surface of the silica skeleton by chemical modification of γ-methacryloxypropyltrimethoxysilane (γ-MAPS). Subsequently, the molecularly imprinted polymer (MIP) coating was copolymerized and anchored onto the surface of the silica monolith. Bovine serum albumin (BSA) and lysozyme (Lyz), which differ greatly in molecular size, isoelectric point, and charge, were representatively selected for imprinted templates to evaluate recognition property of the hybrid silica-based MIP monolith. Some important factors, such as template–monomer molar ratio, total monomer concentration and crosslinking density, were systematically investigated. Under the optimum conditions, the obtained hybrid silica-based MIP monolith showed higher binding affinity for template than its corresponding non-imprinted (NIP) monolith. The imprinted factor (IF) for BSA and Lyz reached 9.07 and 6.52, respectively. Moreover, the hybrid silica-based MIP monolith displayed favorable binding characteristics for template over competitive protein. Compared with the imprinted silica beads for stationary phase and in situ organic polymer-based hydrogel MIP monolith, the hybrid silica MIP monolith exhibited higher recognition, stability and lifetime.  相似文献

We synthesized two kinds of chiral polystyrene‐based monoliths, which are macroporous gel with continuous open‐celled monolith structure. Thus, two chiral styrene monomers, (–)‐p‐[dimethyl(10‐pinanyl)silyl]styrene ((–)‐PSSt) and (–)‐p‐(menthoxycarbonyl)styrene ((–)‐MtSt]), were prepared and subjected to water‐in‐oil emulsion polymerization in the presence of divinylbenzene and AIBN. The macroporous structure of the obtained monoliths was directly confirmed by SEM observation. The obtained monoliths showed an optical resolution ability. That is, in the enantioselective adsorption using trans‐stilbene oxide, poly[(–)‐PSSt] monolith and poly[(–)‐MtSt] monoliths preferentially adsorbed (S,S)‐isomer [poly[(–)‐PSSt] monolith: α(S,S) = 1.49 (0.25 wt % acetone solution); poly[(–)‐MtSt] monolith: α(S,S) = 1.39 (0.25 wt % toluene solution)]. Depinanylsilylation of the poly[(?)‐PSSt] monolith and removal of menthyl groups from the poly[(–)‐MtSt] monolith were achieved by acid‐catalyzed scission of the Si? C bond and base‐catalyzed hydrolysis, respectively. In addition, de‐poly[(–)‐PSSt] and de‐poly[(–)‐MtSt] showed enantioselectivity ((S,S)‐isomer preferentially absorbed) in adsorption using trans‐stilbene oxide in spite of the absence of chiral substituents in the monoliths. © 2005 Wiley Periodicals, Inc. In this study, poly(vinylphosphonic acid‐co‐ethylene dimethacrylate), poly(VPA‐co‐EDMA) capillary monolith was synthesized as a starting material for obtaining a stationary phase for microscale enrichment of phosphopeptides. The chelation of active phosphonate groups with Ti (IV) ions gave a macroporous monolithic column with a mean pore size of 5.4 μm. The phosphopeptides from different sources were enriched on Ti (IV)‐attached poly(VPA‐co‐EDMA) monolith using a syringe‐pump. The monolithic capillary columns exhibited highly sensitive/selective enrichment performance with phosphoprotein concentrations as low as 1.0 fmol/mL. Six different phosphopeptides were detected with high intensity by the treatment of β‐casein digest with the concentration of 1.0 fmol/mL, using Ti (IV)@poly(VPA‐co‐EDMA) monolith. Highly selective enrichment of phosphopeptides was also successfully carried out even at trace amounts, in a complex mixture of digested proteins (molar ratio of β‐casein to bovine serum albumin, 1:1500) and three phosphopeptides were successfully detected. Four highly intense signals of phosphopeptides in human serum were also observed with high signal‐to‐noise ratio and a clear background after enrichment with Ti (IV)@poly(VPA‐co‐EDMA) monolith. It was concluded that the capillary microextraction system enabled fast, efficient and robust enrichment of phosphopeptides from microscale complex samples. The whole enrichment process was completed within 20 min, which was shorter than in the previously reported studies.  相似文献

This study describes stationary counterflow isotachophoresis (ITP) in a poly(acrylamide‐co‐N,N′‐methylenebisacrylamide) monolithic column as a means for improving ITP processing capacity and reducing dispersion. The flow profile in the monolith was predicted using COMSOL's Brinkman Equation application mode, which revealed that the flow profile was mainly determined by monolith permeability. As monolith permeability decreases, the flow profile changes from a parabolic shape to a plug shape. An experimental monolithic column was prepared in a fused‐silica capillary using an ultraviolet‐initiated polymerization method. A monolithic column made from 8% (wt.) monomer was chosen for the stationary counterflow ITP experiments. Counterflow ITP in the monolithic column showed undistorted analyte zones with significantly reduced dispersion compared to the severe dispersion observed in an open capillary. Particularly, for r‐phycoerythrin focused by counterflow ITP, its zone width in the monolithic column was only one‐third that observed in an open capillary. These experiments demonstrate that stationary counterflow ITP in monoliths can be a robust and practical electrofocusing method.  相似文献

A pyridinium‐based immobilized ionic liquid type multifunctional hybrid silica monolith was prepared by the in situ polymerization of 3‐chloropropyl‐silica matrix and 4,4′‐dipyridyl for hydrophilic interaction CEC. The obtained hybrid monolith possessed of high stable skeletal microstructures with obviously hydrophilic retention mechanism under ACN content >50% in the mobile phase. Strong and stable anodic EOF could be observed under a broad pH range from pH 3.0 to 9.0. Due to the immobilized dipyridyl groups bonded to the silica matrix surface, the resulting hydrophilic hybrid monolith possessed multiple separation interactions including hydrogen bond, π–π, and anion exchange. Excellent separations of various polar analytes including electroneutral phenols, charged acid nucleotides, and basic analytes were successfully achieved. The highest column efficiencies up to 120 000, 164 000, and 106 000 plates/m were obtained for nucleotides, nucleic acid bases, and nucleosides and nicotines, respectively. These results demonstrated that the dipyridyl‐immobilized ionic liquid functionalized hybrid monolith possessed highly mechanical stability and good chromatographic performance for hydrophilic interaction electrochromatography.  相似文献

In the present study, a novel poly(acrylamide‐coN‐isopropylacrylamide‐co‐ethylene dimethacrylate) monolithic column modified with saline‐modified delaminated layered double hydroxides was developed and exploited as the stationary phase in the polymer monolith microextraction of β‐agonists. Transmission electron microscopy, Fourier transform infrared spectroscopy, scanning electron microscopy, X‐ray photoelectron spectroscopy, and thermal gravimetric analysis with derivative thermogravimetric determination were employed to characterize the monoliths. By coupling with high‐performance liquid chromatography determinations, extraction parameters affecting the extraction efficiency, including sample volume, flow rate, sample pH, eluent volume, and eluent flow rate were investigated with an orthogonal experiment design, L16 (45). Results demonstrated that under the optimal experimental conditions, low limits of detection in the range of 0.025–0.280 ng/mL were obtained with relative standard deviations in the range of 4.3–7.6% (intraday) and 6.7–8.8% (interday). When the developed method was applied to the analysis of β‐agonists in pork samples, recovery values were obtained in the range of 80.5−108.6%.  相似文献

Chemical characterization was performed in situ after each synthetic step by means of Raman spectroscopy. Good enantioseparations of flavanone enantiomers, chosen as test chiral compound, were achieved under reversed phase conditions by both capillary electrochromatography and nano‐liquid chromatography (nano‐LC) techniques. These results demonstrate the potentiality and usefulness of click chemistry in the preparation of β‐CD containing chiral organic polymer monolith.

In this work, we developed a novel molecularly imprinted solid‐phase microextraction with capillary electrophoresis method for the selective extraction and determination of protoberberines in complicated samples. The imprinted monolith was prepared in a micropipette tip‐based device by using acrylamide as the functional monomer, ethyleneglyoldimethacrylate as the cross‐linker and dimethylsulfoxide as the porogen, and exhibited an imprinting factor of 2.41 to berberine, 2.36 to palmatine and 2.38 to jatrorrhizine. Good capillary electrophoresis separation was achieved by using 20 mM phosphate buffer at pH 7 as running buffer with the addition of organic modifier of 10% methanol. Parameters such as sample pH value, sample flow rate and sample volume were investigated for imprinted monolith‐based solid‐phase microextraction. An imprinted solid‐phase microextraction with capillary electrophoresis method was developed, the method showed a wide linear range (0.3–50 μg/mL), good linearity (R2 ≥ 0.9947) and good reproducibility (relative standard deviations ≤ 0.73%), the limit of detection was as low as 0.1 μg/mL, which was lower than some reported methods based on capillary electrophoresis for protoberberines. The method has been applied for determination of three common protoberberines in Cortex Phellodendri Chinensis, by using a molecularly imprinted monolith as the selective sorbent, most of the matrices in the Cortex Phellodendri Chinensis sample were removed and three protoberberines were selectively enriched and well determined.  相似文献

In this study, polyimide–silica (PI–silica) based hybrid coating compositions were prepared from tetraethoxysilane (TEOS), γ‐glycidyloxypropyl trimethoxy silane (GOTMS), and polyamic acid (PAA) via a combination of sol–gel and thermal imidization techniques. PAA was synthesized from 3,3′,4,4′‐benzophenone tetracarboxylic dianhydride (BTDA) and 3,3'‐Diaminodiphenyl sulfone (DDS) in N‐Methyl‐2‐pyrrolidone (NMP). The silica content in the hybrid coatings was varied from 0 to 20 wt%. The structural characterization of the hybrid coatings was performed using FTIR and 29Si‐NMR spectroscopies. Results from both pendulum hardness and micro indentation test show that the hardness of hybrid coatings improves with the increase in silica content. The tensile tests also demonstrated that the mechanical properties at low silica content are rather striking. Their surface morphologies were characterized by scanning electron microscopy (SEM). SEM studies revealed that inorganic particles were distributed homogenously through the PI matrix. It was also found that, incorporation of the silica domains increased the thermal stability of the hybrid coatings. Copyright © 2007 John Wiley & Sons, Ltd.  相似文献
