We herein describe the development and validation of an HPLC method for the quantitation of 7‐(benzylamino)‐1,3,4,8‐tetrahydropyrrolo [4,3,2‐de]quinolin‐8(1H)‐one (BA‐TPQ), a newly synthesized iminoquinone anticancer agent. BA‐TPQ was extracted from plasma and tissue samples by first precipitating proteins with acetonitrile followed by a liquid–liquid extraction with ethyl acetate. Chromatographic separation was carried out using a gradient flow rate on a Zorbax SB C18 column, and the effluent was monitored by UV detection at 346 nm. The method was found to be precise, accurate, and specific, with a linear range of 3.91–1955.0 ng/mL in plasma, 19.55–1955.0 ng/mL in spleen, brain, and liver homogenates and 19.55–3910.0 ng/mL in heart, lung and kidney homogenates. The method was stable under all relevant conditions. Using this method, we also carried out an initial study determining plasma pharmacokinetics and tissue distribution of BA‐TPQ in mice following intravenous administration. In summary, this simple and sensitive HPLC method can be used in future preclinical and clinical studies of BA‐TPQ.  相似文献

Abstract

A series of new pyrido-cyclopenta[1,2-b]indole derivatives were synthesized via Knoevenagel reaction and followed by reflux with dimethylformamide dimethyl acetal. Their structures were investigated by spectral techniques and elemental analysis. In vitro antibacterial assessment against seven selected microorganisms evidenced that the compounds with halogen substituent have strong inhibitory action than that of the reference drugs. The antioxidant results were apparent that the compounds 5b, 5c, and 6c manifested explicit activity when compared with Butylhydroxyanisole and Vitamin-C. Cytotoxic activity analysis toward HeLa and MCF-7 cell lines was also assessed. Analogs 6c (IC50 values 15.1?μM and 18.6?μM) and 6d (IC50 values 17.4?μM and 20.7?μM) illustrated the interesting cytotoxicity activity. Molecular docking studies against p38 MAP kinase displayed a potential binding affinity with the receptor. Furthermore, in silico pharmacokinetic studies articulated the drug-likeness nature of the target compounds.  相似文献

A series of novel triazolothione, thiadiazole, triazole, and oxadiazole-functionalized pyrido[1,2-a]pyrimidin-4-ones 6a–6l and 7a–7f were prepared, starting from pyridine derivatives 1a and 1b. All the compounds 6a–6l and 7a–7f were screened against four human cancer cell lines (HeLa, COLO205, Hep G2, and MCF 7), among which compounds 6d, 6h, 6j, 7b, and 7e showed promising anticancer activity. Molecular docking studies showed good binding energy and exhibited interactions and better lower free energy values, indicating more thermodynamically favored interaction.  相似文献

A novel, simple, specific, sensitive and reproducible high‐performance liquid chromatography (HPLC) assay method has been developed and validated for the estimation of tofacitinib in rat plasma. The bioanalytical procedure involves extraction of tofacitinib and itraconazole (internal standard, IS) from rat plasma with a simple liquid–liquid extraction process. The chromatographic analysis was performed on a Waters Alliance system using a gradient mobile phase conditions at a flow rate of 1.0 mL/min and C18 column maintained at 40 ± 1 °C. The eluate was monitored using an UV detector set at 287 nm. Tofacitinib and IS eluted at 6.5 and 8.3 min, respectively and the total run time was 10 min. Method validation was performed as per US Food and Drug Administration guidelines and the results met the acceptance criteria. The calibration curve was linear over a concentration range of 182–5035 ng/mL (r2 = 0.995). The intra‐ and inter‐day precisions were in the range of 1.41–11.2 and 3.66–8.81%, respectively, in rat plasma. The validated HPLC method was successfully applied to a pharmacokinetic study in rats. A simple, mild and efficient one‐pot approach for the construction of 2‐aryl‐3‐nitro‐2,9‐dihydrothiopyrano[2,3‐b]indole derivatives has been realized in CH2Cl2 medium at ambient temperature via three‐component tandem reaction of N‐protected‐2‐chloro‐3‐formylindoles, sodium hydrosulfide and β‐substituted nitroolefins/δ‐substituted nitrodienes using DABCO (10 mol%) as an organocatalyst, followed by dehydration in the presence of activated molecular sieves (4 Å). The significant advantages of this protocol are simple operation, shorter reaction time, high atom economy, good to high yields (73% –89%) and wider substrate scope. In addition, all the synthesized compounds have shown the large positive Stokes shift values (5632–6081 cm?1).  相似文献

A simple, sensitive and reproducible high‐performance liquid chromatography (HPLC) assay method was developed for the estimation of 3‐pentylbenzo[c]thiophen‐1(3H)‐one (S5), a potential anti‐ischemic stroke agent, in dog plasma. The analytical procedure involves protein precipitation of S5 and nobiletin (internal standard) from dog plasma with acetonitrile. Chromatographic separation was achieved on Sapphire C18 analytical column with methanol–water (80:20, v/v) as mobile phase. The eluate was monitored using a UV detector set at 260 nm. The calibration curves were linear over the range of 0.2–20 µg/mL. Absolute recoveries of S5 were 79.2–86.1% from dog plasma. The intra‐ and inter‐day relative standard deviation precisions were <7 and 5%, respectively. The method was successfully applied to the pharmacokinetic study of S5 in beagle dogs. A simple, specific, sensitive and reproducible high‐performance liquid chromatography (HPLC) assay method has been developed and validated for the estimation of odanacatib in rat and human plasma. The bioanalytical procedure involves extraction of odanacatib and itraconazole (internal standard, IS) from a 200 μL plasma aliquot with simple liquid–liquid extraction process. Chromatographic separation was achieved on a Symmetry Shield RP18 using an isocratic mobile phase at a flow rate of 0.7 mL/min. The UV detection wave length was 268 nm. Odanacatib and IS eluted at 5.5 and 8.6 min, respectively with a total run time of 10 min. Method validation was performed as per US Food and Drug Administration guidelines and the results met the acceptance criteria. The calibration curve was linear over a concentration range of 50.9–2037 ng/mL (r2 = 0.994). The intra‐ and inter‐day precisions were in the range of 2.06–5.11 and 5.84–13.1%, respectively, in rat plasma and 2.38–7.90 and 6.39–10.2%, respectively, in human plasma. An efficient method for the synthesis of novel benzo[b]pyrido[3',2':4,5] thieno[2,3-e][1,6] naphthyridine-8-one derivatives has been developed using a Pictet-Spengler reaction between 4-(3-aminothieno[2,3-b]pyridin-2-yl)quinoline-2-ones, which could be obtained from the alkylation of 4-bromomethylquinoline-2-ones with 3-cyanopyridine-2-thione followed by a Thorpe-Ziegler isomerization, and aromatic aldehydes under p-TsOH as catalysis in good yields.  相似文献

A solvent-free, one-pot, and operationally simple protocol was adopted to synthesize a new series of multifunctionalized pyrido[2,3-a]carbazole derivatives which were structurally characterized by FTIR, 1H-NMR, 13C-NMR, and elemental analysis. The synthesized compounds were screened in vitro for their antibacterial activity and the compounds showed weak to moderate inhibition of bacterial growth. The antioxidant properties of the compounds were evaluated by their 2,2-diphenyl-1-picrylhydrazyl free radical scavenging activity and total antioxidant capacity. Moreover, the cytotoxic effect of the compounds was examined on cancerous cell lines MCF-7 and A549 under in vitro conditions and the results showed that the compounds exhibited significant anticancer activity. Furthermore, the morphological changes and apoptosis induction were studied by inverted light microscopy, acridine orange/ethidium bromide, and 4′,6-diamidino-2-phenylindole dihydrochloride fluorescence microscopic analyses. The results indicated that among pyridocarbazole compounds, 2-ethoxy-8-chloro-4-(thiophen-2′-yl)-5,6-dihydro-11H-pyrido[2,3-a]carbazol-3-carboxamide 6g could be exploited as significant antioxidant and anticancer agents.  相似文献

An operationally simple, green and efficient procedure for one‐pot synthesis of novel polycyclic heteroaromatic compounds such as benzamidonaphtho[2,1‐b]furans and benzamidobenzo[b]furans has been developed from the reaction of arylglyoxals, benzamide, and phenols. Thieno[2,3‐b]indole derivatives were efficiently prepared via the reaction of 1,3‐dihydro‐2H‐indole‐2‐thiones with α‐bromo‐substituted ketones or aldehydes and in the presence of Et3N (Scheme 2 and Table). The bioanalytical procedure involves extraction of Orteronel and phenacetin (internal standard) from rat plasma with a simple liquid–liquid extraction process. The chromatographic analysis was performed on a Waters Alliance system using a gradient mobile phase conditions at a flow rate of 1 mL/min and a C18 column maintained at ambient room temperature. The eluate was monitored using a photodiode array detector set at 242. Orteronel and internal standard eluted at 4.8 and 6.2 min, respectively and the total run time was 9 min. Method validation was performed as per US Food and Drug Administration guidelines and the results met the acceptance criteria. The calibration curve was linear over a concentration range of 100–3149 ng/mL (r2 = 0.995). The intra‐ and inter‐day precisions were in the ranges of 0.31–7.87 and 3.97–6.35, respectively, in rat plasma. A specific, sensitive and stable high‐performance liquid chromatographic–tandem mass spectrometry (LC‐MS/MS) method was developed and validated for the quantitative determination of methyl 3‐amino‐6‐methoxythieno [2,3‐b]quinoline‐2‐carboxylate (PU‐48), a novel diuretic thienoquinolin urea transporter inhibitor in rat plasma. In this method, the chromatographic separation of PU‐48 was achieved with a reversed‐phase C18 column (100 × 2.1 mm, 3 μm) at 35°C. The mobile phase consisted of acetonitrile and water with 0.05% formic acid added with a gradient elution at flow rate of 0.3 mL/min. Samples were detected with the triple‐quadrupole tandem mass spectrometer with multiple reaction monitoring mode via electrospray ionization source in positive mode. The retention time were 6.2 min for PU‐48 and 7.2 min for megestrol acetate (internal standard, IS). The monitored ion transitions were mass‐to‐charge ratio (m/z) 289.1 → 229.2 for PU‐48 and m/z 385.3 → 267.1 for the internal standard. The calibration curve for PU‐48 was linear over the concentration range of 0.1–1000 ng/mL (r2 > 0.99), and the lower limit of quantitation was 0.1 ng/mL. The precision, accuracy and stability of the method were validated adequately. The developed and validated method was successfully applied to the pharmacokinetic study of PU‐48 in rats.  相似文献

This paper describes a sensitive, specific and rapid high‐performance liquid chromatography (HPLC) method for the determination of curcumin in rat plasma. After a simple step of protein precipitation in 96‐well format using acetonitrile containing the internal standard (IS), emodin, plasma samples were analyzed by reverse‐phase HPLC. Curcumin and the IS emodin were separated on a Diamonsil C18 analytical column (4.6 × 100 mm, 5 µm) using acetonitrile–5% acetic acid (75:25, v/v) as mobile phase at a flow rate of 1.0 mL/min. The method was sensitive with a lower limit of quantitation of 1 ng/mL, with good linearity (r2 ≥ 0.999) over the linear range 1–500 ng/mL. All the validation data, such as accuracy and precision, were within the required limits. A run time of 3.0 min for each sample made high‐throughput bioanalysis possible. Heterocyclization of bis(2‐oxo‐indol‐3‐ylidene)‐galactaric acid hydrazide ( 3 ) with a variety of one‐nitrogen cyclizing agents gave the corresponding 1,4‐bis{1,2,4‐triazino[5,6‐b]indol‐3‐yl}‐galacto‐tetritols 4–8 . Acetylation of the latter double headed acyclo C‐nucleosides with acetic anhydride in the presence of pyridine at ambient temperature resulted in N‐ and O‐acetylation to give the corresponding 1,2,3,4‐tetra‐O‐acetyl‐1,4‐bis{1,2,4‐triazino[5,6‐b]indol‐3‐yl}‐galacto‐tetritols 9–13 which were found to exist in centro‐symmetric zigzag conformations 20 . The assigned structures were corroborated by 1H, 13C NMR as well as mass spectra.  相似文献

Alternating narrow band gap (NBG) conjugated polymers derived from 6,6′,12,12′‐tetraoctylindeno[1,2‐b]fluorene (IF) and 2,3‐dimethyl‐5,7‐dithien‐2‐yl‐thieno[3,4‐b]pyrazine (DTTP), 2,3‐diphenyl‐5,7‐dithien‐2‐yl‐thieno[3,4‐b]pyrazine (DPTP) or 2,3‐dioctyl‐5,7‐dithien‐2‐yl‐thieno[3,4‐b]pyrazine (DOTP), named as PIF‐DTTP, PIF‐DPTP, and PIF‐DOTP, respectively, were synthesized by Suzuki coupling reaction and characterized. The photochemical stabilities of the copolymers and copolymer derived from IF and 5,7‐dithien‐2‐yl‐thieno[3,4‐b]pyrazine (DTP) were investigated by the UV absorptions, PL spectra, FT‐IR spectra, and photovoltaic properties of the copolymers as a function of UV irradiation time. The studies revealed that the degradation of thieno[3,4‐b]pyrazine (TP) ring under UV irradiation can be retarded or eliminated by introducing phenyl group into the 2,3‐positions of TP ring, and indicated that 2,3‐diphenylthieno[3,4‐b]pyrazine could be used as durable electron deficient moiety to achieve donor–acceptor NBG‐conjugated polymers. © 2011 Wiley Periodicals, Inc. A number of ruthenium complexes were prepared and their catalytic activities in three‐component one‐pot condensation of aldehydes, malononitrile and 4‐hydroxycoumarin or dimedone was considered to afford dihydropyrano[3,2‐c]chromenes and tetrahydrobenzo[b]pyran derivatives under optimum reaction conditions.
