Abstract

A fast potentiometric determination method has been reported for pentazocine in human plasma without complicated pretreatments using a coated-wire potentiometric selective electrode. The sensing membrane was made by incorporating of ion-association complexes of pentazocine cation and sodium tetraphenyl borate (NaTPB) in a polyvinyl chloride. The sensor exhibited fast, stable, and linear Nernstian response over the range of 5 × 10?5 to 0.1 mol L?1 pentazocine with a slope of 57.8 mV per decade and with detection limit of 3.2 × 10?5 mol L?1. The proposed sensor has been used for determination of pentazocine in human plasma and urine.  相似文献

采用超高效液相色谱-四极杆飞行时间高分辨质谱(UPLC/Q-TOF MS)联用技术,对大鼠血浆磷脂提取液进行分析,建立了一种快速、简便、可靠的血浆磷脂鉴别方法。以经典的氯仿甲醇法为磷脂的提取方法,以10 mmol/L甲酸铵水溶液(A)-含10 mmol/L甲酸铵的乙腈-异丙醇(1∶1)混合溶液(B)为流动相,采用ESI源,分别在正、负离子模式下进行数据采集。根据实验结果,结合二级质谱裂解数据、元素组成、数据库匹配,鉴定出81个成分,其中包括7个PE、7个PI和1个SM,其余均为PC。所建立的液相色谱-质谱联用分析方法,可为大鼠血浆中磷脂成分的鉴别提供一种快速、简便、可靠的分析手段,以及为血浆磷脂代谢组学的研究奠定基础。  相似文献

Abstract

In this research, new electrodes were prepared by incorporating zeolite KY into sol-gel matrix. Calibration plot with Nernstian slope (58.6 ± 0.5 mV/decade) for cesium ion was observed over a linear range of about four decades of concentration (1.0 × 10?5 to 1.0 × 10?1 M, at 25°C). The sol-gel electrode shows detection limit of 7.3 × 10?6 M. The influences of membrane composition, the pH of the test solution, and the interfering ions on the electrode performance were investigated. The effect of temperature on the electrode response showed that the temperature higher than 60°C deteriorates the electrode performance. The proposed electrode has high mechanical and thermal stability with a low isothermal temperature coefficient, and it is 0.00018 V/°C. It has a short response time (10 s) and can be used for more than 6 months without any considerable divergence in the potentials. The selectivity coefficients for Cs+ ion with respect to alkali, alkaline earth, ammonium, and some heavy-metal ions were investigated. The results of application show that the electrode can be used successfully in determination of Cs+ ions in various water samples.  相似文献

The effects of sample preparation and chromatographic method differences on the classification and recovery of metabolic biomarkers from UPLC‐MS measurements on urine samples of humans exposed to different dietary interventions have been investigated. Eight volunteers consumed three high‐fat meals (rich in saturated, monounsaturated and polyunsaturated fatty acids, respectively) in randomized order with a washout period in between. For each participant, urine samples were obtained prior to and at three timed intervals after each meal. Samples were processed either by dilution (1 : 4) or by liquid–liquid extraction and then run under two different gradient conditions. For each analysis method, a total of 96 observations (eight participants, four time points, three diets) were measured. The total ion count chromatograms were analyzed using partial‐least‐squares discriminant analysis. All three dietary classes could be discriminated irrespective of sample preparation and chromatographic method. However, the main discriminating metabolites varied according to sample preparation, indicating that sample treatment and chromatographic conditions influence the ability to extract biomolecular information. Diluted samples showed higher m/z compounds (ca 400 u) while liquid–liquid extraction samples showed low m/z at the same retention time span. Optimized methods for metabolite identification (e.g. organic acids) were statistically inferior to global screening for mixed compound identification, confirming that multiple compound class‐based metabolic profiles are likely to give superior metabonomic (diagnostic) classification, although great care has to be taken in the interpretation in relation to matrix effects. Nitrate sensing is an important application for potentiometry in environmental applications. Its recognition is by ion-exchangers whose selectivity is governed by the lipophilicity of the ions in solution. Yet, considerable research efforts have been dedicated to the development of such ionophores and some have already been commercialized. This work examines two commercially available nitrate ionophores, nitrate ionophores V and VI, and compares their performance with widely used ion-exchangers by determining the resulting membrane selectivity and complexation to nitrate. Unfortunately, adding a nitrate ionophore to the membrane did not result in improved selectivity. Sandwich membrane experiments indicated that binding interaction is too weak to be measurable, with a logarithmic formation constant of just 1.36±0.14 for ionophore V with nitrate. Ways to improve the reporting of relevant data are suggested.  相似文献

An easy‐to‐handle magnetic dispersive solid‐phase extraction procedure was developed for preconcentration and extraction of cocaine and cocaine metabolites in human urine. Divinyl benzene and vinyl pyrrolidone functionalized silanized Fe3O4 nanoparticles were synthesized and used as adsorbents in this procedure. Scanning electron microscopy, vibrating sample magnetometry, and infrared spectroscopy were employed to characterize the modified adsorbents. A high‐performance liquid chromatography with mass spectrometry method for determination of cocaine and its metabolites in human urine sample has been developed with pretreatment of the samples by magnetic dispersive solid‐phase extraction. The obtained results demonstrated the higher extraction capacity of the prepared nanoparticles with recoveries between 75.1 to 105.7% and correlation coefficients higher than 0.9971. The limits of detection for the cocaine and cocaine metabolites were 0.09–1.10 ng/mL. The proposed magnetic dispersive solid‐phase extraction method provided a rapid, environmentally friendly and magnetic stuff recyclable approach and it was confirmed that the prepared adsorbents material was a kind of highly effective extraction materials for the trace cocaine and cocaine metabolites analyses in human urine.  相似文献

In order to evaluate recent cocaine exposure or its coingestion with ethanol, a simple and sensitive solid-phase microextraction (SPME) procedure for determination of cocaine and cocaethylene in urine was developed and validated. A polydimethylsiloxane fibre (100 microm) was submersed in the urine sample for 20 min under magnetic stirring after alkalinization with solid buffer (NaHCO(3):K(2)CO(3), 2:1). Gas chromatography-mass spectrometry (GC-MS) was used to identify and quantify the analytes in selected ion monitoring mode (SIM). The limits of quantification were 5.0 ng/mL for both analytes. Good inter- and intra-assay precision was also observed (coefficient of variation <9%).  相似文献

Warfarin is routinely monitored by assessing its pharmacologic effects on the international normalized ratio. However, having a patient with INR not responding to increasing warfarin dose mandates a direct measurement of warfarin concentrations (total and free) for better patient clinical management of warfarin therapy. Therefore, a new fully validated specific, precise and accurate ultra-performance liquid chromatography tandem mass spectrometry was developed for the determination of free and total warfarin in human plasma. Free warfarin was measured in plasma filtrate, prepared by ultrafiltration, and sample pretreatment involved protein precipitation with acetonitrile. Linear response (r(2) ≥0.99) was observed over the studied range of free and total warfarin, with the lower limit of detection of 0.25 ng/mL. The intra- and inter-day precision (relative standard deviation) values were <10% and the accuracy (relative error) was ≤6.6 for free and total warfarin. There was no significant difference (p>0.05) between inter- and intra-day studies for the free and total warfarin, which confirmed the reproducibility of the assay method. The mean extraction efficiency was 88.6-107.2% of free and total warfarin. The assay was sensitive to follow warfarin pharmacokinetics (free and total) in a patient with resistance to warfarin up to 24 h after a daily dose of warfarin.  相似文献

A method for the determination of fluoride in coal, coal fly ash, phosphate rock, limestone, mineral clay, fossilised materials, oyster tissue and vegetation using pyrohydrolysis for sample decomposition is proposed. A specific apparatus was constructed and the influence of vanadium pentoxide (V2O5) as a catalyst for the pyrohydrolysis reaction was investigated. It was verified that V2O5 does not influence the release of fluoride from the vegetation, oyster tissue, coal and coal fly ash matrices analysed. However, the catalyst was necessary for the phosphate rock, fossil bone, mineral clay and limestone samples. Certified and noncertified samples were analysed using an ion selective electrode (ISE) for the analyte detection. Precise (relative standard deviation—R.S.D.<7%) and accurate (recovery in accordance to certified values) results were obtained. The limit of quantification (LOQ) of the method was 5.0 μg g−1 of fluoride using 20 mg of sample and a final dilution to 10 ml. The sample frequency was five samples per hour.  相似文献

Matrix-assisted laser desorption/ionisation (MALDI) mass spectrometry was used for the rapid detection of cocaine, benzoylecgonine and cocaethylene in hair. Different MALDI sample preparation procedures have been tested and the employment of a multi-layer 'graphite-sample-electrosprayed alpha-cyano-4-hydroxycinnamic acid (HCCA)' yielded the best results for standard solutions of the target analytes. The same approach was subsequently applied to hair samples that were known to contain cocaine, benzoylecgonine and cocaethylene, as determined by a classical GC-MS method. It was however necessary to extract hair samples by incubating them in methanol/trifluoroacetic acid for a short time (15 min) at 45 degrees C; 1 microl of the obtained supernatant was deposed on a metal surface treated with graphite, and HCCA was electrosprayed on it. This procedure successfully suppressed matrix peaks and was effective in detecting all the target analytes as their protonated species. The results obtained give further confirmation of the effectiveness of the MALDI for detecting drugs and their metabolites in complex biological matrices. The method can be useful as a fast screening procedure to detect the presence of cocaine and metabolites in hair samples.  相似文献

The goal of this work was to develop and validate an analytical method for the detection and quantification of the biogenic amines serotonin (5‐HT), dopamine (DA) and norepinephrine (NE), using microextraction in packed syringe (MEPS) and liquid chromatography coupled to electrochemical detection (HPLC‐ED) in urine. The method was validated according to internationally accepted guidelines from the Food and Drug Administration. Linearity was established between 50 and 1000 ng/mL for 5‐HT and between 5 and 1000 ng/mL for DA and NE, with determination coefficients (R2) >0.99 for all compounds. The limits of quantification and detection were respectively 50 and 20 ng/mL for 5‐HT, and 5 and 2 ng/mL for DA and NE. Within‐ and between‐run precision ranged from 0.84 to 9.41%, while accuracy ranged from 0.79 to 12.76% for all compounds. The intermediate precision and accuracy were 1.50–8.36 and 0.54–13.51%, respectively. The method was found suitable for clinical routine analysis of the studied compounds, using a sample volume of 0.5 mL. This is the first study employing a commercially available MEPS column for the simultaneous detection and quantification of 5‐HT, DA and NE in urine by coulometric detection. Histamine levels in fish, extracted with methanol, were determined by capillary electrophoresis (CE) using phosphate buffer pH 2.5 and U.V. detection at 210 nm. Histamine was well separated from the other co-extracted components under the given CE condition without any cleanup of the methanol extract. The average recovery of spiked histamine in various types of fish samples was 96%. Using the same methanol extracts from various fish samples, we then compared histamine concentration obtained by CE and fluorometric methods.  相似文献

A rapid, sensitive, and simple ultra-performance liquid chromatography-tandem mass spectrometry (UPLC/MS/MS) method for the determination of udenafil and its active metabolite, DA-8164, in human plasma and urine using sildenafil as an internal standard (IS) was developed and validated. Udenafil, DA-8164 and IS from a 100 microL aliquot of biological samples were extracted by protein precipitation using acetonitrile. Chromatographic separation was carried on an Acquity UPLC BEH C(18) column (50 x 2.1 mm, i.d., 1.7 microm) with an isocratic mobile phase consisting of acetonitrile and containing 0.1% formic acid (75:25, v/v) at flow rate of 0.4 mL/min, and total run time was within 1 min. Detection and quantification was performed by the mass spectrometer using multiple reaction-monitoring mode at m/z 517 --> 283 for udenafil, m/z 406 --> 364 for DA-8164 and m/z 475 --> 100 for IS. The assay was linear over a concentration range of 1-600 ng/mL with a lower limit of quantification of 1 ng/mL in both human plasma and urine. The coefficient of variation of this assay precision was less than 13.7%, and the accuracy exceeded 92.0%. This method was successfully applied for pharmacokinetic study after oral administration of udenafil 100 mg to healthy Korean male volunteers.  相似文献

Daptomycin, a lipopeptide antibiotic with excellent activity against Gram‐positive bacteria, is excreted primarily by the kidneys. Development of effective chromatographic methodologies for the determination of daptomycin in human specimens is necessary for clinical use. This study developed a simple and validated ultra‐high‐performance liquid chromatography method coupled to ultraviolet detection for determination of daptomycin in human plasma and urine. After the pretreatments involving protein precipitation, the supernatants were separated using a 2.3 µm particle size octadecylsilyl column, and the run time was 1 min. The calibration curves were linear over the concentration ranges of 2–200 mg/L for plasma and 25–300 mg/L for urine. Intra‐ and inter‐assay precision and accuracy values of plasma were within 13.5 and 92–100% and within 10.7 and 100–107%, respectively. Those of urine were within 5.0 and 101–104% and within 3.7 and 100–101%, respectively. The validated method was applied to the determination of plasma and urine samples in patients receiving 4–6 mg/kg of intravenous daptomycin, resulting in sufficient sensitivity for evaluating the plasma exposure and urinary excretion. In conclusion, the present method with acceptable analytical performance can be helpful for evaluating the pharmacokinetic disposition of daptomycin in clinical settings. Two different micellar electrokinetic chromatographic methods to determine dabrafenib in urine and serum, both using borate buffer (pH 9.2, 20 mM) and SDS as separation electrolyte, are developed and validated. The analyses were carried out in a fused‐silica capillary of 75 μm of internal diameter and total length of 47 and 37 cm for urine and serum determination, respectively. The detection of the target compound was performed at 227 nm in urine samples and at 251 nm in serum samples. The linearity range was from 1 to 21 mg/L of dabrafenib in urine and from 2 to 40 mg/L in serum. In all cases, inter‐ and intraday RSDs were <4%. Sample preparation of serum samples consists of an only step of 1:1 dilution with water before its injection in the electrophoretic system. These simple, sensitive, accurate, and cost‐effective methods can be used in routine clinical practice to monitor dabrafenib concentrations in urine and serum of metastatic melanoma skin cancer patients.  相似文献

It was found that native amino acids enhanced the chemiluminescence (CL) reaction between luminol and BrO(-) in an alkaline aqueous solution. This has led to the development of a facile and highly sensitive CL detection scheme for the determination of amino acids in biological samples after capillary electrophoretic (CE) separation. The CE-CL conditions were optimized. An electrophoretic buffer of 2.5 x 10(-2) M sodium borate (pH 9.4) containing 1 x 10(-4) M luminol was used. The oxidizer solution of 8 x 10(-4) M NaBrO in 0.1 M sodium carbonate buffer solution (pH 12.5) was introduced post-column. Under the optimal conditions, the detection limits were 1.0 x 10(-7) M for glutamic acid (Glu) and 1.3 x 10(-7) M (S/N = 3) for aspartic acid (Asp). The relative standard deviations (RSDs) of peak area and migration time were in the ranges of 3.8-4.3% and 1.4-1.6%, respectively. The present method was applied to the determination of excitatory amino acids (i.e., Asp and Glu) in rat brain tissue and monkey plasma. The levels of these major excitatory amino acids in monkey plasma were quantified for the first time and found to be 1.17 +/- 0.17 x 10(-5) M (mean +/- SD, n = 6) for Glu and 1.64 +/- 0.19 x 10(-6) M for Asp, which were comparable with the levels in human plasma.  相似文献

The elemental content of different rubber samples was analyzed using different analytical methods (inductively coupled atomic emission spectroscopy and non-polarized energy dispersive X-ray fluorescent spectroscopy). A new application of energy dispersive X-ray spectrometry as an analytical method for the determination of elemental content of rubber samples was investigated. Control analyses were also carried out to compare the results by ICP-AES. In one hand four samples contained same quality of elements (Mg, Si, P, S, Ca, Fe, Zn), while another four samples were without phosphorous. On the other hand there were significant differences in the ratio of C/H in the case of each sample. Based on the results, it was concluded that the X-ray spectrometry as an analytical method is well-applicable to determine the elemental composition of rubber samples, but the calibration is a key part of the analysis. A good correlation was observed between the different methods, but the correlation was the function of characteristic of matrices. Significant matrix effect from the presence of phosphor was observed in the case of some samples, while the change of C/H ratio could not result notable matrix effect.  相似文献

The CO2 reduction processes have been discussed as a way of designing an ecologically totally closed technology. An electric current and molecular hydrogen are the two related available agents that can be discussed as ecologically pure reductants. The most important products are liquid and gaseous fuels, the products of large-scale organic synthesis, monomers, and amino acids. For CO2 reduction, the necessary energy consumption and H2 costs were calculated. For complex organic molecules, amino acids for instance, the energy consumption does not make up the main portion of the costs. The biocatalytic systems of CO2 reduction based on cryoimmobilized cells are described. Conversion of CO2 into L-lysine with electrochemical decomposition of water was effected on the laboratory scale. A general unit for diverse technological processes can be a bioelectrosynthetic Index Entries: Bioelectrosynthesis; CO2 reduction; liquid fuels; amino acids; immobilized cells; economic estimates. modulus, an electrochemical hydrogen generator coupled with a biocatalytic converter of hydrogen and oxygen. The systems for bioelectrosynthesis of motor fuels and essential amino acids have been economically estimated and characterized. Abstract

A PVC membrane electrode for dysprosium(III) [Dy(III)] ions was constructed, having its basis on benzoxazoleguanidine (BG) as a suitable ionophore. The sensor presents a linear dynamic range of 1.0 × 10?6–1.0 × 10?1 M, with a Nernstian slope of 19.5 ± 0.4 mV decade?1 and a detection limit of 4.7 × 10?7 M. The response time is quick (less than 10 s). It can be used in the pH range of 3.3–8.4, and its duration is at least 2 mo without any considerable, noticeable potential divergence. The recommended sensor revealed comparatively good selectivity with respect to most alkali, alkaline earth, some transition, and heavy metal ions. It was successfully employed as an indicator electrode in the potentiometric titration of Dy(III) ions with EDTA. The membrane sensor also applied to the determination of concentration of Dy(III) ions in soil and sediment samples. Validation with certified reference materials (CRMs) was also carried out.  相似文献

This work reports a comparative study of two automated flow-through fluorosensors for the determination of digoxin in serum samples: an immunosensor with an anti-digoxin polyclonal antibody as the reactive phase permanently immobilised on controlled-pore glass and a sensor with a selective reaction system based on a methacrylic molecularly imprinted polymer (MIP) synthesised by bulk polymerisation. The variables affecting the sensitivity and dynamic range of the sensors (e.g. the carrier and elution solutions, flow rates, pH and reagent concentrations) were optimized, and the binding characteristics of their reactive phases were compared in a competitive fluorescent assay. Digoxin was reproducibly determined by both sensors at the milligram per litre level (detection limit = 1.20 × 10−3 mg L−1 and RSD = 4–7% for the immunosensor; detection limit = 1.7 × 10−5 mg L−1 and RSD = 1–2% for the MIP sensor). No cross-reactivity with digoxin-related compounds was seen for either sensor at a digoxin/interferent ratio of 1:100. The lifetime of the immunosensor was about 50 immunoassays; its shelf life, when unused, is about 3 months. The lifetime of the MIP sensor was over 18 months. Both sensors were used to determine the digoxin concentration of human serum samples with satisfactory results.  相似文献
