A new validated potentiometric method is described for batch and continuous quality control monitoring of the drug oseltamivir phosphate (Taminil) (OST). The method involves the development of a potentiometric sensor responsive to the drug based on the use of the ion‐association complex of (OST+) cation with phosphomolybdate anion (PMA?) as an electroactive material in a poly(vinyl chloride) matrix membrane plasticized with o‐nitrophenyloctyl ether (o‐NPOE). Optimization of the performance characteristics of the sensor is described. A membrane incorporating the OST‐PMA‐NPOE complex in a tubular flow through detector is used in a two channel flow injection set up for continuous monitoring of the drug at a frequency of ~30 samples h?1. The sensor shows fast near‐Nernstian response for OST over the concentration range 5.2×10?5–0.8×10?2 M (21.34 µg mL?1–3.23 mg mL?1) with a detection limit of 9.1×10?6 M (3.73 µg mL?1) over the pH range 4.6–6.1. The sensor displays good selectivity for OST drug over some basic drugs, inorganic cations, excipients and diluents commonly used in the drug formulations. Validation of the assay method is tested by measuring the lower detection limit, range, linearity, bias, trueness, accuracy, precision, and between‐day‐variability, within day reproducibility, selectivity and ruggedness (robustness). The results reveal good potentiometric performance of the proposed sensor for determination of OST in pharmaceutical capsules and in biological fluid matrices as well as for testing the dissolution profile of the drug and drug homogeneity.  相似文献

A multi‐electrochemical competitive immunosensor for the rapid determination of unmetabolized cocaine (COC) in urine, saliva and human serum matrices is reported. Anti‐cocaine polyclonal antibodies were immobilized in an oriented way onto protein‐G functionalized magnetic beads. The immunosensor is based on an array of eight carbon‐based screen‐printed electrodes for simultaneous electrochemical determinations. The treatments of the biological samples were simplified and optimized for avoiding matrix interferences. The immunosensor was sensitive (EC50≈2.92–3.88 ng mL?1 COC), required a very small volume of sample (200 µL), was reproducible (%RSD was lesser than about 18 %), and accurate (recovery percentages ranged 88–117 %).  相似文献

A glassy carbon electrode was modified with carbon nanotubes and the ionic liquid N‐butyl pyridinium trifluoromethyl methanesulfonate for the determination of methyldopa in urine samples. Methyldopa exhibited a well‐defined anodic signal over a broad pH range of 2–10 and the peak current increased approximately 100 fold over that of the unmodified electrode. Accordingly, a novel method for the determination of methyldopa was proposed using differential pulse voltammetry. The peak current was linear over a methyldopa concentration range from 21 to 2111 ng mL?1 with a LOD of 6.9 ng mL?1 and a LOQ of 7.4 ng mL?1. The method was applied to determine the excretion profile of methyldopa in urine without sample pretreatment.  相似文献

A highly sensitive automated sequential‐injection chemiluminescence (SIA‐CL) method for determination of glucosamine sulphate (GLS) was developed. The goal of the present work is the evaluation of the enhancement effect of the investigated drug glucosamine sulphate on the chemiluminescence reaction between luminol and H2O2 in alkaline medium of 1.0 × 10?2 mol L?1 sodium hydroxide at pH 11. The experimental conditions affecting the CL reaction such as the sequence of the reagents, concentrations, flow rate and aspirated volumes of reactants were systematically investigated and optimized. Under optimum conditions 50 μL of 1.0 × 10?3 mol L?1 luminol, 30 μL of a GLS test solution and 50 μL of 1.0 × 10?2 mol L?1 H2O2 were used and the luminescing zone was pushed into the detector at a flow rate 100 μL s?1. The proposed method recorded high sensitivity, accuracy and simplicity that could be clarified as linear concentration range 1.0‐2000 ng mL?1 with rectilinear part (r = 0.9992, n = 9) and limit of detection 0.3 ng mL?1, along with relative standard deviation 1.3%. It was found that the developed method can be used directly to determine the investigated drug GLS in its pharmaceutical dosage forms and in spiked serum and urine by diluting the samples for a 1000 fold. The obtained results were statistically analyzed and compared with those obtained by the reported method.  相似文献

Abstract

A sensitive and fast flow‐injection (FI) spectrophotometric method for the determination of levofloxacin based on the formation of a colored product upon oxidation with N‐bromosuccinimide (NBS) in acidic medium is proposed. Optimization of chemical and FI variables has been made. Under the optimized conditions, the sampling rate was over 90 h?1, the calibration curve obtained was linear over the range 10–300 µg·mL?1, and the detection limit was 3 µg·mL?1. The proposed method was successfully applied to the determination of levofloxacin in pharmaceuticals and human urine samples. It was also found that the excipients in the commercial tablet preparation did not interfere with the assay. Results are precise (RSD<2.7%; n =10) and in agreement with those found by the reference high pressure liquid chromatography (HPLC) procedure.  相似文献

Acepromazine maleate (Sedalin?) was administered orally to six thoroughbred horses at a dose of 0.15?mg?kg?1. Urine and blood samples were collected up to 412?h post-administration. Plasma and urine were hydrolysed; plasma samples were then processed using liquid–liquid extraction and urine samples using solid-phase extraction. A sensitive tandem mass spectrometric method was developed in this study, achieving a lower limit of quantification for acepromazine of 10?pg?mL?1 in plasma and 100?pg?mL?1 in urine. Acepromazine, hydroxyethylpromazine, hydroxyacepromazine, hydroxyethylpromazine sulphoxide, hydroxyethylhydroxypromazine, dihydroxyacepromazine and dihydroxyhydroxyethylpromazine were detected in the post-administration samples. The parent drug and its metabolites were identified using a combination of UPLC–MS/MS and accurate mass measurement. Separation of the structural isomers hydroxyethylpromazine sulphoxide and hydroxyethylhydroxypromazine was another significant outcome of this work and demonstrated the advantages to be gained from investing in chromatographic method development.  相似文献

A simple, rapid, and sensitive method based on dispersive liquid–liquid microextraction combined with HPLC‐UV detection applied for the quantification of chlordiazepoxide in some real samples. The effect of different extraction conditions on the extraction efficiency of the chlordiazepoxide drug was investigated and optimized using central composite design as a conventional efficient tool. Optimum extraction condition values of variables were set as 210 μL chloroform, 1.8 mL methanol, 1.0 min extraction time, 5.0 min centrifugation at 5000 rpm min?1, neutral pH, 7.0% w/v NaCl. The separation was reached in less than 8.0 min using a C18 column using isocratic binary mobile phase (acetonitrile/water (60:40, v/v)) with flow rate of 1.0 mL min?1. The linear response (r2 > 0.998) was achieved in the range of 0.005–10 μg mL?1 with detection limit 0.0005 μg mL?1. The applicability of this method for simultaneous extraction and determination of chlordiazepoxide in four different matrices (water, urine, plasma, and chlordiazepoxide tablet) were investigated using standard addition method. Average recoveries at two spiking levels were over the range of 91.3–102.5% with RSD < 5.0% (n = 3). The obtained results show that dispersive liquid–liquid microextraction combined with HPLC‐UV is a fast and simple method for the determination of chlordiazepoxide in real samples.  相似文献

In this work, a fast, easy, and efficient dispersive liquid–liquid microextraction method based on solidification of floating organic drop followed by high‐performance liquid chromatography with UV detection was developed for the separation/preconcentration and determination of the drug valsartan. Experimental design was applied for the optimization of the effective variables (such as volume of extracting and dispersing solvents, ionic strength, and pH) on the extraction efficiency of valsartan from urine samples. The optimized values were 250.0 μL ethanol, 65.0 μL 1‐dodecanol, 4.0% w/v NaCl, pH 3.8, 1.0 min extraction time, and 4.0 min centrifugation at 4000 rpm min?1. The linear response (r2 = 0.997) was obtained in the range of 0.013–10.0 μg mL?1 with a limit of detection of 4.0 ng mL?1 and relative standard deviations of less than 5.0 % (n = 6).  相似文献

In present study, a simultaneous derivatization and air‐assisted liquid–liquid microextraction method combined with gas chromatography–nitrogen phosphorous detection has been developed for the determination of some phenolic compounds in biological samples. The analytes are derivatized and extracted simultaneously by a fast reaction with 1‐flouro‐2,4‐dinitrobenzene under mild conditions. Under optimal conditions low limits of detection in the range of 0.05–0.34 ng mL?1 are achievable. The obtained extraction recoveries are between 84 and 97% and the relative standard deviations are less than 7.2% for intraday (n = 6) and interday (n = 4) precisions. Abstract

The potential of high-performance liquid chromatography (HPLC) for the determination of U(VI) in ground waters and urine has been examined under a variety of HPLC experimental conditions. Conventional cross-linked and bonded-phase ion exchangers, both cation and anion, were studied with aqueous mobile phases containing tartrate, citrate, or α-hydroxyisobutyrate. The best chromatography was obtained on bonded-phase cation exchangers with an α-hydroxyisobutyrate eluent. The metal ions were detected either by visible spectrophotometry after a post-column reaction with a complexing reagent, or with a polarographic detector. Dectection after post-column reaction gave the best sensitivity; the detection limit (2 × baseline noise was 6 ng or 60 ng.ml?1 for 100 μl samples. In-line trace enrichment was used to decrease detection limits and linear calibration curves were observed in the ranges studied; 0.5 to 50 ng.mL?1 for ground waters and 25 to 400 ng.mL?1 for artificial urine.  相似文献

A novel generic reverse phase high performance liquid chromatography (RP‐HPLC) method is developed and validated for simultaneous determination of seven pharmaceutically active ingredients, namely, acetaminophen, dextromethorphan, doxylamine, phenylephrine, guaifenesin, caffeine and aspirin. All seven ingredients were quantified in soft gel, syrup and tablet formulations of the over‐the‐counter US‐marketed products, as per the guidelines of the International Conference on Harmonization. The separation was achieved in a 16 min run time on an Agilent Zorbax Phenyl column using a gradient method with two mobile phases. Mobile phase A was 0.15% trifluoro acetic acid in purified water and while mobile phase B was a mixture of acetonitrile and methanol (750:250 v/v) with 0.02% trifluoro acetic acid. The flow rate was 1.0 mL min?1 and injection volume was 10 μL. Detection was performed at 280 nm using a photodiode array detector. As part of the method validation, specificity, linearity, precision and recovery parameters were verified. The concentration and area relationships were linear (R2 > 0.999), over the concentration ranges 20–120 μg mL?1 for acetaminophen, 75–450 μg mL?1 for dextromethorphan, 31.25–187.5 μg mL?1 for doxylamine, 25–150 μg mL?1 for phenylephrine, 25–150 μg mL?1 for aspirin, 6.5–39 μg mL?1 for caffeine and 12–72 μg mL?1 for guaifenesin. The relative standard deviations for precision and intermediate precision were <1.5%. The proposed RP‐HPLC generic method is applicable for routine analysis of cold and cough over‐the‐counter products.  相似文献

Levamisole is the most common adulterant found in cocaine samples and its electrochemical determination in cocaine seized samples is a challenge due to peak overlapping with cocaine. Herein, we propose a deconvolution procedure for levamisole determination in seized cocaine samples using screen‐printed carbon electrodes (SPE). Square‐wave voltammetry in 0.04 mol L?1 Britton Robinson buffered solution (pH 8.0) was selected in combination with optimized SWV parameters (f=8 s?1, a=10 mV and ΔEs=1 mV) to result in the best peak resolution to apply the deconvolution procedure. Deconvoluted responses of levamisole in the presence of cocaine were similar to untreated signals of standard levamisole solutions in absence of cocaine. A linear response was obtained in the range of 20–100 μmol L?1 (r=0.995). The results obtained for the analysis of a seized cocaine sample was statistically similar to that obtained by gas chromatography. Other adulterants found in cocaine street samples (paracetamol, glucose, phenacetin, caffeine, boric acid and lidocaine) did not affect the treated of voltammetric responses of levamisole. A pharmaceutical sample containing levamisole was also analyzed on SPEs and a recovery of 93±2 % was obtained (no deconvolution required for this sample), showing great applicability of SPEs for forensic and pharmaceutical analyses.  相似文献

We examined the feasibility of capillary liquid chromatography/microchip atmospheric pressure photoionization tandem mass spectrometry (capLC/µAPPI‐MS/MS) for the analysis of anabolic steroids in human urine. The urine samples were pretreated by enzymatic hydrolysis (with β‐glucuronidase from Helix pomatia), and the compounds were liquid‐liquid extracted with diethyl ether. After separation the compounds were vaporized by microchip APPI, photoionized by a 10 eV krypton discharge lamp, and detected by selected reaction monitoring. The capLC/µAPPI‐MS/MS method showed good sensitivity with detection limits at the level of 1.0 ng mL?1, good linearity with correlation coefficients between 0.9954 and 0.9990, and good repeatability with relative standard deviations below 10%. These results demonstrate that microchip APPI combined with capLC/MS/MS provides a new potential method for analyzing non‐polar and neutral compounds in biological samples. An analytical procedure for the simultaneous determination in human urine of several thioamphetamine designer drugs (2C‐T and ALEPH series) is reported. The quantitative analysis was performed by liquid chromatography/tandem mass spectrometry and has been fully validated. The mass spectrometer was operated in positive‐ion, selected reaction monitoring (SRM) mode. In order to minimize interferences with matrix components and to preconcentrate target analytes, solid‐phase extraction was introduced in the method as a clean‐up step. The entire method was validated for selectivity, linearity, precision and accuracy. The method turned out to be specific, sensitive, and reliable for the analysis of amphetamine derivatives in urine samples. The calibration curves were linear over the concentration range of 1 to 100 ng mL?1 for all drugs with correlation coefficients that exceeded 0.996. The lower limits of detection (LODs) and quantification (LOQs) ranged from 1.2 to 4.9 ng mL?1 and from 3.2 to 9.6 ng mL?1, respectively. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

In the present studies, a simple rapid ultra performance liquid chromatography (UPLC) method with UV detection for the simultaneous determination of cortisol, cortisone and 6β-hydroxycortisol in human urine was developed. The three analytes and the internal standard dexamethasone were separated on a Waters Acquity UPLC-Tunable UV system with an Acquity UPLC BEH C18 column (50 × 2.1 mm ID, 1.7 μm) using a gradient elution of methanol and water (containing 0.01% formic acid) with a run time of 7 min. The method was accurate and precise, over the ranges of 5–200 ng mL−1 for cortisol, and 10–1,000 ng mL−1 for both cortisone and 6β-hydroxycortisol, and showed good linearity (r 2 > 0.999). This method was applied for the measurement of cortisol, cortisone and 6β-hydroxycortisol in samples collected over different periods as a tool to assess the activity of CYP3A and 11β-hydroxysteroid dehydrogenase type 2 enzymes.

The determination of target molecules in complicated matrices such as biological samples is largely dependent on sample pretreatment. Molecularly imprinted solid-phase extraction (SPE), using molecularly imprinted polymers as the adsorbent, has been demonstrated to be effective for the selective enrichment of target molecules in biological samples. In this study, molecularly imprinted polymeric microspheres were fabricated by two-step swelling polymerization using polystyrene particles as seeds, nicotinamide as the template, methacrylic acid as a functional monomer, and ethylene glycol dimethacrylate as a cross-linker. The molecularly imprinted polymeric microspheres were packed into empty SPE cartridges, and the spiked urine and serum samples were loaded separately. After an initial washing and elution step, the effluents were analyzed by high-performance liquid chromatography (HPLC) using 1:9 methanol/0.05% phosphoric acid. The obtained molecularly imprinted polymeric microspheres were uniform, and the spherical particles were well distributed. The established method was validated, and the results showed that the method was linear from 0.499 to 19.96?µg?mL?1. The limits of detection and quantification for nicotinamide were 0.3 and 0.9?µg?mL?1, respectively. The relative standard deviations were 1.55 and 2.86% in urine and serum, respectively. The spiked recoveries of nicotinamide were 86.0–98.8% and 87.0–96.8% in urine and serum, respectively. The molecularly imprinted SPE and HPLC methods in this study are useful for the pretreatment and determination of the target compounds in these matrices.  相似文献

A sequential online extraction, clean‐up and separation system for the determination of betaine, l ‐carnitine and choline in human urine using column‐switching ion chromatography with nonsuppressed conductivity detection was developed in this work. A self‐packed pretreatment column (50 × 4.6 mm, i.d.) was used for the extraction and clean‐up of betaine, l ‐carnitine and choline. The separation was achieved using self‐packed cationic exchange column (150 × 4.6 mm, i.d.), followed by nonsuppressed conductivity detection. Under optimized experimental conditions, the developed method presented good analytical performance, with excellent linearity in the range of 0.60–100 μg mL−1 for betaine, 0.75–100 μg mL−1 for l ‐carnitine and 0.50–100 μg mL−1 for choline, with all correlation coefficients (R2) >0.99 in urine. The limits of detection were 0.15 μg mL−1 for betaine, 0.20 μg mL−1 for l ‐carnitine and 0.09 μg mL−1 for choline. The intra‐ and inter‐day accuracy and precision for all quality controls were within ±10.32 and ±9.05%, respectively. Satisfactory recovery was observed between 92.8 and 102.0%. The validated method was successfully applied to the detection of urinary samples from 10 healthy people. The values detected in human urine using the proposed method showed good agreement with the measurement reported previously.  相似文献

This paper presents a fast and simple method for the extraction, preconcentration and determination of fluvoxamine, nortriptyline and maprotiline in urine using simultaneous derivatization and temperature‐assisted dispersive liquid–liquid microextraction (TA‐DLLME) followed by gas chromatography–flame ionization detection (GC‐FID). An appropriate mixture of dimethylformamide (disperser solvent), 1,1,2,2‐tetrachloroethane (extraction solvent) and acetic anhydride (derivatization agent) was rapidly injected into the heated sample. Then the solution was cooled to room temperature and cloudy solution formed was centrifuged. Finally a portion of the sedimented phase was injected into the GC‐FID. The effect of several factors affecting the performance of the method, including the selection of suitable extraction and disperser solvents and their volumes, volume of derivatization agent, temperature, salt addition, pH and centrifugation time and speed were investigated and optimized. Figures of merit of the proposed method, such as linearity (r2 > 0.993), enrichment factors (820–1070), limits of detection (2–4 ng mL?1) and quantification (8–12 ng mL?1), and relative standard deviations (3–6%) for both intraday and interday precisions (concentration = 50 ng mL?1) were satisfactory for determination of the selected antidepressants. For the first time, electrospun composite nanofibers comprising polymeric crown ether with polystyrene (PCE‐PS) have been used for the selective extraction of catecholamines – dopamine (DA), norepinephrine (NE) and epinephrine (E) – prior to their analysis by high‐performance liquid chromatography–electrochemical detection. Using a minicartridge packed with PCE‐PS composite nanofibers, the target compounds were extracted effectively from urine samples to which diphenylborinic acid 2‐aminoethyl ester was added as a complexing reagent. The extracted catecholamines could be liberated from the fiber by the addition of acetic acid. A good linearity was observed for catecholamines in the range of 2.0–200 ng mL?1 (NE, E and DA). The detection limits of catecholamines (signal‐to‐noise ratio = 3) were 0.5 ng mL?1 (NE), 0.2 ng mL?1 (E) and 0.2 ng mL?1 (DA), respectively. Under the optimized conditions, the absolute recoveries of the above three catecholamines were 90.6% (NE), 88.5% (E) and 94.5% (DA). The repeatability of extraction performance was from 5.4 to 9.2% (expressed as relative standard deviation). We present a comparison of two sensitive methods, HPLC with fluorescence detector (HPLC/FLD) and UPLC with electrospray tandem mass spectrometry (UPLC/MS/MS), for the determination of indoleamine neurotransmitters (NTs) and their metabolites in sea lamprey plasma samples. Liquid–liquid extraction (LLE) and solid-phase extraction (SPE) were also tested for recovery and matrix effect. The recoveries of SPE determined by HPLC/FLD and UPLC/MS/MS ranged from 75 to 123% and 78 to 105%, respectively, while the recoveries of LLE ranged from 45 to 73% and 48 to 75%, respectively. SPE combined with HPLC/FLD and UPLC/MS/MS to determine the target analytes in plasma samples were validated of the sensitivity, reproducibility, accuracy and precision. Both methods exhibited excellent linearity in the range of 0.2–50 ng mL−1 for all analytes. The limits of detection (LOD) varied from 0.04 ng mL−1 to 0.13 ng mL−1 for HPLC/FLD method and 0.003 ng mL−1 to 0.02 ng mL−1 for UPLC/MS/MS method. The inter-day accuracy ranged from 82.5 to 127.0% for HPLC/FLD and 93.0 to 113.0% for UPLC/MS/MS. The inter-day precision ranged from 9.9 to 32.3% for HPLC/FLD and 5.4 to 13.2% for UPLC/MS/MS. These results demonstrated that the values obtained by both methods were within the satisfactory range and the UPLC/MS/MS method provided more accurate and precise measurements than HPLC/FLD method. The comparison is of great importance to determine the available detectors, considering the complexity and expensiveness versus quality parameters. These two methods were applied to the analysis of four important indoleamine neurotransmitter analytes (5-hydroxytryptamine, 5-hydroxyindole-3-acetic acid, tryptamine and melatonin) in sea lamprey plasma samples.  相似文献
