The cleavage of the pyridazine ring of the acyltetrahydro [1] benzofuro [2,3-d] pyridazones was carried out by hydrolysis, alcoholysis or aminolysis reactions and they affect the lactam 3,4-bond. They lead chiefly to benzofuran derivatives with acid, ester, amide or hydrazide groups in the 2-position and eventually an acylated methylhydrazine group in the 3-position. The cyclization reactions of 3-hydrazinomethylbenzofuran-2-carboxylic acid and its derivatives or ethyl 3-bromome thylbenzofuran-2-carboxylate affords tetrahydrobenzofuropyridazones. The nmr spectra were studied.  相似文献

The 1- and 4-hydrazino[1]benzothiéno[2,3-d]pyridazines have been cyclised to form triazolobenzothienopyridazines, tetrazolobenzothienopyridazines and benzothienopyridazinotriazines. NMR study shows a tautomeric equilibrium in solution between tetrazolo [1,5-b][1]benzothiéno[3,2-d] pyridazine and 1- azido [1]benzothieno[2,3-d]pyridazine. Reactions of the benzothienopyridazinotriazones with hydrazine were studied.  相似文献

The new benzofuro[2,3-b]naphthyridine ring system was prepared. 2,4-Dichloro-3-(o-methoxyphenyl)naphthyridines 3a and 3b were obtained by chlorination of hydroxynaphthyridinones 2a and 2b . Demethylation followed by cyclization of 3a and 3b afforded 11-chlorobenzofuro[2,3-b]naphthyridines 4a and 4b . Hydrogenolysis of these 11-chlorobenzofuronaphthyridines gave the parent benzofuro[2,3-b]naphthyridines 5a and 5b . The analog 4b was also converted to 11-methoxybenzofuro[2,3-b][1,8]naphthyridine ( 6 ).  相似文献

Polymethine dyes — benzofuro[2,3-b]pyridine and selenonaphtheno[2,3-b]pyridine derivatives — were synthesized, and their spectral properties are discussed. Replacement of a vinylene group by an oxygen or selenium atom in one benzene ring of the 2-benzo[h]quinoline residue in the cyanines causes a large bathochromic shift of the absorption maxima of the dyes as compared with replacement by a sulfur atom. The dyes derived from benzofuro[2,3-b]pyridine derivatives are more deeply colored than the derivatives of both thionaphtheno[2,3-b]-pyridine and selenonaphtheno[2,3-b]pyridine.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 12, pp.1611–1614, December, 1972.  相似文献

New pyrrolo[3,4-c]isoxazole derivatives were synthesized from the key intermediates 4-cyanopyrrolidin-3-ones in two steps. Pyrrolo[2,3-d][1,2,3]triazoles and triazolo[4,5-c]pyridazine were obtained from 2-arylhydrazono-4-cyano-1-(4′-methoxyphenyl)-3-oxopyrrolidines by refluxing with phenylhydrazine in either ethanol or glacial acetic acid. [1]Benzofuro[2,3‐d]pyridazines fused with 1,2,4‐triazole ( 6 and 7 ), 1,2,4‐triazine ( 8–10 ) and 1,2,4‐tri‐azepine (12) were prepared by the ring closure of 4‐hydrazino‐[1]benzofuro[2,3‐d]pyridazine ( 5 ), derived from naturally occurring rotenone. Compounds ( la and lb ) exhibited significant inhibitory activity against 12‐O‐tetradecanoylphorbol 13‐acetate (TPA)‐induced Epstein‐Barr virus early antigen (EBA‐EA) activation in Raji cells. In contrast, the fused [1]benzofuro[2,3‐d]pyridazines except 6c and 8 were quite inactive.  相似文献

Polymethine dyes of various types were obtained from quaternary salts of 4-methylbenzofuro- and 4-methylbenzoselenopheno[2,3-b]pyridines. It is shown that replacement of the vinylene group in the benzene ring of the benzo[h]-4-quinoline residue by an oxygen or selenium atom gives rise to a larger hypsochromic shift of the absorption maxima of the corresponding cyanine dyes than replacement by a sulfur atom. The alkylation of o-cyanophenol by 4-chloromethylcoumarins and subsequent intramolecular condensation by the cyano and methylene groups gives substituted 4-(3-amino-2-benzo-furanyl)coumarins. We studied the reactions of these compounds with acylating agents as well as with aldehydes, which lead to the 6H-[1]benzofuro[3,2-b]chromeno[4,3-d]pyridin-6-one system as the result of consecutive transformations.  相似文献

The synthesis of 1,2-dihydro[1]benzofuro[2,3-d]pyridazin-1-one and 3,4-dihydro [1]benzofuro[2,3-d]pyridazin-4-one was accomplished by the eyclization of appropriately carbonyl-substituted benzofuran derivatives. Another successful synthetic route was provided using 1,2,3,4-tetrahydro[1]benzofuro[2,3-d]pyridazine-1,4 dione and 1,2,3,4-tetrahydro[1]benzofuro[2,3-d]pyridazin-4-one. The structure of a nitrobenzofuropyridazin-4-one was established using nmr and the nuclear Overhauser effect.  相似文献

A method is proposed for the synthesis of 5-acylamino-2,3-dimethylthiophene-4-carbaldehydes and 5-acylamino-2,3-tetramethylenethiophene-4-carbaldehydes by the formylation of the corresponding thiophenes using the Vilsmeier reaction. From the formyl derivatives obtained, previously unaccessible derivatives of thieno[2,3-b]pyridine, thieno[2,3-d]pyrimidine, and thieno[3′,2′∶5,6]pyrido[2,3-d]-pyrimidine have been synthesized.  相似文献

4-Cyano-5,6-diphenyl-2,3-dihydropyridazine-3-onc 1 reacts with phosphorous oxychloride to give 70% of the corresponding 3-chloro derivative 2. Treating 2 with anthranilic acid in butanol, 4-cyano-2,3-diphenyl-10H-pyridazino[6,1-b]quinoxaline-10-one, 3 was obtained. Compound 1 reacts with phosphorous pentasulphide to give 3-mercapto derivative 4, which was converted by acrylonitrile to S-(2-cyanoethyl)pyridazine derivative 5. Compound 4 reacts with ethyl bromoacetate and with phenacyl bromide gave the corresponding thieno[2,3-c] pyridazine derivatives 8, 9, Alkylation of 1 with ethyl chloroacetate afforded 3-0-carbethoxymethyl derivative 10. Compound 10 reacts with amines (aniline, hydrazine) to give the corresponding amide and acid hydrazide 13, 12 respectively. Hydrolysis of 10 with sodium hydroxide gave the corresponding acid derivative 11. Treating 1 with methyl iodide, 3-0-methyl derivative 14 was obtained, which was converted by ammonium acetate/acetic acid to 3-amino-4-cyano-5,6-diphenyl pyridazine 15. Compound 1 reacts with methyl magnesium iodide gave 4-acetyl derivative 16, which was reacted with hydrazine, phenyl hydrazine and with hydroxylamine to give the substituted I H pyrazolo [3,4-c] pyridazine 17 a,b and isoxazolo [5,4-c] pyridazine 18 derivatives respectively.  相似文献

Summary A synthetic approach to new 1-benzyl-7-alkyl-2,3-dimethyl-4-oxopyrrolo[2,3-b]pyridine-5-carboxylic acids using 5,6-dimethyl-2,4-dioxopyrrolo[2,3-d][1,3]oxazine as the starting material is reported. The antimicrobial activity of these compound is reported.

Abstract

The synthesis of acetic 2-{[1-methyl-2-(4-oxo-5,6,7,8-tetrahydro-4H-benzo [4,5]-thieno [2,3-d] [1,3-oxazin-2-yl)ethylidene]amino}-4,5,6,7-tetrahydrohenzo[b]thiophene ?3-carboxylic acid anhydride 5 and 2-(oxopropyl)-5,6,7,8-tetrahydro-4H-benzo-[4,5] thieno[2,3-d][1,3]oxazin-4-one 7, has been achieved in three steps from ethyl 2-amino-4,5,6,7-tetrahydrobenzo(b]thiophene-3-carboxlate 1 via the reaction with ethyl acetoacetate followed by hydrolysis and acetic anhydride-induced cyclization. The 2-substituent in compound 5 has two functional groups i.e. active methylene and acid anhydride which are suitably located for intramolecular transformation. Thermal and/or base catalyzed intramolecular cyclization of 5 afforded 2-(4-acetoxy-(hydroxyl)-2-methyl-5,6,7,8-tetrahydrobenzo[4,5] thieno[2,3-b]pyridin-3-yl)-5,6,7,8- tetrahydro-4H-benzo[4,5]thieno[2,3-d] [1,3]oxazin-4-one 10 and 9 respectively. Treatment of 5 with hydrazine hydrate, aromatic and/or heterocyclic amines induced the same intramolecular cyclization with a concomitant oxazine-pyrimidine interconversion to give 3-amino(aryl or heteryl)-2-(4-hydroxy-5,6,7,8-tetrdhydrobenzo-[4,5]thieno[2,3-b]pyridin-3-yl)-3,4,5,6,7,8-hexahydrobenzo[4,5] thieno[2,3-d] pyrimid in-4-one 11–14 respectively.  相似文献
