A convenient method for double Michael additions to quinone systems catalyzed by Al2O3, is reported. The advantages of this method include the use of a cheap and environment-friendly catalyst, a straightforward isolation of the pure product by filtration, high yields, and excellent selectivity, thus providing rapid access to useful building blocks for the preparation of biologically active quinones.  相似文献

An efficient synthesis of unsymmetrically substituted 1,3-pyrazole derivatives has been developed via three-component coupling reaction involving 3-(dimethylamino)-1-phenylprop-2-en-1-one, hydrazine, and aryl halides in one pot process exhibiting high regioselectivity. The pyrazole synthesis proceeds via a sequential series of reactions such as Michael addition, heterocyclization, dehydration, and Ullmann cross-coupling.  相似文献

Solid state Michael addition reaction of indole with α, β-unsaturated carbonyl compounds was carried out, by which a series of compounds containing three different heterocyclic groups binding to one carbon atom were ob-tained. In the presence of Lewis acid, indole could undergo the solid state condensation reaction with aromatic ketones and aldehydes or quinones. The solid state reaction showed higher selectivity and yield than solution reaction. The structures of products were identified by IR,1H NMR, MS. elemental analysis and X-ray crystal analysis. A stereocontrolled total synthesis of methyl (±)-O-methyl podocarpate (4) has been successfully accomplished using the trans-fused diester 21 as a key intermediate. Intramolecular Michael reaction of the enone-diester 18 afforded the cis-fused keto-diester 19 in high yield which was stereoselectively converted into 21 via the enone 20.  相似文献

Levoglucosenone, the ,ß-unsaturated ketone obtained by the pyrolysis of cellulose, reacts with NH-azoles in the presence of bases. In this process not only normal Michael adducts are formed, but in some cases their hydrates (gem-diols) are also formed. In all cases the addition proceeds stereospecifically and with good yields. 3-Nitro-s-triazole, 3-nitro- and 4-nitropyrazoles, 3-methyl-4-nitropyrazole, 5-nitrotetrazole, 4,5-dicarbomethoxy-v-triazole, 3,4-dinitropyrazole, 3(5)-methylpyrazole, pyrazole, and imidazole have been studied in this reaction. An efficient, versatile, enantioselective synthesis of 1,3-disubstituted and 1,3,6-trisubstituted 5-methylidenedihydrouracils applying Horner-Wadsworth Emmons methodology was developed. Starting 1,3-disubstituted 5-diethoxyphosphoryluracils were subjected to reduction of the double bond or addition of various Grignard reagents and obtained Horner-Wadsworth Emmons reagents were used for the olefination of formaldehyde. Enantioselective synthesis of 1,3,6-trisubstituted 5-methylidenedihydrouracils was accomplished by introducing (R,R)- or (S,S)-di(1-phenylethylamino)phosphoryl groups as chiral auxiliary. Additions of Grignard reagents in the presence of these groups were highly and complimentary diastereoselective (de?~?80%). Further separation of the diastereomeric mixtures by column chromatography enabled synthesis of (R)- and (S)-1,3,6-trisubstituted-5-methylidenedihydrouracils with ee?≥?98%. Furthermore, absolute configuration of the adducts and final products was established using single crystal X-ray analysis. Stereochemical course of the addition reactions is also discussed.  相似文献

A simple, practical, and regioselective synthetic protocol for the formation of pyrazoles was developed. Unlike all other previously reported reactions of nitroallylic acetates, this process was initiated by a SN2 reaction at the electrophilic γ site. The preparation of two pyrimido[4,5-c]isoquinoline-7,10-quinones from acylhydroquinones and 1,3-dimethyl-5-aminouracil and their cycloadditions with 1-trimethylsilyloxybutadiene and 1-dimethylamino-3-methyl-1-azabutadiene is described. The remarkable regiocontrol of these cycloadditions that yield stable 1:1 cycloadducts is discussed on the basis of steric interactions into the pyrimido[4,5-c]isoquinoline-7,10-quinones. The access to angucyclinone AB-pyridopyrimidine analogues from Diels-Alder adducts and preliminar evidences on their antitumour activities are also reported.  相似文献

A convenient stereoselective synthesis of β-lactams from thio-Michael/aldol tandem adduct is described. syn-Selective tandem reaction followed by amidation and intramolecular SN2 reaction provided β-lactams in diastereomerically pure form. The tandem reaction with aliphatic aldehydes, on the other hand, afforded a mixture of diastereomers of corresponding tandem adducts in about 3:1 ratio so that the conversion to β-lactams afforded a diastereomeric mixture. As an alternative approach to prepare the tandem adducts, the stereoselective Michael addition of aliphatic thiols to Baylis-Hillman adduct was developed. The stereoselectivity was sensitive to the protective group at the hydroxyl group and TBS protection brought the most successful syn-selective formation of the tandem adducts. The procedure could be applied to the ketone derivatives of the Baylis-Hillman adduct but no selectivity was observed for the nitrile-Baylis-Hillman adducts. A similar conversion of the adduct provided desired β-lactams stereoselectively.  相似文献

Optically active 5-substituted pyrrolidin-2-ones underwent conjugate addition to nitroalkenes to give the corresponding adducts in a diastereoselective manner. The presence of 18-crown-6 was crucial to achieve good stereoselective addition. Addition of 6-substituted piperidin-2-ones also gave the corresponding adduct in a stereoselective manner. The adduct was readily converted into a bicyclic lactam through intramolecular nitroaldol reaction, and the formal synthesis of indolizidine 167B was achieved.  相似文献

A two step efficient and practical synthesis of a variety of 4-chromanones is described. Phenols undergo a Michael addition to acrylonitriles in the presence of catalytic amounts of potassium carbonate and tert-butanol to generate the corresponding 3-aryloxypropanenitriles in 50-93% yields. Treatment of the resulting aryloxypropionitriles with 1.5 equiv of TfOH and 5 equiv of TFA, followed by an aqueous work up afforded 4-chromanones in moderate to excellent yields.  相似文献
