β-Aminopeptidases exhibit both hydrolytic and aminolytic (peptide bond formation) activities and have only been reported in bacteria. We identified a gene encoding the β-aminopeptidase homolog from a genome database of the filamentous fungus Aspergillus oryzae. The gene was overexpressed in A. oryzae, and the resulting recombinant enzyme was purified. Apart from bacterial homologs [β-Ala-para-nitroanilide (pNA)], the enzyme preferred d-Leu-pNA and d-Phe-pNA as substrates. Therefore, we designated this gene as d-stereoselective aminopeptidase A (damA). The purified recombinant DamA was estimated to be a hexamer and was composed of two subunits with molecular masses of 29.5 and 11.5 kDa, respectively. Optimal hydrolytic activity of DamA toward d-Leu-pNA was observed at 50 °C and pH 8.0. The enzyme was stable up to 60 °C and from pH 4.0–11.0. DamA also exhibited aminolytic activity, producing d-Leu-d-Leu-NH2 from d-Leu-NH2 as a substrate. In the presence of 3.0 M NaCl, the amount of pNA liberated from d-Leu-pNA by DamA was 3.1-fold higher than that in the absence of NaCl. Thus, DamA is a halophilic enzyme. The enzyme was utilized to synthesize several hetero-dipeptides containing a d-amino acid at the N-terminus as well as physiologically active peptides. 相似文献

Asporyergosterol (1), a new steroid with an E double bond between C-17 and C-20, was identified from the culture extracts of Aspergillus oryzae, an endophytic fungus isolated from the marine red alga Heterosiphonia japonica. Moreover, four known steroids including (22E,24R)-ergosta-4,6,8(14),22-tetraen-3-one (2), (22E,24R)-3beta-hydroxyergosta-5,8,22-trien-7-one (3), (22E,24R)-ergosta-7,22-dien-3beta,5alpha,6beta-triol (4), and (22E,24R)-5alpha,8alpha-epidioxyergosta-6,22-dien-3beta-ol (5) were isolated. Structures of these compounds were unambiguously established by spectroscopic techniques and by comparison with literature values. All the isolates exhibited low activity to modulate acetylcholinesterase (AChE). 相似文献

The structure of the ligand was characterized by NMR spectra and X-ray diffraction analysis. A novel lipase gene from Aspergillus fumigatus, afl1-1, was cloned and expressed with a molecular mass of 38 kDa in Escherichia coli for the first time. The recombinant lipase had a preference for short carbon chain p-nitrophenyl esters, especially toward C2 p-nitrophenyl ester and exhibited potent hydrolysis activity that had not been observed. The optimum pH and temperature of this new enzyme were 8.5 and 65 °C, respectively. The recombinant lipase (AFL1-1) is an alkaline enzyme which was stable in the pH range 6.0~8.5 for 16 h (at 4 °C) and at 30~50 °C for 1 h. It is an intracellular enzyme which was purified approximately 8.47-fold with an overall yield of 86.1% by single-step Ni-NTA affinity purification, with a very high specific activity of approximately 1.00?×?10(3) U mg(-1) on a standard substrate of p-nitrophenyl acetate. The Michaelis-Menten kinetic parameters V (max) and K (m) of the lipase were 1.37 mM mg(-1) min(-1) and 14.0 mM, respectively. Ca(2+) and other metal ions could not activate the lipase. According to the homology analysis and site-directed mutagenesis assay, the catalytic triad of the recombinant lipase was identified as Ser-165, Asp-260, and His-290 residues. 相似文献

金属有机骨架(Metal-organic Frameworks,MOFs)材料与传统的多孔材料相比,具有结构有序且多样、可调控的孔径尺寸和超高的比表面积等特点,使其在光催化领域得到了广泛的关注。同时,纳米荧光材料量子点(Quantum Dots,QDs)具有良好的光学特性且能够将近红外光转换为可见光,进而可以促进对占太阳光谱较大比例的可见光的吸收,它的出现也推动了光催化领域的发展。但二者作为单一相催化剂本身都存在缺点,限制了其作为光催化剂的进一步应用与发展。近年来,研究者发现将QDs和MOFs材料有效结合形成复合型光催化剂是提高单一相催化剂光催化性能的途径之一,并进行了初步研究,结果表明其催化性能大大提高,在解决环境问题方面有着广阔的应用前景。本文综合国内外现状,围绕QDs@MOFs复合型光催化剂的制备及在光催化降解领域的应用及发展进行了综述。本文还提出了在研究QDs@MOFs复合型光催化剂过程中应注意的关键性问题以及对未来的发展趋势进行了展望。 相似文献

Aspergillus oryzae SBS50 secreted a high titre of phytase in solid-state fermentation (SSF) using wheat bran at 30 °C after 96 h at the initial substrate to moisture ratio of 1:2 and a water activity of 0.95. The production of phytase increased when wheat bran was supplemented with sucrose and beef extract. Further enhancement in enzyme production was recorded when the substrate was supplemented with the surfactant Triton X-100 (145 U/g of DMB). An overall 29-fold improvement in phytase production was achieved owing to optimization. Under optimized conditions, the mould secreted 9.3-fold higher phytase in SSF as compared to submerged fermentation (SmF). The mesophilic mould also secreted amylase, cellulase (CMCase), pectinase and xylanase along with phytase in SSF. Scanning electron microscopy revealed luxuriant growth of A. oryzae on wheat bran with abundant spores. The enzyme dephytinized wheat bran with concomitant liberation of inorganic phosphate. 相似文献

肿瘤标志物在癌症早期诊断与预后监测中具有非常重要意义。前列腺癌作为男性泌尿生殖系统中最常见的恶性肿瘤,其血清前列腺特异性抗原(PSA)是最重要的早期检测指标。该研究以制备的四元Cu-Zn-In-S纳米晶(NCs)作为发光体,金纳米星(AuNSs)作为猝灭探针,利用K2S2O8作为共反应剂,发展了一种基于量子点电化学发光共振能量转移的免疫传感器(ECL-RET)用于血清中PSA的高灵敏检测。固定在传感器上的NCs表现出极强的初始ECL信号和较低的ECL电位。作为固定基底的多壁碳纳米管(MWNTs)不仅加速了NCs与S2O2?8之间的反应,获得了较高的ECL信号,而且作为载体可以固定大量的抗体,提高传感器检测PSA的灵敏度。采用电化学交流阻抗法(EIS)对组装过程进行跟踪,证实了该传感器组装的可行性。采用电化学发光法研究了传感器对底液中不同浓度PSA的响应,结果显示,随着体系中PSA浓度的增加,电极上结合AuNSs的标记抗体(Ab2-AuNSs)越多,触发的猝灭效果越明显,ECL信号降低越明显。因此,该传感器表现出对PSA较好的分析性能,其线性范围为0.01~150 ng/mL,检出限为0.03 ng/mL。用于血清样品的检测,该传感器的测试结果与常用的化学发光法基本吻合,相对误差仅为-7.2%~6.0%,结果准确度良好,为临床检测PSA提供了准确可靠的新方法。 相似文献

详细研究了头孢氯苄在酸,碱介质中的降解反应,建立了酸,碱降解的荧光分析法,线性范围是0.50-100ng/mL(酸降解)和2.0-70ng/mL(碱降解)。从精密度和准确度看,酸降解优于碱降解的荧光分析,将酸降解荧光分析应用于血清和尿样中头氨苄的测定,结果良好。 相似文献

针对g-C3N4有限的可见光吸收能力和容易复合的光生载流子限制其光催化性能发挥的问题,以尿素为前驱体,核黄素(RF)为掺杂试剂,通过一步热聚合制备了新型g-C3N4-RF光催化剂。系列表征测试(FT-IR,XRD,SEM,UV-vis DRS,PL)表明,该新型g-C3N4-RF光催化剂被成功制备,并具有优异的可见光吸收能力和有效的光生电子空穴对分离效率。光催化降解实验表明,该催化剂在可见光下对四环素抗生素具有优异的光催化降解性能。该研究为开发新型氮化碳光催化剂提供了新的思路,为抗生素污染物的高效去除提供了新的线索。

以直接法和蒸干法制样后用液闪仪测量了稀土厂废水的总(α+β)放射性比活度(Bq.L-1,下称比活度),以萃取法制样后测量了废水中氡的比活度。测量结果表明,在本实验条件下,以直接法和蒸干法制样测得结果有较大差异,废水的总比活均高于排放标准(GB8978-1996),由直接法所得结果远高于标准;三种废水中的氡均具有较高的比活度和放射性比重(80%)。指出了常规方法制样测量结果偏低的原因是忽略了废水中氡的放射性"贡献";讨论了废水中氡的来源、迁移和危害性。为综合评估稀土厂"三废"的放射性及其危害性提供了依据。 相似文献

以十二胺(DDA)或十八胺(ODA)为模板剂,采用焙烧或萃洗法去除模板剂制备中孔分子筛(HMS).X射线粉末衍射(XRD)及氮吸附表征结果说明,以十二胺为模板剂且采用焙烧法去除模板剂制备的分子筛具有明显的XRD介孔衍射峰,而且其氮吸附曲线具有典型的IV类等温线特征及H1型脱附滞后环,这属于典型的介孔材料特征.以此分子筛为载体,通过F—C反应将磺酸铁酞菁(FePcS)修饰在HMS上,得到新型光催化剂.催化剂的BET比表面积为675.1m2·g-1,平均孔径为5.78nm,孔容为0.587cm3·g-1,且仍保持着鲜明的介孔特征.最后在模拟可见光照射下应用催化剂处理浓度高达1000mg·L-1的模拟苯酚废水,反应400min后,苯酚的转化率达到85%以上,反应溶液pH值也由4.52降到2.65,表明有酸类降解中间产物生成,反应最终苯酚转化率接近100%,总有机碳(TOC)的去除率达81%以上.催化剂表现出了良好的催化降解有机废水的性能. 相似文献

The water-soluble polysaccharides (APPs) isolated from the edible mushroom Auricularia polytricha were irradiated by γ-ray at doses of 10, 100, and 1000 kGy. The effect of gamma irradiation on the degradation of the polysaccharide was investigated. After irradiation treatment, the viscosity and molecular weight of APPs decreased with the increase in the irradiation dose. The changes in the enthalpy of APPs after irradiation treatment were observed. Meanwhile, SEM showed that R-APPs were crushed into fragments and the surfaces became smooth and wrinkled after irradiation. In further spectrum analysis, it was found that the glycoside bonds of the polysaccharides were broken and accompanied by the formation of double bonds. This suggested that gamma irradiation could cause the depolymerization and oxidation of polysaccharides. In addition, irradiated APPs could reduce the body weight of hyperlipidemia mice. The levels of serum and liver TC, TG, and serum LDH-c significantly decreased in hyperlipidemia mice after treatment by irradiated APPs. It indicated that gamma irradiation significantly improved the anti-hypolipidemic activity of APPs. The relationship between the physicochemical properties and hypolipidemic activity of polysaccharides was interpreted, which provides a theoretical basis for the further development of APP products. Gamma irradiation is a viable technology for macromolecular modification for degradation. 相似文献

分别以溴代甲基咪唑反应型离子液体[C4mim]Br、[C8mim]Br和[C12mim]Br作为溴源和软模板,通过乙二醇溶剂热处理,获得了3种不同形貌结构的溴化氧铋(BiOBr)微粉。 X射线衍射(XRD)、扫描电子显微镜(SEM)和比表面积测定的表征结果证实,产物均属四方晶系,纯度较高,且随着离子液体中阳离子取代基上碳原子数(Cn)的增多,BiOBr晶体沿着(110)晶面择优生长;微米级的BiOBr产物颗粒均是由纳米尺度的花瓣状薄片相互簇集、自组装而形成,呈球心放射状,粒径和表面孔隙均随Cn的增加而减小,但比表面积随之而增大。 可见光降解苯胺的结果表明,3种BiOBr催化剂的催化活性不同,反应280 min时,降解率依次为54.13%、78.67%和95.06%;光生空穴是光降解过程的主要活性物质,羟基自由基所起的作用较小。 相似文献
