Isochrysis is a genus of marine algae without cell wall and capable of accumulating lipids. In this study, the lipid production potential of Isochrysis was assessed by comparing 15 Isochrysis strains with respect to their growth rate, lipid production, and fatty acid profiles. Three best strains were selected (lipid productivity, 103.0~121.7 mg L?1 day?1) and their lipid-producing capacities were further examined under different controlled parameters, e.g., growth phase, medium nutrient, and light intensity in laboratory cultures. Furthermore, the three Isochrysis strains were monitored in outdoor panel photobioreactors with various initial cell densities and optical paths, and the strain CS177 demonstrated the superior potential for outdoor cultivation. A two-stage semi-continuous strategy for CS177 was subsequently developed, where high productivities of biomass (1.1 g L?1 day?1) and lipid (0.35 g L?1 day?1) were achieved. This is a comprehensive study to evaluate the lipid-producing capability of Isochrysis strains under both indoor and outdoor conditions. Results of the present work lay a solid foundation for the physiological and biochemical responses of Isochrysis to various conditions, shedding light on the future utilization of this cell wall-lacking marine alga for biofuel production.  相似文献

Abstract

Brazilian sugarcane spirits were analyzed to elucidate similarities and dissimilarities by principal component analysis. Nine aldehydes, six alcohols, and six metal cations were identified and quantified. Isobutanol (LD 202.9 µg L?1), butiraldehyde (0.08–0.5 µg L?1), ethanol (39–47% v/v), and copper (371–6068 µg L?1) showed marked similarities, but the concentration levels of n-butanol (1.6–7.3 µg L?1), sec-butanol (LD 89 µg L?1), formaldehyde (0.1–0.74 µg L?1), valeraldehyde (0.04–0.31 µg L?1), iron (8.6–139.1 µg L?1), and magnesium (LD 1149 µg L?1) exhibited differences from samples.  相似文献

There is potential in the utilization of microalgae for the purification of wastewater as well as recycling the resource in the wastewater to produce biodiesel. The large-scale cultivation of microalgae requires pretreatment of the wastewater to eliminate bacteria and protozoa. This procedure is costly and complex. In this study, two methods of pretreatment, UV irradiation, and sodium hypochlorite (NaClO), in various doses and concentrations, were tested in the dairy wastewater. Combining the efficiency of biodiesel production, we proposed to treat the dairy wastewater with NaClO in the concentration of 30 ppm. In this condition, The highest biomass productivity and lipid productivity of Chlorella vulgaris reached 0.450 g L?1 day?1 and 51 mg L?1 day?1 after a 4-day cultivation in the dairy wastewater, respectively.  相似文献

Bacillus licheniformis TISTR 1010 was used for glutamic acid-independent production of poly-γ-glutamic acid (γ-PGA). A fed-batch production strategy was developed involving feedings of glucose, citric acid, and ammonium chloride at specified stages of the fermentation. With the dissolved oxygen concentration controlled at ≥50% of air saturation and the pH controlled at ~7.4, the fed-batch operation at 37 °C afforded a peak γ-PGA concentration of 39.9 ± 0.3 g L?1 with a productivity of 0.926 ± 0.006 g L?1 h?1. The observed productivity was nearly threefold greater than previously reported for glutamic acid-independent production using the strain TISTR 1010. The molecular weight of γ-PGA was in the approximate range of 60 to 135 kDa.  相似文献

Yeasts are good candidates to utilize the hydrolysates of lignocellulose, the most abundant bioresource, for bioproducts. This study aimed to evaluate the efficiencies of single-cell protein (SCP) and xylitol production by a novel yeast strain, Candida intermedia FL023, from lignocellulosic hydrolysates and xylose. This strain efficiently assimilated hexose, pentose, and cellubiose for cell mass production with the crude protein content of 484.2 g kg?1 dry cell mass. SCP was produced by strain FL023 using corncob hydrolysate and urea as the carbon and nitrogen sources with the dry cell mass productivity 0.86 g L?1 h?1 and the yield of 0.40 g g?1 sugar. SCP was also produced using NaOH-pretreated Miscanthus sinensis straw and corn steep liquor as the carbon and nitrogen sources through simultaneous saccharification and fermentation with the dry cell productivity of 0.23 g L?1 h?1 and yield of 0.17 g g?1 straw. C. intermedia FL023 was tolerant to 0.5 g L?1 furfural, acetic acid, and syringaldehyde in xylitol fermentation and produced 45.7 g L?1 xylitol from xylose with the productivity of 0.38 g L?1 h?1 and the yield of 0.57 g g?1 xylose. This study provides feasible methods for feed and food additive production from the abundant lignocellulosic bioresources.  相似文献

Botryococcus braunii is a microalga that is regarded as a potential source of renewable fuel because of its ability to produce large amounts of lipid that can be converted into biodiesel. Agro-industrial by-products and wastes are of great interest as cultivation medium for microorganisms because of their low cost, renewable nature, and abundance. In this study, two strategies for low-cost production of B. braunii biomass with high lipid content were performed: (i) the mixotrophic cultivation using molasses, a cheap by-product from the sugar cane plant as a carbon source, and (ii) the photoautotrophic cultivation using nitrate-rich wastewater supplemented with CO2 as a carbon source. The mixotrophic cultivation added with 15 g L?1 molasses produced a high amount of biomass of 3.05 g L?1 with a high lipid content of 36.9 %. The photoautotrophic cultivation in nitrate-rich wastewater supplemented with 2.0 % CO2 produced a biomass of 2.26 g L?1 and a lipid content of 30.3 %. The benefits of this photoautotrophic cultivation are that this cultivation would help to reduce accumulation of atmospheric carbon dioxide and more than 90 % of the nitrate could be removed from the wastewater. When this cultivation was scaled up in a stirred tank photobioreactor and run with semi-continuous cultivation regime, the highest microalgal biomass of 5.16 g L?1 with a comparable lipid content of 32.2 % was achieved. These two strategies could be promising ways for producing cheap lipid-rich microalgal biomass that can be used as biofuel feedstocks and animal feeds.  相似文献

The paper deals with the exploitation of Ipomoea carnea as a feedstock for the production of bioethanol. Dilute acid pretreatment under optimum conditions (3 %H2SO4, 120 °C for 45 min) produced 17.68 g L?1 sugars along with 1.02 g L?1 phenolics and 1.13 g L?1 furans. A combination of overliming and activated charcoal adsorption facilitated the removal of 91.9 % furans and 94.7 % phenolics from acid hydrolysate. The pretreated biomass was further treated with a mixture of sodium sulphite and sodium chlorite and, a maximum lignin removal of 81.6 % was achieved. The enzymatic saccharification of delignified biomass resulted in 79.4 % saccharification with a corresponding sugar yield of 753.21 mg g?1. Equal volume of enzymatic hydrolysate and acid hydrolysate were mixed and used for fermentation with a hybrid yeast strain RPRT90. Fermentation of mixed detoxified hydrolysate at 30 °C for 28 h produced ethanol with a yield of 0.461 g g?1. A comparable ethanol yield (0.414 g g?1) was achieved using a mixture of enzymatic hydrolysate and undetoxified acid hydrolysate. Thus, I. carnea biomass has been demonstrated to be a potential feedstock for bioethanol production, and the use of hybrid yeast may pave the way to produce bioethanol from this biomass.  相似文献

In the present study, the profiling of 17 fatty acids (FAs) in avocado seed and pulp was investigated. The fatty acids were extracted with vortex-assisted extraction, methyl esterified and finally preconcentrated by dispersive liquid–liquid microextraction. The preconcentrated fatty acid methyl esters (FAMEs) were analyzed using gas chromatography–mass spectrometry (GC–MS) to obtain qualitative and quantitative information. The GC–MS data were analyzed using multivariate curve resolution-alternating least squares (MCR-ALS) method to overcome general chromatographic problems such as overlapped peaks, background interference and peak shifts. The calibration data were prepared using pure analytical information obtained by MCR-ALS. The linear dynamic ranges and regression coefficients (R 2) for FAMEs were in the range of 0.19–65 mg L?1 and 0.990–0.999, respectively. The relative standard deviation (RSD%) for determination of FAs in avocado seed and pulp was 0.17–8.84 and 0.64–17.93, respectively. The main FAs in the avocado pulp were: oleic acid (74.25 g Kg?1), linoleic acid (26.87 g Kg?1), palmitic acid (26.02 g Kg?1), palmitoleic acid (1.22 g Kg?1) and stearic acid (0.05 g Kg?1). And, the main FAs in the avocado seed were: linoleic acid (1.09 g Kg?1), palmitic acid (0.47 g Kg?1), oleic acid (0.33 g Kg?1), linolenic acid (0.12 g Kg?1), and palmitoleic acid (0.04 g Kg?1).  相似文献

The aim of this work was to optimize the enzymatic hydrolysis of the cellulose fraction of cashew apple bagasse (CAB) after diluted acid (CAB-H) and alkali pretreatment (CAB-OH), and to evaluate its fermentation to ethanol using Saccharomyces cerevisiae. Glucose conversion of 82?±?2 mg/g CAB-H and 730?±?20 mg/g CAB-OH was obtained when 2% (w/v) of solid and 30 FPU/g bagasse was used during hydrolysis at 45 °C, 2-fold higher than when using 15 FPU/g bagasse, 44?±?2 mg/g CAB-H, and 450?±?50 mg/g CAB-OH, respectively. Ethanol concentration and productivity, achieved after 6 h of fermentation, were 20.0?±?0.2 g L?1 and 3.33 g L?1 h?1, respectively, when using CAB-OH hydrolyzate (initial glucose concentration of 52.4 g L?1). For CAB-H hydrolyzate (initial glucose concentration of 17.4 g L?1), ethanol concentration and productivity were 8.2?±?0.1 g L?1 and 2.7 g L?1 h?1 in 3 h, respectively. Hydrolyzates fermentation resulted in an ethanol yield of 0.38 and 0.47 g/g glucose with pretreated CAB-OH and CAB-H, respectively. Ethanol concentration and productivity, obtained using CAB-OH hydrolyzate, were close to the values obtained in the conventional ethanol fermentation of cashew apple juice or sugar cane juice.  相似文献

Temperature control is a very important factor on triterpene productivity in submerged liquid fermentation. Temperature effects from 23 to 32 °C on triterpene production by Ganoderma lucidum G0119 were investigated in 6-L stirred fermentor. Logistic and Luedeking-Piret equations were used to estimate the mycelial growth and triterpene production kinetics by regression analysis. On that basis, a temperature-shifting fermentation control strategy was established. From 0 to 61 h, culturing was performed at 32 °C to get high specific mycelial growth rate. Between 62 and 127 h, the temperature was decreased stepwise from 31 to 30 °C to maintain high triterpene productivity. After 128 h, temperature was maintained at 29 °C to minimize triterpene production inhibition and sustain high productivity. Elevated triterpene level (0.269 g L?1), yield (0.0101 g g?1), and productivity (0.00207 g (L h)?1) were achieved representing 27.32, 13.94, and 37.11 % higher than submerged liquid fermentation at constant temperature of 29 °C, respectively, feasible for the industrial scale.  相似文献

Accumulated carbohydrate in microalgae is promising feedstock for bioethanol fermentation. Selection of suitable cultivation conditions in semi-continuous cultivation is critical to achieve a high carbohydrate productivity. In the current study, the effects of macro-nutrient (nitrogen, phosphorus, and sulfur) limitations and light intensity were evaluated for the carbohydrate accumulations of Chlorella sp. AE10 under 10% CO2 conditions. It was shown that nitrogen limitation and high light intensity were effective for improving carbohydrate productivity. The average carbohydrate and biomass productivity in semi-continuous cultivation with 1/4 N medium and 1000 μmol photons m?2 s?1 was 0.673 and 0.93 g L?1 day?1, respectively. Sulfur and phosphorus limitations could improve the carbohydrate content but they could not enhance the carbohydrate productivity. The cell cycle progression and chlorophyll a were investigated using flow cytometry (FCM). The results showed that macro-nutrient limitation and high light intensity indeed influenced cell cycle progression and led to the formation of polyploid cells along with the carbohydrate accumulation in a certain range. FCM was rapid and accurate method to investigate the operation conditions why 1/4 N, 2 days as a cycle, and high light intensity were optimal ones. In addition, the remaining high level of photosynthesis activity was also important for achieving a high carbohydrate productivity. Dynamic tracking of carbohydrate accumulation is helpful for establishment of a semi-continuous cultivation for enhancing carbohydrate productivity in microalgae.  相似文献

A novel strain of Bacillus licheniformis P-104 was isolated from Chinese soybean paste to produce a bioflocculant. The bioflocculant was confirmed as ultra-high molecular weight poly-γ-glutamic acid (γ-PGA) using Fourier transform infrared spectrum, high-performance liquid chromatography, and gel permeation chromatography with multi-angle laser light scattering. The production technology and flocculation properties of γ-PGA were investigated. By fed-batch fermentation in a 7-L bioreactor, the maximum γ-PGA yield reached 41.6 g L?1 with a productivity rate of 1.07 g L?1 h?1. The flocculating activity of γ-PGA for kaolin suspension was 33.5?±?1.6 1/OD under the optimized flocculation conditions (6 mM Ca2+, 1.5 mg L?1 γ-PGA, and pH 6.0). The optimized dosage of γ-PGA for flocculation was just about 30 % of that of reported γ-PGA produced by other strains. Moreover, the flocculation activity of γ-PGA produced by strain P-104 was much higher than commercial γ-PGA with the molecular weight ranging 200–500 kDa and 1,500–2,500 kDa. This study provided a promising strain and an efficient method for production of ultra-high molecular weight γ-PGA which could be used as a potential green bioflocculant.  相似文献

The voltammeric behaviour of rimsulfuron herbicide has been studied by square wave stripping voltammetry on static hanging mercury drop electrode. It exhibited a well-defined peak within the pH range of 1.0–6.0, having a maximum peak response at ?600 mV (vs.Ag/AgCl) at pH 3.0. The factors such as accumulation potential (Eacc), accumulation time (tacc), frequency (f), pulse amplitude (ΔE) and step potential (ΔEs) have been optimised. The calibration plot was a straight line in the range of 4.4–134.4 μg L?1 with a detection limit of 1.3 μg L?1. The validity of the method was assessed from the recoveries of spiked lake water, tomato juice and agrochemical formulation of Doncep®. The results of the experiments conducted for five recoveries were 48.8 ± 1.7 and 49.7 ± 1.0 μg L?1, which are very close to the rimsulfuron spiked to lake water and tomato juice (50 μg L?1), with a relative error of –2.4% and ?0.6%, respectively. A brief study on dissolved radionuclides in aquatic environment, especially in ground water, constitutes the key aspect for assessment and control of natural exposure. In the present study the distribution of natural uranium and 226Ra concentration were measured in ground water samples collected within a 10 km radius around the Narwapahar uranium mine in the Singhbhum thrust belt of Jharkhand, India in 2007–2008. The natural uranium content in the ground water samples in this region was found to vary from 0.1 to 3.75 μg L?1 with an average of 0.87 ± 0.73 μg L?1 and 226Ra concentration was found to vary from 5.2 to 38.1 mBq L?1 with an average of 13.73 ± 7.34 mBq L?1. The mean annual ingestion dose due to intake of natural uranium and 226Ra through drinking water pathway to male and female adults population was estimated to be 6.55 and 4.78 μSv y?1, respectively, which constitutes merely a small fraction of the reference dose level of 100 μSv y?1 as recommended by WHO.  相似文献

An anaerobic sequencing batch reactor containing immobilized biomass (AnSBBR) was used to produce biomethane by treating the effluent from another AnSBBR used to produce biohydrogen from glucose- (AR-EPHG) and sucrose-based (AR-EPHS) wastewater. In addition, biomethane was also produced from sucrose-based synthetic wastewater (AR-S) in a single AnSBBR to compare the performance of biomethane production in two steps (acidogenic and methanogenic) in relation to a one-step operation. The system was operated at 30 °C and at a fixed stirring rate of 300 rpm. For AR-EPHS treatment, concentrations were 1,000, 2,000, 3,000, and 4,000 mg chemical oxygen demand?(COD)?L?1 and cycle lengths were 6 and 8 h. The applied volumetric organic loads were 2.15, 4.74, 5.44, and 8.22 g COD L?1 day?1. For AR-EPHG treatment, concentration of 4,000 mg COD L?1 and 4-h cycle length (7.21 g COD L?1 day?1) were used. For AR-S treatment, concentration was 4,000 mg COD L?1 day?1 and cycle lengths were 8 (7.04 g COD L?1 day?1) and 12 h (4.76 g COD L?1 day?1). The condition of 8.22 g COD L?1 day?1 (AR-EPHS) showed the best performance with respect to the following parameters: applied volumetric organic load of 7.56 g COD L?1 day?1, yield between produced methane and removed organic material of 0.016 mol CH4?g COD?1, CH4 content in the produced biogas of 85 %, and molar methane productivity of 127.9 mol CH4?m?3 day?1. In addition, a kinetic study of the process confirmed the trend that, depending on the biodegradability characteristics of the wastewaters used, the two-step treatment (acidogenic for biohydrogen production and methanogenic for biomethane production) has potential advantages over the single-step process.  相似文献

The occurrence of 26 commonly used cytostatic compounds in wastewaters was evaluated using an automated solid-phase extraction (SPE) method with liquid chromatography–high-resolution mass spectrometry (LC–HRMS). Detection was optimized using Oasis HLB SPE cartridges at pH 2. Two hospital effluents and their two receiving wastewater treatment plants were sampled over five days. In hospital effluents, eight cytostatics were detected at levels up to 86.2 μg L?1 for ifosfamide, 4.72 μg L?1 for cyclophosphamide, and 0.73 μg L?1 for irinotecan, the three most relevant compounds identified. Cyclophosphamide and megestrol acetate were found in wastewaters at concentrations up to 0.22 μg L?1 for the latter. The predicted environmental concentrations (PEC) in sewage effluents of ifosfamide (2.4–4.3 ng L?1), capecitabine (11.5–14.2 ng L?1), and irinotecan (0.4–0.6 ng L?1), calculated from consumption data in each hospital, published excretion values for the target compounds, and wastewater elimination rates, were in agreement with experimental values.  相似文献

Growths of Lyngbya limnetica and Oscillatoria obscura were investigated at varying pH, light intensity, temperature, and trace element concentration with a view to optimize these parameters for obtaining the maximum carbohydrate content. The maximum growth for both strains was obtained at pH 9.0 and temperature 20 ± 3 °C using a light intensity of 68.0 μmol m?2 s?1 with continuous shaking. Growth under the nitrogen starvation condition affected the carbohydrate content more compared to the phosphorus starvation, and maximum concentrations were found as 0.660 and 0.621 g/g of dry biomass for L. limnetica and O. obscura, respectively. Under the optimized nitrogen-rich conditions, the specific growth rates for the two strains were found to be 0.187 and 0.215 day?1, respectively. The two-stage growth studies under nitrogen-rich (stage I) followed by nitrogen starvation (stage II) conditions were performed, and maximum biomass and carbohydrate productivity were found as 0.088 and 0.423 g L?1 day?1 for L. limnetica. This is the first ever attempt to evaluate and optimize various parameters affecting the growth of cyanobacterial biomass of L. limnetica and O. obscura as well as their carbohydrate contents.  相似文献

The objective of this study was to set up a method to detect five compounds in fresh smashed apples by HPLC/DAD simultaneously. Different methods have been tested to control browning and ascorbic acid with ultrasonication was adopted. Methanol–water–acetic acid (30:69:1, v/v) containing 2.0 g of ascorbic acid L?1 was chosen as the extract solvent. The method effectively simplified the sample treatment compared with the traditional ways. And primarily, the results were used to identify between different varieties. The chromatographic separation was performed on an Atlantis C18 (250 mm × 4.5 mm, particle size 5 μm) with a gradient elution program using a mixture of acetonitrile and 2% aqueous acetic acid (v/v) as mobile phase within 20 min at 270 nm wavelength. The variation of the content of five compounds was gallic acid (ND ~1.81 μg g?1), protocatechuic acid (ND ~1.79 μg g?1), chlorogenic acid (13.81–189.4 μg g?1), caffeic acid (6.82–45.02 μg g?1) and rutin (0.96–18.55 μg g?1). The results could successfully be used to discriminate between different apple varieties (Gala, Fuji, Delicious, 8th Apple US, Golden Apple, Green Apple and Red Rose); chlorogenic acid and rutin being the polyphenols that contribute most to the differentiation.  相似文献

The effects of aeration and agitation on the properties and production of xanthan gum from crude glycerin biodiesel (CGB) by Xanthomonas campestris mangiferaeindicae 2103 were investigated and optimized using a response surface methodology. The xanthan gum was produced from CGB in a bioreactor at 28 °C for 120 h. Optimization procedures indicated that 0.97 vvm at 497.76 rpm resulted in a xanthan gum production of 5.59 g L?1 and 1.05 vvm at 484.75 rpm maximized the biomass to 3.26 g L?1. Moreover, the combination of 1.05 vvm at 499.40 rpm maximized the viscosity of xanthan at 0.5 % (m/v), 25 °C, and 25 s?1 (255.40 mPa s). The other responses did not generate predictive models. Low agitation contributed to the increase of xanthan gum production, biomass, viscosity, molecular mass, and the pyruvic acid concentration. Increases in the agitation contributed to the formation of xanthan gum with high mannose concentration. Decreases in the aeration contributed to the xanthan gum production and the formation of biopolymer with high mannose and glucose concentrations. Increases in aeration contributed to increased biomass, viscosity, and formation of xanthan gum with greater resistance to thermal degradation. Overall, aeration and agitation of CGB fermentation significantly influenced the production of xanthan gum and its properties.  相似文献

Voltammetric procedures for trace metals analysis in polluted natural waters using homemade bare gold-disk microelectrodes of 25- and 125-μm diameters have been determined. In filtered seawater samples, square wave anodic stripping voltammetry (SWASV) with a frequency of 25 Hz is applied for analysis, whereas in unfiltered contaminated river samples, differential pulse anodic stripping voltammetry (DPASV) gave more reliable results. The peak potentials of the determined trace metals are shifted to more positive values compared to mercury drop or mercury-coated electrodes, with Zn always displaying 2 peaks, and Pb and Cd inversing their positions. For a deposition step of 120 s at ?1.1 V, without stirring, the 25-μm gold-disk microelectrode has a linear response for Cd, Cu, Mn, Pb and Zn from 0.2 μg L?1 (1 μg L?1 for Mn) to 20 μg L?1 (30 μg L?1 for Zn, Pb and 80 μg L?1 for Mn). Under the same analytical conditions, the 125-μm gold-disk microelectrode shows linear behaviour for Cd, Cu, Pb and Zn from 1 μg L?1 (5 μg L?1 for Cd) to 100 μg L?1 (200 μg L?1 for Pb). The sensitivity of the 25-μm electrode varied for different analytes from 0.23 (±0.5%, Mn) to 4.83 (±0.9%, Pb) nA L μmol?1, and sensitivity of the 125-μm electrode varied from 1.48 (±0.7%, Zn) to 58.53 (±1.1%, Pb  nA L μmol?1. These microelectrodes have been validated for natural sample analysis by use in an on-site system to monitor Cu, Pb and Zn labile concentrations in the Deûle River (France), polluted by industrial activities. First results obtained on sediment core issued from the same location have shown the ability of this type of microelectrode for in situ measurements of Pb and Mn concentrations in anoxic sediments.   相似文献
