Three new 19-membered macrolides, amphidinolides T2 (2), T3 (3), and T4 (4), structurally related to amphidinolide T1 (1) have been isolated from two strains of marine dinoflagellates of the genus Amphidinium. The structures of 2-4 were elucidated on the basis of spectroscopic data. The absolute configurations at C-7, C-8, and C-10 of 1-4 were determined by comparison of NMR data of their C-1-C-12 segments with those of synthetic model compounds for the tetrahydrofuran portion. The biosynthetic origins of amphidinolide T1 (1) were investigated on the basis of 13C NMR data of a 13C enriched sample obtained by feeding experiments with [1-(13)C], [2-(13)C], and [1,2-(13)C2] sodium acetates and 13C-labeled sodium bicarbonate in the cultures of the dinoflagellate. These incorporation patterns suggested that amphidinolide T1 (1) was generated from four successive polyketide chains, an isolated C1 unit formed from C-2 of acetates, and three unusual C2 units derived only from C-2 of acetates. Furthermore, it is noted that five oxygenated carbons of C-1, C-7, C-12, C-13, and C-18 were not derived from the C-1 carbonyl, but from the C-2 methyl of acetates.  相似文献

The beneficial effects of Ginkgo biloba extract in the "treatment" of dementia are attributed to its terpene trilactone (TTL) constituents. The interactions between TTLs and amyloid peptide are believed to be responsible in preventing the aggregation of peptide. These interactions have been investigated using infrared vibrational absorption (VA) and circular dichroism (VCD) spectra. Four TTLs, namely ginkgolide A (GA), ginkgolide B (GB), ginkgolide C (GC) and bilobalide (BB) and amyloid Abeta(25-35) peptide, as a model for the full length peptide, are used in this study. GA-monoether and GA-diether have also been synthesized and investigated to help understand the role of individual carbonyl groups in these interactions. The precipitation and solubility issues encountered with the mixture of ginkgolide+Abeta peptide for VA and VCD studies were overcome using binary ethanol-D(2)O solvent mixture. The experimental VA and VCD spectra of GA, GB, GC and BB, GA-monoether and GA-diether have been analyzed using the corresponding spectra predicted with density functional theory. The time-dependent experimental VA and VCD spectra of Abeta(25-35) peptide and the corresponding experimental spectra in the presence of TTLs indicated that the effect of the TTLs in modulating the aggregation of Abeta(25-35) peptide is relatively small. Such small effects might indicate the absence of a specific interaction between the TTLs and Abeta(25-35) peptide as a major force leading to the reduced aggregation of amyloid peptides. It is possible that the therapeutic effect of G. biloba extract does not originate from direct interactions between TTLs and the Abeta(25-35) peptide and is more complex.  相似文献

(13)C NMR chemical shifts and nu(C[double bond]O) frequencies have been measured for several series of phenyl- or acyl-substituted phenyl acetates and for acyl-substituted methyl acetates to investigate the substituent-induced changes in the electrophilic character of the carbonyl carbon. Charge density, bond order, and energy calculations have also been performed. The spectroscopic and charge density results indicate that opposite to the conventional thinking, electron-withdrawing substituents do not increase the electrophilicity of the carbonyl carbon but instead decrease it. On the other hand, reaction energies of the isodesmic reactions designed show that electron-withdrawing substituents destabilize the carbonyl derivatives investigated. So, a significant ground-state destabilization of carboxylic acid esters, and carbonyl compounds in general, due to the decreased resonance stabilization, is proposed as a novel concept to explain both the increase in their reactivity and the changes in the chemical shifts and carbonyl frequencies induced by electron-withdrawing substituents.  相似文献

Ginkgolide B is in great demand worldwide on account of its extensive and excellent pharmacological effects, however, it is difficult to separate and purify ginkgolide B. In this study, ginkgolide B molecularly imprinted polymers were prepared by combining software simulation and molecular imprinting technique, and its characterization and adsorption performed evaluation were performed to understand the adsorption behavior of the polymers. The adsorption equilibrium concentration of molecularly imprinted polymers was 0.70 mg/mL, and the adsorption equilibrium time was 4 h. Meanwhile, the adsorption isotherm of the polymers for ginkgolide B fitted well with the Langmuir model, and the adsorption kinetics was in line with the pseudo‐second‐order kinetics. In contrast, the adsorption capacity of molecularly imprinted polymers on ginkgolide B was higher than that of non‐molecular imprinted polymers, with better selectivity and better adsorption after repeated use for six times. The application experiments showed that molecular imprinted polymers have a good adsorption effect in low purity samples. Therefore, the polymers reported herein can be expected to apply in the adsorption and separation of ginkgolide B samples.  相似文献

A method has been developed for measuring ginkgolide A, B, and bilobalide in ginkgo biloba L. extracts by using a self-assembled microdialysis device coupled on-line to high-performance liquid chromatography. The optimal conditions for dialysis efficiency and chromatographic analysis were investigated. The results show that the dialysis efficiencies for ginkgolide A, B, and bilobalide are 97.8–100.4%. For the spike concentration of 45 mg L–1, the recoveries of ginkgolide A, B, and bilobalide in ginkgo biloba L. extracts were at 97–100.7% with 3.1–4.7% RSD. The proposed method has therefore been shown to be applicable to the analysis of ginkgolide A, B, and bilobalide in ginkgo biloba L. extracts.  相似文献

Ginkgolides from the Ginkgo biloba tree are diterpenes with a cage structure consisting of six five-membered rings and a unique tBu group. They exert a variety of biological properties. In addition to being antagonists of the platelet activating factor receptor (PAFR), it has recently been shown that native ginkgolides are potent and selective antagonists of the inhibitory glycine receptor. Forty new ginkgolide derivatives have been prepared in good to high yields on milligram scales and investigated for their antagonistic properties at homomeric alpha 1 glycine receptors, thus providing the first structure-activity relationship study of ginkgolides at glycine receptors. A high-throughput screening assay showed that native ginkgolide C was the most potent ligand, and that manipulation of any of the hydroxyl groups led to loss of activity at alpha 1 glycine receptors.  相似文献

A comprehensive Raman spectroscopic study of the acetates of potassium, sodium and magnesium in the solid state have been made at both 298 and 77 K. Band separation of the C=O stretching region was not achieved in the 298 K spectra but was in the 77 K spectra. The CO stretching vibration is observed as a single band in both the 298 and 77 K spectra and its frequency is cation dependent. Single C–C stretching bands are observed for the acetates in the 77 K spectra. The OCO deformation vibrations were also cation dependent. Low frequency vibrations of magnesium acetate are observed at 338, 253 and 268 cm−1 and are assigned to the MgO stretching vibration of the magnesium bisacetato complex. Low frequency bands were also observed for sodium acetate at 219, 277 and 288 cm−1.  相似文献

The biosynthetic origins of amphidinoide B (1) were investigated on the basis of 13C-NMR data of 13C-enriched samples obtained by feeding experiments with [1-(13)C], [2-(13)C], and [1,2-(13)C2] sodium acetates in cultures of a dinoflagellate Amphidinium sp. These incorporation patterns suggested that 1 was generated from three successive polyketide chains, an isolated C1 unit from C-2 of acetates, six branched C1 units from C-2 of acetates, and an "m-m" and an "m-m-m" unit derived only from C-2 of acetates. The labeling patterns of amphidinolide B (1) were different from those of amphidinolide H (2), a 26-membered macrolide closely related to 1.  相似文献

The application of reversed-phase high-performance liquid chromatography to the analytical- and preparative-scale separation of sterols has been evaluated. The capacity factors, k', for a number of compounds chromatographed on a muBondapak C18 (LESS THAN 10 MUM) COLUMN ARE PRESENTED. C27, C28 and C29 sterols and also sterols differing in degree of unsaturation could be readily separated as their acetates in this system. The present reversed-phase chromatographic method is apparently not as selective as silver nitrate-silica gel thin-layer chromatography for the position of unsaturation in the sterol molecule.  相似文献

A new method for the preparation and separation of alditol acetates from neutral sugars has been applied to aminosugars. Reduced aminosugars were rapidly acetylated using 1-methylimidazole as the catalyst without removal of borate formed during reduction. The alditol acetates were separated by glass capillary gas chromatography on Silar 10C. The alditol acetates of aminosugars had retention times much longer than those of neutral sugars. However, the alditol acetates of the deamination products of aminosugars had shorter retention times and were resolved from those of neutral sugars. This method was used for the simultaneous detection of neutral and aminosugars in acid hydrolysates of chitin and the glycoproteins, ovalbumin and peroxidase.  相似文献

Second-order rate constants have been measured spectrophotometrically for the nucleophilic reactions of three substituted phenyl acetates with butane-2,3-dione monoximate (Ox(-)) as an alpha-nucleophile and p-chlorophenoxide (ClPhO(-)) as corresponding normal nucleophile, in MeCN-H2O mixtures of varying compositions at 25.0 +/- 0.1 degrees C. The reactivity of Ox(-) toward the aryl acetates decreases upon addition of MeCN to the reaction medium up to ca. 30 mol % MeCN, followed by a gradual increase in rate upon further addition of MeCN. A similar result has been obtained for the reaction of ClPhO(-) with the aryl acetates. However, the decrease in rate is more significant for the less reactive ClPhO(-) than for the more reactive Ox(-). Thus, for all the aryl acetates studied, Ox(-) exhibits a sizable alpha-effect (k(Ox)-/k(ClPhO)-) whose magnitude increases as the mol % MeCN in the reaction medium increases. The relative basicities (DeltapK(a)) of Ox(-) and ClPhO(-) have been determined spectrophotometrically using piperazine as a reference base. The DeltapK(a) values increase on increasing the mol % MeCN in the medium for both Ox(-) and ClPhO(-). The difference in the relative basicities of these nucleophiles (DeltaDeltapK(a)) becomes larger with increasing mol % MeCN. The plots of log k(Ox)-/k(ClPhO)- vs DeltaDeltapK(a) for the three substrates are linear with near-unit slope, indicating that the difference in the relative basicity of the nucleophiles is largely responsible for the increasing alpha-effect with medium composition in this system.  相似文献

Ginkgolide B is a dietary diterpene with multiple pharmacological activities. However, current research on ginkgolide B is not comprehensive. The current study analyzed the metabolic profile of ginkgolide B in vivo and in vitro using ultra-high-performance liquid chromatography quadrupole time-of-flight mass spectrometry. To detect and identify the different metabolites in ginkgolide B, a novel data processing method was used as an assistant tool. A total of 53 different metabolites of ginkgolide B (38 phase I metabolites and 15 phase II metabolites) were detected relative to blank samples. The biotransformation route of ginkgolide B was identified as oxidation, dehydroxylation, hydrogenation, decarbonylation, demethylation, sulfate conjugation, glucose conjugation, methylation, and acetylation. The current study demonstrated a method for rapidly detecting and identifying metabolites and provided useful information to further characterize the pharmacology and mechanism of ginkgolide B. A method for the analysis of other diterpene metabolic components in vivo and in vitro was also established.  相似文献

Ginkgo biloba L. is known to contain the unique terpene trilactone compounds bilobalide and ginkgolides. Capillary gas chromatography is used for the quantitative identification of bilobalide and the main ginkgolides (ginkgolide A, B, and C). The retention indices of these compounds are also determined. Retention indices of bilobalide and ginkgolide A, B, and C substitute the use of their standards at their routine identification. Our procedure does not require temperature-programmed operation.  相似文献

Organomercury derivatives have found widespread use as intermediates in organic syntheses.1-4 The organomercuric halides have been used most extensively due to the ease with which they are prepared, isolated, and manipulated. 5-8 The organomercuric acetates would be expected to be more reactive than the corresponding halide derivatives9 but they have not been utilized extensively because, with the exception of the aryl derivatives, they are not readily available. Arylmercuric acetates can be prepared via electrophilic aromatic substitution reactions1O,l1 but the preparation of vinyl and alkylmercuric acetates is not as straightforward. The few known vinylmercuric acetates were prepared via transmetallation reactions12 or destructive reactions of divinylmercury reagents.9 Except in rare instances, l3,14 the alkylmercuric acetates have not been isolated but have served as intermediates in the synthesis of alkylmercuric halides.7  相似文献

A concise synthesis of pyrazolo[1,5‐a]indole derivatives by copper‐catalyzed aerobic oxygenation and cyclization of indoles with oxime acetates is described. This protocol represents an elegant example of N‐1, C‐2, and C‐3 tri‐functionalization of indoles in one‐pot. Mechanistic studies indicate the reaction proceeds through a radical procedure. Oximes as an internal oxidant have been demonstrated to be a driver to initiate aerobic oxidation, which provides a new oxidative pattern for C‐H functionalization even with high atom‐ and step‐economy.  相似文献
