[reaction: see text] An efficient synthesis of the tricyclic A-B subunit 2 of angelmicin B is described. A formal three-component coupling of aldehydes 4 and 6 with gamma-silylallylborane 7 was employed to assemble the tetrahydrofuranyl core of 2, a strategy highlighted by the stereoselective [3 + 2] annulation of allylsilanes 5a/5b with aldehyde 4. The efficiency of the [3 + 2] annulation was greatly improved by using the allylic benzhydryldimethylsilane 5b compared to the allylic phenyldimethylsilane 5a.  相似文献

An efficient and general one-pot procedure for the synthesis of pyrazoles from acid chlorides, terminal alkynes and hydrazines was described via a coupling and cyclocondensation sequence. Acid chlorides coupled with terminal alkynes to give α,β-unsaturated ynones, and in situ converted into pyrazoles by the cycloaddition of hydrazines. The desired pyrazoles were obtained with 15-85% isolated yields.  相似文献

Both simple Ag(I) and Au(I) are effective catalysts for a tandem [3,3]-sigmatropic rearrangement/formal Myers-Saito cyclization of propargyl esters to form aromatic ketones. A mechanism in which the metal catalyzes both of these processes through alkyne activation is proposed. By using this method a wide range of aromatic structures including naphthyl, anthracenyl and indole ketones are available from readily available propargyl esters.  相似文献

Acyclic enynes undergo a tandem enyne metathesis/cyclopropanation sequence in the presence of Grubbs' 1st generation metathesis catalyst and diazo compounds. In practice, the acyclic substrates in the presence of the ruthenium alkylidene first undergo a ring-closing enyne metathesis to generate cyclic 1,3-dienes; then upon addition of a diazo compound, these products are cyclopropanated selectively at the more accessible olefin. Overall, the reaction sequence converts acyclic enynes into vinyl cyclopropanes in single operation through two unique ruthenium-catalyzed transformations.  相似文献

The sodium salt of hypophosphorous acid is shown to act as a double radical precursor in a double, sequential radical addition on 3-exo-methylenefuranose derivative 14 and 4-ethylenefuranose 10 to furnish phosphinates 18d in good overall yields. A straightforward access to the C10-C20 skeleton of gymnodimine, incorporating a tetrahydrofuran fragment, is described. The elaboration of the THF moiety is based on a stereocontrolled Ueno-Stork cyclization. A Lewis-acid mediated allylation of the resulting acetal at C13 and a Horner-Wadsworth-Emmons olefination on the ketone at C17 complete the synthesis.  相似文献

A computer-aided design of alkoxyl radical precursors is performed. The new precursors should combine the advantages of N-alkoxypyridine-2(1H)thiones (less reactive radicals) and N-alkoxythiazole-2(3H)thiones (stable with respect to daylight). Additionally, the radical liberation process should be initiated by light with a wavelength of around 350 nm. To find promising compounds, 18 test candidates were obtained by a systematic variation of the parent compound N-alkoxythiazole-2(3H)thione. The properties of the test molecules were computed by a protocol that was already successfully used to rationalize the photochemical behavior of N-alkoxypyridine-2(1H)thiones and N-alkoxythiazole-2(3H)thiones. The computations identify two promising new compounds. For N-methoxy-(1,3)dihydro-[1,3]azaphosphole-2-thione (6a), they predict that the fragmentation process will be initiated by an absorption at 348 nm. An analysis of its fragmentation process indicates that the free excess energy of the resulting radicals should more resemble the situation found for N-alkoxypyridine-2(1H)thiones. For N-methoxy-(1,3)dihydro-pyrrole-2-thione (3a), the excitation energy is somewhat higher (330 nm), but the computed fragmentation paths again indicate that the remaining excess energy of the released radicals is quite favorable. The test molecules also contained the experimentally well-known N-methoxypyridine-2(1H)one (1b). For this molecule, our computed data rationalizes nicely the experimental findings.  相似文献

Racemic AB monomers encompassing a secondary hydroxy group and a methyl ester moiety were synthesized and converted to chiral polyesters by iterative tandem catalysis (ITC). The concurrent action of an enantioselective acylation catalyst (Novozym 435) and a racemization catalyst (Ru(Shvo)) results in the high conversion of the racemic monomers to enantio‐enriched polymers. Several factors are important for attaining high ee's and high molecular weights. The enantioselectivities observed for the novel AB monomers by Novozym 435 are high enough at 70 °C (E ratio ≥ 200) for the monomers to be useful for ITC. ITC of methyl 6‐hydroxyheptanoate showed that a catalyst loading of ~1.4 mol % Ru(Shvo), 25 mg Novozym 435/mmol AB monomer, and 0.5 mmol DMP/mmol monomer employing a monomer concentration of 1 mol/L gave a monomer conversion of 94%, an ee of 91%, and an Mp of 6.0 kg/mol. Application of these conditions to the other AB monomers revealed the sensitivity of the system. Reduced enantioselectivities were observed when longer reaction times were required for attaining high conversions. These long reaction times were necessary due to the slow (or absent) racemization activity of the Ru(Shvo) catalyst as a result of catalyst deactivation. Since quantitative conversions are crucial to attain high molecular weight polymers in polycondensation reactions, we could significantly improve the system by switching to isopropyl esters of the AB monomers and/or by strict exclusion of oxygen during the ITC. © 2008 Wiley Periodicals, Inc. A norbornene-mediated palladium-catalyzed sequence is described in which two alkyl-aryl bonds and one alkenyl-aryl bond are formed in one pot with use of microwave irradiation. A variety of symmetrical and unsymmetrical oxygen-, nitrogen-, silicon-, and sulfur-containing tricyclic heterocycles were synthesized from a Heck acceptor and an aryl iodide containing two tethered alkyl halides. The natural chalcone bavachromanol has been prepared through a tandem reaction sequence that joins cationic cyclization of an epoxide to an adjacent MOM-acetal with electrophilic aromatic substitution by a presumed methoxymethylene cation. Only a single regioisomer of the tandem product was observed, with substitution taking place exclusively ortho to the position of the original acetal. This regiocontrol provided a key intermediate from a symmetrical precursor, and allowed preparation of the meroterpenoid through a short reaction sequence.  相似文献
