Reaction of 4-iminothiazolidin-2-one with acetylacetone

By the reaction of acetylacetone and arylazoacetylacetones with 4-iminothiazolidin-2-one thiazolo[4,5-b]pyridines were obtained in good yields. Optimum reaction conditions were chosen and some properties of compounds obtained were studied.     

Reaction of benzothiazol-2-one with 4-hydroxy-4-methyl-2-pentynonitrile

Reaction between the title compounds gave the unexpected 3-[5,5-dimethyl-2,5-dihydrofur-4-yl-2-(3-isopropanol-2-propenonitrile)imino]benzothiazol-2-one, as a result of the addition of two cyanoacetylene molecules.Irkutsk Institute of Organic Chemistry, Siberian Branch, Russian Academy of Sciences, Irkutsk 664033. 2-氨基-4-羰基噻吩的Knoevenagel反应

在碳酸钾等无机碱的存在下,异硫氰酸酯和4-氯乙酰乙酸乙酯反应生成2-氨基-4-羰基-4,5-二氢噻吩-3-甲酸乙酯。该化合物显示了与醛基的反应活性,即Knoevenagel反应,本文对该噻吩化合物的Knoevenagel反应位点选择性进行了研究。运用Gaussian03程序对化合物进行了几何全优化和相应的电荷、轨道能量分析研究。     

Reaction of 3,4,4-Trichloro-1-(4-methylphenyl)-3-buten-1-one with Amines

Reaction of 3,4,4-trichloro-1-(4-methylphenyl)-3-buten-1-one with amines results in replacement of the internal chlorine atom and is accompanied by prototropic allyl rearrangement leading to formation of the corresponding 3-amino-4,4-dichloro-1-(4-methylphenyl)-2-buten-1-ones. The reaction of the title compound with 2,4-dinitrophenylhydrazine yields 3,4,4-trichloro-1-(4-methylphenyl)-3-buten-1-one 2,4-dinitrophenylhydrazone and is not accompanied by allyl rearrangement.

Reaction of 2-trifluoromethyl-4H-3,1-benzoxazin-4-one derivatives with carboethoxymethylenetriphenylphosphorane

Treatment of a series of 2-trifluoromethyl-4H-3,1-benzoxazin-4-one derivatives 1 with carboethoxymethyl-enetriphenylphosphorane 2 yielded the phosphoranes 3 in boiling toluene solution. Thermolysis of these phosphoranes 3 gave esters 6.     

2-Carbethoxymethyl-4H-3,1-benzoxazin-4-one. 4. Reaction with anilines

In a solution of DMF, 2-carbethoxymethyl-4H-3,1-benzoxazin-4-one reacts with primarily aromatic amines basically with the formation of the corresponding 2-carbethoxymetlzyl-3-arylquinazolin-4(3H)-ones. Reaction of 3,3-Dichloro-2-dichloroacetylaminoacrylonitrile with Amines

Polychlorinated enamidonitrile Cl₂CÍC(CN)NHCOCHCl₂ readily reacts with primary aromatic amines, dialkylamines, piperidine, and morpholine. The reaction is accompanied by complete elimination of chlorine atoms as chloride ions with formation of 4-cyanooxazoles having the corresponding amine residue in position 5 of the ring and a CHÍN or diaminomethyl moiety in position 2. The structure of 5-arylamino(dialkylamino)-4-cyano-2-formyloxazoles was confirmed by their acid hydrolysis, as well as by the condensation with phenylhydrazine, N-alkylrhodanines, and ethyl acetoacetate in the presence of urea.     

Reaction of 2-Tetrafluoropropoxy-5,6-benzo-1,3,2-dioxaphosphorinan-4-one with Pyruvonitrile

The reaction of pyruvonitrile with 2-tetrafluoropropoxy-5,6-benzo-1,3,2-dioxaphosphorinan-4-one results mainly in the enlargement of the starting heteroring to form 4-cyano-4-methyl-2-tetrafluoropropoxy-6,7-benzo-1,3,2⁵-dioxaphosphepin-5-one 2-oxide as a mixture of two diastereomers (14:1) along with minor amounts of 3-cyano-3-methyl-2-tetrafluoropropoxy-6,7-benzo-1,4,2⁵-dioxaphosphepin-5-one 2-oxide.

在4-二甲氨基吡啶(DMAP)催化下, 1,3-二酮与3-丁炔-2-酮可以区域选择性地合成四取代的苯, 该方法具有反应条件温和和操作简单等优点, 产物的结构通过IR, 1H NMR, 13C NMR, HRMS确证.     

Reaction of 2-(4-Arylthiazol-2-yl)-4-chloro-3-oxobutyronitriles with Secondary Aliphatic Amines

We have obtained 2-(4-arylthiazol-2-yl)-4-chloro-3-oxobutyronitriles by acylation of 4-aryl-2-cyanomethylthiazoles with -chloroacetyl chloride. We have studied their reaction with secondary aliphatic amines, leading to formation of 4-dialkylamino-2-(4-arylthiazol-2-yl)-3-oxobutyronitriles, and also intramolecular alkylation with formation of 3-aryl-7-cyano-6(5H)-oxopyrrolo[2,1-b]thiazoles. We have determined some aspects of the tautomerism of the synthesized compounds.     

Investigation of the Reaction of 3,5-Diamidino-4-nitropyrazole with Amines

The interaction of 3,5-bis(N,N-dimethylaminomethylene)amino-1-methyl-4-nitropyrazole with amines proceeds regioselectively with the formation of 3-amino-5-(N,N-dimethylaminomethylene)amino-1-methyl-4-nitropyrazole. Amines are converted in this way into N,N-dimethylformamidine derivatives. The structures of the compounds obtained were proved by ¹H and ¹³C NMR spectroscopy and by X-ray crystallographic analysis.     

Unusual Direction of the Reaction of 2-Trifluoroacetoxy-1,3-benzodioxaphosphorin-4-one with N-Methyl-p-Methoxybenzalimine

芳亚甲基丙二腈和4-羟基喹啉-2-酮反应的研究

以芳亚甲基丙二腈和4-羟基喹啉-2-酮为原料,在六氢吡啶催化下以乙醇为溶剂,80℃合成了一系列新的2-氨基-3-氰基-4-芳基5,6-二氢化-4H-吡喃并[3,2-c]喹啉-5-酮衍生物,反应条件温和,产率较高,并通过单晶X射线衍射分析确证产物的结构.     

Reaction of 5,5-dimethyl-4-cyanomethylene-1,3-oxathiolan-2-one with amines and ammonia

A simple method for the synthesis of 5,5-dimethyl-4-cyanomethylene-2-cyanomethyl-2-[1-methyl-1-(carbamoyloxy)ethyl]-1,3-dithiolanes was developed on the basis of the reaction of 5,5-dimethyl-4-cyanomethylene-1,3-oxathiolan-2-one with amines (primary and secondary) and ammonia.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 3, pp. 335–338, March, 1992.     

Reaction of 2-bromo-4-acetyl-2,6-di-tert-butylcyclohexadiene-3,5-one with some nucleophilic agents

Conclusions When 2-bromo-4-acetyl-2,6-di-tert-butylcyclohexadiene-3,5-one is reacted with amines, potassium methylate, and methylmagnesium bromide, the reaction proceeds through a step in which 4-acetyl-2,6-di-tert-butylphenoxyl is formed.Translated from Izvestiya Akademii Nauk SSSR, Seriya Khimicheskaya, No. 10, pp. 2311–2313, October, 1969.The authors express their appreciation to N. Reaction of 2,6-dibutylaminohepta-2,5-dien-4-one with malononitrile

Malononitrile reacted with the title compound to give 6-amino-5-cyano-2-(3,3-dicyano-2-methylallylidene-4-methyl-2H-pyran (3). Treatment of 3 with hot 80% sulfuric acid yielded 4,7-dimethyl-56-hydroxy-2(1H)quinolone. With concentrated aqueous sodium hydroxide, 3 gave 5-amino-3,6-dicyano-4,7-dimethyl-2(1H)quinolone and 5-amino-6-carbamoyl-3-cyano-4,7-dimethyl-2(1H)quinolone. The reaction of 3 with hydrochloric in acetic acid gave a mixture of 6-amino-3,7-dicyano-2,8-dimethyl-4-quinolizone and 3-cyano-4-methyl-6-(3,3-dicyano-2-methylallyl)-2-pyrone. Compound 3 also reacted with methylamine, butylamine and piperidine to give 8-amino-5-cyano-4-methyl-2-pyridone, 6-bulylamino-5-cyano-4-methyl-2-pyridone and 5-eyano-4-methyl-6-piperidino-2-pyridone respectively.     

The reaction of Tanshinones with Amines

The reaction of cryptotanshinone and tanshinone IIA with several biogenic amine metabolites involved in the pathogenic pathways of HE were investigated and eight 1,2,3,4-tetrahydrophenanthrene derivatives,2-6 and 8-10,were obtained.The probable mechanism on reaction was discussed.
