The present study describes the development and validation of a selective liquid chromatographic (LC) method for the analysis of tenofovir disoproxil fumarate (TDF) and its related substances. The gradient method uses a base deactivated C18 column (Hypersil BDS column; 25 cm×4.6 mm I.D.) maintained at a temperature of 30°C. The mobile phases consist of acetonitrile, tetrabutylammonium/phosphate buffer pH 6.0 and water: (A; 2:20:78 v/v/v) and (B; 65:20:15 v/v/v). The flow rate is 1.0 mL/min and UV detection is performed at 260 nm. Good separation of TDF and 21 impurities was achieved. A system suitability test (SST) to check the quality of separation is also specified. The developed method was further validated with respect to robustness, precision, sensitivity and linearity. The method is proved to be robust, precise, sensitive and linear between 0.1 μg/mL and 0.15 mg/mL. The limit of detection and limit of quantification are 0.03 and 0.1 μg/mL, respectively. The method was successfully applied to the quantification of related substances and assay of commercial TDF samples (bulk substances and tablets).  相似文献

A simple, rapid, and precise reversed-phase high-performance liquid chromatographic method for the simultaneous determination of lamivudine, tenofovir disoproxil fumarate and efavirenz in bulk and tablet dosage form has been developed and validated. Chromatography was performed on a 150 mm × 4.6 mm i.d., 5-μm particle, Phenomenex Luna C18 column with 30: 45: 25 (v/v/v) acetonitrile: methanol: water as mobile phase at a flow rate of 0.5 mL/min. UV detection was done at 258 nm; lamivudine, tenofovir disoproxil fumarate and efavirenz were eluted with retention times of 3.27, 4.58 and 10.90 min, respectively. The method was validated in accordance with ICH guidelines. Validation revealed the method is specific, rapid, accurate, precise, reliable and reproducible. Calibration plots were linear over the concentration ranges 1–6 μg/mL for lamivudine and tenofovir disoproxil fumarate and 2–12 μg/mL for efavirenz. Limits of detection were 0.05, 0.09 and 0.11 μg/mL and limits of quantification were 0.15, 0.28 and 0.34 μg/mL for lamivudine, tenofovir disoproxil fumarate and efavirenz, respectively. The high recovery and low coefficients of variation confirm the suitability of the method for the simultaneous determination of these three drugs in bulk and tablets.  相似文献

A CE method was developed and validated for the assessment of the chiral purity of the drug tenofovir applying a quality by design approach. Following selection of a quaternary ammonium β-CD as chiral selector, a fractional factorial resolution V+ design was employed for identification of the critical process parameters, while a central composite design served for method optimization. The final method used a 40/50.2 cm, 50 μm id fused-silica capillary, a BGE composed of a 100 mM sodium phosphate buffer, pH 6.4, containing 45 mg/mL quaternary ammonium β-CD, an applied voltage of 18 kV, and a capillary temperature of 22°C. Robustness was assessed by a Plackett–Burman design. The method was validated according to guideline Q2(R1) of the International Council for Harmonization of Technical Requirements for Pharmaceuticals for Human Use and enabled the determination of the (S)-enantiomer of tenofovir at the 0.1% level.  相似文献

By computer simulation, using both quantum and classical dynamics, we determined the rate constant and the kinetic isotope effect of the rate-determining step in the neutral hydrolysis of p-methoxyphenyl dichloroacetate in aqueous solution. This step involves a proton transfer concerted with the formation of a C O bond. A method of biased sampling was used; the Gibbs free energy of the biased configuration from which proton transfer is likely to occur was determined by a combination of semiempirical quantum calculations and thermodynamic integration. The proton dynamics was modeled with the quantum-dynamical density matrix evolution method that includes nonadiabatic pathways. The proton dynamics is driven by a fluctuating proton potential that was derived from a classical molecular dynamics simulation of the system including solvent. The calculated rate constant of 3×10−2 s−1 agrees within the error of the calculation with the experimentally observed value of 2.78×10−3. The calculated pseudo-first-order kinetic isotope effect of 3.9 is in good agreement with the experimentally observed value of 3.2. The results show the feasibility of computational approaches to slow reactions in complex environments, where proton transfer with an essential quantum-dynamical nature is the rate-limiting step. ©1999 John Wiley & Sons, Inc. Amidoalkyl naphthols are synthesized via a simple, one-pot, three-component reaction between aldehydes, 2-naphthol and amides or ureas using polyphosphate ester (PPE) as a reaction mediator under solvent-free conditions in good to excellent yields. Polysubstituted pyridines are prepared in good yield and with total regiocontrol by the one-pot reaction of an alkynone, 1,3-dicarbonyl compound and ammonium acetate in alcoholic solvents. This new three-component heteroannulation reaction proceeds under mild conditions in the absence of an additional acid catalyst and has been used in the synthesis of dimethyl sulfomycinamate, the acidic methanolysis degradation product of the sulfomycin family of thiopeptide antibiotics.  相似文献

Hydrolytic degradation data from a wide variety of experiments on Estane 5703, a commercial poly(ester urethane), are modeled with an AAC2 mechanism of reversible esterification and hydrolysis together with a cluster model for water concentration. The experimental conditions spanned a range of temperatures from 21 to 95 °C, relative humidities from 0 to 100%, times up to more than 30 years, and different initial molecular weights and acidities. In addition, the experiments were performed in several different laboratories with a number of different instruments and techniques. All the experimental data, both for molecular weight and acidity, are modeled with a single set of concentration‐independent rate coefficients, and the kinetic model fits the data well and constitutes a robust model of the hydrolytic degradation of this polymer. The results also show that ester hydrolysis is the dominant cause of the molecular weight loss of Estane 5703 in indoor storage at ambient temperatures and humidities. © 2003 Wiley Periodicals, Inc. A one-pot synthesis of disubstituted benzimidazoles from 2-nitroanilines is described. Hydrogenation of N-substituted 2-nitroanilines with palladium on carbon as catalyst in the presence of trimethyl orthoformate and catalytic pyridinium p-toluenesulfonate (PPTS) at room temperature provided good to excellent yields of the corresponding disubstituted benzimidazoles.  相似文献

Phosphate ester hydrolysis is a key step in several enzymatic processes, which follow either a dissociative or an associative mechanism. While in the aqueous phase both pathways are favoured to about the same extent, the associative mechanism is relatively rarely observed. In this paper we report on quantum mechanical calculations for three enzymes HIV integrase, β-phosphoglucomutase and dUTPase, and try to find an explanation for the preference of the associative mechanism in a given enzyme. It is reasonable to suppose that the stabilisation of the pentacovalent, trigonal bipyramidal phosphorane moiety by formation of a covalent bond, one or more hydrogen bonds, or by co-ordination of a divalent metal cation with the equatorial oxygen atoms is the key factor. In all three enzymes studied one of the equatorial oxygen atoms is co-ordinated to a magnesium dication, while a second one is involved in a covalent bond. While in HIV integrase the third oxygen atom may only form a weak hydrogen bond with a solvent water molecule, in β-phosphoglucomutase this atom is stabilised by two strong hydrogen bonds with adjacent protein side chains and in dUTPase it is involved in a covalent bond. Hydroxyl-group functional polylactones were prepared and converted to acid- terminated polyesters in a reaction with a series of alkenylsuccinic anhydrides containing 8, 12, or 18 carbons in their alkenyl chains. These polyester precursors were then linked into higher molecular weight poly(ester anhydrides) containing alkenyl moieties in their polyester blocks. The hydrolysis behaviour of the poly(ester anhydrides) was found to depend on the thermal properties of the polyester precursors. For poly(ester anhydrides) prepared from low molecular weight prepolymers with thermal transitions below 37 degrees C, the presence of hydrophobic alkenyl chains in the polyester precursors slowed the rate of weight loss. Poly(ester anhydrides) prepared from higher molecular weight prepolymers showed the opposite weight-loss behaviour; i.e., the crystallinity and thermal transitions of the alkenyl chain-containing poly(ester anhydrides) were low, and the weight loss was faster than for poly(ester anhydrides) without the alkenyl chains. The differences in length of the alkenyl chain, as such, had little effect on the hydrolysis behaviour and thermal properties of the poly(ester anhydrides).  相似文献

In preparation for studying the kinetics of the hydrolysis of Estane® 5703, published hydrolysis data for a similar poly(butylene adipate) (herein called 63 PBA) have been modeled. Poly(ester urethanes) are known to degrade by acid‐catalyzed hydrolysis of the ester links, and the data on 63 PBA show that this degradation is reversible at low relative humidities (RHs). Previous work with these data was unable to fit them with concentration‐independent rate coefficients. It is shown herein that, with the water‐concentration model of the preceding article and the AAC2 mechanism of reversible hydrolysis/esterification, the experimental kinetic data that vary across temperature, RH, and starting molecular weight can be fit with a single set of concentration‐independent rate coefficients. © 2001 John Wiley & Sons, Inc. A sensitive ultra high performance liquid chromatography with tandem mass spectrometry method was developed for the simultaneous determination of darunavir, ritonavir and tenofovir in human plasma. Sample preparation involved a simple liquid–liquid extraction using 200 μL of human plasma extracted with methyl tert‐butyl ether for three analytes and internal standard. The separation was accomplished on an Acquity UPLC BEH C18 (50 mm x 2.1 mm, 1.7 μm) analytical column using gradient elution of acetonitrile/methanol (80:20, v/v) and 5.0 mM ammonium acetate containing 0.01% formic acid at a flow rate of 0.4 mL/min. The linearity of the method ranged between 20.0 and 12 000 ng/mL for darunavir, 2.0 and 2280 ng/mL for ritonavir, and 14.0 and 1600 ng/mL for tenofovir using 200 μL of plasma. The method was completely validated for its selectivity, sensitivity, linearity, precision and accuracy, recovery, matrix effect, stability, and dilution integrity. The extraction recoveries were consistent and ranged between 79.91 and 90.04% for all three analytes and internal standard. The method exhibited good intra‐day and inter‐day precision between 1.78 and 6.27%. Finally the method was successfully applied for human pharmacokinetic study in eight healthy male volunteers after the oral administration of 600 mg darunavir along with 100 mg ritonavir and 100 mg tenofovir as boosters.  相似文献

ABSTRACT

With cholic acid as the core, five multi-arm ester liquid crystals were synthesised with different terminal substituents. B1B5 were mesogenic arms which were linked to multifunctional chiral core cholic acid. The effect of electron-withdrawing and electron-donating groups on the mesogenic behaviour of the compounds was discussed. All products were structurally well characterised by elemental analysis, 1HNMR, and FT-IR. The phase behaviours were investigated by means of polarised optical microscopy and differential scanning calorimetry. B2, B4, B5 and C2, C4, C5 and D2, D4, D5 exhibited typical nematic texture and belonged to thermotropic enantiotropic nematic liquid crystals. B3, C3 and D3 also displayed typical nematic texture of thermotropic monotropic nematic liquid crystal. B1, C1 and D1 did not have mesogenic performance. Wider mesogenic range for 81.6°C of C5 whose terminal substituent is nitro on heating cycle while 121.1°C on cooling cycle. The results indicated that terminal substituents have a pretty important effect on the mesogenic phase and range of multi-arm liquid crystal compounds. The mesomorphic behaviour of compounds with polar groups terminally substituted performance much better than those without polar groups. In this ester multi-arm liquid crystal system, electron-withdrawing groups terminally substituted behave better than those electron-donating groups substituted.  相似文献

To test the potential of heterogeneous catalysts for the nylon-6 synthesis from 6-aminocapronitrile, a number of zeolites, aluminum silicate, and metal oxides were tested as catalysts for the model reaction of pentanenitrile with water and hexylamine to N-hexylpentanamide. All zeolitic and aluminum silicate systems showed an insufficient performance, while the metal oxides (TiO(2), ZrO(2), Nb(2)O(5)) showed very promising results. The kinetic behavior of the metal oxides was further investigated. First the nitrile was catalytically hydrolyzed to the terminal amide and subsequently the amidation of the hexylamine occurred. To polymerize 6-aminocapronitrile into nylon-6, more than 99 % nitrile conversion was required to obtain a high-molecular-weight polymer. Pentanenitrile conversions larger than 99 % can be obtained within six hours, at 230 degrees C, by using ZrO(2) as the catalyst. A kinetic study (by using IR spectroscopy) on the behavior of the metal oxides demonstrated that the adsorbed nitrile was catalytically hydrolyzed at the surface, but remained tightly bound to the surface. Zirconia-catalyzed polymerizations of 6-amino-capronitrile demonstrated that high-molecular-weight nylon-6 is feasible by using this route.  相似文献

Homogeneous precursor/precursor solutions with various compositions were obtained with appreciably high solid contents in N-methyl-2-pyrrolidone from soluble poly(amic diethyl ester) precursors of rodlike poly(p-phenylene biphenyltetracarboximide) (BPDA-PDA) and flexible poly(4,4′-oxydiphenylene biphenyltetracarboximide) (BPDA-ODA), which are hydrolytically more stable as well as more soluble than the corresponding poly(amic acid)s being equilibrated with the constituent monomers. Both optical microscopic and light scattering measurements showed that the dried precursor blend films and resultant polyimide composite films were optically transparent, regardless of compositions and process conditions. The composite films showed a single Tg behavior. However, for the composite of 30 wt % BPDA-PDA dispersed in the matrix of 70 wt % BPDA-ODA, a smectic crystalline-like aggregation of the BPDA-PDA component was detected on wide-angle x-ray diffraction patterns, indicative of microscopic phase separation between the two components. This phase separation was not detected on the optical microscopy, light scattering, and dynamic mechanical thermal analysis because of their resolution limits: Optical microscopy has a resolution of submicrometers, whereas dynamic mechanical thermal analysis and light scattering have a resolution of ca. 50 Å. Therefore, it is speculated that in the composite films BPDA-PDA and BPDA-ODA polyimide molecules have demixed on the scale of a few nanometers. The mean long periodicity, which was estimated from the small-angle x-ray scattering pattern, varied from 134 to 170 Å as the content of BPDA-ODA component increased. The use of I2/AgOAc in dichloromethane constitutes a cheap, mild, and efficient method for the selective iodination of a variety of heterocycles.
