In an effort to develop an efficient synthetic method of highly diastereoselective (2′R)- and (2′S)-2′-deoxy[2′-2H]guanosines, chemoenzymatic conversion was investigated. The synthesis of (2′R > 98% de)-2′-deoxy[2′-2H]guanosine was achieved by biological transdeoxyribosylation using (2′R > 98% de)-2′-deoxy[2′-2H]uridine, 2,6-diaminopurine, and Enterobacter aerogenes AJ-11125, followed by treatment with adenosine deaminase. (2′S > 98% de)-2′-Deoxy[2′-2H]guanosine was synthesized from (2′S > 98% de)-2′-deoxy[2′-2H]uridine and 2,6-diaminopurine using thymidine phosphorylase and purine nucleoside phosphorylase instead of E. aerogenes AJ-11125.  相似文献

A stereoselective synthesis of (1′S,3R,4R)-4-acetoxy-3-(2′-fluoro-1′-trimethylsilyloxyethyl)-2-azetidinone as a new fluorine-containing intermediate towards β-lactams, is described. The synthetic key step relies upon the dynamic kinetic resolution (DKR) of ethyl 2-benzamidomethyl-4-fluoro-3-oxo-butanoate via asymmetric transfer hydrogenation catalyzed by [Ru(η6-arene)(S,S)-R2NSO2DPEN].  相似文献

Optically pure trans-cyclobutane analogues of glutamic acid are prepared by highly selective enzymatic transamination of a single racemic trans-cyclobutane α-ketoglutaric acid derivative 5, which is synthesized in five steps from maleic anhydride. (2S,1′R,2′R)- and (2S,1′S,2′S)-2-(2′-carboxycyclobutyl)glycines L-CBG-II and L-CBG-I are obtained using aspartate aminotransferase (AAT) and branched chain aminotransferase (BCAT), respectively.  相似文献

Semi-empirical (AM1-SCI) calculations have been performed on 2-(2′-hydroxyphenyl)oxazole (HPO), 2-(2′-hydroxyphenyl)imidazole (HPI) and 2-(2′-hydroxyphenyl)thiazole (HPT) to rationalise the photophysical behaviour of the compounds exhibiting intramolecular rotation as well as excited state intramolecular proton transfer (ESIPT). The calculations reveal that there is a gradual variation in the properties from HPO to HPT through HPI so far as the existence of the rotational isomers in the ground state is concerned. While HPO gives rise to two stable rotamers (I and II) in all the common solvents, there is only one stable species for HPT in the S0 state. For HPI, rotamer II is possible only in the isolated state and/or in solvents of low polarity, but in high polar solvents it gives rise to the normal form (I) only. For all the molecules in the series, however, intramolecular proton transfer (IPT) takes place in the lowest excited singlet (S1) and the triplet (T1) states. Combination of the rotamerism and ESIPT gives rise to multiple fluorescence bands for the fluorophores. Theoretical assignments have been made for the excitation, fluorescence and phosphorescence bands. Simulated potential energy curves (PEC) in different electronic states reveal that the IPT process is feasible in either of the S1 and T1 states but not in the ground state. The ESIPT reaction has been found to be favoured both thermodynamically and kinetically in these electronic states compared to the ground state. However, quantum mechanical tunnelling has been proposed for the prototropic reaction to proceed in the excited states.  相似文献

A concise enantioselective total synthesis of (2S,3′R,7′Z)-N-(3′-hydroxy-7′-tetradecenoyl)-homoserine lactone is described. Key feature of this protocol is a catalytic asymmetric hydrogenation and a prophenol-zinc-catalyzed diazo addition to imine reaction as genesis of chirality. Moreover, flexibility is built in the synthesis to generate enantioenriched analogs using catalytic amount of enantioenriched C2-symmetric ligands.  相似文献

Cascade assembly of N,N′-dialkylbarbituric acids and aldehydes in the presence of bromine leads to the selective and efficient formation of substituted 1,5-dihydro-2H,2′H-spiro(furo[2,3-d]pyrimidine-6,5′-pyrimidine)-2,2′,4,4′,6′-(1′H,3H,3′H)-pentones in 70-88% yields via a complex cascade process. Spirobarbiturates containing the furo[2,3-d]pyrimidine framework are a class of compounds with interesting pharmacological and physiological activity.  相似文献

The synthesis of 2′-O,3′-O bicyclic adenosine derivatives is presented as the first examples of a new family of 13-membered ring bicyclic nucleoside analogues. Cyclisation was achieved through ring closing metathesis (RCM) on a diene intermediate using Grubbs’ catalyst. The Z and E isomers were purified and characterised.  相似文献

A new pyrrolidinyl peptide nucleic acid (PNA) comprising of an alternate sequence of 4′-nucleobase-modified proline with (2′R,4′S) configuration and a (1S,2S)-2-aminocyclopentanecarboxylic acid [(2′R,4′S)-acpcPNA] backbone was synthesized and its DNA-, RNA- and self-pairing properties studied. Tm and CD studies suggested that the (2′R,4′S)-acpcPNA forms antiparallel hybrids to DNA and RNA with high sequence and direction specificity. The stability of these hybrids is comparable to those of the (2′R,4′R)-acpcPNA hybrids previously reported by our group. On the other hand, experiments with a self-complementary sequence indicated that the new (2′R,4′S)-acpcPNA forms a more stable antiparallel self-hybrid than (2′R,4′R)-acpcPNA.  相似文献

The condensation of pyrrole with 4-pyridylcarboxyaldehyde and methyl 4-formyl benzoate under Adler-Longo conditions yielded the series of meso-(4′-pyridyl)/(4′-carboxymethylphenyl)porphyrins as a mixture. Careful column chromatography afforded each isomer in pure form. In this paper we focus on the two bis-substituted isomeric meso-porphyrins, 5,10-bis(4′-pyridyl)-15,20-bis(4′-carboxymethylphenyl)porphyrin and 5,15-bis(4′-pyridyl)-10,20-bis(4′-carboxymethylphenyl)porphyrin, respectively, 4′-cis and 4′-transDPyDMeP. The assignment of the geometry of the two isomers was performed by 1H NMR spectroscopy on the trinuclear adducts [(4′-cisDPyDMeP){Ru(TPP)(CO)}2] and [(4′-transDPyDMeP){Ru(TPP)(CO)}2], obtained by selective coordination of [Ru(TPP)(CO)(EtOH)] (TPP=tetraphenylporphyrin) to the peripheral nitrogen atoms. The axially bound ruthenium porphyrins act as chemical shift reagents on the central porphyrin, allowing a clear distinction of the pyrrole proton resonances and consequent unambiguous assignment of the geometry of each isomer based upon symmetry considerations.  相似文献

The highly enantioselective synthesis of (R)-isopropyl 3-(3′,4′-dihydroxyphenyl)-2-hydroxypropanoate and its enantiomer has been achieved starting from 3,4-dihydroxybenzaldehyde. The stereogenic centers were established through asymmetric dihydroxylation of (E)-isopropyl 3,4-bis(benzyloxy) cinnamate. A convenient manipulation in selective catalytic hydrogenation and deprotection was also accomplished in HCl-iPrOH employing 10% Pd/C catalyst.  相似文献

An insecticidal piperidine alkaloid, (2R,3R,6R)-N-methyl-6-(deca-1′,3′,5′-trienyl)-3-methoxy-2-methylpiperidine, was efficiently synthesized in a stereoselective manner starting from d-alanine. Chiral center at C-6 was controlled by hydrogenation of imine and side chain was introduced by Julia olefination. Synergistic liquid-liquid extractive spectrophotometric determination of gold(III) using 1-(2′,4′-dinitro aminophenyl)-4,4,6-trimethyl-1,4-dihydro pyrimidine-2-thiol [2′,4′-dinitro APTPT] has been described. Equal volumes (5 cm3) of the 2′,4′-dinitro APTPT (0.02 mol L−1) in the presence of pyridine (0.5 mol L−1) form an orange-red coloured ternary complex with gold(III) of molar ratio 1:1:1 at pH 1.8-2.4 with 5 min of shaking. The absorbance of coloured organic layer in 1,2-dichloroethane is measured spectrophotometrically at 445 nm against reagent blank. A pronounced synergism has been observed by the binary mixture of 2′,4′-dinitro APTPT and pyridine, which shows that the enhancement in the absorbance is observed in the presence of pyridine by the adduct formation in the organic phase. Beer's law was obeyed in the concentration range 2.5-20.0 μg mL−1, with molar absorptivity and Sandell's sensitivity values of 8.7 × 103 dm3 mol−1 cm−1 and 0.023 μg cm−2 respectively. A repetition of the method was checked by finding relative standard deviation (R.S.D.) (n = 10) which was 0.17%. The composition of the gold(III)-2′,4′-dinitro APTPT-pyridine adduct was established by slope analysis, molar ratio and Job's method. The ternary complex was stable for more than 48 h. The influence of various factors such as pH, 2′,4′-dinitro APTPT concentration, solvent and pyridine on the degree of complexation has been established. A number of foreign ions tested for their interferences and use of suitable masking agents wherever necessary are tabulated, which show that selectivity of the method has been enhanced. The method is successfully employed for the determination of gold(III) in binary, synthetic mixtures and ayurvedic samples. The reliability of the method is assured by inter-comparison of experimental values, using an atomic absorption spectrometer.  相似文献

The absorption and emission properties of benzimidazol-2-yl-quinoline (BIQ) and bis (2-(benzimidazol-2-yl) quinolinato) zinc (ZnBIQ) a new emitter used for organic light emitting device (OLED) were reported. Exciplexes are observed for ZnBIQ with N,N′-bis-(1-naphthyl)-N,N′-diphenyl-1,1′-biphenyl-4,4′-diamine (NPB) system, in both electro- and photoluminescent processes. The identification of exciplex emission in co-evaporated and multi-layer ZnBIQ thin film was reported for the first time. The optical formation of the exciplex involves the excitation of a single molecule, followed by the relaxation of that exciton into a lower energy exciplex state. Both BIQ and ZnBIQ possess very high thermal stabilities and can be purified by subliming under the high vacuum condition. Devices consisting of ZnBIQ as the emitting layer have been fabricated, and the emission spectra of ZnBIQ-base devices gave a voltage-dependent spectrum, with the red emission observed (3-7 V), switching over to strong white emission as the bias was raised.  相似文献

As novel methodology for synthesizing the furan ring, a photoinduced oxidative cyclization of 5-(4′,9′-methanocycloundeca-2′,4′,6′,8′,10′-pentaenylidene)pyrimidine-2,4,6(1,3,5H)-triones (7a-c) and related compounds 9a-c was accomplished to give 5,10-methanocycloundeca[4,5]furo[2,3-d]pyrimidine-2,4(1,3H)-dionylium tetrafluoroborates (8a-c+·BF4) and related compounds 2a-c+·BF4, respectively. In the photoinduced oxidative cyclization, the molecular oxygen in air is used as oxidant and the reaction proceeds under mild conditions to give desired products without byproducts, and thus, it is interesting from the viewpoint of the green chemistry. On the reactions of the mono-substituted derivatives 7d,e and 9e,f, the selectivity of the photoinduced cyclizations were reversed as compared with those of the DDQ-promoted oxidative cyclizations. By the NMR monitoring of the reactions of 7a and deuterated compound 7a-D2 under degassed conditions, the details of the reaction pathway were clarified and rationalized on the basis of the MO calculation by the 6-31G basis set of the MP2 levels as well.  相似文献

Lipase-catalyzed asymmetric acetylation of a mixture of (6R,1′S,4′S,5′R)- and (6R,1′R,4′R,5′S)-7′-norsesquisabinen-4′-ol (3) afforded a separable mixture of the recovered former and the acetate of the latter. The recovered alcohol was oxidized to (6R,1′S,5′R)-sesquisabina ketone (2), whose absolute configuration could be assigned by its CD comparison with (1R,5S)-sabina ketone (4). Conversion of (6R,1′S,5′R)-sesquisabina ketone (2) to the bioactive pheromone revealed the stereostructure of the male aggregation pheromone of the stink bug Erysarcoris lewisi (Distant) to be (2Z,6R,1′S,5′S)-2-methyl-6-(4′-methylenebicyclo[3.1.0]hexyl)hept-2-en-1-ol (sesquisabinen-1-ol, 1).  相似文献

Knoevenagel products formed by the condensation of N-monoalkyl barbituric acids with o-tert-amino benzaldehydes undergo tert-amino effect reactions (T-reactions) yielding 1-alkyl-2,4,6-trioxoperhydropyrimidine-5-spiro-3′-(1′,2′,3′,4′-tetrahydroquinoline) derivatives as a mixture of (S,S)- and (S,R)-diastereomers. Mostly, the major diastereomer has the S,S-configuration. According to X-ray diffraction data in the solid form and NOE data in solution, diastereoselectivity of the T-reactions can be associated with the structure of the Knoevenagel products whose conformation is fixed by the strong intramolecular C-H?π interaction.  相似文献

All of the four stereoisomers of (1′S)-1′-ethyl-2′-methylpropyl 3,13-dimethylpentadecanoate, the major component of the female sex pheromone of Clania variegata, were synthesized by employing olefin cross metathesis as the key reaction and starting from (R)- or (S)-2-methyl-1-butanol, (R)- or (S)-citronellal, and (S)-2-methyl-3-pentanol. Their bioassay revealed the (3R,13R,1′S)-isomer as the bioactive one, whose more efficient synthesis was achieved in two different ways by employing Wittig reaction as the key step.  相似文献

To synthesize (3′R,5′S)-3′-hydroxycotinine [(+)-1], the main metabolite of nicotine (2), cycloaddition of C-(3-pyridyl)nitrones 3a, 3c, and 15 with (2R)- and (2S)-N-(acryloyl)bornane-10,2-sultam [(2R)- and (2S)-8] was examined. The dimeric starting material [Ru(η6-p-cymene)(μ-Cl)Cl]2 reacts with N3,N3′-bis(diphenylphosphino)-2,2′-bipyridine-3,3′-diamine, 1 and P,P′-diphenylphosphinous acid-P,P′-[2,2′-bipyridine]-3,3′-diyl ester, 2 ligands to afford bridged dinuclear complexes [C10H6N2{NHPPh2-Ru(η6-p-cymene)Cl2}2], 3 and [C10H6N2{OPPh2-Ru(η6-p-cymene)Cl2}2], 4 in quantitative yields. These bis(aminophosphine) and bis(phosphinite) based Ru(II) complexes serve as active catalyst precursors for the transfer hydrogenation of acetophenone derivatives in 2-propanol and especially 4 acts as a good catalyst, giving the corresponding alcohols in 99% yield in 20 min (TOF ? 280 h−1).  相似文献
