A series of 3-methoxycarbonylpropoxy-7-(imidazol-4-ylpropinamide)-1,3-dihydrogen-1-methyl-5-phenyl-2H-1,4-benzodiazepin-2-ones,as farnesyltransferase(Ftase) inhibitors,were synthesized. The preparation of the key intermediate,7-amino-3-methoxycabonylpropoxy-1-methyl-5-phenyl-2H-1,4-benzodiazepin-2-one,was improved.  相似文献

Air stable benzodiazepine containing palladacycles were synthesized by a C-H activation reaction and studied by mass spectrometry and X-ray crystallography. Catalytic C-H functionalizations of 1-methyl-5-phenyl-1H-1,4-benzodiazepin-2(3H)-one with diphenyliodonium hexafluorophosphate led to a mixture, which included the starting material and the expected product 1-methyl-5-(2′-biphenyl)-1H-1,4-benzodiazepin-2(3H)-one.  相似文献

Reductive cyclizations of methyl 2-(2-nitrophenylthio)acetate 2 by means of electrochemistry or by catalytic hydrogenation have been used as key steps in the syntheses of the hemiacetals 2-hydroxy-2H-1,4-benzothiazin-3(4H)-one 10 and its 4-hydroxy derivative 9 representing thio analogues of allelo chemicals found in Gramineae and Acanthaceae. In contrast to its natural counterpart 2,4-dihydroxy-2H-1,4-benzoxa-zin-3(4H)-one hydroxamic acid 9 did not undergo degradation by extrusion of formic acid.  相似文献

2-Amino-2H-1,4-benzoxazin-3(4H)-one 3 , 2-amino-2H-1,4-benzothiazin-3(4H)-one 4 , 2-mercapto-2H-1,4-benzoxazin-3(4H)-one 7 , and 2-mercapto-2H-1,4-benzothiazin-3(4H)-one 8 representing aza and thio analogues of the natural product's aglucone Blepharigenin (2-hydroxy-2H-1,4-benzoxazin-3(4H)-one) from Gramineae and Acantnaceae species have been synthesized for the first time from their 2-bromo precursors 1 and 2 . Attempts to similarly prepare the 4-hydroxy derivatives of 7 and 8 , which would represent new thio analogues of the naturally occurring cyclic hydroxamic acid, 2,4-dihydroxy-2H-1,4-benzoxazin-3(4H)-one, have failed.  相似文献

4-Hydroxy-2H-chromen-2-one and 4-hydroxy-2H-chromene-2-thione reacted with allyl bromide, 1,1,3-trichloroprop-1-ene, and 1,3-dichlorobut-2-ene to give the corresponding ethers, which were oxidized to (2-oxo-2H-chromen-4-yloxy)acetic acid with potassium permanganate, and various derivatives of that acid were obtained. 3-(3,3-Dichloroprop-2-enyl)-7-hydroxy-4-methyl-2H-chromen-2-one and 3-(3,3-dichloroprop-2-enyl)-7-hydroxy-4-methyl-2H-chromene-2-thione were synthesized, and some their transformations were studied.  相似文献

An efficient and simple synthesis of 1,4-benzoxazepin-2-one derivatives has been achieved via the reaction of isoquinoline, activated acetylenes, and 1-(6-hydroxy-2-isopropenyl-1-benzofuran-yl)-1-ethanone in water without using any catalyst. To evaluate the feasibility of poly(1,4-dioxan-2-one) (PPDO) as a feed stock recycling material, the pyrolysis kinetics of PPDO were investigated. The pyrolysis of PPDO exclusively resulted in the distillation of 1,4-dioxan-2-one (PDO). From thermogravimetric measurements conducted at different heating rates, the kinetic parameters of the pyrolysis: activation energy, Ea=127 kJ mol−1; order of reaction, n=0; and pre-exponential factor, A=2.3×109 s−1, were estimated by plural analytical methods. The estimates show that the decomposition of PPDO proceeds by unzipping depolymerization as main reaction and random degradation process with lower Ea and A values. Equivalent isothermal degradation curves calculated from the thermogravimetric curves were supported by experimental isothermal degradation data. The calculation that PPDO is converted smoothly into PDO at 270°C agrees with the reported ceiling temperature of PPDO.  相似文献

We report the complete synthesis of a variety of 1,4-disubstituted gamma-carbolines. These compounds are of particular interest for their involvement in many biological processes and are believed to possess various medicinal activities. A large number of N-tosylaldimines were condensed with indoles affording an array of 3-aminomethyl indoles. Subsequent additions, followed by intramolecular cyclization, afforded an array of 1,2-dihydro-3H-gamma-carbolones in good yield. Upon subsequent aromatization, the corresponding fully aromatic functionalized 1-aryl-4-hydroxy-gamma-carbolines resulted.  相似文献

Solid-phase syntheses of 1,3,5-tri-substituted and 1,3,4,5-tetra-substituted 1,4-diazepin-2-ones 15-18 have been accomplished by employing inexpensive commercially available alpha- and beta-amino acids on Wang resin. Reductive amination of the imine formed by condensation of Wang aldehyde resin respectively with beta-alaninate 2 and beta-homophenylalaninate 3, followed by aminoacylation with a set of alpha-N-Boc amino acids (Phe epsilon-( Z)-Lys, and Leu) gave tertiary amide resins 7 and 8. Exposure of resins 7 and 8 to an excess of vinyl magnesium bromide in the presence of copper cyanide gave the corresponding gamma,delta-unsaturated ketone resins 9 and 10 by way of a cascade addition. Diazepinones were made by Boc deprotection and intramolecular reductive amination. To diversify the heterocycle, N-alkylation was performed using a series of alkyl halides. Alternatively, diazepinones 15e-g were obtained from treatment of methyl beta-alaninate resins 4 and 20 under similar copper-catalyzed cascade conditions to afford the gamma,delta-unsaturated ketone 21, which was acylated using alpha-N-Fmoc-amino acids (Phe, Trp, gamma-(t-Bu)-Glu). Formation of diazepinones 15 followed a similar protocol, after Fmoc removal with piperidine. Cleavage of the heterocycles with TFA/TES 95:5 gave the N1-p-hydroxybenzyl diazepinones 15-18 in overall isolated yields from 6 to 24% after purification in purities ranging from 81 to 100% according to LCMS analysis.  相似文献
