3-p-Toluoyl-1,2-dihydro-4H-pyrrolo-[2,1-c][1,4]benzoxazine-1,2,4-trione reacts with benzylamine to yieldZ-3-(1-benzyl-2-hydroxy-4,5-dioxo-2-p-tolyltetrahydropyrrol-3-idene)-3,4-dihydro-2H-1,4-benzoxazin-2-one. The crystal and molecular structure of the latter compound was studied by X-ray structural analysis. Thermal decarboxylation of 3-p-toluoyl-1,2-dihydro-4H-pyrrolo[5,1-c][1,4]benzoxazine-1, 2,4-trione resulted in 2-oxo-2H-1,4-benzoxazin-3-yl(p-toluoyl)ketene; the latter under-goes [4+2]-cycloaddition withp-bromobenzaldehyde to form 1-p-bromophenyl-4-p-toluoyl-1, 3-dihydro-5H-[1,3]oxazino[4,3-c][1,4] benzoxazine-3,5-dione. Reactions of 3-Z-aroylmethylene-6-chloro-3,4-dihydro-2H-1,4-benzoxazine-2-ones with oxalyl chloride afford 3-aroyl-8-chloro-1,2-dihydro-4H-pyrrolo[2,1-c][1,4]benzoxazine-1,2,4-triones and Z-3-(2-aryl-2-chlorovinyl)-6-chloro-2H-1,4-benzoxazine-2-ones. Aroyl(imidoyl)ketenes generated by decarbonylation of pyrrolobenzoxazinetriones undergo dimerization through [4+2]-cycloaddition to form 4-aroyl-3-aroyloxy-2-(2-oxo-2H-1,4-benzoxazin-3-yl)-1H, 5H-pyrido[2,1-c][1,4]benzoxazine-1,5-diones.  相似文献

Thermal decarbonylation of 3-p-toluoylpyrrolo[2,1-c][1,4]benzoxazine-1,2,4-trione affords (2-oxo-1,4-benzoxazin-3-yl)(p-toluoyl)ketene. The latter enters into the [4+2]-cycloaddition reaction withp-bromobenzylidene-p-anisidine to form 1-p-bromophenyl-2-p-methoxyphenyl-4-p-toluoyl-1,2-dihydropyrimidino[4,3-c][1,4]benzoxazine-3,5-dione. A new direction of nucleophilic attack was revealed in the reactions of 3-aroylpyrrolo[2,1-c][1,4]-benzoxazine-1,2,4-triones with o-tolylhydrazine and 2-hydrazinylbenzoic acid hydrochloride, which afforded the corresponding hydrazones at the C2=O carbonyl group, substituted 2-(2-o-tolylhydrazono)pyrrolo[2,1-c]-[1,4]benzoxazine-1,4(2H)-diones and pyrrolo[2,1-c][1,4]benzoxazin-2(4H)-ylidenehydrazinylbenzoic acids. The structure of (Z)-2-{2-[3-benzoyl-1,4-dioxo-1H-pyrrolo[2,1-c][1,4]benzoxazin-2(4H)-ylidene]hydrazinyl}-benzoic acid was determined by X-ray analysis.  相似文献

Thermolysis of 3-aroylpyrrolo[1,2-d][1,4]benzoxazine-1,2,4(4H)-triones generates aroyl(2-oxo-1,4-benzoxazin-3-yl)ketenes which react as dienes at the aroylketene fragment in [4 + 2]-cycloaddition at the C=N bond of N,N??-dicyclohexylcarbodiimide with formation of 3-[6-aryl-4-oxo-3-cyclohexyl-2-cyclohexylimino-3,4-dihydro-2H-1,3-oxazin-5-yl]-2H-1,4-benzoxazin-2-ones. Thermolysis of the latter is accompanied by elimination of N,N??-dicyclohexylcarbodiimide, and aroyl(2-oxo-1,4-benzoxazin-3-yl)ketenes thus generated undergo cyclodimerization to produce 7-aroyl-6,10-dioxo-9-(2-oxo-2H-1,4-benzoxazin-3-yl)-6,10-dihydrobenzo[b]pyrido[1,2-d][1,4]oxazin-8-yl benzoates.  相似文献

A series of substituted hexahydropyrido [2,1-c][1,4]oxazin-4(3H)-ones has been prepared by reaction of the sodio derivative of the appropriate piperidyl carbinol with ethyl chloroacetate. From a study of their NMR spectra the configurations of these compounds have been assigned and the conformation of the perhydro-1, 4-oxazinone moiety shown to be that with the ring oxygen atom and the angular hydrogen on the same side of the plane defined by C3? C(O)? N. The synthesis and NMR spectra of hexahydro-2H-pyrido [1,2-d][1,4]oxazepin-5(4H)-one, 1,6,11,11a-tetrahydro [1,4]-oxazino [4,3-b]isoquinolin-4(3H)-one and of hexahydropyrido [2,1-c][1,4]thiazin-4(3H)-one are also discussed. cis-6,9a-H-6-Methyl-hexahydropyrido[2,1-c][1,4]oxazin-4(3H)-one is shown to exist in a conformation containing a deformed perhydropyridine ring.  相似文献

Reaction of di-4-morpholinylphosphinic chloride on 5,10-dihydro-9H-pyrrolo[2,1-c][1,4]benzodiazepin-11-one 9 afforded 11-(di-4-morpholinylphosphinyloxy)-5H-pyrrolo[2,1-c][1,4]benzodiazepine 10 . Displacement of di-4-morpholinylphosphinyloxy group of 10 by formylhydrazine with concomitant intramolecular cyclization led directly to 9H-pyrrolo[2,1-c]-s-triazolo[4,3-a][1,4]benzodiazepine 7, a novel nitrogen heterocyclic ring of pharmaceutical interest.
