Lutetium texaphyrin, PCI-0123, is a pure, water-soluble photosensitizer with a large broad absorption band centered at 732 nm. The compound was tested for photodynamic therapy (PDT) effectiveness in a murine mammary cancer model. The texaphyrin macrocycle as illustrated by magnetic resonance imaging and 14 C-radiolabeled texaphyrin studies was shown to be tumor selective; a tumor-to-muscle ratio of 10.55 was seen after 5 h. Lutetium texaphyrin, at a drug dose of 20 μmol/kg with irradiation 5 h postinjection at 150 J/cm2 and 150 mW/cm2 , had significant efficacy (P < 0.0001) in treating neoplasms of moderate size (40 ± 14 mm3 ) and also had significant efficacy ( P < 0.0001) in treating larger neoplasms (147 ± 65 mm3 ). The PDT efficacy was correlated with the time interval between PCI-0123 administration and light exposure. A 100% cure rate was achieved when photoirradiation took place 3 h postinjection compared to 50% for 5 h using 10 μmol/kg and 150 J/cm2 at 150 mW/cm2 . The PDT efficacy was attributable to the selective uptakehetention of the texaphyrin photosensitizer in addition to the depth of light penetration achievable at the 732 nm laser irradiation. 相似文献

We have investigated the pharmacokinetics (PK) of Lutetium Texaphyrin (Lu-Tex), a second-generation photosensitizer, in the Syrian hamster cheek pouch early cancer model. Ten male hamsters, five with chemically induced early squamous cell cancer of the left cheek pouch, received an intracardiac injection of a 10 mg/ml Lu-Tex solution, resulting in a dose of 12 mg Lu-Tex per kg of bodyweight. The PK of the dye have been measured during the 24 h following the injection with an optical-fiber-based spectrofluorometer on the ventral skin, the healthy and the tumoral cheek-pouch mucosa. The Lu-Tex fluorescence is excited at 460 nm and detected around 740 nm. All the measurements yield very similar pharmacokinetic curves. The fluorescence intensity reaches a maximum between two and three hours after the injection and, at its maximum, it is consistently higher (up to 1.5 times) on the tumor than on the healthy mucosa. It remains smaller on the skin than on cheek-pouch mucosa. After 24 h, the Lu-Tex fluorescence is no longer detectable either on the skin, on the lesion or on the healthy mucosa. Moreover, Lu-Tex clearly displays a significant fluorescence selectivity between early carcinoma and healthy mucosa in this model. Furthermore, the inter-animal fluctuations of the fluorescence signal are small (+/-16% on the tumor-bearing mucosa). Eight-minute-long skin-irradiation tests have been performed 24 h after the injection of the Lu-Tex on the ventral skin of 16 additional animals with a solar simulator. No reaction is observed, either macroscopically or microscopically, which further demonstrates, as suggested by the fluorescence measurements, that this photosensitizer is significantly cleared from the skin after 24 h. 相似文献

Phototherapies including photodynamic therapy(PDT) and photothermal therapy(PTT) are the most promising and non-invasive cancer treatments. However, the efficacy of mono-therapy of PDT or PTT is often limited by the phototherapeutic defects such as low light penetration depth of photosensitizers and insufficiency of photothermal agents. Peroxynitrite(ONOO~-) has been proved to be an efficient oxidizing and nitrating agent that involves in various physiological and pathological processes. Therefore, ONOO~-produced in tumor site could be an effective treatment in cancer therapy. Herein, a novel cyanine dye-based(Cy7) polymer nanoplatform is developed for enhanced phototherapy by in situ producing ONOO~-. The Cy7 units in the nanoparticles can not only be served as the photosensitizer to produce reactive oxygen species(ROS) including singlet oxygen and superoxide anion for PDT, but also be used as a heat source for PTT and the release of NO gas from N-nitrosated napthalimide(NORM) at the same time. Since NO can react quickly with superoxide anion to generate ONOO~-, the enhanced phototherapy could be achieved by in situ ONOO~-produced by PCy7-NO upon exposure to the near infrared(NIR) light. Therefore, the NIRtriggered Cy7-based nanoplatform for ONOO~--enhanced phototherapy may provide a new perspective in cancer therapy. 相似文献

Gold(III) complexes have recently emerged as new versatile and efficacious metal containing anticancer agents. In an attempt to reconcile the specific affinity of such complexes for target sulfur containing biomolecules with their capability to strongly bind thiol-containing compounds widely distributed in non-tumoral cells, a new series of cyclometalated Au(III)-hydride complexes has been proposed as photoactivatable anticancer prodrugs. Here, the computational exploration of the photophysical properties and reactivity in dark and under light irradiation of the first member of the series, named 1 a , is reported. Complex 1 a low hydricity in dark together with facile hydride substitution leading to H2 elimination under excitation by visible light have been examined by means of DFT and TD-DFT computations. Both singlet and triplet excited states have been characterized, allowing the identification of the active species involved in photoactivation pathways leading to the controlled detachment of the hydride ligand. The pseudobinary systems NaCl—LnCl3 (Ln=Tm, Yb, Lu) were investigated by DTA and X-ray diffraction. Two types of ternary chlorides exist: congruently melting compounds Na3LnCl6 with the cryolite-structure, incongruently melting compounds NaLnCl4 with the NaErCl4-Ln (Ln=Tm) or the NaLnCl4-structure (Ln=Yb, Lu). Human serum albumin (HSA) is one of the key components in human blood that may influence drug distribution. As such, it is important to know the affinity of any drug for albumin. Previously, Photofrina mixture of monomeric, dimeric and oligomeric porphyrins, has been subjected to HSA binding studies. However, due to its complex nature, binding studies on Photofrin or other hematoporphyrin derivatives with HSA are inconclusive. In this report, the binding properties of some components (dimers and trimers) of Photofrin® and the relationship between murine photosensitizing efficacy and those binding properties were investigated. The interaction of these porphyrins with HSA was investigated by direct ultrafiltration and fluorescent titration techniques with fluorescent probes such as dansyl-L-proline (DP), which is known to interact selectively with site II on HSA. Porphyrins also were tested for antitumor activity in a mouse model following intravenous administration and exposure to laser light. Together, the results suggest that the photosensitizers that were preferentially bound to site II of HSA were most effective at controlling murine tumor regrowth 相似文献

The spin lattice model for the spin-gapped layered magnetic solids Na3Cu2SbO6 and Na2Cu2TeO6 was examined by evaluating the three spin exchange interactions of their Cu2MO6 (M = Sb, Te) layers in terms of spin dimer analysis based on extended Hückel tight binding calculations and mapping analysis based on first principles density functional theory electronic band structure calculations. For both compounds, our calculations show that the two strongest spin exchange interactions, that is, the Cu-O...O-Cu super-superexchange (J2) and the Cu-O-Cu superexchange (J1) interactions, form alternating chains that interact weakly through the Cu-O-Cu superexchange (J3) interactions. The dominant one of the three spin exchange interactions is J2, and it is antiferromagnetic in agreement with the fact that both of the compounds are spin gapped. For Na3Cu2SbO6 and Na2Cu2TeO6, the superexchange J1 is calculated to be ferromagnetic, hence, leading to the alternating chain model in which antiferromagnetic and ferromagnetic spin exchange interactions alternate. This picture does not agree with the recent experimental analysis, which showed that the temperature-dependent magnetic susceptibilities of both compounds should be described by the alternating chain model in which two antiferromagnetic spin exchange interactions of different strengths alternate. 相似文献

The biodistribution and excretion of temoporfin (tetra[ m -hydroxyphenyl]chlorin, m -THPC), a recently developed photosensitizer, was investigated in BALB/c mice. [14 C]temoporfin was administered intravenously (0.73 μmol/kg) to tumor-free mice or to mice implanted with the Colo 26 colorectal carcinoma. Blood, tissue and fecal samples were collected for 35 days and 10 days postdose from tumor-free mice and tumor-bearing mice, respectively. Blood concentrations fell rapidly such that at later time points they were indistinguishable from background counts. Tumor concentrations rose to a peak of 0.34 μg temoporfin equivalents/mL at 2 days and then declined in parallel (log plot) with the blood concentrations. Tumor : tissue ratios at 2 days for skin, adipose tissue and skeletal muscle underlying the tumor were 1.5, 2.3 and 3.8, respectively. By 4 days the corresponding values were 1.6, 3.4 and 4.0. Nearly 40% of the administered radioactivity was excreted in the feces in the first 24 h and more than 80% had been excreted by 20 days. Less than 0.2 % of the dose was recovered from the urine. An elimination half-life of 10–12 days was calculated from the excretion data. 相似文献

The conformations of the exocyclic group of N(6)-acetyl- and N(6)-carbamoylsydnoneimines and their salts were studied by means of their vibrational spectra. It is shown that the side chain in compounds that do not contain a bulky substituent in the 4 position of the sydnoneimine ring has primarily a planar syn-s-cis form. The synthesis of Me(6)Te in 1990 stimulated the exploration of hexamethylchalcogen potential energy surfaces. This earlier ab initio work focused only on the D(3) conformers, but it has been noted that the pseudooctahedral X(CH(3))(6) compounds show either D(3) or S(6) symmetry. Here are reported the results of an ab initio molecular orbital study of the hexamethylchalcogens confined to S(6) symmetry. Stationary points were found for each of the three hexamethylchalcogens studied and were shown to be minima for the two larger hexamethylchalcogens. Each of the S(6) stationary points found was energetically higher lying than the earlier reported D(3) counterpart. These energy differences are discussed in terms of nuclear repulsion and molecular orbital bonding considerations. 相似文献

A metal-boron mixed cluster LiB(6) (-) was produced and characterized by photoelectron spectroscopy and ab initio calculations. A number of electronic transitions were observed and used to compare with theoretical calculations. An extensive search for the global minimum of LiB(6) (-) was carried out via an ab initio genetic algorithm technique. The pyramidal C(2v) ((1)A(1)) molecule was found to be the most stable at all levels of theory. The nearest low-lying isomer was found to be a triplet C(2) ((3)B) structure, 9.2 kcal/mol higher in energy. Comparison of calculated detachment transitions from LiB(6) (-) and the experimental photoelectron spectra confirmed the C(2v) pyramidal global minimum structure. Natural population calculation revealed that LiB(6) (-) is a charge-transfer complex, Li(+)B(6) (2-), in which Li(+) and B(6) (2-) interact in a primarily ionic manner. Analyses of the molecular orbitals and chemical bonding of B(6) (2-) showed that the planar cluster is twofold (pi- and sigma-) antiaromatic, which can be viewed as the fusion of two aromatic B(3) (-) units. 相似文献

The 3-hydroxyquinone derivative of the non-psychotrophic phytocannabinoid cannabigerol, so-called VCE-003.2, and some other derivatives have been recently investigated for neuroprotective properties in experimental models of Parkinson’s disease (PD) in mice. The pharmacological effects in those models were related to the activity on the peroxisome proliferator-activated receptor-γ (PPAR-γ) and possibly other pathways. In the present study, we investigated VCE-004.8 (formulated as EHP-101 for oral administration), the 3-hydroxyquinone derivative of cannabidiol (CBD), with agonist activity at the cannabinoid receptor type-2 (CB2) receptor in addition to its activity at the PPAR-γ receptor. Studies were conducted in both in vivo (lesioned-mice) and in vitro (SH-SY5Y cells) models using the classic parkinsonian neurotoxin 6-hydroxydopamine (6-OHDA). Our data confirmed that the treatment with VCE-004.8 partially reduced the loss of tyrosine hydroxylase (TH)-positive neurons measured in the substantia nigra of 6-OHDA-lesioned mice, in parallel with an almost complete reversal of the astroglial (GFAP) and microglial (CD68) reactivity occurring in this structure. Such neuroprotective effects attenuated the motor deficiencies shown by 6-OHDA-lesioned mice in the cylinder rearing test, but not in the pole test. Next, we explored the mechanism involved in the beneficial effect of VCE-004.8 in vivo, by analyzing cell survival in cultured SH-SY5Y cells exposed to 6-OHDA. We found an important cytoprotective effect of VCE-004.8 at a concentration of 10 µM, which was completely reversed by the addition of antagonists, T0070907 and SR144528, aimed at blocking PPAR-γ and CB2 receptors, respectively. The treatment with T0070907 alone only caused a partial reversal, whereas SR144528 alone had no effect, indicating a major contribution of PPAR-γ receptors in the cytoprotective effect of VCE-004.8 at 10 µM. In summary, our data confirmed the neuroprotective potential of VCE-004.8 in 6-OHDA-lesioned mice, and in vitro studies confirmed a greater relevance for PPAR-γ receptors rather than CB2 receptors in these effects. 相似文献

Photoelectron spectroscopy was combined with ab initio calculations to elucidate the structure and bonding in Si6 2- and NaSi6 -. Well-resolved electronic transitions were observed in the photoelectron spectra of Si6 - and NaSi6 - at three photon energies (355, 266, and 193 nm). The spectra of NaSi6 - were observed to be similar to those of Si6 - except that the electron binding energies of the former are lower, suggesting that the Si6 motif in NaSi6 - is structurally and electronically similar to that in Si6 -. The electron affinities of Si6 and NaSi6 were measured fairly accurately to be 2.23+/-0.03 eV and 1.80+/-0.05 eV, respectively. Global minimum structure searches for Si6 2- and NaSi6 - were performed using gradient embedded genetic algorithm followed by B3LYP, MP2, and CCSDT calculations. Vertical electron detachment energies were calculated for the lowest Si6 - and NaSi6 - structures at the CCSD(T)/6-311+G(2df), ROVGF/6-311+G(2df), UOVGF/6-311+G(2d), and time-dependent B3LYP/6-311+G(2df) levels of theory. Experimental vertical detachment energies were used to verify the global minimum structure for NaSi6 -. Though the octahedral Si6 2-, analogous to the closo form of borane B6H6 2-, is the most stable form for the bare hexasilicon dianion, it is not the kernel for the NaSi6 - global minimum. The most stable isomer of NaSi6 - is based on a Si6 2- motif, which is distorted into C2v symmetry similar to the ground state structure of Si6 -. The octahedral Si6 2- coordinated by a Na+ is a low-lying isomer and was also observed experimentally. The chemical bonding in Si6 2- and NaSi6 - was understood using natural bond orbital, molecular orbital, and electron localization function analyses. 相似文献
