From the unripe fruits of Lo Han Kuo (Siraitia grosvenori), a Chinese medicinal plant, two new cucurbitane triterpene glycosides, 20-hydroxy-11-oxomogroside IA(1) (1) and 11-oxomogroside IIE (2), were isolated along with five known cucurbitane glycosides, 11-oxomogroside IA(1) (3), mogroside IIE (4), mogroside III (5), mogroside IVA (6), and mogroside V (7), and two flavonoid glycosides, kaempferol 7-O-alpha-L-rhamnopyranoside (8) and kaempferol 3,7-di-O-alpha-L-rhamnopyranoside (9). Their structures were determined on the basis of detailed analyses of 1D, 2D-NMR spectroscopic methods and by comparing with literature values. This paper describes the first investigation of unripe bitter Lo Han Kuo fruits.  相似文献

建立了高效液相色谱测定罗汉果中罗汉果苷Ⅴ、罗汉果苷ⅣA、罗汉果苷Ⅲ、11-氧化罗汉果苷Ⅲ、罗汉果苷ⅡE和11-氧化罗汉果苷ⅡE等6种苷类化合物含量的方法。采用的色谱条件:ZORBAX SB-C18色谱柱(150 mm×4.6 mm, 5 μm);柱温30 ℃;以水(A相)和乙腈(B相)为流动相,梯度洗脱程序:0~3 min,20%B~30%B;3~8 min,30%B~35%B;8~9 min,35%B;流速0.8 mL/min;检测波长203 nm;进样量10 μL。结果表明,以上6种苷类化合物分别在0.04~1.0 mg/mL、0.011~0.68 mg/mL、0.010~0.80 mg/mL、0.0097~0.58 mg/mL、0.025~1.0 mg/mL、0.013~0.76 mg/mL范围内线性关系良好(r>0.9991)。加标回收率依次为99.65%,101.6%,97.05%,103.1%,99.25%和103.0%;相对标准偏差分别为0.83%,3.1%,1.9%,3.3%,0.59%和2.0%。该方法简便、快速、准确,适用于罗汉果原材料及其制品中苷类化合物的检测和含量测定。  相似文献

The chemical compositions of the essential oils from the peel of ripe and unripe fruits of Hymenaea courbaril L., obtained by hydrodistillation, were analyzed by GC and GC-MS. The main constituents of the essential oil from the peel of the ripe fruits were the sesquiterpenes alpha-copaene (11.1%), spathulenol (10.1%) and beta-selinene (8.2%), while germacrene-D (31.9%), beta-caryophyllene (27.1%) and bicyclogermacrene (6.5%) were the major compounds in the oil from unripe fruits. The essential oils were tested against Aedes aegypti larvae and showed LC50 values of 14.8 +/- 0.4 microg/mL and 28.4 +/- 0.3 microg/mL for the ripe and unripe fruit peel oils, respectively. From the peel of the ripe fruits, the diterpenes zanzibaric acid and isoozic acid were isolated, along with the sesquiterpene caryolane-1,9beta-diol. To the best of our knowledge, this is the first report of this sesquiterpene in the genus. The structures of all compounds isolated were identified on the basis of their spectral data (IR, MS, 1D- and 2D-NMR) and by comparison with literature spectral data.  相似文献

The essential oils of the fruits and the leaves of pistachio (Pistacia vera L.) were analyzed by GC and GC/MS. Fresh unripe pistachio fruits were richer in essential oil (0.5%, w/w) than the leaves (0.1%, w/w). Twenty one compounds were identified in the essential oil of the fruits and the major components were (+)-alpha-pinene (54.6%) and terpinolene (31.2%). The enantiomeric ratio of the major constituents of the essential oil of the fruits was determined using chiral GC/MS and it was found that the (+)/(-)-alpha-pinene ratio was 99.5:0.5, (+)/(-)-limonene 80:20, (+)/(-)-beta-pinene 96:4, and (+)/(-)-alpha-terpineol 0:100. Thirty three compounds were identified in the essential oil of the leaves and the major components were found to be alpha-pinene (30.0%), terpinolene (17.6%) and bornyl acetate (11.3%).  相似文献

Herein, we systematically investigate the origin of astringent mouthfeel when we eat unripe fruits, drink coffee or tea, from the perspective of lubrication by simulating the dynamic weak interaction on the tongue with model protein (mucoprotein, MP) and polyphenolic compounds (tannic acid, TA). Astringency was due to the protein‐mediated lubrication failure when encountering polyphenolic molecules that normally exist, for example in unripe fruits, coffee, tea. The underlying molecular mechanism of oral tribology is widely present in nature and enables us to engineer a tongue‐like polyacrylamide composite hydrogel that exhibits high TA sensitivity and to develop a scientific strategy for catching slippery fish using TA‐containing gloves. These results provide novel and useful insights into the failure of biological boundary lubrication on soft tissue surface with the adsorbed proteins.  相似文献

In this study, the total phenolic contents and total antioxidant capacities of some commercial bitter melon products (powder, packaged powder, capsule, paste in olive oil), and of unripe and ripe fruits were determined by spectrophotometric and chromatographic methods. The total antioxidant capacities of unripe and ripe bitter melon samples, determined by using the CUPRAC (cupric reducing antioxidant capacity assay) and ABTS (2,2′-azino-bis(3-ethylbenzthiazolin-6-sulfonic acid))/HRP (horseradish peroxidase) methods, were 42.5 and 36.3 µmol TRE (Trolox equivalent) g–1, and 8.7 and 7.0 µmol TRE g–1, respectively. The TAC (total antioxidant capacity) order of the studied samples using the same 2 methods were determined as follows: capsule (CUPRAC value, 140.8; ABTS/HRP value, 143.6 µmol TRE g–1) > packaged powder (129.6; 126.1) > powder (52.3; 64.3) > unripe fruit (42.5; 36.3) > paste in olive oil (17.6; 14.4) > ripe fruit (8.7; 7.0). The order of phenolic content was found as follows: unripe fruit (193.2 µmol GAE (gallic acid equivalent) g-1) > capsule (162.0) > packaged powder (160.6) > powder (83.6) > paste in olive oil (38.3) > ripe fruit (14.6).  相似文献

This study aimed to evaluate the polyphenolic composition along with the biological activity of guabiroba (Campomanesia xanthocarpa Berg.) fruits using comprehensive two-dimensional liquid chromatography (LC × LC). A simplex centroid design comprising three solvents (methanol, 2% acetic acid, and acetonitrile) was used to optimize the extraction mixture for polyphenols from ripe and unripe guabiroba fruits. A quantitative LC × LC platform was proposed to characterize the guabiroba extracts using a RP-Amide column and a C18 column in the first and second dimensions, respectively. Antidiabetic properties, using in vitro enzyme assay models and in vivo antioxidant activity with the eukaryote model Saccharomyces cerevisiae, was measured. Total phenolics compounds were more efficiently extracted with 2% acetic acid solution and acetonitrile (50:50, v/v). A total of 37 different compounds were identified and quantified using the proposed LC × LC method (linearity ranging from 0.9990 to 0.9994, intra- and interday precision from 0.40 to 10.57% and, accuracy from 81.89 to 108.98%). Significant differences were observed between ripe and unripe guabiroba fruits, especially for the compounds geraldone and methyl galangin isomer. Guabiroba fruits showed significant antidiabetic and antioxidant properties and may be potentially adopted as part of dietary strategies in the management of early stages of type 2 diabetes and associated complications.  相似文献

Fruits of Bromelia hieronymi, a tropical South American plant, possess a high content of peptidases with potential biotechnological uses. Total RNA was extracted from unripe fruits and peptidase cDNA was obtained by 3'RACE-PCR. The consensus sequence of the cysteine peptidase cDNA contained 875 bp, the 690 first ones codifying for a hypothetical polypeptide chain of the mature peptidase, named Bh-CP1 (molecular mass 24.773 kDa, pI 8.6, extinction molar coefficient 58,705 M(-1) cm(-1)). Bh-CP1 sequence shows a high percentage of identity with those of other cysteine plant proteases. The presence of highly preserved residues is observed, like those forming the catalytic site (Gln19, Cys25, His159, and Asn175, papain numbering), as well as other six Cys residues, involved in the formation of disulfide bounds. Molecular modeling results suggest the enzyme belongs to the α?+?β class of proteins, with two disulfide bridges (Cys23-Cys63 and Cys57-Cys96) in the α domain, while the β domain is stabilized by another disulfide bridge (Cys153-Cys203). Additionally, peptide mass fingerprints (PMFs) of the three peptidases previously isolated from B. hieronymi fruits (namely hieronymain I, II, and III) were performed and compared with the theoretical fingerprint of PMF of Bh-CP1, showing a partial matching between the in silico-translated protein and hieronymain II.  相似文献

Degradative and other data described below have shown that the tannin of unripe fruits of persimmon (Diospyros virginiana) actually contains (+)-gallocatechin unit in the structure and must be regarded as condensed flavonoid pigments, probably of similar structure to the polymeric proanthocyanidin from avocado seed. The possible structure of the proanthocyanidin from persimmon fruits is proposed tentatively as a polymer of 5, 7, 3′, 4′, 5′-pentahydroxyflavanonol.  相似文献

Streptococcus zoonotic bacteria cause serious problems in aquaculture with clinical effects on humans. A structure-antibacterial activity relationships analysis of 22 isoflavones isolated from M. tricuspidata (leaves, ripe fruits, and unripe fruits) against S. iniae revealed that prenylation of the isoflavone skeleton was an important key for their antibacterial activities (minimum inhibitory concentrations: 1.95–500 μg/mL). Through principal component analysis, characteristic prenylated isoflavones such as 6,8-diprenlygenistein (4) were identified as pivotal compounds that largely determine each part’s antibacterial activities. M. tiricuspidata ripe fruits (MTF), which showed the highest antibacterial activity among the parts tested, were optimized for high antibacterial activity and low cytotoxicity on fathead minnow cells using Box–Behnken design. Optimized extraction conditions were deduced to be 50%/80 °C/7.5 h for ethanol concentration/extraction temperature/time, and OE-MTF showed contents of 6,8-diprenlygenistein (4), 2.09% with a MIC of 40 µg/mL. These results suggest that OE-MTF and its active isoflavones have promising potential as eco-friendly antibacterial agents against streptococcosis in aquaculture.  相似文献

Systematic phytochemical studies of the extract of Luo Han Go (Siraitia grosvenorii) furnished six known cucurbitane glycosides, namely, mogrosides IVa, mogrosides V and VI, isomogroside V, 11-oxomogroside V, and siamenoside I. The structures of all the isolated compounds were characterized on the basis of extensive spectral and chemical studies. Though the partial NMR spectral data has been reported for some of the isolated compounds previously, the complete 1H and 13C NMR spectral assignments of all the isolated compounds are reported herewith for the first time based on COSY, HSQC, HMBC, and NOESY spectroscopic data.  相似文献

A phytochemical study of the CHCl3 extract of the dried unripe fruits of Evodia rutaecarpa (Juss. ) Benth. (Rutaceae) resulted in the isolation of three new alkaloids, evollionines A–C ( 1 – 3 , resp.), together with two known compounds, evodianinine and wuzhuyumide I. The structures of the new compounds were elucidated on the basis of detailed spectroscopic evidence and confirmed in the case of compound 1 by single‐crystal X‐ray analysis.  相似文献

Abstract

In Brazilian folk medicine, Protium heptaphyllum is used to treat inflammatory conditions and to hasten wound repair. This paper aims to investigate the chemical composition and the in vitro antibacterial effects of the essential oils (EOs) obtained from P. heptaphyllum leaves and ripe and unripe fruits against a representative panel of oral pathogens. The GC-FID and GC-MS analysis revealed that the major components determined in P. heptaphyllum essential oils were myrcene (59.0%), β-elemene (17.2%), limonene (12.9%), spathulenol (12.6%), α-cubebene (11.6%), germacrene D (10.6%), trans-nerolidol (9.8%), and α-cadinol (8.8%). The essential oils of the ripe and unripe fruits showed the strongest antibacterial activity against the anaerobic bacteria Prevotella nigrescens (MIC?=?50?µg/mL). The leaf essential oil displayed very promising activity against Streptococcus mutans (MIC?=?50?µg/mL) and Streptococcus mitis (MIC?=?62.5?µg/mL). The antibacterial activity of EOs against oral pathogens is also described for the first time.

Triterpenoids and steroids are considered to be important for the fruit quality and health-promoting properties for the consumers. The aim of the study was the determination of the changes in triterpenoid and steroid biosynthesis and the accumulation in hypanthium and achenes of rugosa rose (Rosa rugosa Thunb.) hip during fruit development and ripening at three different phenological stages (young fruits, fully developed unripe fruits, and matured fruits). Triterpenoids and steroids were also determined in the peel and the pulp of the matured hips. The obtained results indicated that the distribution of the analyzed compounds in different fruit tissues is a selective process. The increased rate of hydroxylation of triterpenoids, the deposition of hydroxylated acids in fruit surface layer, and the continuous biosynthesis of phytosterols in achenes versus its gradual repression in hypanthium accompanied by the accumulation of their biosynthetic intermediates and ketone derivatives seem to be characteristic metabolic features of maturation of rugosa rose accessory fruit. These observations, apart from providing the important data on metabolic modifications occurring in developing fruits, might have a practical application in defining fruit parts, particularly rich in bioactive constituents, to enable the development of novel functional products.  相似文献

The essential oils from the aerial parts of Pimpinella anagodendron Bolle and Pimpinella rupicola Svent., two endemic species growing in Tenerife, Canary Islands, Spain, were studied by gas chromatography and gas chromatography-mass spectrometry. The major components of the flowering tops (flowers+unripe fruits) of P. rupicola (PRFT) were found to be beta-bisabolene (34.8%), limonene (10.9%) and alpha-zingiberene (10.5%), whereas in the flowering tops of P. anagodendron (PAFT), the main constituents were alpha-zingiberene (32.9%), beta-bisabolene (17.9%), beta-pinene (15.8%) and ar-curcumene (11.5%). The major compounds found in the stems+leaves of P. rupicola (PRSL) were beta-bisabolene (31.6%), alpha-zingiberene (11.4%) and limonene (10.8%), whereas those of P. anagodendron (PASL) were alpha-zingiberene (32.3%), beta-bisabolene (14.0%) and ar-curcumene (12.6%). In all the oils were found the characteristic constituents of genus Pimpinella, the pseudoisoeugenol esters. In accordance with the morphological, chorological and chemical differences between both species, we suggest that P. rupicola Svent. is a good taxon and not a synonym of P. anagodendron.  相似文献

The flavonoid and furocoumarin composition was investigated of peel and pulp tissues of unripe fruits of Citrus myrtifolia Rafinesque, an ingredient of the popular soft drink "chinotto". Compound separation and identification was made using an HPLC-PDA detector coupled to ESI/MS/MS in positive and negative mode. Eighteen compounds (3-hydroxy-3-methylglutaryl-, C- and O-glycosyl flavonoids, furocoumarins and polymethoxylated flavones) were identified and quantified. Data indicated that the overall amount of flavonoids and furocoumarins in peel was higher than in the pulp, even though their relative distribution did not significantly change, apart from a different distribution of flavones and a lower content of naringin in the peel.  相似文献

The compositions of the essential oils from unripe (UFr) and ripe (RFr) fruits of Schinus terebinthifolius were analyzed by GC and GC/MS. The major compound identified in both oils was limonene (UFr = 44.1 +/- 1.3%; RFr = 31.8 +/- 1.2%), followed by alpha-phellandrene (15.7 +/- 0.4%) in the UFr oil and thujene (21.7 +/- 0.9%) in the RFr oil. Repellent and toxicity activities of the two oils were also evaluated, and the results compared with eugenol. The UFr oil was more active in the fumigation tests (LC50 = 1.46 microL/L of air), whereas the RFr oil was more active in the sealed dishes (SD) than open dishes (OD) contact assay (LC50 = 3.04 microL/cm2) and not significantly different from eugenol. Both oils exhibited significant repellent activity comparable with that of eugenol. The results suggest that the repellent activity of these oils in association with its toxicity could be a great advantage for the integrated management of T. urticae.  相似文献

The current study was designed to find out how olive maturity indices (2.5, 3.5, and 4.5) affect the individual phenolic compounds and antioxidant potencies of olive oils produced from cv. Nizip Yaglik olives. Liquid chromatography coupled to diode array detection and electrospray ionization tandem mass spectrometry in multiple reaction monitoring mode was utilized for the determination of phenolic composition qualitatively and quantitatively. Findings asserted a quite similar phenolic profile (14 phenols) depending on the various phenolic groups in all oils, while the concentration of total and individual phenolic compounds revealed significant differences between the samples statistically (p < 0.05). Among the individual phenolic classes in all samples, secoiridoids were the most prevailing group and their total content showed a clear significant decline as the olive fruits get ripened. Antioxidant potency values showed a clear diminution attitude during the maturation of the olives. The principal component analysis revealed that oils were discriminated from each other according to phenolic compounds and antioxidant potencies. Moreover, oils obtained from the unripe and medium‐ripe fruits possessed a very good quality marked by their elevated phenolic levels.  相似文献

Eight extraction agents (water, methanol–water mixtures in various ratios, methanol, a 20 mmol l?1 ammonium phosphate buffer, and a methanol–phosphate buffer) were tested for the extraction of arsenic compounds from fruits, stems + leaves, and roots of pepper plants grown on soil containing 17.2 mg kg?1 of total arsenic. The arsenic compounds in the extracts were determined using high‐performance liquid chromatography–hydride generation inductively coupled plasma mass spectrometry. Whereas pure water was the most effective extraction agent for fruits (87 ± 3.3% extraction yield) and roots (96 ± 0.6% extraction yield), the 20 mM ammonium phosphate buffer at pH 6 extracted about 50% of the arsenic from stems + leaves. Decreasing extractability of the arsenic compounds was observed with increasing methanol concentrations for all parts of the pepper plant. In pepper fruits, arsenic(III), arsenic(V), and dimethylarsinic acid (DMA) were present (25%, 37%, and 39% respectively of the extractable arsenic). Arsenic(V) was the major compound in stems + leaves and roots (63% and 53% respectively), followed by arsenic(III) representing 33% and 42% respectively, and small amounts (not exceeding 5%) of DMA and methylarsonic acid were also detected. This study is designed to examine the chemical composition and antioxidant activity of the essential oil of Juniperus phoenicea L. ripe and unripe berries. GC and GC/MS analyses resulted in the detection of 42 components representing approximately 96.50-99.57% of the oils. Major components of the oils were α-pinene (58.61-77.39%), camphene (0.67-9.31%), δ-3-carene (0-10.01%) and trans-verbenol (0-5.24%). Antioxidant activities were determined by two different test systems, DPPH and ABTS radical scavenging activities. In both systems ripe berries exhibited better activity potential than the unripe ones.  相似文献
