The photodynamic effects of the cationic TMPyP (meso‐tetrakis [N‐methyl‐4‐pyridyl]porphyrin) and the anionic TPPS4 (meso‐tetrakis[4‐sulfonatophenyl]porphyrin) against PC/CL phosphatidylcholine/cardiolipin (85/15%) membranes were probed to address the influence of phorphyrin binding on lipid damage. Electronic absorption spectroscopy and zeta potential measurements demonstrated that only TMPyP binds to PC/CL large unilamellar vesicles (LUVs). The photodamage after irradiation with visible light was analyzed by dosages of lipid peroxides (LOOH) and thiobarbituric reactive substance and by a contrast phase image of the giant unilamellar vesicles (GUVs). Damage to LUVs and GUVs promoted by TMPyP and TPPS4 were qualitatively and quantitatively different. The cationic porphyrin promoted damage more extensive and faster. The increase in LOOH was higher in the presence of D2O, and was impaired by sodium azide and sorbic acid. The effect of D2O was higher for TPPS4 as the photosensitizer. The use of DCFH demonstrated that liposomes prevent the photobleaching of TMPyP. The results are consistent with a more stable TMPyP that generates long‐lived singlet oxygen preferentially partitioned in the bilayer. Conversely, TPPS4 generates singlet oxygen in the bulk whose lifetime is increased in D2O. Therefore, the affinity of the porphyrin to the membrane modulates the rate, type and degree of lipid damage.  相似文献

Steady-state absorption and emission spectroscopy and time-resolved fluorescence measurements were employed in the study of meso-tetrakis(4-N-methylpyridinium)porphine (TMPyP) interactions with half-generation carboxyl-terminated poly(amidoamine) (PAMAM) dendrimers in water. TMPyP experiences a less polar environment and a strong fluorescence quenching effect upon dendrimer association. The tertiary amine functional groups in PAMAM dendrimers are likely to be responsible for the fluorescence quenching of TMPyP through an electron-transfer mechanism. The Stern-Volmer plots achieve a plateau at high dendrimer concentrations that was attributed to full porphyrin-dendrimer association, and an average fluorescence quantum yield of 15-20% relative to aqueous TMPyP was estimated. The association constant for the 1:1 complex with generation 2.5 at dendrimer-porphyrin ratio D/P = 1 is 5.75 x 10(7) M(-1), indicating a strong binding affinity. The dissociation of the complex with increasing ionic strength reinforces the role of electrostatic forces in porphyrin-dendrimer association. Comparison of Stern-Volmer plots obtained from quantum yields or lifetimes showed the importance of a static effect in these systems. The fluorescence decays of the porphyrin-dendrimer complex were fitted with a dispersed kinetics model. At intermediate dendrimer-porphyrin ratios (D/P approximately 1), diffusional quenching processes between free porphyrin and dendrimer were modeled with the Sano-Tachiya pair survival probability equation. Transient diffusional effects were dismissed as a possible explanation for the static effect detected.  相似文献

We report the formation of host-guest complexes between water-soluble calix[n]arene-p-tetrasulfonates (n = 4, 6, 8) or 2-hydroxypropyl-cyclodextrins (alpha-, beta-, gamma-) and the tetratosylate salt of 5,10,15,20-tetrakis(4-N-methylpyridyl)porphyrin (TMPyP). The binding constants ranging between 10(2) and 10(5) M-1 were calculated from the absorption and fluorescence changes. Calix[4]arene-p-tetrasulfonate has a high binding affinity and forms with TMPyP a 1:1 complex, whereas other calixarenes bind two molecules of TMPyP. Electrostatic attraction is the dominating binding mode. Binding to calixarenes leads to a considerable decrease of the quantum yields of the triplet and excited singlet states and to shortening of the singlet and triplet lifetimes of TMPyP. The quenching mechanism is attributed to electron transfer between calixarene phenolates and excited TMPyP. Photoinduced electron transfer within a novel supramolecular complex calixarene/TMPyP (electron donor)/methyl viologen (electron acceptor) has been proven by absorption and fluorescence measurements. Electrostatic attraction between the cationic donor and cationic acceptor, on the one hand, and the anionic host, on the other, overcomes the electrostatic repulsion forces. In contrast, the interaction of cyclodextrin with TMPyP is hydrophobic in nature and only slightly influences the photophysical properties of TMPyP. The different behavior of TMPyP bound to either of the hosts has been assigned to the specific effects of the dominant binding modes, viz. the electrostatic attraction for calixarenes and the hydrophobic interactions for inclusion complexes with cyclodextrins.  相似文献

The luminescence behaviors of Cu(II) meso-tetrakis (4-N-methyl-pyridiniumyl) porphyrin (Cu(II)TMPyP) and Co(II) meso-tetrakis (4-N-methyl-pyridiniumyl) porphyrin (Co(II)TMPyP) are investigated, and their interactions with calf thymus DNA (ctDNA) are studied by UV-vis, fluorescence and resonance light scattering (RLS) and based on the changes of UV-vis spectra, fluorescence and RLS spectra, the intrinsic binding constants of Cu(II)TMPyP/Co(II)TMPyP with ctDNA are obtained in the case of phosphate buffer solution (pH 7.2), respectively. According to the experimental results, it can be inferred that the interaction model of Cu(II)TMPyP with ctDNA is intercalative binding, while Co(II)TMPyP is the long-range assembly on the molecular surface of ctDNA.  相似文献

The interaction between positively charged porphyrin TMPyP(4) (tetrakis‐(1‐methyl‐4‐pyridiniyl) porphyrin) and negatively charged surface of colloidal CdS nanoparticles has been studied by absorption spectrum, fluorescence spectroscopy, and time‐correlated single‐photon‐counting methods. Addition of CdS colloid to a TMPyP(4) solution leads to TMPyP(4) adsorption onto the surface of CdS colloid with an apparent association constant of 4643 L/mol. This adsorption results in distinct changes in absorption spectrum of TMPyP (4) and the quenching of its′ fluorescence, but the biexponential fluorescence decay changes hardly. Nearly 90% of the fluorescence of 5 × 10?6 mol/L TMPyP(4) can be quenched with 1.5 × 10?4 mol/L CdS colloid. These spectral changes are attributed to the formation of a certain form complex TMPyP(4)‐CdS.  相似文献

Fluorescence quenching of meso-tetrakis-4-sulfonatophenyl (TPPS(4)) and meso-tetrakis-4-N-methylpyridil (TMPyP) porphyrins is studied in aqueous solution and upon addition of micelles of sodium dodecylsulfate (SDS), cetyltrimethylammonium chloride (CTAC), N-hexadecyl-N,N-dimethyl-3-ammonio-1-propanesulfonate (HPS) and t-octylphenoxypolyethoxyethanol (Triton X-100). Potassium iodide (KI) was used as quencher. Steady-state Stern-Volmer plots were best fitted by a quadratic equation, including dynamic (K(D)) and static (K(S)) quenching. K(S) was significantly smaller than K(D). Frequency-domain fluorescence lifetimes allowed estimating bimolecular quenching constants, k(q). At 25 degrees C, in aqueous solution, TMPyP shows k(q) values a factor of 2-3 higher than the diffusional limit. TPPS(4) shows collisional quenching with pH dependent k(q) values. For TMPyP quenching results are consistent with reported binding constants: a significant reduction of quenching takes place for SDS, a moderate reduction is observed for HPS and almost no change is seen for Triton X-100. Similar data were obtained at 50 degrees C. For CTAC-TPPS(4) system an enhancement of quenching was observed as compared to pure buffer. This is probably associated to accumulation of iodide at the cationic micellar interface. The attraction between CTAC headgroups and I(-), and repulsion between SDS and I(-), enhances and reduces the fluorescence quenching, respectively, of porphyrins located at the micellar interface. The small quenching of TPPS(4) in Triton X-100 is consistent with strong binding as reported in the literature.  相似文献

The spectroscopic properties of the self-associated complexes formed between the anionic surface docking site of spinach plastocyanin and the cationic metalloporphyrins, in which the tyrosine 83 (Y83) moiety is placed just below the docking site, tetrakis(N-methyl-4-pyridyl)porphyrin (Pd(II)TMPyP(4+) and Zn(II)TMPyP(4+)), have been studied and reported herein. The fluorescence quenching phenomenon of the self-assembled complex of Zn(II)TMPyP(4+)/plastocyanin has also been discovered. The observed red-shifting of the Soret and Q-bands of the UV-visible spectra, ca. 9 nm for Pd(II)TMPyP(4+)/plastocyanin and ca. 6 nm for the Zn(II)TMPyP(4+)/plastocyanin complexes, was explained in terms of exciton theory coupled with the Gouterman model. Thus, the hydroxyphenyl terminus of the Y83 residue of the self-associated plastocyanin/cationic porphyrin complexes was implicated in the charge-transfer ligation with the central metal atoms of these metalloporphyrins. Moreover, ground-state spectrometric-binding studies between Pd(II)TMPyP(4+) and the Y83 mutant plastocyanin (Y83F-PC) system proved that Y83 moiety of plastocyanin played a critical role in the formation of such ion-pair complexes. Difference absorption spectra and the Job's plots showed that the electrostatic attractions between the cationic porphyrins and the anionic patch of plastocyanin, bearing the nearby Y83 residue, led to the predominant formation of a self-associated 1:1 complex in the ground-state with significantly high binding constants (K = (8.0 +/- 1.1) x 10(5) M(-1) and (2.7 +/- 0.8) x 10(6) M(-1) for Pd(II)TMPyP(4+) and zinc variant, respectively) in low ionic strength buffer, 1 mM KCl and 1 mM phosphate buffer (pH 7.4). Molecular modeling calculations supported the formation of a 1:1 self-associated complex between the porphyrin and plastocyanin with an average distance of ca. 9 A between the centers of mass of the porphyrin and Y83 positioned just behind the anionic surface docking site on the protein surface. The photoexcited singlet state of Zn(II)TMPyP(4+) was quenched by the Y83 residue of the self-associated plastocyanin in a static mechanism as evidenced by steady-state and time-resolved fluorescence experiments. Even when all the porphyrin was complexed (more than 97%), significant residual fluorescence from the complex was observed such that the amplitude of quenching of the singlet state of uncomplexed species was enormously obscured.  相似文献

Aggregation behavior of water soluble porphyrins, 5-(1-(4-carboxybutyl) pyridinum-4-yl) 10,15,20-tris (1-methylpyridinium-4-yl) porphyrin (5-CBPyP) in the presence of various concentrations of calf thymus DNA (ct-DNA) and sodium chloride were studied in comparison with meso-tetrakis (4-N-methyl pyridinum) porphyrin (TMPyP), by optical absorption, fluorescence and resonance light scattering (RLS) spectroscopies. Both porphyrins obey Beer’s law in extended range of concentration. Optical absorption and RLS measurements demonstrated nonaggregation for both porphyrins under increasing concentration of ct-DNA and NaCl. However, in comparison, 5-CBPyP had less tendency for aggregation that may be taken as an advantage for its probable application in photodynamic therapy of cancer. The trend of changes in absorption spectra of both porphyrins in the presence of ct-DNA indicates the homogeneous intercalation binding mode. The values of (2.81 ± 0.28) × 106 M?1 and (0.95 ± 0.09) × 106 M?1 were obtained for apparent binding constant of TMPyP and 5-CBPyP from analysis of optical absorption data, respectively. This indicates the less affinity of 5-CBPyP to ct-DNA in comparison with TMPyP. The binding of both porphyrins to ct-DNA quenches fluorescence emission of Ethidium bromide (EB) that is bound to ct-DNA. The quenching process obeys linear Stern-Volmer relationship indicating the displacement of EB from its binding sites by these porphyrins. The results of this technique also represent the intercalation mode of binding for both porphyrins and higher binding affinity of TMPyP compared with 5-CBPyP.  相似文献

The Raman and fluorescence spectroscopic properties of water‐soluble oxo‐titanium(IV) mesotetrakis (1‐methyl pyridium‐4‐yl) porphyrin (O=Ti(TMPyP)4+) bound with calf thymus DNA and artificial DNAs such as double stranded poly[d(A‐T)2] and poly[d(G‐C)2] have been investigated on the single DNA molecule basis by AFM‐correlated confocal scanning microscope (CSM)‐coupled Raman and fluorescence spectroscopic techniques as well as the ensemble‐averaged spectroscopy. The ensemble‐averaged spectroscopic studies imply that the porphyrin interacts with DNA in different groove binding patterns depending on the base pairs. AFM‐images of the different DNAs bound with O=Ti(TMPyP)4+ were measured, and their morphologies are found to depend on kind of base pairs interacting with O=Ti(TMPyP)4+. Being correlated with the AFM images, the CSM‐coupled Raman and fluorescence spectral properties of the three different single O=Ti(TMPyP)4+‐DNA complexes were observed to be highly resolved and sensitive to base pair‐dependent axial ligation of Ti‐O bond as compared to the corresponding ensemble‐averaged spectral properties, which affect the groove binding and its strength of the O=Ti(TMPyP)4+ with DNA. The axial ligation was found to be accompanied by vibration structural change of the porphyrin ring, leading to keep the shape of double stranded poly[d(A‐T)2] rigid while poly‐[d(G‐C)2] and calf thymus DNA flexible after binding with the oxo‐titanyl porphyrin. The base pair dependence of the fluorescence decay times of the DNA‐bound porphyrins was also observed, implying that an excited‐state charge transfer takes place in the G‐C rich major groove in calf thymus DNA. These results suggest that binding of O=Ti(TMPyP)4+ is more preferential with the G‐C rich major groove than with the A‐T rich minor groove in calf thymus DNA so that the morphology of DNA is changed.  相似文献

采用快速混合停流技术,考察了pH=7.4,V(CH~3CN):V(H~2O)=1:1的混合溶剂中水溶性锰卟啉Mn^I^I^I(TMPyP)与单氧给体NaOCl及KHSO~5构建的细胞色素P-450模拟酶体系催化氧化活性物种的生成及催化烯烃DPBD环氧化过程。实验表明,在反应进行中存在着两种中间体:oxo-Mn^V(Por.)(1)和oxo-Mn^I^V(Por.)(2),但两者的催化活性有差异,在催化烯烃DPBD环氧化反应中,对于Mn^I^I^I(TMPyP)-NaOCl体系起催化作用的主要作用的主要是中间体1,而对于Mn^I^I^I(TNPyP)-KHSO~5体系两种中间体均与烯烃配位生成环氧化产物,并且该体系催化活性较高。  相似文献

Aggregation of two porphyrin derivatives with carboxylic groups, 4-oxo-4-((4-(10,15,20-triphenyl-21H,23H-porphin-5-yl)phenyl)amino)butanoic acid (MAC) and 4,4',4',4'-[21H,23H-porphine-5,10,15,20-tetrayltetrakis(4,1-phenyleneimino)]tetrakis(4-oxo-butanoic acid) (TA4C), and their affinity to bovine serum albumin were investigated via absorption spectrometry, (1)H NMR and fluorescence spectrometry. MAC and its complexes with beta-cyclodextrin could form aggregates in an aqueous solution while TA4C was self-associated loosely. From the absorbance profiles of MAC in the titration of bovine serum albumin, hypochromicity was observed without any shift of the maximum absorbance wavelength. In both absorption spectra of TA4C in aqueous solutions and in solid state, three Q bands appeared in the visible region. In the measurements of absorption and fluorescence spectra upon titration of BSA, some spectral changes of TA4C were observed. The whole procedure of titration could be divided into three successive stages. The three-banded profiles of TA4C might be explained according to a loose dimer model.  相似文献

采用稳态吸收和荧光光谱、圆二色谱和皮秒时间分辨荧光光谱手段, 研究了5,10,15,20-四[4-(N-甲基吡啶)]卟啉(TMPyP4)与腺嘌呤(A)、鸟嘌呤(G)、胸腺嘧啶(T)和胞嘧啶(C)等4种碱基, 以及相应的核苷、核苷酸和单链DNA的结合能力和光谱学性质. 研究结果发现, 嘌呤与TMPyP4的结合能力比嘧啶的强. 对于某一碱基系列, 结合能力强弱顺序依次为: 碱基~核苷<核苷酸<单链DNA. 时间分辨荧光谱研究发现, 除鸟嘌呤外, 核酸和TMPyP4复合物的荧光动力学均含有快(1~2 ns)和慢(约10 ns)两个衰减过程, 它们分别是由激基复合体和环境极性对激发态TMPyP4分子的影响所致. 单链DNA能诱导TMPyP4产生诱导圆二色信号, 而单分子(碱基、核苷、核苷酸)则无此功能.  相似文献

Noncovalent interaction of two water-soluble synthetic macromolecules, Cucurbit[7]uril (CB7) and 5,10,15,20-tetrakis(4- N-methylpyridyl)porphyrin (TMPyP), has been studied from the viewpoint of organizing them through supramolecular interactions and thereby modulating their functional activities. Steady-state and time-resolved fluorescence measurements along with NMR results establish that CB7 crowns the N-methylpyridyl group of the TMPyP in a 1:4 stoichiometry. The overall binding constant was evaluated to be approximately 4.5 x 10 (19) M (-4). The high binding affinity, promoting a stable and extendable molecular assembly in aqueous solution, could open new frontiers in the design and synthesis of higher-order supramolecular structures with photofunctional moieties and project their utility in therapeutic applications.  相似文献

In this work, a new approach for TMPyP [5,10,15,20-tetrakis (1-methylpyridinium-4-yl) porphyrin] and TMAPP [5,10,15,20-tetrakis (N,N,N-trimethyl-anilinium-4-yl) porphyrin] intercalation into α-phase of zirconium hydrogen phosphate (α-ZrP) was described: porphyrins were inserted through exchanging pre-intercalated alkylamine. Pre-intercalated n-butylamine (BA) could form either a mobile monolayer or a stable bilayer in α-ZrP. The exchange speed between porphyrins and BA in mobile monolayer is obviously faster than that in stable bilayer. Therefore mobility of spacers is one important intercalation factor. In addition, we investigated the interlayer state of TMPyP by XRD, visible spectrum, fluorescence spectrum and molecular modeling. The results collectively revealed that the porphyrin was orderly arranged with their planes inclined to the host lamella and was presented as monomer instead of aggregation in the gallery of α-ZrP.  相似文献

Mesoporous organosilica-porphyrin composites were obtained by entrapment of 5,10,15,20-tetraphenylporphyrin (TPP) and 5,10,15,20-tetrakis(1-methyl-4-pyridinio)porphyrin (TMPyP) into three polysilsesquioxanes prepared by the sol-gel method from 1,2-bis(triethoxysilyl)ethane, 1,6-bis(triethoxysilyl)hexane, and 1,8-bis(triethoxysilyl)octane. The materials were characterised by their texture and optical properties (fluorescence and absorbance) and by light-induced antimicrobial activity against E. coli BL21(DE3) (pET16bDsRed) strain.  相似文献

The fluorescence and solid substrate room temperature phosphorescence (SS-RTP) properties of Pd(II) meso-tetrakis (4-N-methyl-pyridiniumyl) porphyrin (Pd(II)TMPyP) were studied. The factors influencing the SS-RTP emission, such as filter type, inorganic salt sort, drying temperature, pre-drying time and drying time were investigated in detail. Strong SS-RTP signal can be induced on the slow speed filter paper in the presence of the external inorganic salt, Ca(NO3)2, with the maximum excitation and emission wavelengths at 421 nm and 675 nm, respectively. The interaction between calf thymus DNA (ctDNA) and Pd(II)TMPyP was investigated at pH 7.2 using SS-RTP, fluorescence and UV-vis spectroscopy. The SS-RTP intensity of Pd(II)TMPyP was enhanced efficiently with the increasing amount of ctDNA. This phenomenon demonstrates that the intercalated porphyrin is shielded by ctDNA to avoid collision quenching. This result was supported by SS-RTP lifetime measurement, SS-RTP anion quenching experiment and fluorescence polarization measurement. Furthermore, with the addition of ctDNA, the UV-vis spectra of Pd(II)TMPyP shows apparent hypochromicity (40%) at the Soret maximum of 417 nm and a red shift of Δλ = 15 nm, also indicating that Pd(II)TMPyP intercalates into ctDNA bases. The binding constant of Pd(II)TMPyP to ctDNA was calculated to be 4.41 × 105 L/mol based on the derivative McGhee-von Hippel plots.  相似文献

The heteroleptic supramolecular double-decker porphyrin 1 was synthesized with DABCO as a guest between two cofacial porphyrin units as characterized by (1)H NMR and ESI-MS. While DABCO is not seen to tumble inside the cavity, even at higher temperatures (80 °C), such motion was triggered upon addition of various coordinating ligands (quinuclidine, 4-bromopyridine, or excess of DABCO). Different stoichiometric amounts were needed depending on the n donor quality of the added ligands to initiate tumbling of the "inside" DABCO. As demonstrated in an example with excess DABCO, the tumbling was stopped by lowering the temperature to -50 °C.  相似文献

In this work we explore the structure of a G-rich DNA aptamer termed AT11-L2 (TGGTGGTGGTTGTTGTTGGTGGTGGTGGT; derivative of AT11) by evaluating the formation and stability of G-quadruplex (G4) conformation under different experimental conditions such as KCl concentration, temperature, and upon binding with a variety of G4 ligands (360A, BRACO-19, PDS, PhenDC3, TMPyP4). We also determined whether nucleolin (NCL) can be a target of AT11-L2 G4. Firstly, we assessed by circular dichroism, UV and NMR spectroscopies the formation of G4 by AT11-L2. We observed that, for KCl concentrations of 65 mM or less, AT11-L2 adopts hybrid or multiple topologies. In contrast, a parallel topology predominates for buffer containing 100 mM of KCl. The Tm of AT11-L2 in 100 mM of KCl is 38.9 °C, proving the weak stability of this sequence. We also found that upon titration with two molar equivalents of 360A, BRACO-19 and PhenDC3, the G4 is strongly stabilized and its topology is maintained, while the addition of 3.5 molar equivalents of TMPyP4 promotes the disruption of G4. The KD values between AT11-L2 G4, ligands and NCL were obtained by fluorescence titrations and are in the range of µM for ligand complexes and nM when adding NCL. In silico studies suggest that four ligands bind to the AT11-L2 G4 structure by stacking interactions, while the RBD1,2 domains of NCL interact preferentially with the thymines of AT11-L2 G4. Finally, AT11-L2 G4 co-localized with NCL in NCL-positive tongue squamous cell carcinoma cell line.  相似文献

The DNA-binding affinities and DNA photocleavage abilities of cationic porphyrin, 5-(4-carboxyphenyl)-10,15,20-tris(4-methylpyridiniumyl)porphyrin (CTMPyP), and its reference compound meso-tetrakis(N-methyl-4-pyridiniumyl)porphyrin (H2TMPyP) have been investigated. The DNA-binding behaviors of the two compounds in NaH2PO4 buffer were compared systematically by using absorption, fluorescence and circular dichroism (CD) spectra, thermal denaturation as well as viscosity measurements. The experimental results show that CTMPyP binds to DNA in an outside binding mode, while H2TMPyP in an intercalative mode. Photocleavage experiments reveal that both two compounds employ 1O2-mediated mechanism in cleaving DNA and H2TMPyP can cleave DNA more efficiently than CTMPyP. Theoretical calculations were carried out with the density functional theory (DFT), and the calculated results indicate that the character and energies of some frontier orbitals of CTMPyP are quite different from those of H2TMPyP. These theoretical results can be used to explain their different DNA-binding modes and affinities to a certain extent.  相似文献
