Distinctions between liquid chromatography of synthetic polymers under critical conditions (LC CC) and liquid chromatography under limiting conditions (LC LC) are elucidated. Surface adsorption retention mechanism of macromolecules is employed in the chromatographic systems composed of 10 and 30 nm pore diameter bare silica gels, poly (methyl methacrylate)s of different molar masses, and mixed eluents acetonitrile/dichloromethane of different compositions, and at different temperatures. Increased robustness of the LC LC methods compared to LC CC is confirmed: the LC LC elution behavior is much less sensitive to eluent composition changes compared to LC CC. Still, the LC CC system under study is, surprisingly robust in terms of temperature variations. LC LC methods produce narrow, focused polymer peaks while a peak broadening is observed in LC CC. The results demonstrate importance of sample solvent applied in the isocratic coupled polymer HPLC methods.  相似文献

Monolithic materials have quickly become a well‐established stationary phase format in the field of capillary electrochromatography (CEC). Both the simplicity of their in situ preparation method and the large variety of readily available chemistries make the monolithic separation media an attractive alternative to capillary columns packed with particulate materials. This review summarizes the contributions of numerous groups working in this rapidly growing area, with a focus on monolithic capillary columns prepared from synthetic polymers. Various approaches employed for the preparation of the monoliths are detailed, and where available, the material properties of the resulting monolithic capillary columns are shown. Their chromatographic performance is demonstrated by numerous separations of different analyte mixtures in variety of modes. Although detailed studies of the effect of polymer properties on the analytical performance of monolithic capillaries remain scarce at this early stage of their development, this review also discusses some important relationships such as the effect of pore size on the separation performance in more detail.  相似文献

A novel high performance liquid chromatographic (HPLC) method viz. “enthalpic partition assisted size exclusion chromatography” deliberately combines entropic and enthalpic partition mechanisms. It enables separation of homopolymers according to their molar mass with increased selectivity, as well as discrimination of polymer species differing in their nature/composition. Enthalpic partition of macromolecules takes place between the mobile phase and the stationary “liquid” of a different chemical nature, which is immobilized within pores of an appropriate carrier (a bonded phase). The extent of enthalpic partition depends on the accessibility of bonded phase for macromolecules and on the difference of polymer solubility in the mobile phase and in the solvated bonded phase. The enthalpic partition in favor of column packing arises from better solubility of polymer solutes in the solvated stationary phase compared to the mobile phase. Macromolecules are “pushed” into the solvated stationary phase and their retention volumes (VR) increase. In the area of high molar masses, the extent of enthalpic partition as rule raises with the increasing size of macromolecules. However, under properly chosen experimental conditions the enthalpic partition may rapidly diminish with the sample molar mass (M), likely due to the solubility changes and/or due to partial exclusion of macromolecules from the pores. As result, the corresponding retention volumes sharply drop within a narrow range of M with the increasing size of macromolecules. This results in the log M vs. VR dependences, which resemble in their form that for size exclusion chromatography but are much more flat indicating highly selective separations of homopolymers according to their molar masses. In this way, enthalpic partition “assists” entropic partition (size exclusion). Polymer species, which do not undergo enthalpic partition, elute from the HPLC column in the conventional size exclusion mode and can be discriminated from the partitioning species. Enthalpic partition assisted size exclusion chromatography can be utilized in separation and characterization of various homopolymers, and polymer blends.  相似文献

 以溶质计量置换保留模型 (SDM R)为依据 ,通过研究 3种正链醇同系物溶剂置换剂对 14种正链醇同系物溶质色谱保留行为的影响 ,发现计量置换参数Z(对应 1mol溶质被吸附时从溶质与固定相接触处释放出的溶剂的总摩尔数 )、lgI(与 1mol溶质对固定相亲和势有关的常数 )和 j(与 1mol溶剂对固定相亲和势有关的常数 )均随着同系物置换剂相对分子质量的增大而减小 ,并呈现出线性变化 ,表明溶剂强度与溶剂分子的大小有关 ,分子愈大 ,溶剂洗脱能力愈强 ,并遵循同系物规律。  相似文献

最常用的测试高聚物的分子量和分子量分布的体积排除色谱(SEC)是高效液相色谱 (HPLC)的一个重要分支,HPLC的另一个重要分支是相互作用液相色谱, 它是20世纪90年代开始用于高分子分离和表征的研究领域。相互作用液相色谱可以根据高分子的化学结构(如共混物组成、共聚物组成、端基)来分离,它比SEC 有更高的分离效率。本文介绍了高聚物液相色谱的分离模式,并就高聚物体积排除色谱、相互作用液相色谱、临界液相色谱和全二维液相色谱用于分离和表征高聚物的研究进展进行了较系统的综述,并对该技术目前存在的问题和今后可能的发展前景进行了探讨。  相似文献

A chiral liquid chromatographic method is described for the determination of the enantiomers of tamsulosin hydrochloride and its synthetic intermediates. Enantioseparation was achieved on a Chiralcel OD-R column (250 mm × 4.6 mm, 10 m) using a mobile phase consisting of a mixture of 0.5 mol L–1 sodium perchlorate and acetonitrile (80:20, v/v, pH 4.0). The flow rate was 0.4 mL min–1 and detection was at 223 nm. Excellent enantiomer separations were achieved for tamsulosin hydrochloride and its synthetic intermediates. No other methods are available for the separation of these enantiomers. The method developed in this study has been successfully applied for purity control.  相似文献

Liquid adsorption chromatography, in combination with full adsorption–desorption and precipitation–redissolution techniques, is a powerful procedure for the fractionation of mixtures in the synthesis of star polymers. An easy separation of 4‐ and 6‐arm star poly(methyl methacrylate)s from their single arm precursors could be carried out by both procedures, with differences of separation efficiency also depending on the molecular weight of the precursor and the number of arms.

建立了反相高效液相色谱法同时测定鱼子酱中的柠檬黄、日落黄、偶氮玉红、苋菜红、胭脂红、赤藓红、红色2G和诱惑红8种合成色素的方法.样品中的合成色素经甲醇-4 mol/L尿素溶液(体积比1 ∶ 1)提取,固相萃取聚酰胺柱净化后,采用XDB-C18色谱柱(4.6 mm×250 mm×5 μm),以甲醇-乙腈(体积比1 ∶ 1)和0.01 mol/L乙酸钠溶液为流动相,在最佳梯度洗脱条件下,8种合成色素在17 min内实现分离.各色素在0.05 ~50 mg/L质量浓度范围内具有良好的线性关系.将该法用于实际样品的检测,8种合成色素的加标回收率为89% ~110%,相对标准偏差为1.4% ~4.7%.方法简单、高效,可用于鱼子酱中合成色素的日常监督检测.  相似文献

With four kinds of mobile phases,methanol-water,ethanol-water,2-propanol and acetonitrile-water(all containing 0.1%rifluroacetic acid),the displacement between solute and solvent in RPLC was proved to be universal in frontal analysls(FA).Based on the measured Z value in usual RPLC to be a constant and the quantitative determination of methanol increment in mobile phase in FA,the stoichiometric displacement(SD)between insulin and methanol was directly proved by the experiment.The SD was also proved to occur only on about the one-fourth of the maximum amount of adsorbed methanol in the bonded phase layer(BPL)without any dynamic problem of mass transfer,while in FA,the SD firstly occurs on the surface of the BPL and then gradually sinks into the deeper sites companied with a dynamic problem.Although the displaces soplvent by the same solute is less in the former case,the SD is independent of how deep of the solute enters the BPL.In addition,the adsorbed amount of solute on and adsorbent not only depends on the numbers of the adsorbed layer on the adsorbent surface,but also on the extent of the complete removal of the displace able solvent in the BPL.The pyhsical fundamental of the SD and the methodoloby for investigation were also discussed.  相似文献

合成一种了氨基酸衍生物:4,4′-二苯亚甲基桥联-二[2-(1-咪唑基)-3-苯基丙醇氨基甲酸酯](ImPh-Carb),并将其键合到硅胶上制备了一种新的氨基甲酸酯功能化的离子液体HPLC固定相(ImPh-Carb-Silica)。 利用1H NMR、13C NMR、MS和FTIR对ImPh-Carb进行了表征;通过FTIR和元素分析对ImPh-Carb-Silica固定相进行了表征,根据N含量计算得到ImPh-Carb-Silica的键合量为0.19 mmol/g。 以5种芳烃、5种酚类化合物和4种有机磷农药为分析物,分别在正相和反相色谱模式下对固定相的色谱分离性能进行了评价,同时考察了流动相的变化与溶质保留因子lg k之间的关系。 结果表明,该固定相与溶质分子间存在多重作用力,如疏水、氢键、π-π和偶极-诱导偶极作用等,使其能同时在正相和反相色谱模式下使用;在正相色谱条件下固定相对酚类化合物和有机磷农药表现出较好的分离选择性。  相似文献

LC of polymers under limiting conditions of adsorption (LC LCA) is a novel method based on different mobility of (pore excluded) macromolecules compared to (pore permeating) solvent molecules. Polymer sample is injected in a solvent preventing its adsorption within the column. Eluent promotes sample adsorption. Under these conditions, macromolecules cannot leave its initial solvent and elute from the column independently of their molar mass. In contrast, a less interactive simultaneously injected polymer leaves its initial solvent zone and is eluted in the size exclusion mode. As a result, chemically different polymer species can be discriminated. The effect of selected experimental conditions was studied on the LC LCA behavior of poly(methyl methacrylate)s eluted from bare silica gel columns. The parameters were packing pore diameter, injected sample volume and concentration, as well as column temperature. The size independent elution was only little affected by the above parameters and LC LCA produced well-focused peaks. The LC LCA mechanism was operative even at a very large sample of both volume and concentration. This makes LC LCA a robust and user-friendly method, likely suitable also for characterization of minor components of polymer mixtures.  相似文献

Reduced sample recovery is a frequent feature of LC of macromolecules under critical conditions of enthalpic interactions (LC CC). Several methods of assessment of LC CC sample recovery are compared. A novel approach is based on an online combination of the. The LC CC column with a noninteractive SEC column. It provides not only the amount but also the molar mass of the eluted/withheld polymer. The procedure was tested with poly(methyl methacrylate), bare silica gel column packings, and “critical eluent” tetrahydrofuran/toluene. It was shown that macromolecules with higher molar masses were preferentially trapped within the LC CC column packing so that the eluted part of the sample was no longer representative. The incomplete polymer elution can make the LC CC polymer analyses susceptible to significant experimental errors.  相似文献

有效地确定了反相高效液相色谱分离儿茶酚化合物的最佳条件。在水和甲醇的二元流动相里分别加入乙酸缓冲液,利用基于ln k=ln kw +SF, k=A+B/F, ln k=L+MF+NF2 (F是流动相中有机物甲醇的体积分数)等保留因子的一次或二次方程式的塔板理论得到色谱分离结果;利用保留原理得到等度和梯度洗脱的最佳条件。得出最佳初始流动相是含0.1%乙酸的水和含0.1%乙酸的甲醇(体积比为75∶25)的混合溶液;梯度洗脱条件:初始流动相保持15 min,然后用10 min的时间将上述二元流动相的体积比线性变换成50∶50,直到完成全部分离。通过实验证实该计算结果与实验值相近。  相似文献

With the combination of the the stoichiometric displacement model for retention (SDM-R) in reversed phase liquid chromatography (RPLC) and the stoichiometric displacement model for adsorption (SDM-A) in physical chemistry,the total number of moles of the re-solvated methanol of stationary phase side.nr,and that of solute side in the mobile phase,q,corresponding the one mole of the desorbing solute,were separately determined and referred as the characterization parameters of the contributions of the adsorption mechanism and partition mechanism to the solute retention,respectively.A chromatographic system of insulin,using mobile phase consisting of the pseudo-homologue of alcohols(methanol,ethanol and 2-propanol)-water and trifluoroacetic acid was employed.The maximum number of the methanol layers on the stationary phase surface was found to be 10.6,only 3 of which being valid in usual RPLC,traditionally referred as a volume process in partition mechanism.However,it still follows the SDM-R.Both of q and nr of insulin were found not to be zero,indicating that the retention mechanism of insulin is a mixed mode of partition mechanism and adsorption mechanism.When methanol is used as the organic modifier,the ratio of q/nr was 1.13,indicating the contribution to insulin retention due to partition mechanism being a bit greater than that due to adsorption mechanism.A linear relationship between q,or nr and the carbon number of the pseudo-homologue in the mobile phase was also found.As a methodology for investigating the retention mechanism retention and behavior of biopolymers.a homologue of organic solvents as the organic modifier in mobile phase has also been explored.  相似文献
