The effect of irradiance (40 and 840 micromol photons m(-2) s(-1)) of short-term (48 h) irradiation on photosynthetic activity (estimated as oxygen evolution and as chlorophyll fluorescence), specific absorption and fluorescence excitation spectra, photosynthetic pigment accumulation (chlorophyll a and biliproteins) and UV-absorbing compounds (mycosporine-like amino acids, MAAs) was investigated in sun and shade species of the red algal genus Porphyra collected in Trondheimsfjord (Norway). In the sun type, high irradiance exposure (840 micromol photons m(-2) s(-1)) did not alter the Chl a concentration, however, exposure to a lower irradiance (40 micromol photons m(-2) s(-1)) for 48 h significantly increased the chlorophyll concentration. The content of MAAs was significantly higher in the suntype than in the shade type algae. Porphyra-334 is the main MAA in this species followed by shinorine. The total content of MAAs significantly (P<0.05) increased in the sun type after 48 h exposure to both high and low irradiances. However, in the shade type, porphyra-334 significantly decreased (P<0.05) after both high and low irradiance exposure. Photosynthetic activity (as oxygen evolution) and the optimal quantum yield (F(v)/F(m)), as an indicator of photoinhibition, decreased under low and high irradiance in the shade type algae and no full recovery was observed when the algae were transferred to very low irradiation.The sun type algae presented a higher capacity of acclimation to increased irradiance than the shade type algae. This high acclimation of sun type algae to short term high irradiance exposure (48 h) is explained by the higher thermal dissipation. This was estimated as the ratio of nonphotochemical quenching related to the light dose (q(N):dose) and by the accumulation of MAAs.  相似文献

Hydrated thalli of two lichen species--Umbilicaria antarctica and Lasallia pustulata--were exposed to high light (1800 micromol m-2s-1) for 30 min. High light exposure led to a decrease of total glutathione in both species, while de-epoxidation state of xanthophyll cycle pigments and non-photochemical quenching increased. In the subsequent recovery, the values of de-epoxidation state of xanthophyll cycle pigments decreased towards initial values. Glutathione (GSH) was resynthetised slowly. In conclusion, zeaxanthin-related protection is probably more involved than GSH-related protection in short-term response to high light stress in U. antarctica and L. pustulata. Faster recovery from photoinhibition in L. pustulata than U. antarctica is mainly due to faster conversion of zeaxanthin to violaxanthin and larger GSH pool of former species.  相似文献

Seasonal variation in maximal photochemical quantum yield (F(V)/F(M)) of photosystem II (PS II), light adapted quantum yield (Phi(II)) of PS II, non-photochemical quenching (NPQ), contents of chlorophylls, and xanthophyll cycle pigments (VAZ) was studied in Xanthoria parietina repeatedly sampled in one location in S Norway during one year. The seasonal course in the susceptibility to photoinhibition was evaluated as high light-induced changes (1,800 micromol photons m(-2) s(-1) for 24h) in F(V)/F(M), Phi(II), and NPQ, measured as the ability to recover after 2 and 20 h at low light in control thalli with a natural cortical parietin screen, and in thalli from which parietin had been removed prior to high light exposures. F(V)/F(M), Phi(II), chlorophyll content, and the conversion state of VAZ (DEPS) reached minimum in spring. At the same time, yearly maxima of VAZ content and NPQ were recorded. Thereafter, F(V)/F(M), Phi(II), and chlorophyll content increased gradually, reaching maximum values in late autumn. DEPS peaked already in summer. Similarly, VAZ and NPQ decreased from early summer until winter. All data show that the X. parietina photobiont acclimates to seasonal changes in solar radiation, consistent with the lichen's preference for well-lit habitats. However, a comparison with a study of seasonal acclimation in the X. parietina mycobiont shows that in order to understand the seasonal photobiont acclimation, one has to consider the seasonal variation in internal screening caused by the fungal regulation of the PAR-absorbing parietin. A joint effort of both bionts seems to be required to avoid serious photoinhibition.  相似文献

The photosynthetic performance of Enteromorpha linza (L.) J. Agardh-Chlorophyceae was determined with a portable PAM instrument in situ and under seminatural radiation conditions in Patagonia, Argentina. Solar radiation was measured in parallel with a three-channel radiometer, ELDONET (Real Time Computer, M?hrendorf, Germany), in three wavelength ranges, UV-B (280-315 nm), UV-A (315-400 nm), and PAR (400-700 nm). The effective photosynthetic quantum yield decreased after 15-min exposure to solar radiation when the thalli were kept in a fixed position but recovered in the subsequent shade conditions within several hours. A 30-min exposure of free floating thalli, however, caused less photoinhibition. The photosynthetic quantum yield of E. linza was also followed over whole days under clear sky, partly cloudy and rainy conditions in a large reservoir of water (free floating thalli) and in situ (thalli growing in rock pools). Most of the observed effect was due to visible radiation; however, the UV wavelength range, and especially UV-B, caused a significant reduction of the photosynthetic quantum yield. Fluence rate response curves indicated that the species is a typical shade plant which showed non-photochemical quenching at intermediate and higher irradiances. This is a surprising result since these algae are found in the upper eulittoral where they are exposed to high irradiances. Obviously they utilize light only during periods of low irradiances (morning, evening, high tide) while they shut down the electron transport chain during intensive exposure. Fast induction and relaxation kinetics have been measured in these algae for the first time and indicated a rapid adaptation of the photosynthetic capacity to the changing light conditions as well as a fast decrease of PS II fluorescence upon exposure to solar radiation. There was a strong bleaching of chlorophyll due to exposure to solar radiation but less drastic bleaching of carotenoids.  相似文献

The marine diatom Haslea ostrearia was cultured under light of different qualities, white (WL), blue (BL), green (GL), yellow (YL), red (RL), and far-red (FRL) and at two irradiance levels, low and high (20 and 100 micromolphotonsm(-2)s(-1), respectively). The effects of the different light regimes were studied on growth, pigment content, and photosynthesis, estimated by the modulated fluorescence of chlorophyll, as relative electron transport rate (rETR). For all the light qualities studied, growth rates were higher at high irradiance. Compared to the corresponding WL controls, growth was higher in BL and lower in YL at low irradiance, and lower in YL and GL at high irradiance. Except for YL, almost all the pigment contents of the cells were lower at high irradiance. At low irradiance, cell pigment contents (chlorophyll a and c, fucoxanthin) and pigment ratios (in function of chlorophyll a) were lower in YL, RL, and FRL. Whatever the irradiance level, the maximum PSII quantum efficiency (F(v)/F(m) remained almost constant for WL, BL, and GL. Other fluorescence parameters (photochemical quenching, rETR(max), and alpha, the maximum light utilization coefficient) were lower in GL, YL, RL, and FRL, at low irradiance. Although not statistically significant, BL caused an increase in these fluorescence parameters. These findings are interpreted as evidence that inverse chromatic acclimation occurs in diatoms.  相似文献

The filamentous rhodophytes Callithamnion gaudichaudi Agardh and Ceramium sp. were utilized to study the effects of solar radiation (PAR, 400-700 nm, UV-B, 280-315 nm and UV-A, 315-400 nm) on the photosynthetic performance in situ in Patagonia waters (Argentina). A pulse amplitude modulated (PAM) fluorometer was used to determine the fluorescence parameters. The two species grew in different habitats in the eulittoral: Ceramium sp. was found only in rock pools while C. gaudichaudii grew on exposed rocks and fell dry during low tide. Both species differed in their fluorescence parameters and their sensitivity to solar radiation exposure. The photosynthetic quantum yield had its lowest values at noon, but it recovered in the afternoon/evening hours, when irradiances were lower. PAR (irradiance of about 400 W m(-2) at noon) was responsible for most of the decrease in the yield on clear days, especially in Ceramium sp., but UVR (280-400 nm) also accounted for a significant decrease. Fluence rate response curves indicated that both species were adapted to low fluence rates and showed a pronounced non-photochemical quenching at intermediate and higher irradiances. Both species showed a rapid adaptation during measurement of fast induction kinetics but differed significantly in their fluorescence components. All photosynthetic pigments were bleached after 8 h exposure to solar radiation over a full day. Strong absorption in the UV-A range, most likely due to mycosporine-like amino acids, was detected in both strains. The pronounced sensitivity to solar radiation in situ and the recovery capacity of these two filamentous Rhodophyte species, as well as the presence of protective compounds, suggests that these algae have the ability to adapt to the relatively high radiation levels and changes in irradiance found in the Patagonia waters.  相似文献

The steps of protochlorophyllide (Pchlide) photoreduction and subsequent chlorophyllide (Chlide) transformations which occur in the seconds to minutes time-scale were studied using a diode array spectrofluorometer in dark-grown barley leaves. The intensity of the excitation light was varied between 3 and 2,500 micromol m(-2) s(-1) and a series of fluorescence spectra were recorded at room temperature in the seconds and minutes time scales. In certain experiments, 77-K emission spectra were measured with the same equipment. The high quality of the spectra allowed us to run spectral resolution studies which proved the occurrence, at room temperature, of multiple Pchlide and Chlide forms found previously in 77-K spectra. The comparison of the 77-K and room-temperature spectra showed that the fluorescence yields of the nonphotoactive 633-nm Pchlide form and of the Chlide product emitting at 678 nm were temperature independent. The fluorescence intensity of aggregated NADPH-pigment-POR complexes (photoactive 656-nm Pchlide and 693-nm Chlide forms) were strongly increased at 77 K, while that of the NADP(+)-Chlide-POR (684-686-nm Chlide form) was much less affected by temperature. Information was obtained also about the dynamics of the transformation of pigment forms in the light at different photon densities. At low light intensities, the phototransformation of the 642-644-nm Pchlide form was faster than that of the 654-656-nm form. The relative amplitudes of Gaussian components related to different Chlide forms found after exposure to a constant amount of photons strongly depended on the light intensity used. Strong quenching of all Chlide components occurred upon prolonged exposure to high intensity light. These effects are discussed by considering the interconversion processes between different forms of the pigment-protein complexes, their relative fluorescence yields and energy migration processes.  相似文献

Abstract— Photoinhibition of photosynthesis was investigated in the Mediterranean green alga Halimeda tuna measuring pulse amplitude modulation (PAM) chlorophyll fluorescence and oxygen evolution in situ under solar radiation. Exposure to solar radiation at the surface caused a drastic decline in the photosynthetic quantum yield accompanied by a decline in the photochemical quenching, while the nonphotochemical quenching dramatically increased. The algae recovered from these effects within a few hours indicating that these are mainly due to reversible photoinhibition and only to a smaller extent to nonreversible photodamage. Individuals harvested from deeper waters were more affected than those from shallower waters. Photoinhibition occurs in this alga even in its natural habitat when the sun is at high angles as shown by measuring the fluorescence parameters at hourly intervals during the course of the day. Photoinhibition was less pronounced when the short wavelength band was increasingly removed from solar radiation using cut-off filters. After exposure of thalli to solar radiation at the water surface, oxygen production decreased drastically within 30 min; this inhibition was even more pronounced in algae harvested from deeper waters. Oxygen measurements at different depths showed optimal photosynthesis at a depth of 1 m. Also for photosynthetic oxygen production inhibited by high solar irradiance at least partial recovery could be observed within several hours. Despite the fact that UVB accounts for a very small fraction of solar radiation, it has a considerable effect on photosynthesis, whereas UVA seems to contribute only little to photoinhibition in H. tuna.  相似文献

The effect of oxygen concentration on both absorption and chlorophyll fluorescence spectra was investigated in isolated pea thylakoids at weak actinic light under the steady-state conditions. Upon the rise of oxygen concentration from anaerobiosis up to 412 microM a gradual absorbance increase around both 437 and 670 nm was observed, suggesting the disaggregation of LHCII and destacking of thylakoids. Simultaneously, an increase in oxygen concentration resulted in a decline in the Chl fluorescence at 680 nm to about 60% of the initial value. The plot of normalized Chl fluorescence quenching, F(-O(2))/F(+O(2)), showed discontinuity above 275 microM O(2), revealing two phases of quenching, at both lower and higher oxygen concentrations. The inhibition of photosystem II by DCMU or atrazine as well as that of cyt b(6)f by myxothiazol attenuated the oxygen-induced quenching events observed above 275 microM O(2), but did not modify the first phase of oxygen action. These data imply that the oxygen mediated Chl fluorescence quenching is partially independent on non-cyclic electron flow. The second phase of oxygen-induced decline in Chl fluorescence is diminished in thylakoids with poisoned PSII and cyt b(6)f activities and treated with rotenone or N-ethylmaleimide to inhibit NAD(P)H-plastoquinone dehydrogenase. The data suggest that under weak light and high oxygen concentration the Chl fluorescence quenching results from interactions between oxygen and PSI, cyt b(6)f and Ndh. On the contrary, inhibition of non-cyclic electron flow by antimycin A or uncoupling of thylakoids by carbonyl cyanide m-chlorophenyl hydrazone did not modify the steady-state oxygen effect on Chl fluorescence quenching. The addition of NADH protected thylakoids against oxygen-induced Chl fluorescence quenching, whereas in the presence of exogenic duroquinone the decrease in Chl fluorescence to one half of the initial level did not result from the oxygen effect, probably due to oxygen action as a weak electron acceptor from PQ pool and an insufficient non-photochemical quencher. The data indicate that mechanism of oxygen-induced Chl fluorescence quenching depends significantly on oxygen concentration and is related to both structural rearrangement of thylakoids and the direct oxygen reduction by photosynthetic complexes.  相似文献

Acclimation of the photosynthetic apparatus of chlorophyll b-less barley mutant chlorina f2 to low light (100 micromolm(-2)s(-1); LL) and extremely high light level (1000 micromolm(-2)s(-1); HL) was examined using techniques of pigment analysis and chlorophyll a fluorescence measurements at room temperature and at 77 K. The absence of chlorophyll b in LL-grown chlorina f2 resulted in the reduction of functional antenna size of both photosystem II (by 67%) and photosystem I (by 21%). Chlorophyll fluorescence characteristics of the LL-grown mutant indicated no impairment of the utilization of absorbed light energy in photosystem II photochemistry. Thermal dissipation of excitation energy estimated as non-photochemical quenching of minimal fluorescence (SV(0)) was significantly higher as compared to the wild-type barley grown under LL. Despite impaired assembly of pigment-protein complexes, chlorina f2 was able to efficiently acclimate to HL. In comparison with chlorina f2 grown under LL, HL-grown chlorina f2 was characterized by unaffected maximal photochemical efficiency of photosystem II (F(V)/F(M), doubled content of both beta-carotene and the xanthophyll cycle pigments and considerably reduced efficiency of excitation energy transfer from carotenoids to chlorophyll a. The enormous xanthophyll cycle pool size was however associated with reduced SV(0) capacity. We suggest that the substantial part of the xanthophyll cycle pigments is not bound to the remaining pigment-protein complexes and acts as filter for excitation energy, thereby contributing to the efficient photoprotection of chlorina f2 grown under HL.  相似文献

The marine diatom Thalassiosira weissflogii was cultured under a light regime simulating the daily rise and fall of the sun. The light regime caused a daily cycle in non-photochemical quenching. Remarkable were the changes in fluorescence directly after a light-to-dark transition that occurred in addition to the changes induced by non-photochemical quenching. A transient non-photochemical reduction of PQ and of Q(A) was indicated by a transient increase in apparent F(o) and by changes in the shape of the fluorescence induction curve. The observed changes developed approximately the first 100-120 s after a light-to-dark transition and could be reversed by the application of far-red illumination. Chlororespiration is thought to cause the reduction of PQ and, as the PQ-pool is in equilibrium with Q(A), also a reduction of Q(A). The function and ecological relevance of chlororespiration are discussed.  相似文献

We have used chlorophyll fluorescence, delayed luminescence and thermoluminescence measurements to study the influence of an artificial DeltapH in the presence or absence of zeaxanthin on photosystem II reactions. Energization of the pea thylakoid membranes induced non-photochemical fluorescence quenching and an increase in the overall luminescence emission of PSII during delayed luminescence and thermoluminescence measurements. This DeltapH-induced overall luminescence increase was caused by a strongly enhanced delayed luminescence in the seconds range before sample heating. In the subsequent thermoluminescence measurements the intensity of the B-band decreased after one and increased after two or more single turnover flashes. We propose that strong membrane energization shifted the redox potential of photosystem II radical pairs to more negative values causing the high delayed luminescence. The zeaxanthin-dependent non-photochemical fluorescence quenching component, however, did not alter thermoluminescence B-bands but decreased the delayed luminescence intensity by 30%. To our knowledge this is the first report that the radiative radical pair recombination, exhibited as delayed luminescence but not thermoluminescence emission, is sensitive to the antenna located zeaxanthin related non-photochemical fluorescence quenching. Our data can be interpreted within the frame of the exciton/radical pair equilibrium model that describes photosystem II as a shallow trap and incorporates the transfer of energy from the re-excitated reaction centre to the antenna of photosystem II.  相似文献

We monitored chlorophyll (Chl) fluorescence, pigment concentration and the de-epoxidation state of the xanthophyll cycle (DPS(1)) in two warm temperate broad-leaved evergreen species (Quercus myrsinaefolia and Machilus thunbergii). Reduction of the maximal quantum yield of Photosystem II (PSII) (calculated from Fv/Fm, variable to maximal Chl a fluorescence) and retention of a high DPS were observed in both species in the winter, and can be interpreted as acclimation to winter. In particular, the acclimation of PSII in these species can be chiefly attributed to thermal dissipation, which is correlated with the retention of high zeaxanthin. Furthermore, we attempted to divide the fate of the absorbed light energy by the PSII antennae into three components: (i) PSII photochemistry (represented by its quantum yield, ΦPSII), (ii) dissipation by down-regulation via non-photochemical quenching (ΦNPQ) and (iii) other non-photochemical processes (ΦONP). The estimated energy allocation of the absorbed light indicated that the proportion of ΦPSII decreased, whereas that of ΦNPQ+ΦONP increased during winter. This result suggests that the excess energy absorbed in the PSII complexes is safely dissipated from the PSII antennae. Based on these results, we conclude that thermal dissipation from the PSII antennae plays an important role in two overwintering broad-leaved evergreen trees growing in Japan.  相似文献

Leaves of Secale cereale seedlings were exposed to high light illumination (1200micromolm(-2)s(-1)) and Cd ions at 5 or 50microM concentrations. Influence of these stress factors on violaxanthin cycle pigments content was analysed chromatographically. Chlorophyll a fluorescence induction was used to analyse response of PSII to stress conditions and contribution of light-harvesting complex (LHCII) in non-photochemical quenching of excitation energy. The Cd-induced all-trans violaxanthin isomerization was analysed by HPLC technique in acetonitrile:methanol:water (72:8:3, v/v) solvent mixture. Interestingly, in the control and Cd-treated leaves subjected to high light, photochemical utilization of absorbed energy increased. This indicates plant adaptation to high light stress. In control plants high light caused zeaxanthin formation, however, the presence of Cd in the nutrient solution resulted in reduction of the second step of violaxanthin de-epoxidation process and anteraxanthin accumulation. In this study we have also shown, that non-photochemical quenching can be independent of anteraxanthin and zeaxanthin content. The particular increase in the cis isomers fraction in Cd-treated leaves has been explained in terms of a direct metal-pigment interaction as confirmed by Cd-induced all-trans violaxanthin isomerization in organic solvent, leading to formation of 13-cis, 9-cis and 15-cis isomers.  相似文献

The effect of solar radiation on photosynthesis and chlorophyll fluorescence associated to photosystem II (PS II) was determined in the Phaeophyta Macrocystis pyrifera, the Rhodophyta Chondrus crispus and the Chlorophyta Ulva lactuca by oxygen evolution and pulse-amplitude-modulated (PAM) fluorescence. The algae were maintained in 1.2 m3 outdoor tanks with constant aeration and at 8, 26 and 100% incident irradiance (E(o)). All three species showed a decrease in deltaF/F'm values during solar noon compared to values in the morning and afternoon, suggesting a photoinhibition of photosynthesis. In general, photoinhibition was negatively correlated to increasing daily irradiance in all three species. Photoinhibition in C. crispus occurred in tissue incubated at 8, 26 and 100% E(o), while in M. pyrifera and U. lactuca a decrease in deltaF/F'm values was only observed in tissue incubated at 100% E(o). This suggests that species that naturally grow at greater depths might be more susceptible to excessive light when cultured in shallow waters compared to species that naturally inhabit shallower depths. In M. pyrifera, deltaF/F'm values were lower in the afternoon than those in the morning, suggesting slower repair mechanisms of the photosystem II compared to the other species. The results suggest that photoinhibition could be reduced by reducing incident irradiance to culture systems or increasing of biomass to promote self-shading. Gross oxygenic photosynthesis increased linearly at low electron transport rates after which it saturated in all three species. This suggests that chlorophyll fluorescence could be used as an indicator of the physiological status of macroalgae maintained in dense aquaculture systems.  相似文献

Photomovement measurements were carried out with swarmers of the brown algae Scytosiphon lomentaria (Lyngb.) Link and Petalonia fascia (O. F. Müll.) as a function of irradiance direction, photon irradiance, spectral composition and ultraviolet radiation (UVR, lambda=280-400 nm) dose. Swarmers from both species showed similar photomovement patterns: negative phototaxis occurred under photon irradiances of 10-90 micromol photons m(-2) s(-1), and no movement was observed at 190 micromol photons m(-2) s(-1). The translocational velocity measured between 10 and 90 micromol m(-2) s(-1) ranged from 100 to 200 microm s(-1). The accumulation of swarmers presented a peak at 450 nm (waveband of 50 nm), and smaller peaks at 400 and 500 nm; no effect was observed at wavelengths of 550 nm and above. The decline in phototactic index (an estimator of photomovement response) of swarmers was linearly correlated with the logarithm of UVR doses. These data were correlated with levels of natural solar radiation in the field. It is hypothesized that motility of swarmers could be a critical factor in the survival of these species under a scenario of increased UVR.  相似文献

Isothermal microcalorimetry can be used to investigate the photosynthetic energy conversion of autotrophic organisms. In this study, for the first time a diatom alga was used to compare the calorimetrically measured heat flux with measurements of the photosynthetic performance by oxygen evolution and pulse-amplitude modulated fluorescence. The presented experimental setup proved suitable to compare calorimetric data with those of conventional methods of the determination of photosynthesis rates. Special attention was paid to the contribution of energy dissipation via non-photochemical quenching (NPQ) of chlorophyll fluorescence to the metabolic energy balance. This was achieved by a combination of different light conditions and the use of an inhibitor of NPQ. Although NPQ is an important photoprotective mechanism in diatoms, the inhibition of NPQ resulted in an activation of alternative, energy dissipating pathways for absorbed radiation which completely compensated for the fraction of energy dissipation by NPQ.  相似文献

Abstract— Intact leaves, previously adapted to darkness for a prolonged period of time, were suddenly illuminated with a strong, photosynthetically saturating, white light (ca 1500 μmol m−2 s_1), resulting in the rapid establishment of a large energy-dependent chlorophyll fluorescence quenching (qE) as shown by in vivo fluorescence measurements with a pulse amplitude modulation technique. Two different photothermal methods, photoacoustic spectroscopy and photothermal deflection spectroscopy, were used to monitor thermal deactivation of excited pigments during the dark-light transitions. The in vivo photothermal signals measured with both techniques were shown to remain constant during induction of photosynthesis under high light conditions, suggesting that, in contrast to current hypotheses, energy-dependent quenching qE is not associated with significant changes in thermal dissipation of absorbed light energy in the chloroplasts. When photosynthesis was induced with a low-intensity modulated light, a noticeable decrease in the heat emission yield was observed resulting from the progressive activation of the competing photochemical processes.  相似文献

本实验采用Wittig方法制得了未封端和封端的超支化聚三苯胺-对苯乙烯撑型聚合物,对两种共轭聚合物进行了表征和性能测试.聚合物溶液和固体膜在紫外光照射下均发出较强的绿光.首次对这类聚合物在硝基芳烃化合物荧光猝灭能力进行了初步研究,结果表明:与未封端产物相比,封端后的超支化共轭聚合物在邻硝基甲苯(o-NT)的荧光猝灭效率上有明显提高,当o-NT浓度为21.5×10-3mol/L时,荧光猝灭效率达到97%.这类共轭聚合物不仅合成操作较为简便,猝灭效率也较高,是一种很有潜力的硝基芳烃化合物荧光检测材料.  相似文献

The effect of different wavebands of artificial UV (UVB and UVA) and photosynthetically active radiation (PAR) was assessed in two species of the genus Ulva, U. olivascens and U. rotundata, from southern Spain in order to test for possible differences in acclimation of photosynthesis. Both species share similar morphology but are subject to different light environments: U. rotundata is an estuarine alga, inhabiting subtidal locations, while U. olivascens is an intertidal, sun-adapted organism. Algae were exposed to three different UV conditions, PAR+UVA+UVB, PAR+UVA and PAR for 7 d. Short-term exposure (6 h) was also carried out, using two PAR levels, 150 and 700 micromolm(-2)s(-1). Pigment contents and photosynthesis vs. irradiance curves from oxygen evolution were used to contrast sun- and shade adaptation between these species. O2-based net photosynthesis (Pmax) and PAM-chlorophyll fluorescence (optimal quantum yield, Fv/Fm) were used as parameters to evaluate photoinhibition of photosynthesis in the experiments. The results underline different photobiological characteristics among species: the subtidal U. rotundata had higher contents of pigments (Chl a, Chl b and carotenoids) than the sun-adapted U. olivascens, which resulted in higher thallus absorptance and P-I parameters characterized by higher photosynthetic efficiency at limiting irradiances (alpha) and lower saturating points for photosynthesis (Ek). After 7 d exposure, photoinhibition of Fv/Fm was close to 40-45% in both species. Differences between UV treatments were seen in U. rotundata after 5 d and after 7 d in U. olivascens, in which PAR+UVA impaired strongly photosynthesis (80%). Such patterns were correlated with a progressive decrease in pigment contents, specially chlorophylls. In short-term (6 h) exposures, combinations of UVA+UVB and high PAR level resulted in high rates of photoinhibition of chlorophyll fluorescence (68-92%) in U. rotundata, whereas in U. olivascens photoinhibition ranged between 42% and 53%. Photoinhibition under low PAR combined to UV radiation was lower than observed under high PAR. Net O2-Pmax revealed similar response among the species, with maximal photoinhibition rates close to 60% in algae incubated under high PAR+UVA+UVB. In the case of UV exposure in combination with low PAR, the highest photoinhibition rates were measured in U. rotundata.  相似文献
