Metabolic studies of ipriflavone (TC-80) in rats by gas-liquid chromatography-mass spectrometry led to the characterization of the following metabolites: the parent compound, 7-hydroxy-3-phenyl-4H-1-benzopyran-4-one, 7-hydroxy-3-(4-hydroxyphenyl)-4H-1-benzopyran-4-one, 3-(4-hydroxyphenyl)-7-isopropoxy-4H-1-benzopyran-4-one, 2-(3-phenyl-4-oxo-4H-1-benzopyran-7-yl)oxypropionic acid, 2-[3-(4-hydroxyphenyl)-4-oxo-4H-1-benzopyran-7-yl]oxypropionic acid and 2-[3-(3-hydroxyphenyl)-4-oxo-4H-1-benzopyran-7-yl]oxypropionic acid. From the metabolites identified, TC-80 was shown to be metabolized primarily by oxidation. In vitro study using tissue slices of rats indicated that the above metabolic changes occurred exclusively in the liver. It was also demonstrated that the compound did not undergo metabolic conversion by gut flora of rats.  相似文献

Di- and tri-substituted [(4-oxo-4H-1-benzopyran-7-yl)oxy]acetic acids, and 4-oxo-3-phenyl-4H-furo[2,3-h]-[1]benzopyran-8-carboxylic acid were synthesized and tested for natriuretic and uricosuric activities. Among the compounds tested, 3,5-disubstituted [(4-oxo-4H-1-benzopyran-7-yl)oxy]acetic acids (6c-f, h, n and x) showed potent natriuretic and uricosuric activities, whereas 4-oxo-3-phenyl-4H-furo[2,3-h][1]benzopyran-8-carboxylic acid (6dd) possessed only potent natriuretic activity. This paper describes synthesis of novel 4H-Chromeno[2,3-d] pyrimidin-2-one compounds by green chemistry method using l-proline as the catalyst. Firstly, a one-pot reaction of resorcinol, malononitrile, and benzaldehyde with proline in water at room temperature on reaction gave 2-amino-7-hydroxy-4-phenyl-4H-1-benzopyran-3-carbonitrile and in next step (2-amino-7-hydroxy-4-phenyl-4H-1-benzopyran-3-carbonitrile was alkylated in the presence of K2CO3 in dry acetone at 70–80 °C to give substituted benzopyran 3-carbonitriles. The amino and carbonitrile parts in the substrate were converted into different novel substituted chromeno derivatives on cyclization with urea. The presence of a pyrimidine ring makes this moiety highly biologically efficient.  相似文献

The new 2H,4H-[1]benzopyrano[3,4-b]pyridine-1,3,5-trione derivatives 10a-f were prepared in the following three steps: first the preparation of new N-(tert-butoxycarbonyl)-3-amino-2H-1-benzopyran-2-one derivatives 5a-f by reaction of coumarin-3-carboxylic acids and diphenylphosphorylazide, then hydrolysis of 5a-f with gaseous hydrogen chloride to give the corresponding amines 7a-f , and finally the preparation of 10a-f by reaction of 7a-f and carbon suboxide in the presence of a Lewis acid.  相似文献

Structural and spectroscopic elucidation of merocyanine dye, 2,5-[1-metyl-4-[2-(4-hydroxyphenyl)ethenyl)]piridinium]-hexane tetraphenylborate, is performed in gas and condense phase by means of solution and solid-state conventional and linear-polarized IR-spectroscopy of oriented colloids in nematic liquid crystal suspension, UV-vis and fluorescence methods, HPLC MS/MS tandem and ESI mass spectrometry, (1)H, (13)C and (1)H-(1)H COSY NMR, TGV and DSC methods. Daphwazirin, a new biscoumarin glycoside (1), along with four known compounds (2), (3), (4) and (5) has been isolated from the roots of Daphne oleoides. Abstract

Treatment of 3-(2-bromoacetyl)coumarins with ammonium dithiocarbamate provides 3-2-mercapto-4-thiazolyl)-2H-1-benzopyran-2-one (11) but not 4-hydroxy-4-(3-coumaririnyl) thiazolidine-2-thione (I). II undergoes smooth condensation with alkyl, aralkyl, phenacyl and acid halides to give corresponding thioethers and thioesters (III) respectively. The extraction of (135)Cs from high activity waste arising from reprocessing of spent fuel can be achieved by using calix[4]arene crown compounds. The radiolytic degradation of calix[4]arene crowns as well as their solvent, o-nitrophenyloctyl ether (NPOE), was studied using electrospray ionization mass spectrometry (ESI-MS) (that formed Cs(+) or Na(+) adducts) in nitric acid, as well as by chemical ionization tandem mass spectrometry (MS/MS) experiments. The structures of major degradation products were identified with MS and specifically labelled nitric acid. Although NPOE and calix[4]arene crowns alone are relatively stable, under simulated conditions resembling the real industrial processes involving radiolysis in the presence of nitric acid, several products resulting from nitration and oxidation were observed.  相似文献

Condensation of methyl 7-methylcoumarin-4-acetate ( 2 ) with primary amines and with anthranilic acid gave 7-methyl-2-oxo-N-aryl-2H-[1]-benzopyran-4-acetamide ( 4a—d ) and (7), respectively. Compound 7 underwent cyclization to give 2-(7-methyl-2-oxo-2H-[1]-benzopyran-4-yl)-methyl-4H-3,1-benzoxazin-4-one ( 3 ). The reaction of 3 with aromatic amines gave the corresponding quinazolone derivatives 5 which tautomerises to the thermodynamically more stable isomer 6 , whereas its reaction with Grignard reagents and aromatic aldehydes gave 8a, 8b , and 9a, 9b , respectively.  相似文献

[reaction: see text] 5,7-Dialkyl-4,6-dioxo-4,5,6,7-tetrahydroisothiazolo[3,4-d]pyrimidine-3-carbonitriles 4, prepared from 6-amino-1,3-dialkyluracils 3 and 4,5-dichloro-5H-1,2,3-dithiazolium chloride (Appel's salt) 1, are utilized for the preparation of new derivatives of 4 bearing amino, alkylthio, amido, thioamido, tetrazolyl, and carboximidic acid ethyl ester groups at position 3. Similarly, the reactions of 6-methyl-4-oxo-4H-1-aza-5-oxa-2-thiaindene-3-carbonitrile 8, prepared from 4-amino-6-methyl-2-pyrone 6 and 1, with alkyl- and arylamines in DMF at 50 degrees C and reflux afforded different isothiazole derivatives 11 and 17, respectively. On the other hand, treatment of 8 with 1,3-diaminopropane in THF at room temperature, followed by chromatography on silica gel, gave 3-(2-oxopropyl)-6,7,8-trihydro-4H-1-thia-2,5,9-triazacyclopentacyclononene-4,10-dione 12 in 59% yield.  相似文献

Two new compounds, 7'-(3',4'-dihydroxyphenyl)-N-[(4-methoxyphenyl)ethyl]propenamide (4), and 7'-(4'-hydroxy,3'-methoxyphenyl)-N-[(4-butylphenyl)ethyl]propenamide (5) have been isolated from Cuscuta reflexa along with five known compounds, 6,7-dimethoxy-2H-1-benzopyran-2-one (1), 3-(3,4-dihydroxyphenyl)-2-propen-1-ethanoate (2), 6,7,8-trimethoxy-2H-1-benzopyran-2-one (3), 3-(4-O-beta-D-glucopyranoside-3,5-dimethoxyphenyl)-2-propen-1-ol (6), 2-(3-hydroxy-4-methoxyphenyl)-3,5-dihydroxy-7-O-beta-D-glucopyranoside-4H-1-benzopyrane-4-one (7), reported for the first time from this species. Structures of these compounds were determined by spectral analysis. These compounds showed strong inhibitory activity against alpha-glucosidase.  相似文献

4-Amino-5-(4,6-diphenyl-2-pyrimidinyl)-3,4-dihydro-2H-1,2,4-trazole-3-thione is formed from the reaction of 4,6-diphenylpyrimidinecarboxylic acid or its ethyl ester with thiocarbonyl hydrazide. Alkylation of the product leads to S-alkyl derivaties or 6-substituted 3-(4,6-diphenyl-2-pyriimidinyl)-7H-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazine. Under microwave irradiation,2,2'-alkoxy-bridging or 4,4'-alkoxy-bridging dibenzaldehydes reacted with 4-hydroxycoumarin in DMF to give a series of 3,3',3″,3'″-o-and 3,3',3″,3'″-p-phenylenedimethylidinetetrakis-(4-hydroxy-2H-1-benzopyran-2-ones) in moderate yields.The structures of the synthetic coumarin derivatives were characterized with IR,1H NMR and MS spectroscopy as well as X-ray single crystallography.  相似文献

The reactivities of 3-acetyl-2H-1-benzo-pyran-2-one (2) and 3-acetyl-4-methyl-2H-1-benzopy-ran-2-one (3)are compared in the condensation with aromatic aldehydes. It is established that the benzopyrones 2 and 4 condense with various aromatic aldehydes in chloroform containing some drops of piperidine by boiling for 7 hours and azeotropic distillation of water, yielding 3-cinnamoyl-2H-1-benzopyran-2-ones (3a-g) and 4-(2-arylvinyl)-3-acetyl-2H-1-benzopyran-2-ones (6a-f), respectively. Evidence is provided that benzopyrone 4 participates in the condensation with the methyl group in position 4. A probable mechanism of the process is proposed.  相似文献
