Abstract— The photosynthetic accessory pigments R-phycoerythrin and C-phycocyanin were extracted from a red sea-weed and a filamentous blue-green alga. Investigation of quenching of phycoerythrin fluorescence by phycocyanin at room and liquid nitrogen temperatures yielded rates for the SternVolmer quenching constants ( Kq From these values, rate constants k(D–A) were calculated for the transfer of energy from phycoerythrin to phycocyanin.
The transfer rate constants obtained were such that a collisional transfer mechanism was ruled out, and were of similar order of magnitude as the rate constants for transfer of electronic energy in mixed organic crystals.  相似文献

Abstract— A radiation-resistant Micrococcus sp. and a colorless mutant subject to gamma radiation were found to have identical killing curves. Similar results were obtained with ultraviolet light. However when exposed to visible light in the presence of the photosensitizer, toluidine blue, the pigmented wild-type was unaffected whereas the colorless mutant was killed. Sarcina lutea and a colorless mutant were killed at similar rates following exposure to x-rays and U.V. light, whereas previous studies have demonstrated that the pigmented wild-type is protected against lethal photosensitizations by visible light. It is concluded that the effect of carotenoid pigments in radiation damage is limited to protecting cells against the lethal effects of visible light and is without effect at shorter wavelengths.  相似文献

Abstract— The isolation of a mutant, strain PM-9, of Rhodopseudonionus spheroides with an abnormal complement of carotenoid pigments is described.
PM-9 accumulates phytoene, phytofluene, ζ-carotene and neurosporene. Semi-aerobic cultures form more ζ-carotene and neurosporene relative to total carotenoids than do photo-synthetic cultures.
PM-9 is killed on exposure to light and oxygen.
By making use of the effect of oxygen on the nature of the carotenoids in PM-9, we have shown that these pigments do not directly influence the in vivo spectrum of bacteriochlorophyll.
Diphenylamine inhibits the synthesis of coloured carotenoids in Rhodopseudonionos gelatinosa but does not change the bacteriochlorophyll spectrum.  相似文献

Abstract
Linear dichroism and fluorescence depolarization measurements on chlorophyll a and chlorophyll b molecules incorporated into macroscopically oriented multibilayers of the plant lipid digalactosyldiacylglycerol (DGDG) are presented. The results are combined with measurements of fluorescence anisotropy to obtain both the order parameters describing the orientational statistics and the directions of the absorption and emission transition moments in the frame of the molecules. The problem presented by the overlapping nature of the absorption and emission bands is overcome by determining the fluorescence depolarization at the two maxima of the emission spectrum.  相似文献

A review of the chemical education research literature suggests that the term constructivism is used in two ways: experience-based constructivism and discipline-based constructivism. These two perspectives are examined as an epistemology in relation to the teaching and learning of the concept of idealization in chemistry. It is claimed that experience-based constructivism is powerless to inform the origin of such concepts in chemistry and while discipline-based constructivism can admit such theoretical concepts as idealization it does not offer any unique perspectives that cannot be obtained from other models. Chemical education researchers do not consistently appeal to constructivism as an epistemology or as a teaching/learning perspective and it is shown that, while it draws attention to worthwhile teaching/learning strategies, it cannot be considered as foundational to chemical education research and tends to be used more as an educational label than as an undergirding theory.  相似文献

Abstract. Analysis of the effect of multiple excitations on chlorophyll a fluorescence yields in the green alga Chlorella reveals several distinct reactions. The first excitation in dark-z-adapted units produces photochemistry and the high yield state with a rise time of 35 ns. It is ascribed to a change in coupling between the antenna pigments and the photochemical trap. The second hit produces with the same quantum yield a quenched state which changes to the high yield state with a rise time of 4 μ s. This is ascribed to the formation and the decay of a particular carotenoid triplet state near the funnel or antenna-trap junction. Further hits produce enhanced quenching assigned to mobile triplets with lifetimes in the order of 100 ns. The fluorescence yield decreases monotonically with increasing excitations during the 7 ns pulse. This effect can be adequately ascribed to annihilation of excitations with lifetimes longer than the trapping time, or by a unique model of a multi-trapped unit. The latter model is favored by arguments based both on the absence of a local maximum in the graph of fluorescence yield vs excitation energy and on the fact that the high yield state shows a different behaviour on multiple excitation, fit by a single-trapped unit. This analysis is related to that used in experiments with ps flashes and is applied to the qualitatively different bacterial system.  相似文献

Abstract— The intensity of chlorophyll a fluorescence during the early part of fluorescence induction at O , initial fluorescence, and P, peak fluorescence, was higher during the day phase of the circadian cycle than during the night phase in continuous light (LL) conditions and was positively correlated with the rate of oxygen evolution. The circadian rhythm in fluorescence in LL persisted in the presence of 10μM 3–(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU), which blocks electron flow from photo-system (PS) II in photosynthesis. The rhythmic changes in fluorescence intensity are consistent with a lower rate constant for radiationless transitions during the day phase than during the night phase of the circadian rhythmicity. The circadian changes in the intensity of fluorescence were abolished at 77K, which may indicate the importance of structural changes in membranes in circadian oscillations.  相似文献

Abstract— As an artificial model compound of the chlorophyll-protein complex in vivo , the chlorophyll/water-soluble macromolecular complexes were prepared by using synthetic linear polymers of polyvinylpyrrolidone (PVP), polyvinyl alcohol (PVA), polyethylene glycol (PEG), and a natural polymer of bovine serum albumin (BSA). By the method described here, it is possible to prepare an aqueous chlorophyll (Chl)-macromolecular complex solution of a desired Chi aggregate, such as: Chi a (670), Chi a (740) and Chi b ; and with a desired relative content and concentration. These procedures for preparing such complexes will have wide applicability for technical use in Chi studies. For example, extremely diluted aqueous complex solutions of at least 1 × 10-4% wt Chi a (670 or 740)-macromolecular complex / wt can be obtained without changing the spectral features. From viscosity measurements, the structures of the complexes were inferred: (1) for a linear macromolecular (PVA or PVP) complex, a Chi species is tightly surrounded by a chain of the polymer causing shrinkage of the chain; (2) globular BSA molecules surround Chi species to form a large complex. The mechanism of stabilization of Chi aggregates in thylakoid membrane was discussed concerning an analogy to the complexes studied here.  相似文献

Abstract— –The presumed photoreceptor for phototaxis, the paraflagellar body, in the flagellate Euglena gracilis , was isolated still attached to the flagellum. After solubilization, fast protein liquid chromatography (FPLC) analysis yielded four major protein fractions with the chromophoric groups still attached. Fluorescence spectra showed that three fractions had excitation peaks at 380 nm and emission peaks around 450 nm indicative of pterins, while the fourth chromoprotein had a fluorescence emission at 520 nm and an excitation peak at 450 nm, indicative of a flavin. The separated proteins were analyzed by gel electrophoresis: the pterin binding proteins have apparent molecular masses between 27 000 and 31 600 and the flavin binding protein has an apparent molecular mass of 33 500.  相似文献
