A simple HPLC-MS/MS method for simultaneous determination of loureirin A and loureirin B in rat urine, feces, and bile after oral administration of 10.6 g/kg of longxuejie (one rare traditional Chinese medicinal herb) was developed for the first time. The analytes and buspirone (internal standard) were separated on a C5 column with acetonitrile–water (containing 0.1% formic acid) as mobile phase at a flow rate of 0.4 min/mL. The detector was a Q-trap™ mass spectrometer with an electrospray ionization interface operating in the multiple reaction monitoring mode. Calibration curves of loureirin A in rat urine, feces, and bile were linear over the concentration range of 1.00–5,000 ng/mL. Loureirin B in rat urine, feces, and bile ranged between 0.08 and 20, 0.20 and 20, and 0.10 and 500 ng/mL, respectively. Validation revealed that the method was specific, accurate, and precise. The fully validated method was applied to the excretion study of loureirin A and loureirin B in rats. After oral administration of 10.6 g/kg longxuejie, cumulative excretion amount of loureirin A and loureirin B in rat urine were 2.94 ± 0.81 and 0.36 ± 0.16 μg at 72 h, respectively. Of the total dose, 5.35% of loureirin A and 5.46% of loureirin B were excreted from feces at 60 h. The cumulative amounts of loureirin A and loureirin B in rat bile reached 4.49 ± 0.98 and 5.11 ± 0.83 μg, respectively, at 36 h after dosing, accounting for 0.054% and 0.056% of the total dose.  相似文献

The effectiveness of N,N-dimethyl-2-amino-1,2-diphenylethanol as a chiral ligand was significantly improved via simple hydrogenation of the phenyl rings. Both the catalytic activity and the enantioselectivity of the resulting new ligand in the asymmetric conjugate addition of diethylzinc to enones were found to be superior to the original phenyl analog.  相似文献

A rapid and sensitive reversed-phase high-performance liquid chromatography (HPLC) method is developed to determine the concentration of ibuprofen in human plasma. Ibuprofen is isolated from plasma by adding 0.50 mL of acetonitrile to 1.0 mL of plasma. The endogenous substances precipitated by acetonitrile are separated by centrifugation. The supernatant is saturated with ammonium sulfate to salt-out the acetonitrile. The salted-out acetonitrile is injected directly into the HPLC system. A 150-mm x 4.6-mm column packed with 3-microns reversed-phase octadecylsilane particles (C18) is used for the method finally developed. The mobile phase is a 1:1 ratio of acetonitrile-phosphoric acid (pH 2.2). Ibuprofen is monitored with a UV-visible detector at 220 nm and 0.10-0.002 absorbance units full scale (A.U.F.S.). The mean percent of relative standard deviations for within-day and between-day analyses are less than 3. The limit of detection for ibuprofen (in human plasma) is 25 ng/mL for a 100-microL injection volume. Quantitation of ibuprofen in human plasma at 100 ng/mL can be achieved with a relative standard deviation of less than 5%. The completion time of assay is less than 20 minutes.  相似文献

This study presents a high‐performance liquid chromatography–tandem mass spectrometry (HPLC–MS/MS) method for the simultaneous determination of antofloxacinin and its main metabolite – N ‐demethylated metabolite (N‐ DM) – in human urine. Ornidazole was used as the internal standard. This was a clinical urine recovery study, in which 10 healthy Chinese volunteers were intravenously administered a single 200 mg dose of antofloxacin hydrochloride. Compounds were extracted by albumen precipitation, after which samples were isocratically eluted using a Poroshell 120 SB‐C18 column, and were analysed using HPLC–MS/MS under electronic spray ionization positive ion mode. The method was successfully applied in a urine pharmacokinetic study of antofloxacinin, with a detection range of 0.02/0.01 to 200/100 μg/mL (for antofioxacin/N‐ DM).The average percentages of antofioxacin/N‐ DM measured in urinary excretion frp, 10 volunteers were 54.9 ± 5.7/8.2 ± 2.5% in 120 h duration.  相似文献

A variety of electrophiles (anhydrides, acid chlorides, carbonochloridates, sulfonyl chlorides, and alkyl bromides) react with 3-methoxy-2H-indazole (1a), benzoxazin[3,2-b]indazole (1d), and oxazolino[3,2-b]indazole (1e)?-?substrates available by the Davis-Beirut reaction?-?to yield a diverse set of N(1),N(2)-disubstituted-1H-indazolones. With certain electrophiles, an AERORC (Addition of the Electrophile, Ring Opening, and Ring Closure) process on indazole 1d results in indazoloindazolone formation. An intriguing aspect of these N(1),N(2)-disubstituted-1H-indazolones is that they are poised for diversification through, for example, azide-alkyne cycloaddition chemistry reported here.  相似文献

Metabonomic screening of human urine samples using 1H NMR spectroscopy has revealed the presence of signals resulting from the excretion of ethyl glucoside. Experiments in volunteers have demonstrated that this ethyl glucoside results from dietary exposure to the compound, which is present in beverages such as rice wine and sake, rather than representing a new route for the metabolism of ethanol by humans. The limited studies undertaken in volunteers indicate that ethyl glucoside has a longer biological half life than ethanol itself. The potential problems associated with using this glucoside metabolite as a marker of ethanol consumption are considered.  相似文献

First synthesis of C2-symmetric chiral O,N,N,O-tetradentate 2,2-bipyridyldiolpropane ligands is described. The Mukaiyama–Michael reaction was applied as an important reaction for the synthesis of 2,2-bipyridylpropane 9. Among the ligands synthesized, ligand 11 exhibits excellent chiral induction (up to 97% ee) in diethylzinc addition to various aldehydes. The use of additional Lewis acid such as Ti(OiPr)4 in diethylzinc addition reaction is not required for the present catalytic system.  相似文献

The axially chiral 2-substituted N,N-diisopropyl-1-naphthamides 1 and 2 were resolved by HPLC over a chiral stationary phase to provide enantiomerically pure atropisomers. The absolute stereochemistry of (−)-syn-1 was determined by X-ray crystallographic analysis of the corresponding (1S)-camphanic acid ester derivative. Desymmetrization of cyclic meso anhydrides 5a and 5b using (−)-syn-1 gave a single diastereomer in good yield.  相似文献

According to the density functional theory calculations, the X···H···N (X?N, O) intramolecular bifurcated (three‐centered) hydrogen bond with one hydrogen donor and two hydrogen acceptors causes a significant decrease of the 1hJ(N,H) and 2hJ(N,N) coupling constants across the N? H···N hydrogen bond and an increase of the 1J(N,H) coupling constant across the N? H covalent bond in the 2,5‐disubsituted pyrroles. This occurs due to a weakening of the N? H···N hydrogen bridge resulting in a lengthening of the N···H distance and a decrease of the hydrogen bond angle at the bifurcated hydrogen bond formation. The gauge‐independent atomic orbital calculations of the shielding constants suggest that a weakening of the N? H···N hydrogen bridge in case of the three‐centered hydrogen bond yields a shielding of the bridge proton and deshielding of the acceptor nitrogen atom. The atoms‐in‐molecules analysis shows that an attenuation of the 1hJ(N,H) and 2hJ(N,N) couplings in the compounds with bifurcated hydrogen bond is connected with a decrease of the electron density ρH···N at the hydrogen bond critical point and Laplacian of this electron density ?2ρH···N. The natural bond orbital analysis suggests that the additional N? H···X interaction partly inhibits the charge transfer from the nitrogen lone pair to the σ*N? H antibonding orbital across hydrogen bond weakening of the 1hJ(N,H) and 2hJ(N,N) trans‐hydrogen bond couplings through Fermi‐contact mechanism. An increase of the nitrogen s‐character percentage of the N? H bond in consequence of the bifurcated hydrogen bonding leads to an increase of the 1J(N,H) coupling constant across the N? An interlaboratory study was conducted to evaluate a method for determining total soy isoflavones in dietary supplements, dietary supplement ingredients, and soy foods. Isoflavones were extracted using aqueous acetonitrile containing a small amount of dimethylsulfoxide (DMSO) and all 12 of the naturally occuring isoflavones in soy were determined by high-performance liquid chromatography (HPLC) with UV detection using apigenin as an internal standard. Fifteen samples (6 pairs of blind duplicates plus 3 additional samples) of soy isoflavone ingredients, soy isoflavone dietary supplements, soy flour, and soy protein products were successfully analyzed by 13 collaborating laboratories in 6 countries. For repeatability, the relative standard deviations (RSDr) ranged from 1.07 for samples containing over 400 mglg total isoflavones to 3.31 for samples containing 0.87 mg/g total isoflavones, and for reproducibility the RSDR values ranged from 2.29 for samples containing over 400 mg/g total isoflavones to 9.36 for samples containing 0.87 mg/g total isoflavones. HorRat values ranged from 1.00 to 1.62 for all samples containing at least 0.8 mg/g total isoflavones. One sample, containing very low total isoflavones (< 0.05 mg/g), gave RSDR values of 175 and a HorRat value of 17.6. This sample was deemed to be below the usable range of the method. The method provides accurate and precise results for analysis of soy isoflavones in dietary supplements and soy foods.  相似文献

A rapid and simple reverse‐phase high‐performance liquid chromatography (RP‐HPLC) was developed and validated for the quantification of kirenol in rat plasma after oral administration. Kirenol and darutoside (internal standard, IS) were extracted from rat plasma using Cleanert™ C18 solid‐phase extraction (SPE) cartridge. Analysis of the extraction was performed on a Thermo ODS‐2 Hypersil C18 reversed‐phase column with a gradient eluent composed of acetonitrile and 0.1% phosphoric acid. The flow rate was 1.0 mL/min and the detection wavelength was set at 215 nm. The calibration curve was linear over the range of 9.756–133.333 µg/mL (r2 = 0.9991) in rat plasma. The lower limits of detection and quantification were 2.857 and 9.756 µg/mL, respectively. The intra‐ and inter‐day precisions (relative standard deviation, RSD) were between 2.24 and 4.46%, with accuracies ranging from 91.80 to 102.74%. The extraction recovery ranged from 98.16 to 107.62% with RSD less than 4.81%. Stability studies showed that kirenol was stable in preparation and analytical process. The present method was successfully applied to the pharmacokinetic study of kirenol in male Sprague–Dawley rats after oral administration at a dose of 50 mg/kg. We use the path integral ground state method to study the energetic and structural properties of small para-H2 clusters of sizes ranging from 2 to 20 molecules. A fourth order formula is used to approximate the short imaginary-time propagator and two interaction potentials are considered. Our results are compared to those of exact basis set calculations and other quantum Monte Carlo methods when available. We find that for all cluster sizes considered, our results show a lower ground state energy than literature values obtained by diffusion Monte Carlo and variational Monte Carlo. For the dimer and trimer, ground state energies are in good agreement with exact results obtained using the discrete variable representation. Structural properties are found to be insensitive to the choice of interaction potential. We explore the use of Pekeris coordinates to analyze the importance of linear arrangement in trimers and for trimers within clusters of larger size.  相似文献

This paper reports a novel methodology for the conformational analysis of [2]rotaxanes. It combines NMR spectroscopic (COSY, NOESY and the recently reported paramagnetic line-broadening and suppression technique) and electrochemical techniques to enable a quantitative analysis of the co-conformations of interlocked molecules and the conformations of their components. This methodology was used to study a model [2]rotaxane in solution. This [2]rotaxane consists of an axle that incorporates an electron-poor, doubly positively charged viologen that threads an electron-rich crown ether. It has been shown that the axle of the [2]rotaxane in its dicationic state adopts a folded conformation in solution and the crown ether is localised at the viologen moiety. Following a one-electron reduction of viologen, the paramagnetic radical cation of the [2]rotaxane retains its folded conformation in solution. The data also demonstrate that in the radical cation the crown ether remains localised at the viologen, despite its reduced affinity for the singly reduced viologen. The combined quantitative NMR spectroscopic and electrochemical characterisation of the electromechanical function of the model [2]rotaxane in solution provides an important reference point for the study of switching in structurally related bistable [2]rotaxanes, which is the subject of the second part of this work.  相似文献

An ultra‐performance liquid chromatography (UPLC) method coupled with photodiode array (PDA) detection has been developed and validated for simultaneous quantification of 12 active components (berberine, palmatine, jatrorrhizine, aloe‐emodin, rhein, emodin, chrysophanol, physcion, baicalin, baicalein, wogonoside and wogonin) in Yiqing granule. Optimum separation were achieved on a C18 column (50 × 2.1 mm i.d., 1.7 µm particle) through a 7.5 min gradient delivery of a mixture of A (acetonitrile) and B (0.1% aqueous phosphoric acid containing 1.8 mmol/L sodium dodecyl sulfonate and 10% acetonitrile, v/v) at a flow rate of 0.3 mL/min at 30°C. Because of the different UV characteristics of these components, three detection wavelengths were used for quantitative analysis. All of the analytes showed good linearity (r of >0.999). The method was validated for repeatability, precision, stability, accuracy and selectivity. The validated method was applied to quality control of Yiqing granule from different production batches. A rapid, simple and sensitive liquid chromatography–tandem mass spectrometry (LC/MS/MS) was developed for the determination of an antiepileptic drug, lacosamide, in rat plasma. The method involves the addition of acetonitrile and internal standard solution to plasma samples, followed by centrifugation. An aliquot of the supernatant was diluted with water and directly injected into the LC/MS/MS system. The separations were performed on column packed with octadecylsilica (5 µm, 2.0 × 50 mm) with 0.1% formic acid and acetonitrile as mobile phase, and the detection was performed on tandem mass spectrometry by the multiple‐reaction monitoring via an electrospray ionization source. The standard curve was linear over the concentration range from 0.3 to 1000 ng/mL. The lower limit of quantification was 0.3 ng/mL using 50 μL of rat plasma sample. The intra‐ and inter‐assay precision and accuracy were found to be less than 11.7 and 8.8%, respectively. An interlaboratory study was conducted for evaluation of a method to determine the flavonol aglycones quercetin, kaempferol, and isorhamnetin in Ginkgo biloba products. The method calculates total glycosides based on these aglycones formed after acid hydrolysis. Twelve matrixes were chosen for study by 12 collaborating laboratories in 2 countries. Test materials included crude leaf material, standardized dry powder extract, single and multiple entity finished products, ethanol and glycerol tinctures, and National Institute of Standards and Technology (NIST) standard reference materials (SRMs). Results from 11 laboratories were used for the final calculations. Eight of the 12 matrixes evaluated produced acceptable results for total flavonol glycosides, with HorRat scores ranging from 1.31 to 2.05; repeatability relative standard deviations (RSDr) from 1.46 to 4.14; and reproducibility relative standard deviations (RSDR) from 4.67 to 9.69. These 8 matrixes consisted primarily of simple dosage forms (e.g., dry powder extracts, crude leaf samples, liquid extracts, and SRMs) and a single tablet product (Ginkgo Awareness). Four additional matrixes, consisting of 3 tablets and 1 soft gel product (Ginkgold, Ginkoba, Ginkogen, and Ginkgo Phytosome, respectively), showed greater total flavonol glycoside HorRat scores in comparison, ranging from 2.39 to 5.13, with RSDr values from 2.83 to 8.16, and RSDR values from 8.53 to 20.4. Based on the results presented here, the method is recommended for Official First Action for determination of total flavonol glycosides calculated from quercetin, kaempferol, and isorhamnetin in dry powder extracts, crude leaf material, liquid extracts, and a select finished product, Ginkgo Awareness.  相似文献

Structural and thermodynamic characteristics of heteroelement inorganic heterocycles M1M2E1E2H8 (M1, M2 = Al, Ga, In; E1, E2 = N, P, As) were calculated by the density functional theory B3LYP/LANL2DZ(d,p) method. It was shown that energetic characteristics of heterocycle dissociation processes can be calculated by simple a additive scheme with the use of the average M-E bond energy. Dissociation of heteroelement heterocycles into monomeric H2MEH2 molecules proceeds according to the hardsoft acid-base (HSAB) concept.  相似文献

A specific and sensitive LC‐MS/MS assay was developed to simultaneously quantify three structurally similar flavonoid glycosides – hyperin, reynoutrin and guaijaverin – in mouse plasma. Biosamples were prepared by solid‐phase extraction. Isocratic chromatographic separation was performed on an AichromBond‐AQ C18 column (250 × 2.1 mm, 5 μm) with methanol–acetonitrile–water–formic acid (20:25:55:0.1) as the mobile phase. Detection of hyperin, reynoutrin, guaijaverin and internal standard [luteolin‐7‐Oβ‐d ‐apiofuranosyl‐(1 → 6)‐β‐d ‐glucopyranoside] was achieved by ESI‐MS/MS in the negative ion mode using m/z 463 → m/z 300, m/z 433 → m/z 300, m/z 433 → m/z 300 and m/z 579 → m/z 285 transitions, respectively. Linear concentration ranges of calibration curves were 4.0–800.0 ng/mL for hyperin and reynoutrin and 8.0–1600.0 ng/mL for guaijaverin when 100 μL of plasma was analyzed. We used this validated method to study the pharmacokinetics of hyperin, reynoutrin and guaijaverin in mice following oral and intravenous administration. All three quercetin‐3‐O‐glycosides showed poor oral absorption in mice, and the absolute bioavailability of hyperin after oral administration of 100 mg/kg was 1.2%. Pretreatment with verapamil increased the peak concentration and area under the concentration–time curve of hyperin, which were significantly higher than the control values.
