This study uses atomic force microscopy (AFM) force-deformation (F-Δ) curves to investigate for the first time the Young's modulus of a phospholipid microbubble (MB) ultrasound contrast agent. The stiffness of the MBs was calculated from the gradient of the F-Δ curves, and the Young's modulus of the MB shell was calculated by employing two different mechanical models based on the Reissner and elastic membrane theories. We found that the relatively soft phospholipid-based MBs behave inherently differently to stiffer, polymer-based MBs [Glynos, E.; Koutsos, V.; McDicken, W. N.; Moran, C. M.; Pye, S. D.; Ross, J. A.; Sboros, V. Langmuir2009, 25 (13), 7514-7522] and that elastic membrane theory is the most appropriate of the models tested for evaluating the Young's modulus of the phospholipid shell, agreeing with values available for living cell membranes, supported lipid bilayers, and synthetic phospholipid vesicles. Furthermore, we show that AFM F-Δ curves in combination with a suitable mechanical model can assess the shell properties of phospholipid MBs. The "effective" Young's modulus of the whole bubble was also calculated by analysis using Hertz theory. This analysis yielded values which are in agreement with results from studies which used Hertz theory to analyze similar systems such as cells.  相似文献

Atomic force microscope (AFM) imaging and nanoindentation measurements in water were used to probe the mechanical properties of retinal pigment epithelium melanosomes isolated from 14-year-old and 76-year-old donors. Topographic imaging reveals surface roughness similar to previous measurements on dry melanosomes. Force-indentation measurements show different types of responses that were catalogued into four different categories. In these measurements no permanent surface damage of melanosomes was observed as revealed by imaging before and after indentation measurements. The indentation measurements that exhibited nearly elastic responses were used to determine the Young's modulus of melanosomes. The average Young's modulus values are similar for 14-year-old and 76-year-old melanosomes with a somewhat narrower distribution for the 14-year-old sample. These elastic modulus values are considerably higher than the modulus of organelles with cytoplasm (<1 MPa) and approaching values of the modulus of protein crystals (approximately 100 MPa) indicating rather high packing density of biologic material in melanosomes. The width of the Young's modulus distributions is considerable spanning from few megapascals to few tens of megapascals indicating large heterogeneity in the structure. A fraction of the force curves cannot be described by the homogeneous elastic sample model; these force curves are consistent with approximately 10 nm structural heterogeneity in melanosomes. The approach-withdraw hysteresis indicates a significant viscoelasticity, particularly in the samples from the 14-year-old sample. Adhesion of the AFM probe was detected on approximately 3% and approximately 20% of the surface of 14-year-old and 76-year-old samples, respectively. In light of previous studies on these same melanosomes using photoelectron emission microscopy, this adhesion is attributed to the presence of lipofuscin on the surface of the melanosomes. This suggestion indicates that part of the difference in photochemical properties between the old and young melanosomes originates from surface lipofuscin.  相似文献

Investigation of the interactions between cells and material surfaces is important not only for the understanding of cell biology but also for the development of smart biomaterials. In this study, we investigated the substrate-related effects on the interaction between cell and polymeric ultrathin film (nanosheet) by modulating the mechanical properties of the nanosheet with a metal substrate or mesh. A freestanding polymeric nanosheet with tens-of-nanometers thickness composed of poly(L-lactic acid) (PLLA nanosheet) was fabricated by combination of a spin-coating technique and a water-soluble sacrificial layer. The freestanding PLLA nanosheet was collected on a stainless steel mesh (PLLA-mesh) and subsequently used for cell adhesion studies, comparing the results to the ones on a control SiO(2) substrate coated with an ultrathin layer of PLLA (PLLA-substrate). The adhesion of rat cardiomyocytes (H9c2) was evaluated on both samples after 24 h of culture. The PLLA-mesh with the tens-of-nanometers thick nanosheets induced an anisotropic adhesion of H9c2, while H9c2 on the PLLA-substrate showed an isotropic adhesion independent from the nanosheet thickness. Interestingly, an increment in the nanosheet thickness in the PLLA-mesh samples reduced the cellular anisotropy and led to a similar morphology to the PLLA-substrate. Considering the huge discrepancy of Young's modulus between PLLA nanosheet (3.5-4.2 GPa) and metal substrate (hundreds of GPa), cell adhesion was mechanically regulated by the Young's modulus of the underlying substrate when the thickness of the PLLA nanosheet was tens of nanometers. Modulation of the stiffness of the polymeric nanosheet by utilizing a rigid underlying material will allow the constitution of a unique cell culture environment.  相似文献

Cables insulated with plasticized poly(vinyl chloride) were aged in air at temperatures between 80 °C and 180 °C and their conditions were assessed by indenter modulus measurements, tensile testing, infrared (IR) spectroscopy and differential scanning calorimetry (DSC). Electrical testing of oven-aged cable samples was performed in order to relate the electrical functionality during a high-energy line break (HELB) to the mechanical properties and to establish a lifetime criterion. The mechanical data taken at room temperature after ageing could be superimposed with regard to ageing time and temperature. The ageing-temperature shift factor showed an Arrhenius temperature dependence. The jacketing material showed an immediate increase in stiffness (indenter modulus and Young's modulus) and a decrease in the strain at break on ageing; these changes were dominated by loss of plasticizer by migration which was confirmed by IR spectroscopy and DSC. The core insulation showed smaller changes in these mechanical parameters; the loss of plasticizer by migration was greatly retarded by the closed environment, according to data obtained by IR spectroscopy and DSC, and the changes in the mechanical parameters were due to chemical degradation (dehydrochlorination). A comparison of data obtained from this study and data from other studies indicates that extrapolation of data for the jacketing insulation can be performed according to the Arrhenius equation even down to service temperatures (20-50 °C). The low-temperature deterioration of the jacketing is, according to this scheme, dominated by loss of plasticizer by migration.  相似文献

The roughness and softness of interacting surfaces are both important parameters affecting the capillary condensation of water in apolar media, yet are poorly understood at present. We studied the water capillary adhesion between a cellulose surface and a silica colloidal probe in hexane by AFM force measurements. Nanomechanical measurements show that the Young's modulus of the cellulose layer in water is significantly less (~7 MPa) than in hexane (~7 GPa). In addition, the cellulose surface in both water and hexane is rather rough (6-10 nm) and the silica probe has a comparable roughness. The adhesion force between cellulose and silica in water-saturated hexane shows a time-dependent increase up to a waiting time of 200 s and is much (2 orders of magnitude) lower than that expected for a capillary bridge spanning the whole silica probe surface. This suggests the formation of one or more smaller bridges between asperities on both surfaces, which is confirmed by a theoretical analysis. The overall growth rate of the condensate cannot be explained from diffusion mediated capillary condensation alone; thin film flow due to the presence of a wetting layer of water at both the surfaces seems to be the dominant contribution. The logarithmic time dependence of the force can also be explained from the model of the formation of multiple capillary bridges with a distribution of activation times. Finally, the force-distance curves upon retraction show oscillations. Capillary condensation between an atomically smooth mica surface and the silica particle show less significant oscillations and the adhesion force is independent of waiting time. The oscillations in the force-distance curves between cellulose and silica may stem from multiple bridge formation between the asperities present on both surfaces. The softness of the cellulose surface can bring in additional complexities during retraction of the silica particle, also resulting in oscillations in the force-distance curves.  相似文献

Polypropylene/hemp or sisal fibre composites exhibit interesting recyclability [Bourmaud A, Baley C. Investigations on the recycling of hemp and sisal fibre-reinforced polypropylene composites. Polymer Degradation and Stability 2007;92(6):1034-45]. The obtained results prove that the tensile modulus of these polypropylene/vegetal fibre composites is well conserved with the number of reprocessing cycles. In this work, we investigated the relationship between the mechanical properties of the fibres and those of the composites by taking the influence of the recycling into account.In the first part of this study we carried out nanoindentation and tensile tests on vegetal fibres to obtain transversal and longitudinal Young's moduli. The experimental values were then introduced into micromechanical models, taking the aspect ratio changes into account, to estimate the stiffness of the PP/vegetal fibre injected composites before and after recycling. The first results show an interesting correlation between experimental results and model predictions; however a general underestimation of tensile stiffness of composites can be noticed.  相似文献

We report on the study of the nanomechanical properties of a lactate oxidase (LOx) monolayer immobilized on gold substrates by atomic force microscopy techniques operating under buffer conditions. Topographical contact mode imaging evidenced the protein deformation under the applied tip load. We performed approaching force curves with both stiff and soft cantilevers by imposing maximum loads of 1.6 nN and 400 pN, respectively. We found that the experimental data were well fitted by the Hertz model for a conical indenter. The use of two types of cantilevers allowed us to check further the consistency of the applicability of the Hertz model to the experimental data. After analyzing 180 curves, we obtained an average value of Young's modulus for the LOx layer in the 0.5-0.8 GPa range. These results agreed with those obtained for LOx submonolayer deposits on mica substrates, which allows discarding any important contribution from the underlying substrate on the measured properties. This range of values is closer to those obtained by other techniques on other globular proteins in comparison with those reported in previous AFM studies on similar systems. We found that for our experimental conditions the force curves can be, in principle, well fitted by the Hertz model for both conical and spherical indenter geometries. However, as the Young's modulus obtained for both geometries can differ appreciably, it becomes necessary to assess which indenter geometry is more adequate to explain the experimental data. For such purpose a systematic study of the indentation versus applied force curves obtained from both fittings for all the experimental curves was done.  相似文献

Mechanical properties of model and natural gels have recently been demonstrated to play an important role in various cellular processes such as adhesion, proliferation, and differentiation, besides events triggered by chemical ligands. Understanding the biomaterial/cell interface is particularly important in many tissue engineering applications and in implant surgery. One of the final goals would be to control cellular processes precisely at the biomaterial surface and to guide tissue regeneration. In this work, we investigate the substrate mechanical effect on cell adhesion for thin polyelectrolyte multilayer (PEM) films, which can be easily deposited on any type of material. The films were cross linked by means of a water-soluble carbodiimide (EDC), and the film elastic modulus was determined using the AFM nanoindentation technique with a colloidal probe. The Young's modulus could be varied over 2 orders of magnitude (from 3 to 400 kPa) for wet poly(L-lysine)/hyaluronan (PLL/HA) films by changing the EDC concentration. The chemical changes upon cross linking were characterized by means of Fourier transform infrared spectroscopy (FTIR). We demonstrated that the adhesion and spreading of human chondrosarcoma cells directly depend on the Young's modulus. These data indicate that, besides the chemical properties of the polyelectrolytes, the substrate mechanics of PEM films is an important parameter influencing cell adhesion and that PEM offer a new way to prepare thin films of tunable mechanical properties with large potential biomedical applications including drug release.  相似文献

Sonicated small unilamellar egg yolk phosphatidylcholine (EggPC) vesicles were investigated using atomic force microscopy (AFM) imaging and force measurements. Three different topographies (convex, planar, and concave shape) of the EggPC vesicles on the mica surface were observed by tapping mode in fluid, respectively. It was found that the topography change of the vesicles could be attributed to the interaction force between the AFM tip and vesicles. Force curves between an AFM tip and an unruptured vesicle were obtained in contact mode. During approach, two breaks corresponding to the abrupt penetration of upper and lower bilayer of vesicle were exhibited in the force curve. Both breaks spanned a distance of around 4 nm close to the EggPC bilayer thickness. Based on Hertz analysis of AFM approach force curves, the Young's modulus (E) and the bending modulus (kc) for pure EggPC vesicles were measured to be (1.97 +/- 0.75) x 10(6)Pa and (0.21 +/- 0.08) x 10(-19)J, respectively. The results show that the AFM can be used to obtain good images of intact and deformed vesicles by tapping mode, as well as to probe the integrity and bilayer structure of the vesicles. AFM force curve compare favorably with other methods to measure mechanical properties of soft samples with higher spatial resolution.  相似文献

Indentation is a comparatively simple and virtually nondestructive method of determining mechanical properties of material surfaces by means of an indenter inducing a localized deformation. The paper present experimental results of the load-displacement curves, the hardness and the elastic modulus data, and associated analysis for poly(methyl methacrylate) (PMMA) surfaces as a function of contact displacement. The experimental results include continuous stiffness indentations performed using constant loading rate and constant displacement rate experiments. The continuous stiffness indentation involves continuous calculation of a material stiffness, and hence hardness and elastic modulus of surfaces, during discrete loading-unloading cycles, as in a conventional indentation routine, and in a comparatively smaller time constant. The dependence of the compliance curves, the hardness, the elastic modulus and the plasticity index upon the imposed penetration depth, the applied normal load and the deformation rate are described. Tip area and load frame calibrations for the continuous stiffness indentation are also reported. The paper includes practical considerations encountered during indentation of polymers specifically at low penetration depths. The experimental results show a peculiarly harder response of PMMA surfaces at the submicron (near to surface) layers.  相似文献

We have measured the relaxation modulus in the temperature range 150–220°C of two samples of poly(vinyl chloride) resin with different molecular weights. The data were treated by the principle of reduced variables to yield composite curves. The shift factors (aT) when plotted against reciprocal temperature gave good straight lines from which apparent activation energies were obtained. An apparent activation energy of 50 kcal/mole was obtained for both samples. A relaxation spectrum for each resin was calculated from the relaxation modulus data. These spectra showed a marked molecular weight dependence. The spectra were in the range characteristic of the terminal zone of the entanglement plateau. Zero-shear-rate viscosities were obtained from the integration of relaxation modulus plots. From extrapolation of capillary viscosity data it is shown that the viscosity of the higher molecular weight resin used in this study does not approach its zero-shear value until shear rates less than 10?3 sec?1 are reached. The effect of supermolecular flow units is briefly discussed.  相似文献

Van der Waals equation of state as well as power laws and critical exponent theories are prototypes to study the cubic shape, asymmetries and “flatness” of the vapor–liquid equilibrium curves near the critical point. In this work we study two similar methods to determine the phase curves in analytical form, which differ from each other by simplicity of mathematical calculation. We analyze temperature dependence of the coexistence curves asymptotically close to the vapor–liquid critical point. We explain the novelty of our method with respect to the standard thermodynamic limit discussed in the literature. Therefore we show that the shape of the coexistence curves can strongly influence the accepted value of the critical exponent. The results of theoretical studies have been compared with the ones obtained by experimental methods.  相似文献

A method to measure interfacial mechanical properties at high temperatures and in a controlled atmosphere has been developed to study anodized aluminum surface coatings at temperatures where the interior aluminum alloy is molten. This is the first time that the coating strength has been studied under these conditions. We have investigated the effects of ambient atmosphere, temperature, and surface finish on coating strength for samples of aluminum alloy 7075. Surprisingly, the effective Young's modulus or strength of the coating when tested in air was twice as high as when samples were tested in an inert nitrogen or argon atmosphere. Additionally, the effective Young's modulus of the anodized coating increased with temperature in an air atmosphere but was independent of temperature in an inert atmosphere. The effect of surface finish was also examined. Sandblasting the surface prior to anodization was found to increase the strength of the anodized coating with the greatest enhancement noted for a nitrogen atmosphere. Machining marks were not found to significantly affect the strength.  相似文献

Atomic force microscopy (AFM) has the advantage of obtaining mechanical properties as well as topographic information at the same time. By analyzing force-distance curves measured over two-dimensional area using Hertzian contact mechanics, Young's modulus mapping was obtained with nanometer-scale resolution. Furthermore, the sample deformation by the force exerted was also estimated from the force-distance curve analyses. We could thus reconstruct a real topographic image by incorporating apparent topographic image with deformation image. We applied this method to carbon black reinforced natural rubber to obtain Young's modulus distribution image together with reconstructed real topographic image. Then we were able to recognize three regions; rubber matrix, carbon black (or bound rubber) and intermediate regions. Though the existence of these regions had been investigated by pulsed nuclear magnetic resonance, this paper would be the first to report on the quantitative evaluation of the interfacial region in real space.  相似文献

The Johnson-Kendall-Roberts (JKR) theory of elastic contact, extended to take viscoelastic effects into account, is used to evaluate work of adhesion and modulus of elastomeric films. In this paper, we present a comparison of five approaches to analyze quasi-static and dynamic JKR force curve data obtained using instrumented indentation. The load-displacement experiments were performed using a 200-microm radius borosilicate glass sphere against poly(dimethyl siloxane) (PDMS). By applying a small oscillation to the tip during indentation, dynamic stiffness vs load data were also obtained for frequencies between 25 and 160 Hz. Direct curve fitting as well as simplified 2- and 3-point analysis methods were used to compare modulus values obtained from load-displacement and stiffness-load data. Fit methods not requiring determination of the initial point of tip-sample contact ("zero" displacement) provided modulus values closest to those obtained by direct curve fitting. The dynamic stiffness-load data revealed a frequency dependent modulus; load-displacement measurements obtained simultaneously were consistent with the relaxed, or low-frequency, modulus of the PDMS sample. These experiments demonstrate that both the frequency dependent and relaxed modulus can be obtained from a single experiment.  相似文献

The influence of the thermal history on the morphology and mechanical behavior of PET was studied. The degree of crystallinity (density measurements) and the morphological structure (electron microscopy and small-angle x-ray diffraction) depend on the crystallization temperature. The viscoelastic parameters obtained from the modulus–temperature curves are mainly determined by the morphology of the samples. The glass-transition temperature, Ti, is a function of the crystallinity and the crystallization temperature. It is maximum for a crystallinity between 0.34 and 0.39 for a sample crystallized isothermally between 120 and 150°C. This dependence on crystallization conditions is ascribed to the conformation of the amorphous chain segments between the crystalline lamellae as well as the concentration and the molecular weight of the polymer material rejected during isothermal crystallization. Both factors are supposed to be temperature-dependent. The value of the rubbery modulus is a function of both the volume concentration of the crystalline lamellae and the structure of the interlamellar amorphous regions (chain folds, tie molecules, chain ends, and segregated low molecular weight material). Annealing above the crystallization temperature of isothermally crystallized samples has a marked influence on their morphology and mechanical behavior. The morphological structure and the viscoelastic properties of annealed PET samples are completely different from those obtained with samples isothermally crystallized at the same temperature.  相似文献

Central to the biological function of microtubules is their ability to modify their length which occurs by addition and removal of subunits at the ends of the polymer, both in vivo and in vitro. This dynamic behavior is strongly influenced by temperature. Here, we show that the lateral interaction between tubulin subunits forming microtubule is strongly temperature dependent. Microtubules deposited on prefabricated substrates were deformed in an atomic force microscope during imaging, in two different experimental geometries. Microtubules were modeled as anisotropic, with the Young's modulus corresponding to the resistance of protofilaments to stretching and the shear modulus describing the weak interaction between the protofilaments. Measurements involving radial compression of microtubules deposited on flat mica confirm that microtubule elasticity depends on the temperature. Bending measurements performed on microtubules deposited on lithographically fabricated substrates show that this temperature dependence is due to changing shear modulus, implying that the lateral interaction between the protofilaments is strongly determined by the temperature. These measurements are in good agreement with previously reported measurements of the disassembly rate of microtubules, demonstrating that the mechanical and dynamic properties of microtubules are closely related.  相似文献

The thermal properties and fire behaviour of polypropylene (PP) nanocomposites were investigated using differential scanning calorimetry, dynamic-mechanical analysis, thermogravimetric analysis and glow wire test. In order to study the morphological structure of the materials obtained, TEM and XRD analyses were also carried out. The nanocomposites were prepared using the melt intercalation technique in a co-rotating intermeshing twin screw extruder. Particular attention was given to studying the influence of different processing conditions (barrel temperature profile and screw rate) and compositions of PP-nanoclay blends (clay content, use of compatibiliser) on the thermal properties of the nanocomposites.The results show that all the properties analysed were strongly influenced by the nanocomposite composition; instead, the processing conditions greatly affect only the dynamic-mechanical properties. DSC curves show that the crystallinity is deeply influenced by the presence of the clay in the matrix, owing to the fact that the filler acts as nucleating agent. DMA curves show that materials processed at low temperature profile and high shear stress, i.e. when a good clay dispersion is achieved, are characterised by an enhanced modulus, thus indicating that the incorporation of clay into the PP matrix remarkably enhances its stiffness and has good reinforcing effects. TGA traces in oxidizing atmosphere show a drastic shift of the weight loss curve towards higher temperature and no variation of the onset temperature (i.e. the temperature at which degradation begins). The TGA analyses in inert atmosphere show instead marked increase of this parameter (about 200 °C) and no shift of weight loss curves. Glow wire results highlight that polymer nanocomposites are characterised by enhanced fire behaviour.  相似文献
