A fluorescent nanoprobe is reported for rapid detection of nitrites (NO2?) in plant cells. The probe is fabricated by linking neutral reds (NR) to the surface of upconversion fluorescent core/shell nanocrystalline with the bridging of polyethylene glycol (PEG) molecules. The fluorescence of upconversion nanoparticles (UCNPs) is stored by NR through fluorescence resonance energy transfer (FRET) under 980 nm excitation that can be released by further linking to NO2?. It is observed that the intensity rate of green to red emission of NR‐modified UCNPs changes linearly with increasing the amount of NO2?. So that concentration of NO2? can be accordingly addressed. Worth mentioning is that, comparing with bare core upconversion nanoparticles (NPs), core/shell UCNPs can greatly reduce the surface quenching of the fluorescence induced by solvents instead of NR and thus leading to the enhancement of signal‐to‐noise ratios. Moreover, excitation of core/shell UCNPs requires only a much lower power (0.06 W cm?2) than bare cores which is beneficial to reducing the decomposition of NR to stabilize the FRET processes. Under the optimum conditions, the detection limit of nitrite in plant cells was 0.1 µg mL?1.  相似文献

Pure metal iron nanoparticles are unstable in the air. By a coating iron on nanoparticle surface with a stable noble metal, these air-stable nanoparticles are protected from the oxidation and retain most of the favorable magnetic properties, which possess the potential application in high density memory device by forming self-assembling nanoarrays. Gold-coated iron core-shell structure nanoparticles (Fe/Au) synthesized using reverse micelles were characterized by transmission electron microscopy (TEM). The average nanoparticle size of the core-shell structure is about 8 nm, with about 6 nm diameter core and 1∼2 nm shell. Since the gold shell is not epitaxial growth related to the iron core, the morié pattern can be seen from the overlapping of iron core and gold shell. However, the gold shell lattice can be seen by changing the defocus of TEM. An energy dispersive X-ray spectrum (EDS) also shows the nanoparticles are air-stable. The magnetic measurement of the nanoparticles also proved successful synthesis of gold coated iron core-shell structure. The nanoparticles were then assembled under 0.5 T magnetic field and formed parallel nanobands with about 10 μm long. Assembling two dimensional ordered nanoarrays are still under going. A simple, sensitive, and accurate spectrofluorimetric method was developed for the determination of citalopram in bulk and pharmaceutical preparations. The method is based on the enhancement of the weak fluorescence signal (FL) of the Tb (III)-citalopram system in the presence of silver nanoparticles. Fluorescence intensities were measured at 555 nm after excitation at 281 nm. Prepared silver nanoparticles (AgNPs) were characterized by UV-Visible spectra and transmission electron microscopy (TEM). Various factors affecting the formation of citalopram-Tb (III)-AgNPs complexes were studied and optimized. The fluorescence intensity versus concentration plot was linear over the range 0.02–14 μg?mL?1, with an excellent correlation coefficient of 0.9978. The limit of detection (LOD) and limit of quantification (LOQ) were found to be 7.15?×?10?6?μg?mL?1 and 2.38?×?10?5?μg?mL?1 respectively. The proposed method was found to have good reproducibility with a relative standard deviation of 3.66 % (n?=?6). The interference effects of common excipients found in pharmaceutical preparations were studied. The developed method was validated statistically by performing recoveries studies and successfully applied for the assay of citalopram in bulk powder and pharmaceutical preparations. Percent recoveries were found to range from 98.98 % to 100.97 % for bulk powder and from 96.57 % to 101.77 % for pharmaceutical preparations.  相似文献

A kind of novel gold (Au)/hydroxylated poly (styrene-b-butadiene-b-styrene) triblock copolymer (HO-SBS) composite nanoparticles was synthesized by reduction of tetrachloroaurate ions in HO-SBS micelle. The Au–HO-SBS composite nanoparticles are composed by gold core about 35 nm in diameter and polymer shell about 7 nm in thickness. Formation of the Au/HO-SBS nanoparticles is indentified by infrared (IR) and UV-visible absorption spectroscopies. Transmission electron microscopy (TEM) result shows that the composite nanoparticles tend to aggregate into an ordered hexagonal array on carbon-coated grid.  相似文献

Indium phosphide (InP) quantum dots (QDs) are ideal substitutes for widely used cadmium-based QDs and have great application prospects in biological fields due to their environmentally benign properties and human safety. However, the synthesis of InP core/shell QDs with biocompatibility, high quantum yield (QY), uniform particle size, and high stability is still a challenging subject. Herein, high quality (QY up to 72%) thick shell InP/GaP/ZnS core/shell QDs (12.8 ± 1.4 nm) are synthesized using multiple injections of shell precursor and extension of shell growth time, with GaP serving as the intermediate layer and 1-octanethiol acting as the new S source. The thick shell InP/GaP/ZnS core/shell QDs still keep high QY and photostability after transfer into water. InP/GaP/ZnS core/shell QDs as fluorescence labels to establish QD-based fluorescence-linked immunosorbent assay (QD-FLISA) for quantitative detection of C-reactive protein (CRP), and a calibration curve is established between fluorescence intensity and CRP concentrations (range: 1–800 ng mL−1, correlation coefficient: R2 = 0.9992). The limit of detection is 2.9 ng mL−1, which increases twofold compared to previously reported cadmium-free QD-based immunoassays. Thus, InP/GaP/ZnS core/shell QDs as a great promise fluorescence labeling material, provide a new route for cadmium-free sensitive and specific immunoassays in biomedical fields.  相似文献

The combination of fluorescence microscopy and electron microscopy promises a deeper insight into the ultrastructural features of cell organelles, e.g., after drug administration. Both methods complement each other and provide, as a correlative approach, a keen insight into the fate of nanoparticles within the cell. Moreover, it represents a promising tool to determine alterations of the cellular environment as a response to particle uptake. However, the availability of suitable correlative markers is mandatory for such correlative approaches. In this contribution, the utilization of poly(ethylene imine) based metal–polymer hybrid particles labeled with small gold nanoparticles and Rhodamine B facilitating the observation of the particles by means of fluorescence as well as by transmission electron microscopy is suggested. Correlative light and electron microscopy is used to study uptake and intracellular fusion processes of endosomal/lysosomal structures.  相似文献

Gold nanoparticles (~40 nm in diameter) were encapsulated by a hydrogel overlayer generated by the free radical polymerization of N-isopropylacrylamide-co-acrylic acid (NIPAM-co-AA), which was promoted by a specifically designed radical initiator covalently anchored to the surface of the gold core. The size and morphology of the shell/core nanoparticles were characterized by scanning electron microscopy and transmission electron microscopy. In addition, the optical properties of the nanoparticles were characterized by UV–Visible spectroscopy. Separately, the particle size was evaluated as a function of temperature and pH using dynamic light scattering. The shell/core hydrogel nanoparticles undergo reversible volume changes in water at a lower critical solution temperature (LCST) of ~34 °C as well as at pH values between 3 and 4. The unique composition and properties of these shell/core hydrogel nanoparticles make them attractive for use as nanoscale drug-delivery vehicles.  相似文献

In this work, the fungus Penicillium was used for rapid extra-/intracellular biosynthesis of gold nanoparticles. AuCl4 ions reacted with the cell filtrate of Penicillium sp. resulting in extracellular biosynthesis of gold nanoparticles within 1 min. Intracellular biosynthesis of gold nanoparticles was obtained by incubating AuCl4 solution with fungal biomass for 8 h. The gold nanoparticles were characterized by means of visual observation, UV–Vis absorption spectroscopy, X-ray diffraction (XRD), transmission electron microscopy (TEM), scanning electron microscopy (SEM), and energy-dispersive X-ray spectroscopy (EDX). The extracellular nanoparticles exhibited maximum absorbance at 545 nm in UV–Vis spectroscopy. The XRD spectrum showed Bragg reflections corresponding to the gold nanocrystals. TEM exhibited the formed spherical gold nanoparticles in the size range from 30 to 50 nm with an average size of 45 nm. SEM and TEM revealed that the intracellular gold nanoparticles were well dispersed on the cell wall and within the cell, and they are mostly spherical in shape with an average diameter of 50 nm. The presence of gold was confirmed by EDX analysis.  相似文献

Submicron rod LiFePO4/C has been synthesized via a facile hydrothermal process. The morphology, crystal structure, and charge–discharge performance of the prepared samples were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), and galvanostatic charge–discharge testing. The SEM and TEM illustrate that submicron rods with a width of about 140 nm and a length of up to 400 nm have been obtained. The TEM test also indicates a “core–shell” structure with a 1.5–2 nm carbon shell on the LiFePO4 core. Even though the separate carbon-coated procedure is not used in this method, the electrochemical behavior results are satisfied. It displays that LiFePO4/C has highly crystalline and a desirable core–shell structure with uniform carbon film. Galvanostatic battery testing shows that LiFePO4/C delivers 104 mAh g?1 at 5 C rates. The highest specific capacity of 166 mAh g?1 is achieved at 0.1 C rate, and 99.8 % of the initial specific capacitance remained after 30 cycles.  相似文献

Superparamagnetic iron oxide (SPIO) nanoparticles were synthesized by coprecipitation technique and further functionalized with amino-group to obtain amino-group functionalized (amino-SPIO) nanoparticles. The X-ray diffraction results reveal the structure of amino-SPIO nanoparticles, from which the average iron core diameter is approximately 10 nm by calculation; while Zetasizer reveals their hydrodynamic diameter are mainly distributed in the range of 40?C60 nm. These nanoparticles can be taken up by liver tissue, resulting in dramatically darkening of liver tissue under T2-magnetic resonance imaging (MRI). The spin?Cspin relaxivity coefficient of these nanoparticles is 179.20 mM?1 s?1 in a 1.5 T magnetic resonance system. In addition, amino-SPIO nanoparticles were conjugated to Tat (FITC) peptide and incubated with neural stem cells in vitro, the authors can detect the positive-labeling (labeled) neural stem cells showing green fluorescence, which indicates Tat (FITC) peptide-derivated amino-SPIO nanoparticles are able to enter cells. Furthermore, it was also find significant negative T2 contrast enhancement when compared with the non-nanoparticles-labeled neural stem cells in T2-weighted MRI. The amino-SPIO nanoparticles show promising potential as a new type of labeling probes, which can be used in magnetic resonance-enhanced imaging and fluorescence diagnosis.  相似文献

Irradiation effect of low-energy ion beam has been investigated on nanocoating developed with silica, titania and silica–titania core–shell nanoparticles embedded in an organic binder for nanopaint application. In this work, we have taken polyurethane as a model organic binder. Silica nanoparticles have been prepared through sol–gel synthesis with a particle size of 85?nm. Titania and core–shell nanoparticles have been prepared through both sol–gel and peptization process. Particle sizes obtained were 107?nm for titania and 240?nm for core–shell nanoparticles prepared through sol–gel process and 75?nm for TiO2 and 144?nm for core–shell nanoparticles prepared through peptization process. The coating formulations were developed with the above nanoparticles individually and nanoparticle concentration was varied from 1 to 6?wt% and the best performance in terms of hydrophobicity was obtained with 4?wt % of the nanoparticles in polyurethane coating formulation. All the coating formulations prepared were applied on a glass substrate and dried at 100°C. The dry film thickness obtained was around 100?µm in each case. These films dried on glass substrate were irradiated by nitrogen and argon ion beam with energy of 26?keV at fluences of 1014 to 1016?ions/cm2. The anti-algal property of the irradiated samples was improved and hydrophobicity was reduced.  相似文献

In core–shell nanoparticle analysis, the determination of size distributions of the different particle parts is often complicated, especially in liquid media. Density matching is introduced as a method for analyzing core–shell nanoparticles using Analytical Ultracentrifugation (AUC), making it possible to obtain the core size distribution in liquid dispersions. For this approach, the density of the dispersion is adjusted to the density of the shell. Oil filled nanocapsules are utilized with component densities of around 1 g mL−1 to demonstrate this technique. The shell size distribution is calculated supposing the particle size distribution as a convolution of the shell- and core size distributions. Finally, the distributions of core size, shell thickness, particle size, and particle density and thus particle composition are obtained. To clarify the effect of swelling, AUC measurements are combined with further size characterization methods like Particle Tracking Microscopy and Dynamic Light Scattering.  相似文献

利用新合成的复合纳米结构银/二氧化钛核壳纳米颗粒,研究了金属银纳米颗粒对碲化镉纳米晶层荧光的增强情况.结果表明,这种新型复合金属纳米结构能极大地增强发光纳米晶层的荧光强度.银/二氧化钛核壳纳米颗粒是以水合肼、硝酸银和四异丙氧基钛为原材料,利用胶体化学法在水溶液中合成.透射电子显微镜图片表明这种新合成的银/二氧化钛纳米材料基本上呈球形,有较为明显的核壳结构,中间黑色的核是银纳米颗粒,外层颜色较浅部分是二氧化钛壳层.另外,包裹二氧化钛壳层后,银纳米颗粒的表面等离子吸收带从409 nm红移至430 nm,也证实了这种新型核壳纳米材料的形成.将此合成方法得到的银/二氧化钛纳米颗粒和碲化镉纳米晶用旋转涂覆方法进行直接组合后,得到了银纳米颗粒对碲化镉纳米晶荧光的明显增强,并对其增强的物理过程进行了讨论.这种能够增强荧光团发光的新型复合银纳米结构将在发光器件、荧光成像、生物探测等方面具有一定的应用价值.  相似文献

Hollow core-shell silica nanoparticles (HCSNs) are being considered as one of the most favorable drug carriers to accomplish targeted drug delivery. In the present study, we developed a simple two-step method, employing polystyrene (PS) nanoparticles (150?±?20 nm) as a sacrificial template for the synthesis of microporous HCSNs of size 230?±?30 nm. PS core and the wall structure directing agent cetyl trimethyl ammonium bromide (CTAB) were removed by calcination. Monodispersed spherical HCSNs were synthesized by optimising the parameters like water/ethanol volume ratio, PS/tetraethyl orthosilicate (TEOS) weight ratio, concentration of ammonia, and CTAB. Transmission electron microscopy (TEM) revealed the formation of hollow core-shell structure of silica with tunable thickness from 15 to 30 nm while tailoring the concentration of silica precursor. The results obtained from the cumulative release studies of doxorubicin loaded microporous HCSNs demonstrated the dependence of shell thickness on the controlled drug release behavior. HCSNs with highest shell thickness of 30 nm and lowest surface area of 600 m2/g showed delay in the doxorubicin release, proving their application as a drug carrier in targeted drug delivery systems. The novel concept of application of microporous HCSNs of pore size ~?1.3 nm with large specific surface area in the field of drug delivery is successful.  相似文献

Stable gold nanoparticles have been prepared by using soluble starch as both the reducing and stabilizing agents; this reaction was carried out at 40 °C for 5 h. The obtained gold nanoparticles were characterized by UV–Vis absorption spectroscopy, transmission electron microscopy (TEM) and z-scan technique. The size of these nanoparticles was found to be in the range of 12–22 nm as analyzed using transmission electron micrographs. The optical properties of gold nanoparticles have been measured showing the surface plasmon resonance. The second-order nonlinear optical (NLO) properties were investigated by using a continuous-wave (CW) He–Ne laser beam with a wavelength of 632.8 nm at three different incident intensities by means of single beam techniques. The nonlinear refractive indices of gold nanoparticles were obtained from close aperture z-scan in order of 10?7 cm2/W. Then, they were compared with diffraction patterns observed in far-field. The nonlinear absorption of these nanoparticles was obtained from open aperture z-scan technique. The values of nonlinear absorption coefficient are obtained in order of 10?1 cm/W.  相似文献

Fe3O4@ZnO binary nanoparticles were synthesized by a simple two-step chemical method and characterized using various analytical instruments. TEM result proved the binary nanoparticles have core/shell structures and average particle size is 60 nm. Photocatalytic investigation of Fe3O4@ZnO core/shell nanoparticles was carried out using rhodamine B (RhB) solution under UV light. Fe3O4@ZnO core/shell nanoparticles showed enhanced photocatalytic performance in comparison with the as prepared ZnO nanoparticles. The enhanced photocatalytic activity for Fe3O4@ZnO might be resulting from the higher concentration of surface oxygen vacancies and the suppressing effect of the Fe3+ ions on the recombination of photoinduced electron–hole pairs. Magnetization saturation value (5.96 emu/g) of Fe3O4@ZnO core/shell nanoparticles is high enough to be magnetically removed by applying a magnetic field. The core/shell photocatalyst can be easily separated by using a commercial magnet and almost no decrease in photocatalytic efficiency was observed even after recycling six times.  相似文献

The ferrihydrite mineral core of ferritin is a semi-conductor capable of catalyzing oxidation/reduction reactions. This report shows that ferritin can photoreduce AuCl4 to form gold nanoparticles (AuNPs). An important goal was to identify innocent reaction conditions that prevented formation of AuNPs unless the sample was illuminated in the presence of ferritin. TRIS buffer satisfied this requirement and produced AuNPs with spherical morphology with diameters of 5.7 ± 1.6 nm and a surface plasmon resonance (SPR) peak at 530 nm. Size-exclusion chromatography of the AuNP–ferritin reaction mixture produced two fractions containing both ferritin and AuNPs. TEM analysis of the fraction close to where native ferritin normally elutes showed that AuNPs form inside ferritin. The other peak eluted at a volume indicating a particle size much larger than ferritin. TEM analysis revealed AuNPs adjacent to ferritin molecules suggesting that a dimeric ferritin–AuNP species forms. We propose that the ferritin protein shell acts as a nucleation site for AuNP formation leading to the AuNP-ferritin dimeric species. Ferrihydrite nanoparticles (~10 nm diameter) were unable to produce soluble AuNPs under identical conditions unless apo ferritin was present indicating that the ferritin protein shell was essential for stabilizing AuNPs in aqueous solution.  相似文献

Gold-coated magnetic nanoparticles were synthesized with size ranging from 15 to 40 nm using sodium citrates as the reducing agent. Oxidized magnetites (Fe3O4) fabricated by co-precipitation of Fe2+ and Fe3+ in strong alkaline solution were used as magnetic cores. The structures of gold (Au) shell and magnetic core (Au–Fe) were studied by transmission electron microscopy (TEM) image and energy dispersive spectroscopy (EDS) spectrum. Results from high-resolution X-ray diffraction (HR XRD) show that the Au–Fe oxide nanoparticles have a face-centered cubic shape with the crystalline faces of {1 1 1}. The Au-coated magnetic nanoparticles exhibited a surface plasmon resonance peak at 528 nm. The nanoparticles are well dispersed in distilled water. A 3000 G permanent magnet was successfully used for the separation of the functionalized nanoparticles. Magnetic properties of the nanoparticles were determined by magnetic force microscope (MFM) in nanometric resolution and vibrating sample magnetometer (VSM). Magnetic separation of biological molecules using Au-coated magnetic oxide composite nanoparticles was examined after attachment of protein immunoglobulin G (IgG) through electrostatic interactions. Using this method, separation was achieved with a maximum yield of 35% at an IgG concentration of 400 ng/ml.  相似文献

The preparation and application of rod-shaped core–shell structured Fe3O4–Au nanoparticles for immunomagnetic separation and sensing were described for the first time with this study. To synthesize magnetic gold nanorod particles, the seed-mediated synthetic method was carried out and the resulting nanoparticles were characterized with transmission electron microscopy (TEM), ultraviolet visible spectroscopy (UV–Vis), energy-dispersive X-ray (EDX), and X-ray diffraction (XRD). Magnetic properties of the nanoparticles were also examined. Characterization of the magnetic gold nanorod particles has proven that the resulting nanoparticles were composed of Fe3O4 core and the gold shell. The rod-shaped gold-coated iron nanoparticles have an average diameter of 16 ± 2 nm and an average length of about 50 ± 5 nm (corresponding aspect ratio of 3). The saturation magnetization value for the magnetic gold nanorod particles was found to be 37 emu/g at 300 K. Rapid and room temperature reaction synthesis of magnetic gold nanorod particles and subsequent surface modification with E. coli antibodies provide immunomagnetic separation and SERS application. The analytical performance of the SERS-based homogenous sandwich immunoassay system with respect to linear range, detection limit, and response time is also presented.  相似文献

A scalable synthesis of magnetic core–shell nanocomposite particles, acting as a novel class of magnetic resonance (MR) contrast agents, has been developed. Each nanocomposite particle consists of a biocompatible chitosan shell and a poly(methyl methacrylate) (PMMA) core where multiple aggregated γ‐Fe2O3 nanoparticles are confined within the hydrophobic core. Properties of the nanocomposite particles including their chemical structure, particle size, size distribution, and morphology, as well as crystallinity of the magnetic nanoparticles and magnetic properties were systematically characterized. Their potential application as an MR contrast agent has been evaluated. Results show that the nanocomposite particles have good stability in biological media and very low cytotoxicity in both L929 mouse fibroblasts (normal cells) and HeLa cells (cervical cancer cells). They also exhibited excellent MR imaging performance with a T2 relaxivity of up to 364 mMFe?1 s?1. An in vivo MR test performed on a naked mouse bearing breast tumor indicates that the nanocomposite particles can localize in both normal liver and tumor tissues. These results suggest that the magnetic core–shell nanocomposite particles are an efficient, inexpensive and safe T2‐weighted MR contrast agent for both liver and tumor MR imaging in cancer therapy.  相似文献
