A single-step, environmentally friendly sample treatment was developed and used in combination with liquid chromatography–tandem mass spectrometry (LC–MS/MS) for the quantitation of hexabromocyclododecane (HBCD) stereoisomers in fish. It was based on the microextraction of the stereoisomers with a supramolecular solvent (SUPRAS) made up of reverse aggregates of decanoic acid (DeA). The procedure involved the stirring of the fish sample (750 mg) with 600 μL of SUPRAS for five minutes, subsequent centrifugation for extract separation from matrix components and direct analysis of the extract after dilution 1:1 with methanol. Individual enantiomers of α-, β- and γ-HBCD were separated on a chiral stationary phase of β-cyclodextrin and quantified by monitoring of the [M−H] → Br transition at m/z 640.9→80.9. Driving forces for the microextraction of HBCD in the SUPRAS involved both dispersion and dipole–dipole interactions. Quantitation limits for the determination of individual HBCD enantiomers in hake, cod, sole, panga, whiting and sea bass were within the intervals 0.5–3.4 ng g−1, 0.9–2.5 ng g−1, 0.6–1.4 ng g−1, 1.0–5.6 ng g−1, 0.8–1.3 ng g−1 and 0.5–3.5 ng g−1, respectively. Recoveries for fish samples fortified at the ng g−1 level ranged between 87 and 114% with relative standard deviations from 1 to 10%. The sample treatment proposed greatly simplifies current procedures for extraction of HBCD stereoisomers and is a useful tool for the development of a large scale database for their presence in fish.

An in situ coacervative extraction (IS-CAE) based on a double-solvent supramolecular system coupled to liquid–liquid microextraction is investigated for extraction and enrichment of triazole fungicides. The formation of a double-solvent supramolecular system was generated by in situ formation and used as an extraction solvent for the coacervative extraction method. No disperser solvent was required. This new double-solvent supramolecular system has a higher extraction ability than any of its components alone. The different factors that could affect the extraction capability were studied and optimized, including the type of double extractant and its volume, salt addition, vortex time, and centrifugation time. Under optimum extraction conditions, this method provides high enrichment factors (EFs) of 73–318 with low limits of detection (LODs) of 0.3–1 μg L−1 and limits of quantitation (LOQs) of 1–3 μg L−1. In addition, the proposed method was prosperously applied for the determination of triazole fungicides in water, fruit juice, and soy milk samples.  相似文献

New analytical approaches to the simultaneous identification and quantification of 94 pesticides and 13 polycyclic aromatic hydrocarbons (PAHs) in five representative matrices (pepper, apple, lettuce, wheat, and soil) were developed. The analyses were based on gas chromatography coupled with triple quadrupole tandem mass spectrometry (GC-MS/MS). The procedure was optimized by changing the solvent used during the extraction, from acetonitrile to the acetone: n-hexane mixture at a volume ratio of 1:4 (v/v), as well as the use of a reduced amount of water during the extraction of compounds from cereals. An additional modification was the use of florisil instead of GCB in the sample cleanup step. A full method validation study was performed, at two concentration levels (LOQ and 1000 × LOQ), which showed satisfactory results for all analytes from the PAHs group, with recoveries ranging from 70.7–115.1%, and an average RSD of 3.9%. Linearity was tested in the range of 0.001–1.000 mg/kg and showed coefficients of determination (R2) ≥ 0.99 for all PAHs. Satisfactory recovery and precision parameters (LOQ and 100 × LOQ) were achieved for almost all analytes from the pesticide group in the range of 70.1–119.3% with the mean RSD equal to 5.9%. The observed linearity for all analytes in the concentration range of 0.005–1.44 mg/kg was R2 ≥ 0.99, with the exception of famoxadone, chizalofop-p-ethyl, prothioconazole, spirodiclofen, tefluthrin, and zoxamid. The extended uncertainties were estimated, using a top-down approach of 9.9% (average) and 15.3% (average) for PAHs and pesticides samples, respectively (the coverage factor k = 2, the 95% confidence level). Ultimately, the method was successfully applied to determine pesticide residues in commercial samples of fruit, vegetables and grain, and soil samples for PAHs, which were collected from selected places in the Podkarpacie region. A total of 38 real samples were tested, in which 10 pesticides and 13 PAHs were determined. Proposed changes allow us to shorten the sample preparation time (by 20%) and to reduce the consumption of organic solvents (by 17%). The use of florisil for sample cleanup, instead of GCB, improves the recovery of compounds with flat particles.  相似文献

Quality control materials (QCMs) to be used in daily quality control in environmental laboratories are required to be similar to real samples in terms both of composition and of physical state. The present paper describes in detail the procedure of preparation and the results of homogeneity and stability studies performed on wet sediments QCMs. For this purpose, two sediments with different matrix composition were selected and the main factors involved in the preparation of the material were carefully studied and established with the aim of guaranteeing long-term stability of the QCM: pre-treatment steps, homogenisation, bottling and storage. The results obtained from the homogeneity and from 12 months of stability studies for Cd, Cr, Cu, Ni, Pb and Zn following the aqua regia extraction procedure (ISO 11466), showed that the wet sediment QCMs we have developed are feasible, constituting a new and useful material among the aquatic sediments for quality control purposes in monitoring.  相似文献

A restricted access supramolecular solvent (SUPRAS-RAM) made up of tetradecanoic acid reverse micelles is proposed as a wide-scope and low-cost strategy for the treatment of agrifood samples prior to enzyme-linked immunosorbent assays (ELISA). The approach was assessed for the determination of ochratoxin A (OTA) in wines and spices and aflatoxin B1 (AFB1) in cereals, two ubiquitous mycotoxins that were selected as representative contaminants for this study. The samples were selected to cover a variety of matrices in terms diverse composition and high complexity. Macromolecules such as proteins and carbohydrates were not-co-extracted due to the restricted access properties of the SUPRAS that are provided by chemical and physical mechanisms. In this sense, analyte extraction and clean-up were carried out in a single step. Parameters determining the extraction efficiency were studied and optimized. Certified reference materials were used for method validation. Recoveries of OTA ranged between 83% and 96% in wines (with relative standard deviation, RSD, of about 10%) and between 81% and 93% in spices (RSD 7%). Recoveries for AFB1 in wheat ranged from 75% to 85% (RSD 8%). The detection limits were all below the maximum levels established for OTA and for AFB1 by EU directives. This method offers a green and low-cost alternative to the organic solvent-based extraction and/or immunoaffinity columns-based cleanup of complex samples prior to ELISA.  相似文献

Novel procedures for the determination of florfenicol in freshwater, sediments and bryophyte Fontinalis antipyretica, using reversed-phase high-performance liquid chromatography are described. Liquid chromatography was performed on a 5 µm PuroSpher RP-18E® column using methanol and 0.05 M phosphate buffer (18/82 v/v, pH 7.3) as mobile phase (0.8 ml min?1) and fluorescence detection (excitation wavelength 265 nm and emission wavelength 295 nm). Florfenicol was determined in centrifuged freshwater samples. Florfenicol was extracted from sediments and bryophytes samples by using a solid-liquid extraction step followed by a solid phase extraction step. Linearity was confirmed over the concentration range 25–1000 ng mL?1 water and 50-1000 ng g?1 sediment or bryophyte. Limits of detection and quantitation were 8 and 25 ng mL?1 water and 17 and 50 ng g?1 sediment or bryophyte respectively. Mean extraction recoveries of florfenicol from sediments and bryophyte were from 85.9 to 109.1%.  相似文献

Industrial processing of raspberry juice and wine generates considerable byproducts of raspberry pomace. Ellagic acids/ellagitannins, being characterized by their antioxidant and antiproliferation properties, constitute the majority of polyphenolics in the pomace and are valuable for recovery. In the present study, we developed a novel procedure with sodium bicarbonate assisted extraction (SBAE) to recover ellagic acid from raspberry wine pomace. Key parameters in the procedure, i.e., sodium bicarbonate concentration, temperature, time and solid/liquid (S/L) ratio, were investigated by single factor analysis and optimized subsequently by Response Surface Methodology (RSM). Optimal parameters for the SBAE method here were found to be 1.2% (w/v) NaHCO3, 1:93 (w/v) S/L ratio, 22 min and 100 °C. Under these conditions, the ellagic acid yield was 6.30 ± 0.92 mg/g pomace with an antioxidant activity of 79.0 ± 0.96 μmol Trolox eq/g pomace (DPPH assay), which are 2.37 and 1.32 times the values obtained by extraction with methanol–acetone–water solvent, respectively. The considerable improvement in ellagic acid extraction efficiency could be highly attributed to the reactions of lipid saponification and ellagitannin hydrolysis resulted from sodium bicarbonates. The present study has established an organic solvent-free method for the extraction of ellagic acid from raspberry wine pomace, which is feasible and practical in nutraceutical applications.  相似文献

Glucosinolates (GSLs) are important precursor compounds with anticancer activities in Brassicaceae vegetables and are readily hydrolyzed by myrosinase. Given the diversity of these species, establishing an accurate and universal method to quantify intact GSLs in different plant tissues is necessary. Here, we compared and optimized three tissue disruption methods for sample preparation. After microwave treatment for 90 s, 13 GSLs in homogenized Chinese cabbage samples were recovered at 73–124%. However, a limitation of this method was that different tissues could not be processed under the same microwave conditions. Regarding universality, GSLs in Brassicaceae vegetables could be extracted from freeze-dried sample powder with 70% methanol (v/v) or frozen-fresh sample powder with 80% methanol (v/v). Moreover, heating extraction is necessary for GSLs extracted from frozen-fresh sample powder. Average recoveries of the two optimized methods were 74–119% with relative standard deviations ≤ 15%, with the limits of quantification 5.72–17.40 nmol/g dry weight and 0.80–1.43 nmol/g fresh weight, respectively. Notably, the method for analyzing intact GSLs was more efficient than that for desulfo-GSLs regarding operational complexity, detection speed and quantification accuracy. The developed method was applied to identify the characteristic GSLs in 15 Brassicaceae vegetables, providing a foundation for further research on GSLs.  相似文献

In the present study, a microwave-assisted extraction method has been investigated for the extraction of persistent organochlorine pesticides from sediment in comparison to the classical Soxhlet extraction. The extraction procedure has been screened by a complete factorial design for statistically significant parameters. Thereafter, the identified parameters, composition of extraction solvent (mixtures of n-hexane-acetone), extraction temperature and time were optimized by a Box-Behnken design. Finally, the optimized microwave-assisted procedure has been validated by extraction of two different matrix reference materials, a sediment and a mallow powder. The data obtained for both materials were in good agreement with those obtained by Soxhlet extraction and published values. Special emphasis has been given to an accurate determination of p,p′-DDT, since DDT is known as instable during GC-injection. In order to evaluate the DDT-degradation, 13C12-p,p′-DDT-solutions were analyzed and the degradation rates during GC-injection were calculated. Furthermore, 13C12-p,p′-DDT was added to the sediment samples prior to extraction and the degradation rates during the optimization experiment have been investigated.  相似文献

A novel HPLC-ESI-MS/MS method for simultaneous gonadotropin-releasing hormone (GnRH) analogs and somatostatin analog quantitation was developed and validated. The developed method was successfully applied to pharmacokinetic studies. The sample preparation process included solid-phase extraction (SPE). Effective chromatographic separation of the analytes and internal standard (dalargin) was achieved with a C18 column, using a gradient elution with two mobile phases: 0.1% v/v formic acid (aqueous solution) and 0.1% v/v formic acid (acetonitrile solution). The linearity of the method was demonstrated within a concentration range of 0.5–20 ng/mL, with correlation coefficients between 0.998–0.999 for goserelin, buserelin, triptorelin, and octreotide, respectively. The relative standard deviation (RSD, %) values for method accuracy and precision did not exceed 20% at the lower level of quantitation (LLOQ) or 15% at other concentration levels.  相似文献

A new, efficient, and environmental friendly hollow fiber liquid phase microextraction (HF-LPME) method based on supramolecular solvents was developed for extraction of five benzodiazepine drugs. The supramolecular solvent was produced from coacervation of decanoic acid aqueous vesicles in the presence of tetrabutylammonium (Bu4N+). In this work, benzodiazepines were extracted from aqueous samples into a supramolecular solvent impregnated in the wall pores and also filled inside the porous polypropylene hollow fiber membrane. The driving forces for the extraction were hydrophobic, hydrogen bonding, and π-cation interactions between the analytes and the vesicular aggregates. High-performance liquid chromatography with photodiode array detection (HPLC-DAD) was applied for separation and determination of the drugs. Several parameters affecting the extraction efficiency including pH, hollow fiber length, ionic strength, stirring rate, and extraction time were investigated and optimized. Under the optimal conditions, the preconcentration factors were obtained in the range of 112–198. Linearity of the method was determined to be in the range of 1.0–200.0 μg L−1 for diazepam and 2.0–200.0 μg L−1 for other analytes with coefficient of determination (R2) ranging from 0.9954 to 0.9993. The limits of detection for the target benzodiazepines were in the range of 0.5–0.7 μg L−1. The method was successfully applied for extraction and determination of the drugs in water, fruit juice, plasma and urine samples and relative recoveries of the compounds studied were in the range of 90.0–98.8%.  相似文献

Cannabis sativa L. is an herbaceous plant belonging to the family of Cannabaceae. It is classified into three different chemotypes based on the different cannabinoids profile. In particular, fiber-type cannabis (hemp) is rich in cannabidiol (CBD) content. In the present work, a rapid nano liquid chromatographic method (nano-LC) was proposed for the determination of the main cannabinoids in Cannabis sativa L. (hemp) inflorescences belonging to different varieties. The nano-LC experiments were carried out in a 100 µm internal diameter capillary column packed with a C18 stationary phase for 15 cm with a mobile phase composed of ACN/H2O/formic acid, 80/19/1% (v/v/v). The reverse-phase nano-LC method allowed the complete separation of four standard cannabinoids in less than 12 min under isocratic elution mode. The nano-LC method coupled to ultraviolet (UV) detection was validated and applied to the quantification of the target analytes in cannabis extracts. The nano-LC system was also coupled to an electrospray ionization–mass spectrometry (ESI-MS) detector to confirm the identity of the cannabinoids present in hemp samples. For the extraction of the cannabinoids, three different approaches, including dynamic maceration (DM), ultrasound-assisted extraction (UAE), and an extraction procedure adapted from the French Pharmacopeia’s protocol on medicinal plants, were carried out, and the results achieved were compared.  相似文献

An efficient and reliable method based on gas chromatography–mass spectrometry (GC–MS) was developed for the extraction and analysis of six phenolic endocrine disrupting chemicals (EDCs), such as 4-nonylphenol (4-NP), nonylphenol-mono-ethoxylate (NP1EO), nonylphenol-di-ethoxylate (NP2EO), 4-tert-octylphenol (4-t-OP), bisphenol A (BPA) and 4-cumylphenol (4-CP) in surface water and sediment. The method was developed by using microwave-assisted extraction (MAE), solid phase extraction (SPE) and derivatization procedure. The MAE procedures were performed by optimizing three key process factors, consisted of extraction solvent, extraction temperature and holding time, affecting the extraction efficiency from sediment samples. For SPE, various parameters that may affect the recovery efficiency of water samples, such as SPE phase cartridge, elution solvent, as well as pH of water samples, were investigated. A series of derivatization conditions, such as derivatization reagent, reaction temperature and reaction time, were improved. The method achieved good repeatability and reproducibility with relative standard deviations <13% for all target EDCs in the both samples. Satisfactory recoveries for spiked water and sediment samples ranged from 85 to 101% and 74 to 105%, respectively. The limits of quantification varied from 0.20 (4-t-OP) to 11.50 ng L?1 (NP2EO) and from 0.31 (4-t-OP) to 9.50 ng g?1 dry weight (dw) (NP2EO) for water samples and sediment samples, respectively. The established method was successfully applied to the analysis of target EDCs in surface water and sediment samples collected from Caohai site of Dianchi Lake, China. The results showed that NP1EO, NP2EO and BPA were the three dominant phenolic EDCs in the site, reaching 114, 97 and 149 ng L?1 in surface water, while 444, 186 and 178 ng g?1 dw in surface sediment, respectively.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are a class of persistent organic pollutants of water, and their determination at trace levels in the aquatic ecosystems is essential. In this work, an ultrasound-assisted dispersive liquid–liquid microextraction (DLLME) procedure was suggested utilizing a binary dispersive agent for recovery of different molecular weight polycyclic aromatic hydrocarbons (PAHs) from waters. The detection was carried out by gas chromatography–mass spectrometry (GC-MS) as well as high-performance liquid chromatography with fluorescence and diode-array detection (HPLC-FD/PDA). The method was optimized for the extraction of analytes with respect to the mixture composition, ratios of components, ultrasonication time and centrifugation parameters. The analytical schemes for PAHs extraction from water samples using different ratios of extraction and dispersive solvents are reported. The mixture consisting of chloroform and methanol was applied for the extraction of PAHs containing two or three fused aromatic rings; the mixture of chloroform and acetonitrile is suitable for PAHs containing more than four aromatic rings. The mixture of chloroform:acetone + acetonitrile was applied in the universal scheme and allowed for the simultaneous extraction of 20 PAHs with different structures. The developed sample preparation schemes were combined with GC-MS and HPLC-FD/PDA, which allowed us to determine the analytes at low concentrations (from 0.0002 µg/L) with the recoveries exceeding 80% and relative standard deviations of about 8%. The developed methods for the determination of 20 PAHs were applied to the analysis of water samples from the Karasun Lake (Krasnodar), Azov Sea (Temryuk) and Black Sea (Sochi).  相似文献

The residue after sieving (“dust”) from the willow gentian underground parts is an unexploited herbal tea by-product, although it contains valuable bioactive compounds. Cyclodextrins as efficient green co-solvents, cage molecules, and multifunctional excipients could improve the extraction and contribute to the added value of the resulting extracts. The objective of this study was to determine the optimal conditions for the extraction of gentiopicroside, isogentisin, and total phenolics (TPC) from willow gentian “dust” using ultrasound-assisted water extraction coupled with hydroxypropyl-β-cyclodextrin (HPβCD). The influence of extraction temperature (X1: 20–80 °C), time (X2: 20–50 min), and HPβCD concentration (X3: 2–4% w/v) was analyzed employing the response surface methodology (RSM). The optimal extraction conditions for simultaneously maximizing the extraction yield of all monitored responses were X1: 74.89 °C, X2: 32.57 min, and X3: 3.01% w/v. The experimentally obtained response values under these conditions (46.96 mg/g DW for gentiopicroside, 0.51 mg/g DW for isogentisin, and 12.99 mg GAE/g DW for TPC) were in close agreement with those predicted, thus confirming the suitability and good predictive accuracy of the developed RSM models. Overall, the developed extraction system could be an applicable alternative strategy to improve the extraction of bioactive compounds from the underutilized “dust” of willow gentian underground parts.  相似文献

A rapid and sensitive method has been developed for the simultaneous determination of four avermectins and one milbemycin residues in bovine tissue. The isolation of the analytes from muscle and liver samples was accomplished utilizing a pressurized solvent extractor. The optimized extraction procedure using acetonitrile/water (40:60, v/v) as extraction solvent, was automatically carried out at 100 °C and 10 MPa, applying two static cycles for 3 min. The extracts were cleaned up on a C18 solid-phase extraction cartridge and analyzed by liquid chromatography with fluorescence detection after derivatization. Mean recoveries of the five analytes from fortified samples were between 84.8 and 101.8%, with relative standard deviations lower than 10.8%. The limit of detection and quantification were in the ranges of 0.1–0.2 and 0.5–0.6 μg kg–1, respectively. The application of the newly developed method was demonstrated by analyzing bovine meat samples from market.  相似文献

An analytical chemical method has been developed for the simultaneous determination of 32 different pharmaceuticals in soils and sediments. The pharmaceuticals cover a varity of different compound groups. Soil samples were extracted with different solvents with the help of pressurized-liquid extraction (PLE) followed by clean-up using a solid-phase extraction (SPE) procedure. The purified extracts were analyzed by LC-MS/MS. The extraction method was evaluated by testing the following variables: extraction solvents, solvent pH, and temperature. Applying 20 g of soil/sediment and extracting with a mixture of methanol with aqueous ammonia solution (0.1 mol L?1) at 80?°C for 5 min in five cycles provided satisfactory recoveries between 66 and 114% with SD of between 1 and 14%. For preconcentration and purification tandem MAX-HLB cartridges were used. The volume and composition was optimized and the highest recoveries were obtained with a combination of methanol—aqueous ammonia solution. The limits of quantification (LOQs) were between 0.2 and 2 ng g?1 and linearity higher than 0.98 for the majority of the selected pharmaceuticals. The method was successfully applied to soil samples collected from the Jerez de la Frontera agricultural region, irrigated with treated wastewater, and to sediment samples from the River Guadalete. The detection of nine pharmaceuticals including stimulants, antirheumatics, analgesics, anti-inflammatories, tranquilizers, and veterinary medicines at ng g?1 concentration levels was achieved.  相似文献

Solvents used in microextraction require high solubilising capability to efficiently extract the target compounds. In this article, nanostructured solvents made up of alkyl carboxylic acids (ACAs) aggregate are proposed for the efficient microextraction of acidic pesticides from natural waters. The target compounds were chlorophenoxy acid herbicides (CPAHs) widely used in agriculture, forestry and gardening (viz. 2,4-D, MCPA, MCPP, 2,4,5-T and MCPB). The supramolecular solvents (SUPRASs) tested were generated from solutions of reverse micelles of octanoic (OcA), decanoic (DeA) and dodecanoic (DoA) acid in THF by the addition of water, which acted as the coacervating agent. The DeA-based SUPRAS was the most efficient extractant for CPAHs; actual concentration factors (ACFs) of 260 for 2,4-D, 290 for MCPA, and 400 for MCPP, 2,4,5-T and MCPB were obtained. The explanation for so high ACFs can be found in the extremely efficient retention mechanisms that the DeA-based SUPRAS provides for CPAHs (i.e. formation of hydrogen bonds and hydrophobic interactions), and the high number of binding sites that it contains (i.e. the concentration of biosurfactant in the SUPRAS was 0.56 mg μL−1). Both characteristics permitted to effectively extract the target analytes in a low volume of solvent (about 2 μL of solvent per mL of sample). Others assets of the proposed supramolecular solvent-based microextraction (SUSME) approach included recoveries no dependent on matrix composition, rapidity (sample treatment spent about 15 min), use of low volume of sample (63 mL per analysis) and simplicity (no special lab equipments was needed). Combination with liquid chromatography/ion–trap mass spectrometry [LC–(IT)MS] afforded method quantitation limits for CPAHs within the interval 22–30 ng L−1. The precision of the method, expressed as relative standard deviation (n = 11, [CPAH] = 200 ng L−1), was in the range 2.9–5.8%. The applicability of the method to the analysis of natural waters was assessed by determining the target analytes in fortified river and underground water samples.  相似文献
