A qualitative method, involving supported liquid–liquid extraction (SLE) and ultra high pressure liquid chromatography coupled to tandem mass spectrometry (UHPLC-MS–MS), was developed for the rapid tentative identification of various drugs of abuse in urine. In this study, 28 drugs and metabolites were covered by the screening procedure. Before analysis, urine samples were extracted by SLE and good extraction recoveries were obtained for most investigated compounds. The UHPLC strategy was then selected for the rapid separation of amphetamines, cocaine, opiates and related compounds in urine. Using columns packed with sub-2 µm particles, analysis time was reduced down to 2 min, while maintaining acceptable performance. Finally, the detection was by tandem MS operating in the single reaction monitoring (SRM) mode. The most intense transition was selected for the different drugs and SRM dwell times set at 5 ms, to maintain sufficient data points across the narrow UHPLC peaks. The tentative identification of the drugs of interest, including amphetamines, opiates and cocaine, was based on both, retention times and mass spectrometry information. With the proposed method, limits of detection were estimated at about 1 ng mL ?1 and the applicability was assessed by successfully analyzing several samples of drug abusers. A sensitive, rapid and reproducible LC–MS/MS method for the determination of olmesartan (OLM), amlodipine (ALM) and hydrochlorothiazide (HCZ) in rat plasma and urine has been developed and validated. Irbesartan (IRB) was used as an internal standard. The analytes were separated on a Waters XTerra-C 18 column using gradient elution with acetonitrile and 10 mM ammonium acetate buffer (pH 3.5, adjusted with acetic acid) at a flow rate of 1.0 mL min ?1. The three analytes were ionized by positive ion electrospray using multiple-reaction monitoring (MRM) mode to monitor precursor?→?product ion transitions m/ z 447.31?→?234.97 for OLM, 408.87?→?238.18 for AML and 290.1?→?204.85 for HCZ. The specificity, matrix effect, recovery, sensitivity, linearity, accuracy, precision, and stabilities were all validated over the concentration range 0.4–100 ng mL ?1 for AML, 0.2–100 ng mL ?1 for OLM, 0.1–100 ng mL ?1 for HCZ. The mean concentrations ( Cmax) are 10.32, 587, and 3.4 for OLM, ALM, and HCZ, respectively, by the oral administration of 15 mg kg ?1 of each analyte. 相似文献

A method for screening DNA adducts with unknown chemical structures was developed; it involves the use of liquid chromatography–electrospray ionization-tandem mass spectrometry (LC–ESI–MS–MS). In electrospray ionization (ESI) product ion mass spectra of guanine adducts, fragment ions were observed at m/ z 152 and 135. Precursor ion scan analysis of these fragment ions indicated that the screening of DNA adducts would be possible. The developed method was used for the analysis of DNA adducts derived from acrylamide, which is not only a constituent of many commonly consumed foods but also a carcinogenic compound. We successfully discovered new guanine adducts. The results of this study indicate that the developed method is useful for screening new DNA adducts. 相似文献

In the framework of developing analyses for exogenous contaminants in food matrices such as honey, we have compared data obtained
by high-performance liquid chromatography coupled with mass spectrometry (LC–MS) to those provided by high-performance liquid
chromatography and tandem mass spectrometry (LC–MS–MS). Initial results obtained with LC–MS showed that the technique lacked
selectivity, which is why the method was validated by LC–MS–MS. This method involves a solid-phase extraction (SPE) of nitrofuran
metabolites and nitrofuran parent drugs, a derivatization by 2-nitrobenzaldehyde for 17 h, and finally a clean-up by SPE.
The data obtained show that the limits of detection varied between 0.2 and 0.6 μg kg −1 for the metabolites and between 1 and 2 μg kg −1 for nitrofuran parent drugs. The method was applied to different flower honeys. The results showed that nitrofurans (used
as antibiotics) are consistently present in this matrix, the predominant compound being furazolidone.

A method for the determination of catecholamine derivatives in human urine is proposed that includes the derivatization of target compounds on a solid-phase extraction cartridge and determination of the analytes by a UHPLC method with tandem mass-spectrometric detection. 9-Fluorenyl-methoxycarbonyl chloride was used as the derivatization agent. The limits of detection for the analytes were 2.5 ng mL ?1 for 9-fluorenyl-methoxycarbonyl-adrenaline, 5 ng mL ?1 for 9-fluorenyl-methoxycarbonyl-octopamine, and 25 ng mL ?1 for 9-fluorenyl-methoxycarbonyl-dopamine. Melarsoprol is the only currently available drug for treatment of the late stage of African trypanosomiasis (sleeping sickness). Unfortunately, the arsenic-containing drug causes serious side effects, for which the mechanisms have not been elucidated so far. This investigation describes the study of the melarsoprol biotransformation processes by electrochemical (EC) techniques. Based on EC, potential oxidation reactions of melarsoprol are examined. Moreover, the reactivity of melarsoprol, its metabolite melarsen oxide, and their oxidation products toward the tripeptide glutathione and the proteins hemoglobin and human serum albumin is evaluated. The combination of different analytical techniques allows the identification as well as the quantification of the biotransformation products. The hyphenation of liquid chromatography (LC) and electrospray ionization mass spectrometry (ESI–MS) is applied for identification and structure elucidation, which implies the determination of exact masses and fragmentation patterns. For the selective detection of arsenic containing metabolites, LC coupled to inductively coupled plasma mass spectrometry is utilized. Based on the obtained data, the oxidative biotransformation of melarsoprol can be predicted, revealing novel species which have been suspected, but not been identified up to now. The results of the protein studies prove that melarsen oxide, the active derivative of melarsoprol, strongly binds to human hemoglobin and forms different adducts via the free cysteinyl groups of the hemoglobin α- and β-chain. 相似文献

A stability indicating RP-HPLC method for cephalexin has been developed and validated to identify and characterize potential degradation products. Drug was subjected to hydrolytic (acidic, basic, and neutral), oxidative, thermal, and photolytic stresses as per ICH guidelines Q1A (R2) and Q1B. Chromatographic separation was achieved on C8 column with mixture of ammonium acetate buffer pH 4.5 and acetonitrile in gradient mode as a mobile phase with PDA detection. Specificity of the method was established by peak purity studies. Method was validated as per ICH guideline Q2 (R1) for accuracy, precision, linearity, sensitivity, and robustness. Kinetics for each degradation condition was studied with respect to order of reaction and rate constant. Method was found to comply with acceptance criteria of validation parameters with respect to specificity (peak purity greater than 0.999) linearity ( r 2 greater than 0.99), accuracy (% recovery in the range of 98–102%), and precision (% RSD not more than 2). A total of six degradation products were generated in different stress conditions; these were identified and structures were proposed using LC–MS/MS. Cephalexin undergoes degradation in almost all the conditions. The developed stability indicating method is suitable for analysis of stability samples as it adequately separates all degradation products. Degradation products generated in photolytic and oxidative conditions are reported for the first time. 相似文献

A sensitive, high-throughput and economic liquid chromatographic method for determination of fenofibric acid in human plasma was developed and validated by ultraviolet detection and tandem mass spectrometry, then applied in pharmacokinetic study to investigate Lipanthyl™ 200 mg MC bioavailability under food and fasting conditions. Fenofibric acid with 2-chloro fenofibric acid-d6 (internal standard) was extracted from 100 µL of human plasma by acetonitrile in a single extraction step. 25 and 2 µL from supernatant were injected onto ACE column, 50 mm, 5 micron with 4.6 mm inner diameter for LC–UV and 2.1 mm for LC–MS/MS, and both systems were eluted isocratically by water:methanol:formic acid (35:65:0.1, v/v/v), with a constant flow rate of 1 mL min−1. The established calibration curve was linear between 0.05–20 µg mL−1, and the within- and between-day precisions were all below 13 % in both LC–MS/MS and LC–UV systems during validation, and accuracies ranged between 91 and 112 %. Twenty-eight healthy adult subjects participated in this clinical study, and the pharmacokinetic parameters including coefficient of variation were calculated and discussed. A dramatic decrease in C

Here, we report a rapid and specific method based on high-performance liquid chromatography coupled with tandem mass spectrometry (LC–MS–MS) capable of quantifying six CYP450-specific probe substrates in human liver microsomal incubation mixtures simultaneously. These analytes were prepared by single-step extraction and detected in one run by switching polarity of electrospray ionization mode three times. Following optimization of the chromatographic conditions, the peaks were well separated, and retention times ranged between 2.0 and 8.4 min. The total run time for a single injection was within 9 min. This method was fully validated over linear range of 18.8–3,000.0 ng mL ?1 for diclofenac, 0.8–3,000.0 ng mL ?1 for dapson, 1.5–3,000.0 ng mL ?1 for dextromethorphan, 2.0–4,000.0 ng mL ?1 for omeprazole, 75.0–3,000.0 ng mL ?1 for chlorzoxazone and 0.8–3,000.0 ng mL ?1 for phenacetin using diazepam as internal standard. Samples were prepared by protein precipitation and analyzed on the LC–MS–MS equipped with ESI interface. For each analyte, inter- and intra-day precision (RSD%) were <15 % and accuracy was within 85–115 %. The specificity, precision, accuracy, stabilities and matrix effect were evaluated. 相似文献

A simple, selective and highly sensitive method was developed and optimized to determine the most commonly used UV filters with endocrine-disrupting potential in water, namely benzophenone-3 (BP-3), octocrylene (OC), ethylhexyl dimethyl p-aminobenzoate (OD-PABA), ethylhexyl methoxycinnamate, ethylhexyl salicylate (EHS) and homosalate (HMS). Samples were extracted by stir bar sorptive extraction followed by liquid desorption (SBSE-LD). The important factors influencing SBSE-LD were optimized. Under optimal conditions, assays were performed on 50 mL of water sample using stir bars (0.5 mm in film thickness, 10 mm in length) at room temperature. The analytes were determined by liquid chromatography–tandem mass spectrometry with triple quadrupole analyzer using atmospheric pressure chemical ionization. The main parameters in HPLC–APCI–MS/MS were also optimized to provide the best performances for all analytes. Moreover, matrix effect was investigated using two methods the post-column infusion system and the method of spiked matrices after extraction. As a result, no significant matrix effect on the analysis was observed. The method showed good linearity (R
2 coefficients greater than 0.996 in different water samples after SBSE-LD). Recoveries of the analytes were close to 90%, except for BP-3 (64%) and OC (76%) with relative standard deviation lower than 11%. Detection limits were between 0.6 and 3.3 ng L−1 for all the analytes except for HMS (94 ng L−1) and EHS (114 ng L−1). A simple and specific method for the simultaneous determination of eleutherosides B and E in powdered rhizomes of Eleutherococcus senticosus extract and in solid and liquid dietary supplements was developed and validated. E. senticosus extracts, often mixed with other plants or herbal extracts, are widely used in food supplements because of the tonic and adaptogenic activities referred to the eleutherosides B and E. In this study, samples were analyzed by a liquid chromatography-electrospray tandem mass spectrometry (LC–ESI-MS/MS) method operated in single reaction monitoring (SRM). Validation was carried out in terms of limit of detection (LOD), limit of quantitation (LOQ), linearity, precision and trueness. LOD and LOQ values were fixed at 3 μg L ?1 and 10 μg L ?1, respectively, whereas linearity was established within 10–1,000 μg L ?1 range for both compounds. Good precision was obtained for both eleutherosides in terms of intra-day precision (RSD % lower than 4 %) and inter-day precision (RSD % lower than 6 %). Good percentage recoveries were obtained for both eleutherosides (91.5–103.6 %). Finally, the developed method was successfully applied to analyze a number of solid and liquid commercial dietary supplements containing E. senticosus extracts, also mixed with other herbal extracts. 相似文献

The feasibility of using cloud-point extraction as a simple and effective means of recovery of memantine from rat plasma before LC–MS analysis has been demonstrated. A non-ionic surfactant Triton X-114 was used for extraction of the memantine. On increasing the temperature to the cloud point, phase separation occurred, resulting in an aqueous phase, and a surfactant-rich phase containing most of the analytes. The extraction conditions, for example amount of surfactant, temperature, NaOH concentration, and time of incubation, were optimized. Chromatographic separation was accomplished on a C 18 analytical column with 56:44 ( v/ v) methanol–0.2% aqueous formic acid as isocratic mobile phase at a flow rate of 0.8 mL min ?1. Under the optimum experimental conditions recovery was satisfactory (91–101%) without interference from the surfactant. The method was shown to be reproducible and reliable with intraday precision below 6.6%, interday precision below 14.3%, and linear range from 1 to 400 ng mL ?1. Benzene, toluene, ethylbenzene, and xylene (BTEX) are a group of volatile organic compounds that are ubiquitous in the environment due to numerous anthropogenic sources. Exposure to BTEX poses a health hazard by increasing the risk for damage to multiple organs, neurocognitive impairment and birth defects. Urinary BTEX metabolites are useful biomarkers for the evaluation of BTEX exposure, because of the ease of sampling and their longer physiological half-lives compared with parent compounds. A method that utilizes LC–MS/MS was developed and validated for simultaneously monitoring of 10 urinary BTEX metabolites. During the sample preparation an aliquot of urine was diluted with an equal volume of 1% formic acid; internal standard solution was added, and then the sample was centrifuged and analyzed. The analytes were separated on the Kinetex-F5 column by applying a linear gradient, consisting of 0.1% formic acid and methanol. The method was validated according to the FDA Bioanalytical Method Validation Guidance for Industry. The mean method's accuracies of the spiked matrix were 81–122%; the inter-day precision ranged from 4 to 20%; the limits of quantitation were 0.5–2 μg/L. Cationic polysaccharides containing N-hydroxypropyl- N, N, N-trimethylammonium substituents are widely used as conditioning agents for hair-care products. A sensitive method has been developed for the quantitation of these polymers. After acidic extraction from hair the polysaccharides are hydrolyzed using trifluoroacetic acid. The cationic monoglycosides are determined using liquid chromatography–tandem mass spectrometry (LC–MS–MS). The developed method is independent of hair treatment. Even hair cut from test persons after customary hair wash can be analyzed. After treatment of natural and bleached hair tresses using a real-life treatment procedure 180 g and 300 g of polymer per gram hair were quantified, respectively. Additionally the fragmentation mechanism of the cationic N-hydroxypropyl- N, N, N-trimethylammonium group during electrospray ionization was investigated. A mass loss of 60 Da in combination with loss of a single charge is observed and associated with cleavage of trimethylamine and a proton. It is assumed that this process is promoted by the anionic counter-ion which might be hydroxide in an aqueous environment. 相似文献

Summary A sensitive and selective liquid chromatographic method coupled with electrospray ionization tandem mass spectrometry (LC–ESI–MS–MS) has been developed for quantification of felodipine in human and dog plasma. Compounds were separated on a 2.0 mm × 150 mm, 5.0 m particle, C 8 column with 1 m m ammonium acetate–acetonitrile, 20:80, pH 6.0, as mobile phase at a flow rate of 200 L min –1. Nifedipine was used as internal standard. Plasma samples were extracted with diethyl ether, the centrifuged upper layer was evaporated, the residue was reconstituted with mobile phase, and the reconstituted samples were injected. The analytical column lasted for at least 1000 injections. By use of multiple reaction monitoring (MRM) mode in MS–MS felodipine and nifedipine were detected without severe interference from the human or dog plasma matrix. Felodipine produced a protonated precursor ion ([M + H] +) at m/z 384 and a corresponding product ion at m/z 338. And internal standard (nifedipine) produced a protonated precursor ion ([M + H] +) at m/z 347 and a corresponding product ion at m/z 315. Detection of felodipine in human and dog plasma was accurate and precise, with a limit of quantification of 0.05 ng mL –1. The method has been successfully applied to preliminary pharmacokinetic study of felodipine in human and dog plasma. 相似文献
