We report the development of cell-penetrating quantum dots (QDs) based on the use of multivalent and endosome-disrupting (endosomolytic) surface coatings. Hyperbranched copolymer ligands such as polyethylene glycol (PEG) grafted polyethylenimine (PEI-g-PEG) are found to encapsulate and solubilize luminescent quantum dots through direct ligand-exchange reactions. Because of the positive charges and a "proton sponge effect" associated with multivalent amine groups, this class of ligand-exchanged QDs is able to penetrate cell membranes and is also able to disrupt endosomal organelles in living cells. The grafted PEG segment is essential for reducing the cytotoxicity of PEI as well as for improving the overall nanoparticle stability and biocompatibility. In comparison with previous QDs encapsulated with amphiphilic polymers, the cell-penetrating QDs are smaller in size and are considerably more stable in acidic environments. Cellular uptake and imaging studies reveal that the number of PEG grafts per PEI molecule has a pronounced effect on the intracellular pathways of internalized QDs. In particular, QDs coated with PEI-g-PEG2 are rapidly internalized by endocytosis, and are initially stored in vesicles, followed by slow endosomal escape and release into the cytoplasm. These insights are important toward the design and development of nanoparticle agents for intracellular imaging and therapeutic applications.  相似文献

The motion of vesicles created through Langmuir monolayer collapse has been investigated. The vesicles grow only in a narrow molecular area range, and they exhibit remarkable, various biological cell-like behaviors such as division (cell division in cell biology, cytokinesis) and self-propulsion (motility). The vesicle division includes some dynamic modes: (i) an expulsion of a single satellite vesicle from an initial vesicle, (ii) a hierarchical and a sequential expulsion of a satellite vesicle, and (iii) a successive expulsion of two satellite vesicles from an initial vesicle. Two neighboring vesicles often show alternate fusion and division between them. Strong shape fluctuations dominate through vesicle division. The vesicles created exhibit distinct motions depending on the molecular area. At a large molecular area where most initial vesicles are created, they show a continuous, random motion on a few tens of micrometers length scale with a strong shape fluctuation and a constant velocity fluctuation profile. At a small molecular area they cease to move and shape fluctuations also become suppressed. At an intermediate molecular area there coexist vesicles with different dynamic modes: some vesicles show random motion similar to that at a large molecular area, but in a less fluctuating manner, while others exhibit a directional motion with an intermittent velocity jump. The directional motion is characterized by three distinct steps, i.e., extension, adhesion, and retraction. The characteristic motion is discussed from the viewpoint of haptotaxis, or the motion driven by adhesion gradients on the monolayer created by the local transfer of charged surfactant molecules between the vesicle and the monolayer, which the vesicle adheres to.  相似文献

Cell-penetrating peptides have been widely used to improve cellular delivery of a variety of proteins and antisense agents. However, recent studies indicate that such cationic peptides are predominantly entering cells via an endosomal pathway. We now show that the nuclear antisense effect in HeLa cells of a variety of peptide nucleic acid (PNA) peptide conjugates is significantly enhanced by addition of 6 mM Ca(2+) (as well as by the lysosomotrophic agent chloroquine). In particular, the antisense activities of Tat(48-60) and heptaarginine-conjugated PNAs were increased 44-fold and 8.5-fold, respectively. Evidence is presented that the mechanism involves endosomal release. The present results show that Ca(2+) can be used as an effective enhancer for in vitro cellular delivery of cationic peptide-conjugated PNA oligomers, and also emphasize the significance of the endosomal escape route for such peptides.  相似文献

Understanding cellular signaling mediated by cell surface receptors is key to modern biomedical research and drug development. The discovery of a growing number of potential molecular targets and therapeutic compounds requires downscaling and accelerated functional screening. Receptor-mediated cellular responses are typically investigated on single cells or cell populations. Here, we show how to monitor cellular signaling reactions at a yet unreached miniaturization level. On the basis of our observations, cytochalasin induces mammalian cells to extrude from their plasma membrane submicrometer-sized native vesicles. They comprise functional cell surface receptors correctly exposing their extracellular ligand binding sites on the outer vesicle surface and retaining cytosolic proteins in the vesicle interior. As a prototypical example, ligand binding to the ionotropic 5-HT(3) receptor and subsequent transmembrane Ca(2+) signaling were monitored in single attoliter vesicles. Thus, native vesicles are the smallest autonomous containers capable of performing cellular signaling reactions under physiological conditions. Because a single cell delivers about 50 native vesicles, which can be isolated and addressed as individuals, our concept allows multiple functional analyses of individual cells having a limited availability and opens new vistas for miniaturized bioanalytics.  相似文献

Copper nanoparticle (CuNPs) were successfully synthesized within the confined volume of niosomal vesicles. Metallic copper nanoparticles have been prepared in niosomal vesicles. The nanoparticle characteristics are guided by the specific properties of the niosomes. It has been found that the hydrophile: lipophile balance (HLB), area per molecule and gel-fluid transition temperature of the surfactants forming the niosome are important factors affecting nanoparticle characteristics. Entrapment ability, hydration volume, vesicle size and “leakiness” are the niosomal parameters that need to be optimized for nanoparticle formation. The synthesized nanoparticles function as very effective catalysts for reduction of Methylene Blue (MB) dye. This report gives a first hand account of how the particle characteristics of the CuNPs synthesized in niosomal vesicles can be related to their efficiency as catalysts. Since use of, niosomes for drug delivery and in cosmetic formations is well documented, the present work indicates the potential for prospective delivery of CuNPs via niosomes for various applications in future.  相似文献

Polymeric vesicles have attracted considerable attention in recent years, since they are a model for biological membranes and have versatile structures with several practical applications. In this study, we prepare vesicles from polystyrene-b-poly(acrylic acid) block copolymer in dioxane/water and dioxane/THF/water mixtures. We then examine the ability of additives (such as NaCl, HCl, or NaOH), solvent composition, and hydrophilic block length to control vesicle size. Using turbidity measurements and transmission electron microscopy (TEM) we show that larger vesicles can be prepared from a given copolymer by adding NaCl or HCl, while adding NaOH yields smaller vesicles. The solvent composition (ratio of dioxane to THF, as well as the water content) can also determine the vesicle size. From a given copolymer, smaller vesicles can be prepared by increasing the THF content in the THF/dioxane solvent mixture. In a given solvent mixture, vesicle size increases with water content, but such an increase is most pronounced when dioxane is used as the solvent. In THF-rich solutions, on the other hand, vesicle size changes only slightly with the water concentration. As to the effect of the acrylic acid block length, the results show that block copolymers with shorter hydrophilic blocks assemble into larger vesicles. The effect of additives and solvent composition on vesicle size is related to their influence on chain repulsion and aggregation number, whereas the effect of acrylic acid block length occurs because of the relationship among the block length, the width of the molecular weight distribution, and the stabilization of the vesicle curvature.  相似文献

The conformations of vesicles deformed by microtubules are studied within the framework of the curvature energy. The phenomenon in which the destruction of a microtubule is followed by the formation of peristaltic shapes on a protrusion created by the microtubule is investigated. The influence of the spontaneous curvature on the conformations of vesicles is examined, and the results are compared to existing experiments. The elastic properties of a vesicle deformed by the microtubule are studied.  相似文献

We had previously developed surface-modified poly (D,L-lactide-co-glycolide) (PLGA) nanoparticles (NPs) for use as a cellular drug delivery system. The cellular uptake of PLGA-NPs was mediated predominantly by endocytosis, and this uptake was increased by surface modifications with polymers, such as chitosan (CS) and polysorbate 80 (P80). In the present study, we prepared a cell-sized giant unilamellar vesicle (GUV) that mimics a cell membrane to investigate the interaction between cell membranes and NPs. Endocytosis-like uptake of NPs into a GUV was observed when the NPs were modified with nonionic surfactant P80 probably due to change in viscoelasticity and enhanced fusion activity of the membrane induced by P80. In contrast, unmodified NPs and those modified with CS were not internalized into a GUV. These results suggest that surface properties of PLGA-NPs are an important formulation parameter for their interaction with lipid membranes.  相似文献

Using two of the most commonly synthesized noble metal nanoparticle preparations, citrate-reduced Au and Ag, the impacts of short-term accidental nanoparticle exposure are examined in primary culture murine adrenal medullary chromaffin cells. Transmission electron microscopy (TEM), inductively coupled plasma atomic emission spectroscopy (ICP-AES) and Alamar Blue viability studies revealed that nanoparticles are taken up by cells but do not decrease cell viability within 48 hours of exposure. Carbon-fiber microelectrode amperometry (CFMA) examination of exocytosis in nanoparticle-exposed cells revealed that nanoparticle exposure does lead to decreased secretion of chemical messenger molecules, of up to 32.5% at 48 hours of Au exposure. The kinetics of intravesicular species liberation also slows after nanoparticle exposure, between 30 and 50% for Au and Ag, respectively. Repeated stimulation of exocytosis demonstrated that these effects persisted during subsequent stimulations, meaning that nanoparticles do not interfere directly with the vesicle recycling machinery but also that cellular function is unable to recover following vesicle content expulsion. By comparing these trends with parallel studies done using mast cells, it is clear that similar exocytosis perturbations occur across cell types following noble metal nanoparticle exposure, supporting a generalizable effect of nanoparticle-vesicle interactions.  相似文献

We used correlative transmission electron microscopy (TEM) and nanoscale secondary ion mass spectrometry (NanoSIMS) imaging to quantify the contents of subvesicular compartments, and to measure the partial release fraction of 13C-dopamine in cellular nanovesicles as a function of size. Three modes of exocytosis comprise full release, kiss-and-run, and partial release. The latter has been subject to scientific debate, despite a growing amount of supporting literature. We tailored culturing procedures to alter vesicle size and definitively show no size correlation with the fraction of partial release. In NanoSIMS images, vesicle content was indicated by the presence of isotopic dopamine, while vesicles which underwent partial release were identified by the presence of an 127I-labelled drug, to which they were exposed during exocytosis allowing entry into the open vesicle prior to its closing again. Demonstration of similar partial release fractions indicates that this mode of exocytosis is predominant across a wide range of vesicle sizes.  相似文献

Asymmetric cell division is common in biology and plays critical roles in differentiation and development. Unicellular organisms are often used as model systems for understanding the origins and consequences of asymmetry during cell division. Although basic as compared to mammalian cells, these are already quite complex. We report complete budding and asymmetric fission of very simple nonliving model cells to produce daughter vesicles that are chemically distinct in both interior and membrane compositions. Our model cells are based on giant lipid vesicles (GVs, 10-30 μm) encapsulating a polyethylene glycol (PEG)/dextran aqueous two-phase system (ATPS) as a crowded and compartmentalized cytoplasm mimic. Ternary lipid compositions were used to provide coexisting micrometer-scale liquid disordered (L(d)) and liquid ordered (L(o)) domains in the membranes. ATPS-containing vesicles formed buds when sucrose was added externally to provide increased osmotic pressure, such that they became not only morphologically asymmetric but also asymmetric in both their interior and their membrane compositions. Further increases in osmolality drove formation of two chemically distinct daughter vesicles, which were in some cases connected by a lipid nanotube (complete budding), and in others were not (fission). In all cases, separation occurred at the aqueous-aqueous phase boundary, such that one daughter vesicle contained the PEG-rich aqueous phase and the other contained the dextran-rich aqueous phase. PEGylated lipids localized in the L(o) domain resulted in this membrane domain preferentially coating the PEG-rich bud prior to division, and subsequently the PEG-rich daughter vesicle. Varying the mole ratio of lipids resulted in excess surface area of L(o) or L(d) membrane domains such that, upon division, this excess portion was inherited by one of the daughter vesicles. In some cases, a second "generation" of aqueous phase separation and budding could be induced in these daughter vesicles. Asymmetric fission of a simple self-assembled model cell, with production of daughter vesicles that harbored different protein concentrations and lipid compositions, is an example of the seemingly complex behavior possible for simple molecular assemblies. These compartmentalized and asymmetrically dividing ATPS-containing GVs could serve as a test bed for investigating possible roles for spatial and organizational cues in asymmetric cell division and inheritance.  相似文献

In this paper, we report a novel approach using peptide CALNN and its derivative CALNNGGRRRRRRRR (CALNNR(8)) to functionalize gold nanoparticles for intracellular component targeting. The translocation is effected by the nanoparticle diameter and CALNNR(8) surface coverage. The intracellular distributions of the complexes are change from the cellular nucleus to the endoplasmic reticulum by increasing the density of CALNNR(8) at a constant nanoparticle diameter. Additionally, increasing the nanoparticle diameter at a constant density of CALNNR(8) leads to less cellular internalization. These translocations of the complexes cause unique colorimetric expressions of the cell structure. The cell viability is affected by the internalized gold nanoparticle-peptide complexes in terms of quantities of particles per cell. In addition, the intracellular distribution of the fluorescence quenching is investigated by a fluorescent confocal scanning laser microscopy, which also gives further evidence of intracellular distribution of the gold nanoparticle-peptide complexes.  相似文献

本文报告了用于检测单囊泡及其粒径分析的石英纳米通道电阻-脉冲分析方法. 采用圆柱形石英纳米通道可检测粒径为100~300 nm的单磷脂囊泡和直径为170~400 nm的聚苯乙烯纳米颗粒. 单囊泡和纳米颗粒的迁移可通过检测各自产生的方波电流脉冲信号, 并由此确定颗粒尺寸. 结果表明,采用石英纳米通道电阻-脉冲分析方法得到的颗粒/囊泡粒径与采用动态光散射法和扫描电子显微法得到的结果完全一致. 这种基于电子的分析方法具有快速简单的特点,所用的自制微传感器廉价耐用. 石英通道的应用还可与其它分析方法如电流分析法和荧光显微法联用,以获得生物囊泡及人工囊泡更完全的信息.  相似文献

Lipid bilayer membranes are known to form various structures such as large sheets or vesicles. When the two leaflets of the bilayer have an equal composition, the membrane preferentially forms a flat sheet or a spherical vesicle. However, a difference in the composition of the two leaflets may result in a curved bilayer or in a wide variety of vesicle shapes. Vesicles with different shapes have already been shown in experiments and diverse vesicle shapes have been predicted theoretically from energy minimization of continuous curves. Here we present a molecular dynamics study of the effect of small changes in the phospholipid headgroups on the spontaneous curvature of the bilayer and on the resulting vesicle shape transformations. Small asymmetries in the bilayers already result in high spontaneous curvature and large vesicle deformations. Vesicle shapes that are formed include ellipsoids, discoids, pear-shaped vesicles, cup-shaped vesicles, as well as budded vesicles. Comparison of these vesicles with theoretically derived vesicle shapes shows both resemblances and differences.  相似文献

Vesicle fusion has long provided an easy and reliable method to form supported lipid bilayers (SLBs) from simple, zwitterionic vesicles on siliceous substrates. However, for complex compositions, such as vesicles with high cholesterol content and multiple lipid types, the energy barrier for the vesicle-to-bilayer transition is increased or the required vesicle–vesicle and vesicle–substrate interactions are insufficient for vesicle fusion. Thus, for vesicle compositions that more accurately mimic native membranes, vesicle fusion often fails to form SLBs. In this paper, we review three approaches to overcome these barriers to form complex, biomimetic SLBs via vesicle fusion: (i) optimization of experimental conditions (e.g., temperature, buffer ionic strength, osmotic stress, cation valency, and buffer pH), (ii) α-helical (AH) peptide-induced vesicle fusion, and (iii) bilayer edge-induced vesicle fusion. AH peptide-induced vesicle fusion can form complex SLBs on multiple substrate types without the use of additional equipment. Bilayer edge-induced vesicle fusion uses microfluidics to form SLBs from vesicles with complex composition, including vesicles derived from native cell membranes. Collectively, this review introduces vesicle fusion techniques that can be generalized for many biomimetic vesicle compositions and many substrate types, and thus will aid efforts to reliably create complex SLB platforms on a range of substrates.  相似文献

Vesicles form spontaneously in a aqueous mixture of sodium bis(2-ethylhexyl) sulfosuccinate (Aerosol OT) and lauryl sulfonate betaine (LSB). Different from catanionic vesicles, the formation or disaggregation of such zwitterionic/anionic vesicles may be easily controlled by adjusting the relative amount of LSB and salinity. The participation of LSB reduces the polydispersity of the vesicles and even results in the formation of monodispersed vesicles at a certain salinity. But as LSB exceeds a certain proportion, vesicles cannot form at any concentration and salinity, making convenient the study of the structural transitions. We applied pyrene as a fluorescence probe and monitored the transition among the monomer, micelle, and vesicle through the variation of I(1)/I(3), accompanied by conductivity and turbidity measurements. In LSB solution and LSB-rich mixture, an abrupt change of the ratio of I(1)/I(3) was found in the transition from monomer to micelle with increasing concentration, as well as in the transition from micelle to vesicle with increasing salinity, which shows that a difference of the polarity of the microenvironment between the micelle and the vesicle bilayer resulted from the composition change. But in AOT solution and AOT-rich mixture, only a gradual change in the transition is observed due to the existence of intermediate structures, which have different microenvironments from micelles and vesicles. So the formation of vesicle experiences a process of monomer to premicelle to micelle to bilayer segment with increasing concentration by combining the conductivity method. The ratio of I(1)/I(3) is independent of the vesicle size once formed.  相似文献

Combined dynamic and static light scattering (DLS, SLS) and cryogenic transmission electron microscopy (cryo-TEM) were used to investigate extruded cationic vesicles of dioctadecyldimethylammonium chloride and bromide (DODAX, X being Cl(-) or Br(-)). In salt-free dispersions the mean hydrodynamic diameter, D(h), and the weight average molecular weight, M(w), are larger for DODAB than for DODAC vesicles, and both D(h) and M(w) increase with the diameter (varphi) of the extrusion filter. NaCl (NaBr) decreases (increases) the DODAB (DODAC) vesicle size, reflecting the general trend of DODAB to assemble as larger vesicles than DODAC. The polydispersity index is lower than 0.25, indicating the dispersions are rather polydisperse. Cryo-TEM micrographs show that the smaller vesicles are spherical while the larger ones are oblong or faceted, and the vesicle samples are fairly polydisperse in size and morphology. They also indicate that the vesicle size increases with phi and DODAB assembles as larger vesicles than DODAC. Lens-shaped vesicles were observed in the extruded preparations. Both light scattering and cryo-TEM indicate that the vesicle size is larger or smaller than phi when phi is smaller or larger than the optimal phi approximately 200 nm.  相似文献

The novel and burgeoning technique of surfaced-enhanced cellular fluorescence imaging has tremendous potential in the monitoring and investigation of intracellular processes at the single-molecular level, for instance, high-resolution cellular imaging, long-term in vivo observation of cell trafficking, tumor targeting, and diagnostics. The success hinges on the development and fabrication of plasmonic nanostructured surfaces with size and shape compatible with cell interactions because they are crucial to enhanced cellular imaging. In this review, the mechanism of surface-enhanced cellular fluorescence imaging is discussed in view of metal-enhanced fluorescence. The design of nanostructured surfaces with evenly distributed plasmonic fields suitable for enhanced cellular fluorescence imaging such as nanoparticle superlattice coatings, lithographically-based substrates, and alumina-templated surface are described.  相似文献

An artificial glycocalix self-assembles when unilamellar bilayer vesicles of amphiphilic β-cyclodextrins are decorated with maltose- and lactose-adamantane conjugates by host-guest interactions. The maltose-decorated vesicles aggregate in the presence of lectin concanavalin A whereas the lactose-decorated vesicles aggregate in the presence of lectin peanut agglutinin. The kinetics of the orthogonal multivalent interfacial interactions present in this ternary system of vesicles, carbohydrates, and lectins were studied by time-dependent measurements of the optical density at 400 nm. The average vesicle and vesicle aggregate sizes were monitored by dynamic light scattering. The aggregation process was evaluated as a function of lectin concentration, vesicle concentration, and surface coverage of the vesicles by the carbohydrate-adamantane conjugates. The initial rate of vesicle aggregation scales linearly with the lectin as well as the cyclodextrin vesicle concentration. Furthermore, each lectin requires a characteristic critical density of carbohydrates at the vesicle surface. These observations allow a prediction of the response of the ternary supramolecular system at different concentrations of its components. Also, the effective binding site separation in a multivalent receptor such as a multiple binding site protein can be accurately determined. This methodology can be extended to multivalent noncovalent interactions in other ligand-receptor systems at interfaces.  相似文献

The fusion and fission behaviors of exosomes are essential for the cell-to-cell communication. Developing exosome-mimetic vesicles with such behaviors is of vital importance, but still remains a big challenge. Presented herein is an artificial supramolecular vesicle that exhibits redox-modulated reversible fusion-fission functions. These vesicles tend to fuse together and form large-sized vesicles upon oxidation, undergo a fission process and then return to small-sized vesicles through reduction. Noteworthy, the aggregation-induced emission (AIE) characteristics of the supramolecular building blocks enable the molecular configuration during vesicular transformation to be monitored by fluorescence technology. Moreover, the presented vesicles are excellent nanocarrier candidates to transfer siRNA into cancer cells.  相似文献
