Wittig reactions of 2-furaldehyde (20) [and thiophene-2-carbaldehyde (21)] with (3-guaiazulenylmethyl)triphenylphosphonium bromide (19) in ethanol containing NaOEt at 25 °C for 24 h under argon give (E)-1-(2-furyl)-2-(3-guaiazulenyl)ethylene (22E) and (E)-1-(2-thienyl)-2-(3-guaiazulenyl)ethylene (23E) in 53 and 36% yields. Similarly, Wittig reactions of 3-furaldehyde (29) [and thiophene-3-carbaldehyde (30)] with 19 under the same reaction conditions as for 20 and 21 afford (E)-1-(3-furyl)-2-(3-guaiazulenyl)ethylene (31E) and (E)-1-(3-thienyl)-2-(3-guaiazulenyl)ethylene (32E) in 32 and 46% yields. Molecular structures and characteristic properties as well as preparation of the title E (i.e., one of the geometrical isomers) forms, with a view to comparative study, are reported. Moreover, reactions of those conjugated π-electron systems with TCNE (=tetracyanoethylene) in benzene [and in DMF (=N,N-dimethylformamide)] at 25 °C for 24 h under argon yield unique products, possessing interesting molecular structures, respectively, whose characteristic properties and crystal structures are documented, also.  相似文献

An efficient synthesis of both enantiomers of dimethyl δ-amino-β-hydroxyethylphosphonate 6 has been achieved starting from anthranilic acid, through the resolution of dimethyl (±)-2-(2-N,N-dibenzylaminophenyl)-2-hydroxyethylphosphonate 9 with (S)-O-methylmandelic acid. The absolute configuration of the enantiomers 9 was assigned by the Dale and Mosher approach using the extended Newman projections and molecular mechanics.  相似文献

Racemic 8-ethoxy-2-(4-fluorophenyl)-3-nitro-2H-chromene (S14161) was recently identified as a potent inhibitor of phosphoinositide 3-kinase (PI3K). In order to investigate the effects of its two enantiomers on tumor cell lines, we designed a novel synthesis for (R)-S14161 and (S)-S14161 using a chemical resolution and derivation strategy. The readily available 3-(tert-butyldimethylsilyloxy)-salicylaldehyde underwent a tandem oxa-Michael–Henry reaction with trans-4-fluoro-β-nitrostyrene in the presence of a catalytic amount of l-proline and triethylamine to give the 3-nitro-2H-chromene. Upon removal of the TBS protecting group, the resolution of the resulting racemic 2-(4-fluorophenyl)-8-hydroxy-3-nitro-2H-chromene was achieved via diastereomeric ester formation using (S)-(+)-α-methoxyphenylacetic acid as the derivatizing agent, followed by aminolysis. Finally, the ethyl ether formation of each enantiomer furnished (R)-S14161 and (S)-S14161 in enantiomerically pure forms. The absolute configurations of these chiral molecules were determined by a circular dichroism method. The two enantiomers showed no marked differences in inhibition of growth of human myeloma LP1 and OPM-2 cells.  相似文献

Fischer indolisation of 2-aminophenyl ethyl ketone phenylhydrazone using glacial acetic acid saturated with hydrogen chloride as catalyst affords 3-methylindolo(l′:2′-3:4)2-methylquinazoline and 2-(2-aminophenyl)-3-methylindole. The latter compound is autoxidised to 2-(2-amino-phenyl)-3-hydroxy-3-methyl-3H-indole, a reaction which is shown to be dependent upon the presence of the primary amino group at the 2-position of the 2-phenyl substituent and which is much slower than the corresponding autoxidation of 2-(2-hydroxyphenyl)-3-methylindole to 3-hydroxy-2-(2-hydroxyphenyl)-3-methyl-3H-indole previously reported.Nitration of isopropyl phenyl ketone occurs preferentially at the ortho- rather than the meta- positions of the benzene nucleus.  相似文献

An efficient synthesis of tert-butyl-(2S)-2-[(tert-butoxycarbonyl)amino]-4-(2-oxiranyl) butanoate (5), the key intermediate for preparation of collagen cross-links (+)-pyridinoline (Pyd, 1) and (+)-deoxypyridinoline (Dpd, 2) was described from (4S)-5-(tert-butoxy)-4-[(tert-butoxycarbonyl)amino]-5-oxopentanoic acid (6) in six steps. Spectral and photochemical properties of 1-(9-phehanthryl)-2-(2-quinolyl)ethylene (9Ph2QE) in neutral and protonated forms have been investigated. It has been found that both isomers of 9Ph2QE are photoactive. The quantum yield of transcis photoisomerization (? tc = 0.47) in the neutral form is typical of the diabatic photoisomerization; on passing to the protonated form, ? tc increases up to 0.70. Thus, the double annelation of the 2-styrylquinoline phenyl group to form 9Ph2QE makes it possible to conserve the α-effect, which consists in an increase in the quantum yield to ? tc > 0.5 on passing from the neutral to protonated form, whereas the effect disappears for other types of annelation (naphthylquinolylethylenes, 1-(9-anthryl)-2-(2-quinolyl)ethylene).  相似文献

An efficient synthetic method for the preparation of (E)- and (Z)-3(5)-(2-hydroxyphenyl)-4-styrylpyrazoles has been developed. The reaction of (E)- and (Z)-3-styrylchromones with hydrazine hydrate afforded the corresponding (E)- and (Z)-4-styrylpyrazoles, respectively, saved 4′-nitro-derivatives where both (E)- and (Z)-4′-nitro-3-styrylchromones afforded (E)-3(5)-(2-hydroxyphenyl)-4-(4-nitrostyryl)pyrazoles. The reaction mechanism for these transformations was discussed and the stereochemistry of all products was assigned by NMR experiments.  相似文献

2-Methoxy-2-(9-phenanthryl)propionic acid was synthesized as a novel chiral resolving agent. The absolute configuration of (+)-2-methoxy-2-(9-phenanthryl)propionic acid was determined to be S by using X-ray structural analysis of the (1R,2S,5R)-menthyl ester. In the crystal, the methoxyl and carbonyl groups of the ester are in a syn-periplanar position. The syn-periplanar conformations of (1R,2S,5R)-menthyl esters were also observed by the NMR analyses in CDCl3. The utility of (S)-(+)-2-methoxy-2-(9-phenanthryl)propionic acid was exemplified by the resolution of (±)-3-octanol.  相似文献

2-(2-Oxo-2-arylethylidene)-2,3-dihydropyrimidine-4(1H)-ones react with aromatic and heteroaromatic aldehydes to form the unsaturated ketones, whereas in the case of 3- and 4-benzaldehydes the corresponding trans-2-styrylpyrimidine-4(3H)-ones were obtained. A possible mechanism of hydrolytic cleavage of the product of condensation of 2-(2-oxo-2-phenylethylidene)-2,3-dihydropyrimidine-4(1H)-one with paraformaldehyde under acid catalysis and mechanochemical activation has been discussed.  相似文献

NMR spectral data of some (E)-α-phenyl-β-(2-thienyl) acrylic acids indicate that these compounds exist in the preferred s-trans conformation. In the case of (E)-α-phenyl-β-(2-furyl)acrylic acids and their methyl esters the presence of only s-cis rotamer has been established.  相似文献

The compounds (E)-1,2-diphenyl-2-(phenylimino)ethanol, (E)-1,2-diphenyl-2-(p-tolylimino)ethanol, and (E)-2-((4-chlorophenyl)imino)-1,2-diphenylethanol) were synthesized by reaction of a p-substituted aniline with benzoin then oxidized with chromium trioxide–triethylamine in chloroform to give (E)-1,2-diphenyl-2-(phenylimino)ethanone, (E)-1,2-diphenyl-2-(p-tolylimino)ethanone, and (E)-2-((4-chlorophenyl)imino)-1,2-diphenylethanone in very high yield. The preparation and resolution of two new axially chiral quinazoline-containing phosphinamine ligands, 2-(2-pyridyl)-Quinazolinap and 2-(2-pyrazinyl)-Quinazolinap, is described. The ligands were synthesised in good yield over eight steps and included two Pd-catalysed reactions, a Suzuki coupling to form the biaryl linkage and the introduction of the diphenylphosphino group, as the key transformations. The racemic ligands were resolved via the fractional crystallisation of diastereomeric palladacycles derived from (+)-di-μ-chlorobis{(R)-dimethyl[1-(1-naphthyl)ethyl]aminato-C2,N}dipalladium (II) X-ray crystal structures of the (S,R)-2-pyridyl- and (S,R)-2-pyrazinyl-palladacycles are included. Displacement of the resolving agent by reaction with 1,2-bis(diphenylphosphino)ethane gave enantiopure 2-(2-pyridyl)-Quinazolinap and 2-(2-pyrazinyl)-Quinazolinap, new atropisomeric phosphinamine ligands for asymmetric catalysis. These ligands were applied in the palladium-catalysed allylic substitution of 1,3-diphenylprop-2-enyl acetate resulting in moderate conversions and enantioselectivities of up to 81%.  相似文献

This paper describes the selective syntheses of two cis-isomer-enriched cyclopentanone fragrances: (Z)-3-(2-oxopropyl)-2-(pent-2-en-1-yl)cyclopentanone (four steps, 62% overall yield, 67% cis) and Magnolione® (five steps, 60% overall yield, 55% cis). In addition, the asymmetric synthesis of (3aR,7aS)-5-methyl-2,3,3a,4,7,7a-hexahydro-1H-inden-1-one as well as (3a′R,7a′S)-5′-methyl-2′,3′,3a′,4′,7′,7a′-hexahydrospiro[[1,3]dioxolane-2,1′-indene] has been realized by an efficient kinetic resolution, which enables the selective synthesis of the 2S,3R-isomer-enriched 3 and 4.  相似文献

The recently discovered [4+1]-spirocyclization of nitroalkenes to indoles provided a convenient new approach to 2-(1H-indol-2-yl)acetonitriles. However, this reaction was complicated by the formation of inert 3-(2-nitroethyl)-1H-indole byproducts. Herein, we offer a workaround this problem that allows for effective transformation of the unwanted byproducts into acetonitrile target molecules.  相似文献

Complexes of 5-(phenylazo)-2-thiohydantoin (L1) and 5-(2-hydroxyphenylazo)-2-thiohydantoin (HL2) with Co(II), Ni(II) and Cu(II) salts have been synthesised and characterized by elemental analysis, conductivity, magnetic susceptibility, UV-Vis, IR, ESR and TG studies. The magnetic and spectral data suggested octahedral geometry for [L1M(OAc)2(H2O)2xH2O {M=Nill and Cull} and [L1CuCl2(H2O)]·H2O (dimeric form for the latter), trigonal bipyramidal geometry for [L2Co(OAc)(H2O)]·2H2O, square pyramidal geometry for [L2Ni(OAc)(H2O)]·H2O and square planar geometry for [L2CuCl]·2H2O. TG studies confirmed the chemical formulations of these complexes and showed that their thermal degradation takes place in three to five steps, depending on the type of the ligand and the geometry of the complex. The kinetic parameters (n, E#, A, ΔH#, ΔS# and ΔG#) of the thermal decomposition stages were computed using the Coats-Redfern and other standard equations and are discussed.  相似文献
