A reliable ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) method for simultaneous determination of aflatoxins B1, B2, G1, G2, M1 and M2 in peanuts and their derivative products was developed. The sample was extracted by 84% of acetonitrile aqueous solution and the extract was purified by a reliable solid phase extraction-based clean-up method. Then, the analytes were separated on Acquity UPLC HSS T3 column (100 mm × 2.1 mm, 1.8 μm particle size), and eluted with a mobile phase consisting of (A) water containing 0.1% formic acid and (B) acetonitrile/methanol (50/50, v/v). The separated compounds were detected with a Waters Micromass Quattro Ultima Pt tandem quadrupole mass spectrometer operating in positive electro-spray ionization using multiple reaction monitoring mode. The established method was extensively validated by determining the linearity (R2 ≥ 0.9990), average recovery (74.7-86.8%) and precision (relative standard deviation ≤ 10.9%). It was shown to be a suitable method for simultaneous determination of the six aflatoxins in peanuts and their derivative products. Finally, a total of 73 samples randomly collected from different areas in Zhejiang province were screened for aflatoxins with the proposed method. The results showed that 31 samples of peanut butter, 14 samples of fresh peanut and 5 samples of musty peanut were contaminated with aflatoxins. Meanwhile, this was the first report on aflatoxins M1 and M2, which were found in unprocessed peanuts and their derivative products.  相似文献

An ion-pairing reversed-phase liquid chromatography–mass spectrometry (IP-RP-LC–MS) was developed for the determination of nucleotides, nucleosides and their transformation products in Cordyceps. Perfluorinated carboxylic acid, namely pentadecafluorooctanoic acid (PDFOA, 0.25 mM), was used as volatile ion-paring agent and a reversed-phase column (Agilent ZORBAX SB-Aq column) was used for the separation of three nucleotides namely uridine-5′-monophosphate (UMP, 0.638–10.200 μg/mL), adenosine-5′-monophosphate (AMP, 0.24–7.80 μg/mL) and guanosine-5′-monophosphate (GMP, 0.42–13.50 μg/mL), seven nucleosides including adenosine (0.55–8.85 μg/mL), guanosine (0.42–6.75 μg/mL), uridine (0.33–10.50 μg/mL), inosine (0.21–6.60 μg/mL), cytidine (0.48–15.30 μg/mL), thymidine (0.20–6.30 μg/mL) and cordycepin (0.09–1.50 μg/mL), as well as six nucleobases, adenine (0.22–6.90 μg/mL), guanine (0.26–4.20 μg/mL), uracil (0.38–12.15 μg/mL), hypoxanthine (0.13–4.20 μg/mL), cytosine (0.39–12.45 μg/mL) and thymine (0.26–8.25 μg/mL) with 5-chlorocytosine arabinoside as the internal standard. The overall LODs and LOQs were between 0.01–0.16 μg/mL and 0.04–0.41 μg/mL for the 16 analytes, respectively. The contents of 16 investigated compounds in natural and cultured Cordyceps were also determined and compared after validation of the developed IP-RP-LC-MS method. The transformations of nucleotides and nucleosides in Cordyceps were evaluated based on the quantification of the investigated compounds in three extracts, including boiling water extraction (BWE), 24 h ambient temperature water immersion (ATWE) and 56 h ATWE extracts. Two transformation pathways including UMP → uridine → uracil and GMP → guanosine → guanine were proposed in both natural Cordyceps sinensis and cultured Cordyceps militaris. The pathway of AMP → adenosine → inosine → hypoxanthine was proposed in natural C. sinensis, while AMP → adenosine → adenine in cultured C. militaris. However, the transformation of nucleotides and nucleosides was not found in commercial cultured C. sinensis.  相似文献

A simple hydrophilic-interaction chromatography (HILIC) method was developed for the identification and quantification of 14 nucleosides and nucleobases, namely cytosine, uracil, cytidine, guanine, hypoxanthine, xanthine, uridine, thymine, inosine, guanosine, thymidine, 2′-deoxyadenosine, 2′-deoxyinosine and 2′-deoxyuridine in two traditional Chinese medicines, Geosaurus and Leech. The separation was achieved on a TSKgel Amide-80 column (150 mm × 2.0 mm, 3.0 μm) with a mixture of acetonitrile and 10 mM aqueous ammonium acetate as the mobile phase at a flow rate of 0.2 mL/min. The temperature was set at 30 °C and UV detection wavelength was set at 260 nm. All calibration curves showed good linearity (R2 > 0.9957) within the test ranges. The overall intra- and inter-day RSD ranged from 0.4 to 3.4% and from 0.7 to 3.3%, respectively. The LOD and LOQ were in the range of 0.07-30.49 ng/mL and 0.26-60.98 ng/mL, respectively. The repeatability of the method was in the range of 2.2-5.8% for Geosaurus and 1.4-5.5% for Leech. The recoveries of the samples were in the range of 91.4-100.9% for Geosaurus, and 91.9-99.3% for Leech. The established method was applied successfully for the analysis of nucleosides and nucleobases in 22 commercially available samples collected from different regions in China and Japan. Our data showed that HILIC had advantages as a useful tool for the study of the bioactive components in Geosaurus and Leech as well as their quality control, and could therefore be used for the determination of the analytes in pharmaceutical products and biological fluids.  相似文献

A new method based on accelerated solvent extraction (ASE) followed by ultra performance liquid chromatography (UPLC) analysis has been developed for the identification and quantification of major alkaloids in extracts of Coptis chinensis Franch. The UPLC system consisted of a dual detection system of photodiode array detector (PDA) and positive ion electrospray ionization-tandem mass spectrometry (ESI-MS/MS) in sequential configuration. The operational parameters of ASE including extraction solvent, extraction temperature, static extraction time and extraction cycles were optimized. UPLC analysis was performed on an ACQUITY UPLC BEH C18 column eluted by a mobile phase of acetonitrile spiked with a buffer solution consisting of 0.50% acetic acid and 20 mmol L−1 ammonium acetate. A tandem quadrupole spectrometer operating in either full scan mode or in MS/MS mode for multiple reaction monitoring (MRM) was used for the identification and quantitative analysis of eight major alkaloids in C. chinensis Franch extracts. The samples were also analyzed on a high-performance liquid chromatography-electrospray ionization-time-of-flight mass spectrometry (HPLC-ESI-TOF-MS) system to confirm the identification results. Three of the eight major alkaloids, berberine, palmatine and jatrorrhizine were quantified by UPLC-PDA and UPLC-MS/MS. The results indicated that both UPLC-PDA and UPLC-MS/MS methods were simple, sensitive and reliable for the determination of alkaloids in C. chinensis Franch. Seven Huanglian samples from different locations were analyzed using the established methods. UPLC fingerprints based on the distribution of the eight major alkaloids can serve as a rapid and reliable method for the authentication and quality evaluation of traditional Chinese medicine (TCM) herbs.  相似文献

A simple and sensitive method for determination of the O-demethylation activity of rat, dog, minipig, and human liver micrsomes toward paeonol using ultra-performance liquid chromatography with mass detection (UPLC-MS) has been developed. The method uses chemically synthesized O-demethylated metabolite of paeonol (2,4-dihydroxyacetophenone, DHA) as a standard for method validation. Validation was done with respect to specificity, linearity, detection limit, recovery, stability, precision and accuracy. The chromatographic separation was achieved on a UPLC BEH C18 column (50 mm × 2.1 mm i.d., 1.7 μm), with phase of acetonitrile-water (ratio 30:70). Selective ion reaction (SIR) monitor was specific for paeonol, DHA and I.S. The method was specific since there were no interference peaks from the reaction matrix. The calibration curve for DHA was linear from 0.5-100 μM with r2 = 0.9999. The newly developed method has good precision and accuracy. The method was successfully used to determine the kinetics of DHA activities toward paeonol in liver microsomes from different species. Dog liver microsomes (DLMs) were the most active in paeonol O-demethylation (709.7 pmol/min/mg protein) followed by rat liver microsomes (RLMs) (579.6 pmol/min/mg protein), HLMs (569.3 pmol/min/mg protein), and then minipig liver microsomes (PLMs) (417.3 pmol/min/mg protein). The developed method was appropriated for rapid screening paeonol O-demethylation activity in liver microsomes from different species.  相似文献

We present a comparison of two sensitive methods, HPLC with fluorescence detector (HPLC/FLD) and UPLC with electrospray tandem mass spectrometry (UPLC/MS/MS), for the determination of indoleamine neurotransmitters (NTs) and their metabolites in sea lamprey plasma samples. Liquid–liquid extraction (LLE) and solid-phase extraction (SPE) were also tested for recovery and matrix effect. The recoveries of SPE determined by HPLC/FLD and UPLC/MS/MS ranged from 75 to 123% and 78 to 105%, respectively, while the recoveries of LLE ranged from 45 to 73% and 48 to 75%, respectively. SPE combined with HPLC/FLD and UPLC/MS/MS to determine the target analytes in plasma samples were validated of the sensitivity, reproducibility, accuracy and precision. Both methods exhibited excellent linearity in the range of 0.2–50 ng mL−1 for all analytes. The limits of detection (LOD) varied from 0.04 ng mL−1 to 0.13 ng mL−1 for HPLC/FLD method and 0.003 ng mL−1 to 0.02 ng mL−1 for UPLC/MS/MS method. The inter-day accuracy ranged from 82.5 to 127.0% for HPLC/FLD and 93.0 to 113.0% for UPLC/MS/MS. The inter-day precision ranged from 9.9 to 32.3% for HPLC/FLD and 5.4 to 13.2% for UPLC/MS/MS. These results demonstrated that the values obtained by both methods were within the satisfactory range and the UPLC/MS/MS method provided more accurate and precise measurements than HPLC/FLD method. The comparison is of great importance to determine the available detectors, considering the complexity and expensiveness versus quality parameters. These two methods were applied to the analysis of four important indoleamine neurotransmitter analytes (5-hydroxytryptamine, 5-hydroxyindole-3-acetic acid, tryptamine and melatonin) in sea lamprey plasma samples.  相似文献

Present study described a simple, sensitive, and viable method for the determination of bisphenol A, 4-n-nonylphenol and 4-tert-octylphenol in water samples using temperature-controlled ionic liquid dispersive liquid-phase microextraction coupled to high performance liquid chromatography-fluorescence detector. In this experiment, 1-octyl-3-methylimidazolium hexafluorophosphate ([C8MIM][PF6]) was used as the extraction solvent, and bisphenol A, 4-n-nonylphenol and 4-tert-octylphenol were selected as the model analytes. Parameters affecting the extraction efficiency such as the volume of [C8MIM][PF6], dissolving temperature, extraction time, sample pH, centrifuging time and salting-out effect have been investigated in detail. Under the optimized conditions, good linear relationship was found in the concentration range of 1.0-100 μg L−1 for BPA, 1.5-150 μg L−1 for 4-NP, and 3-300 μg L−1 for 4-OP, respectively. Limits of detection (LOD, S/N = 3) were in the range of 0.23-0.48 μg L−1. Intra day and inter day precisions (RSDs, n = 6) were in the range of 4.6-5.5% and 8.5-13.3%, respectively. This method has been also successfully applied to analyze the real water samples at two different spiked concentrations and excellent results were obtained.  相似文献

A novel method, dispersive liquid-liquid microextraction (DLLME) coupled with high-performance liquid chromatography-variable wavelength detector (HPLC-VWD), has been developed for the determination of three phthalate esters (dimethyl phthalate (DMP), diethyl phthalate (DEP), and di-n-butyl phthalate (DnBP)) in water samples. A mixture of extraction solvent (41 μL carbon tetrachloride) and dispersive solvent (0.75 mL acetonitrile) were rapidly injected into 5.0 mL aqueous sample for the formation of cloudy solution, the analytes in the sample were extracted into the fine droplets of CCl4. After extraction, phase separation was performed by centrifugation and the enriched analytes in the sedimented phase were determined by HPLC-VWD. Some important parameters, such as the kind and volume of extraction solvent and dispersive solvent, extraction time and salt effect were investigated and optimized. Under the optimum extraction condition, the method yields a linear calibration curve in the concentration range from 5 to 5000 ng mL−1 for target analytes. The enrichment factors for DMP, DEP and DnBP were 45, 92 and 196, respectively, and the limits of detection were 1.8, 0.88 and 0.64 ng mL−1, respectively. The relative standard deviations (R.S.D.) for the extraction of 10 ng mL−1 of phthalate esters were in the range of 4.3-5.9% (n = 7). Lake water, tap water and bottled mineral water samples were successfully analyzed using the proposed method.  相似文献

Ultrasound-assisted ionic liquid dispersive liquid-phase micro-extraction was developed for the determination of four aromatic amines such as 2,4-dichloroaniline, 1-naphthylamine, 6-chloroanline and N,N-dimethylaniline. High-performance liquid chromatography coupled with UV detector was used for the determination of aromatic amines. In the novel procedure, 1-hexyl-3-methylimidazolium hexafluorophosphate [C6MIM] [PF6] was dispersed into the aqueous sample solution as fine droplets by ultrasonication, and which promoted the analytes more easily migrate into the ionic liquid phase. Variable affecting such as the volume of [C6MIM] [PF6], sample pH, ultrasonication time, extraction time, centrifuging time have been investigated in detail. The proposed method has been found to have excellent detection sensitivity with limits of detection (LOD, S/N = 3) in the range of 0.17–0.49 μg L−1 and precisions in the range of 2.0–6.1% (RSDs, n = 6). This method has been also successfully applied to analyze the real water samples and good spiked recoveries over the range of 92.2–119.3% were obtained.  相似文献

By using ionic liquid as membrane liquid and tri-n-octylphosphine oxide (TOPO) as additive, hollow fiber supported liquid phase microextraction (HF-LPME) was developed for the determination of five sulfonamides in environmental water samples by high-performance liquid chromatography with ultraviolet detection The extraction solvent and the parameters affecting the extraction enrichment factor such as the type and amount of carrier, pH and volume ratio of donor phase and acceptor phase, extraction time, salt-out effect and matrix effect were optimized. Under the optimal extraction conditions (organic liquid membrane phase: [C8MIM][PF6] with 14% TOPO (w/v); donor phase: 4 mL, pH 4.5 KH2PO4 with 2 M Na2SO4; acceptor phase: 25 μL, pH 13 NaOH; extraction time: 8 h), low detection limits (0.1–0.4 μg/L, RSD ≤ 5%) and good linear range (1–2000 ng/mL, R2 ≥ 0.999) were obtained for all the analytes. The presence of humic acid (0–25 mg/L dissolved organic carbon) and bovine serum albumin (0–100 μg/mL) had no significant effect on the extraction efficiency. Good spike recoveries over the range of 82.2–103.2% were obtained when applying the proposed method on five real environmental water samples. These results indicated that this present method was very sensitive and reliable with good repeatabilities and excellent clean-up in water samples. The proposed method confirmed hollow fiber supported ionic liquid membrane based LPME to be robust to monitoring trace levels of sulfadiazine, sulfamerazine, sulfamethazine, sulfadimethoxine and sulfamethoxazole in aqueous samples.  相似文献

A flow injection on-line sorption system was developed for the separation and preconcentration of traces of Ag, Cd, Co, Ni, Pb, U and Y from natural water samples with subsequent detection by ICP TOF MS. Simultaneous preconcentration of the analytes was achieved by complexation with the chelating reagent 1-phenyl-3-methyl-4-benzoylpyrazol-5-one immobilized on the inner walls of a (200 cm × 0.5 mm) PTFE knotted reactor. The analytes were eluted and transported to an axial ICP TOF MS system with 1% (v/v) HNO3 containing 0.3 μg l−1 of Rh as an internal standard using ultrasonic nebulization. The detection limits (3σ) varied from 0.3 ng l−1 for Y to 15.2 ng l−1 for Ni and the precision (R.S.D.) was better than 4%. Using a loading time of 90 s and a sample flow rate of 4.5 ml min−1, enhancement factors of 3-14 were obtained for the different analytes in comparison with their direct determination by ICP TOF MS with ultrasonic nebulization without preconcentration. The accuracy of the method was demonstrated by analysis of water based certified reference materials.  相似文献

A confirmatory method for the determination of illicit drugs based on micro-solid phase extraction with modified tips, made of a functionalized fiberglass with apolar chains of octadecylsilane into monolithic structure, has been developed in this study. Drugs belonging to different chemical classes, such as amphetamine, methamphetamine, methylenedioxyamphetamine, methylenedioxyethylamphetamine, methylenedioxymethylamphetamine, cocaine, benzoylecgonine, ketamine, mescaline, phencyclidine and psilocybine were analyzed. The quantitation was performed by liquid chromatography-tandem mass spectrometry and the analytes were detected in positive ionization by means of an electrospray source. The limits of quantification ranged between 0.3 ng mL−1 for cocaine and 4.9 ng mL−1 for psilocybine, with coefficients of determination (r2) >0.99 for all the analytes as recommended in the guidelines of Society of Forensic Toxicologists-American Association Forensic Sciences.  相似文献

In present study, a convenient, sensitive and environmentally friendly method for the determination of five benzoylurea insecticides (BUs) in water and juice samples was developed. To extract trace benzoylurea insecticides effectively, poly(methacrylic acid-co-ethylene dimethacrylate) monolith was prepared and used as the sorbent of multiple monolithic fiber solid-phase microextraction (MMF-SPME). The influences of preparation conditions of monolith and extraction parameters of MMF-SPME on BUs were studied thoroughly. Under the optimized conditions, the combination of MMF-SPME with high performance liquid chromatography-diode array detection (MMF-SPME-HPLC-DAD) showed expected analytical performance for target analytes. The limits of detection (S/N = 3) of the developed method were 0.026–0.075 μg L−1 in water and 0.053–0.29 μg L−1 in juice samples. Good linearity was obtained for analytes with the correlation coefficients (R2) above 0.99. Satisfactory repeatability and reproducibility was achieved, with relative standard deviations (RSD) of both less than 10%. Finally, the established MMF-SPME-HPLC-DAD method was successfully applied for the determination of BUs residues in juice and environmental water samples. Recoveries obtained for the determination of BUs in spiking samples ranged from 65.1% to 118%, with RSD below 10% in all cases.  相似文献

A new, efficient, and environmental friendly hollow fiber liquid phase microextraction (HF-LPME) method based on supramolecular solvents was developed for extraction of five benzodiazepine drugs. The supramolecular solvent was produced from coacervation of decanoic acid aqueous vesicles in the presence of tetrabutylammonium (Bu4N+). In this work, benzodiazepines were extracted from aqueous samples into a supramolecular solvent impregnated in the wall pores and also filled inside the porous polypropylene hollow fiber membrane. The driving forces for the extraction were hydrophobic, hydrogen bonding, and π-cation interactions between the analytes and the vesicular aggregates. High-performance liquid chromatography with photodiode array detection (HPLC-DAD) was applied for separation and determination of the drugs. Several parameters affecting the extraction efficiency including pH, hollow fiber length, ionic strength, stirring rate, and extraction time were investigated and optimized. Under the optimal conditions, the preconcentration factors were obtained in the range of 112–198. Linearity of the method was determined to be in the range of 1.0–200.0 μg L−1 for diazepam and 2.0–200.0 μg L−1 for other analytes with coefficient of determination (R2) ranging from 0.9954 to 0.9993. The limits of detection for the target benzodiazepines were in the range of 0.5–0.7 μg L−1. The method was successfully applied for extraction and determination of the drugs in water, fruit juice, plasma and urine samples and relative recoveries of the compounds studied were in the range of 90.0–98.8%.  相似文献

A novel method was developed for the determination of amiodarone and desethylamiodarone by high-performance liquid chromatography (HPLC) coupled with chemiluminescent (CL) detection. The procedure is based on the post-column photolysis of the analytes into photoproducts which are active in the tris(2,2′-bipyridyl)ruthenium(III) [Ru(bpy)33+] CL system. Ru(bpy)33+ was on-line generated by photo-oxidation of the Ru(II) complex in the presence of peroxydisulfate. The separation was carried out on a Mediterranea C18 column with isocratic elution using a mixture of methanol and 0.017 mol L−1 ammonium sulfate buffer of pH 6.8. Under the optimum conditions, analytical curves, based on standard solutions, were linear over the range 0.1-50 μg mL−1 for amiodarone and 0.5-25 μg mL−1 for desethylamiodarone. The detection limits of amiodarone and desethylamiodarone were 0.02 and 0.11 μg mL−1, respectively. Intra- and inter-day precision values of 0.9% relative standard deviation (R.S.D.) (n = 10) and 1.6% R.S.D. (n = 15), respectively, were obtained. The method was applied successfully to the determination of these compounds in serum and pharmaceutical formulations.  相似文献

Homogeneous ionic liquid microextraction (HILME) was developed for the extraction of schizandrin, schisantherin A and deoxyschizandrin from Schisandra chinensis and Schisandra sphenanthera. 1-Butyl-3-methylimidazolium tetrafluoroborate ([C4MIM][BF4]) aqueous solution was used as extraction solvent, and ammonium hexafluorophosphate ([NH4][PF6]) was used as ion-pairing agent. 1-Butyl-3-methylimidazolium hexafluorophosphate ([C4MIM][PF6]), which is barely soluble in water, was formed in situ, and was used as sample solution. High-performance liquid chromatography (HPLC) was employed for separation and determination of the analytes. The calibration curve showed good linear relationship (r > 0.9998). The recoveries were between 69.71% and 88.33% with RSDs lower than 4.86%. External standard method was adopted in the proposed method, and internal standard method was applied for the evaluation of the proposed method. The two methods were compared and the results indicated that the proposed method was acceptable and simple. The HILME is free of volatile organic solvents, and represents lower expenditures of sample, extraction time and solvent, compared with ultrasonic and Soxhlet extraction. There was no obvious difference in the extraction yields of active constitutions obtained by the three extraction methods.  相似文献

A simple and rapid method for the direct determination of Cd, Cr, Cu, Pb and Zn in soil was developed. The method was developed using three certified reference materials of soil: Eutric Cambisol, Orthic Luvisols and Rendzina, which differed in their matrix composition. Chemical modifiers were essential to achieve reproducible and interference-free signals for the analytes studied. The best results were obtained with a Pd/Mg(NO3)2 admixture for the determination of Cd, Pb and Zn and NH4F for Cu. The combination of W (as a permanent modifier) and Mg(NO3)2 provided well-defined signal profiles for Cr. The following spectral lines were used: Cd 228.8 nm, Cr 520.6 nm, Cu 218.2 nm, Pb 205.3 nm and Zn 307.6 nm. The limit of detection was 4.2 ng g− 1 for Cd, 1.1 μg g− 1 for Cr, 0.5 μg g− 1 for Cu, 1.3 μg g− 1 for Pb and 8.6 μg g− 1 for Zn for the maximum sample mass used. Under optimized conditions, the analyte and matrix were separated effectively in situ, and aqueous standards could be used for calibration.  相似文献

A new biochemical oxygen demand (BOD) sensing method employing a double-mediator (DM) system coupled with ferricyanide and a lipophilic mediator, menadione and the eukaryote Saccharomyces cerevisiae has been developed. In this study, a stirred micro-batch-type microbial sensor with a 560 μL volume and a two-electrode system was used. The chronamperometric response of this sensor had a linear response between 1 μM and 10 mM hexacyanoferrate(II) (r2 = 0.9995, 14 points, n = 3, average of relative standard deviation and R.S.D.av = 1.3%). Next, the optimum conditions for BOD estimation by the DM system (BODDM) were investigated and the findings revealed that the concentration of ethanol, used to dissolve menadione, influenced the sensor response and a relationship between the sensor output and glucose glutamic acid concentration was obtained over a range of 6.6-220 mg O2 L−1 (five points, n = 3, R.S.D.av 6.6%) when using a reaction mixture incubated for 15 min. Subsequently, the characterization of this sensor was studied. The sensor responses to 14 pure organic substances were compared with the conventional BOD5 method and other biosensor methods. Similar results with the BOD biosensor system using Trichosporon cutaneum were obtained. In addition, the influence of chloride ion, artificial seawater and heavy metal ions on the sensor response was investigated. A slight influence of 20.0 g L−1 chloride ion and artificial seawater (18.4 g L−1 Cl) was observed. Thus, the possibility of BOD determination for seawater was suggested in this study. In addition, no influence of the heavy metal ions (1.0 mg L−1 Fe3+, Cu2+, Mn2+, Cr3+ and Zn2+) was observed. Real sample measurements using both river water and seawater were performed and compared with those obtained from the BOD5 method. Finally, stable responses were obtained for 14 days when the yeast suspension was stored at 4 °C (response reduction, 93%; R.S.D. for 6 testing days, 9.1%).  相似文献

A simple and rapid pressurized liquid extraction (PLE) and high performance liquid chromatography-electrospray ionization tandem mass spectrometry (HPLC-ESI-MS/MS) method was developed for qualitative and quantitative determination of nucleosides, bases and their analogues in natural and cultured Cordyceps. The samples were extracted using PLE. The separation was achieved on a ZORBAX Eclipse XDB-C18 column with gradient elution of methanol and 5 mM aqueous ammonium acetate as mobile phase. Target compounds were identified by characterizing their product ions, precursor ions and retention times. Quantitative analysis of investigated compounds were performed using time programmed selective ion monitoring (SIM) or selective reaction monitoring (SRM) with 10 segments in positive (negative for uridine) ion mode. The results showed that 43 bases, nucleosides and their analogues were detected in Cordyceps, of these 16 compounds were identified. The simultaneous determination of seven nucleosides and six bases in Cordyceps was achieved using PLE and HPLC-ESI-MS/MS method described above, which afforded good linearity, selectivity, precision, recovery, short analysis time as well as LOD and LOQ in the ng/ml range.  相似文献

Different analytical methods for the determination of lead in Orujo spirits by electrothermal atomic absorption spectrometry (ETAAS) were developed using permanent modifiers (W, Ir, Ru, W-Ir and W-Ru) thermally deposited on platforms inserted in pyrolitic graphite tubes and Pd-Mg(NO3)2 conventional modifier mixture. In all cases, the Pb determination was performed without any sample pretreatment or preconcentration steps. The comparison between the chemical modifiers employed has been made in terms of pyrolysis and atomization temperatures, characteristic masses, detection limits, and atomization and background signal shapes. The limits of detection obtained were 0.375, 0.387, 0.109, 0.251 and 0.267 ng mL−1 for W, Ir, Ru, W-Ir and W-Ru, respectively and 0.710 ng mL−1 for Pd-Mg(NO3)2. The characteristic masses were 14.1, 11.2, 5.6, 8.3 and 9.3 pg for W, Ir, Ru, W-Ir and W-Ru, respectively and 22.2 pg for Pd-Mg(NO3)2. For all the developed procedures using the different modification systems, the relative standard deviations (<10%) and the analytical recoveries (95-103%) were acceptable. The more suitable methods for Pb determination in distillate spirits were those using permanent modifiers in contrast with classical Pd-Mg(NO3)2. The best analytical performance was achieved for W, Ir and W-Ir methods, which were applied to lead determination in Orujo spirit samples from Galicia (NW Spain). The Pb concentrations found in the analyzed samples were comprised in the range (<LOD to 1.5 μg mL−1).  相似文献
