A simple method to identify and determine six sulfonamides (sodium sulfacetamide, sulfamethizole, sulfaguanidine, sulfamerazine, sulfathiazole and sulfamethoxazole) in milk by micellar liquid chromatography (MLC) is reported. The assay makes use of a precolumn diazotisation-coupling derivatisation including the formation of an azo dye that can be detected at 490 nm. Furthermore, the use of MLC as an analytical tool allows the direct injection of non-purified samples. The separation was performed with an 80 mM SDS-8.5% propanol eluent at pH 7. Analysis times are below 16 min with a complete resolution. Linearities (r > 0.9999), as well as intra- and inter-day precision (below 2.7%), were studied in the validation of the method. The limits of detection and quantification ranged from approximately 0.72 to 0.94 and 2.4 to 3.1 ng mL−1, respectively. The detection limit was below the maximum residue limit established by the European Community. Finally, recoveries in spiked milk samples were in the 83-103% range.  相似文献

In this study, a metal‐organic framework (MOF), [Mn(cam)(bpy)], was synthesized and characterized by thermogravimetric analysis, scanning electron microscopy, and Fourier transform infrared spectrometry. An open‐tubular capillary column was fabricated from [Mn(cam)(bpy)] via the amide coupling method. Ten types of sulfonamides were separated through the fabricated capillary column, which showed a good limits of detection (<0.07 μg/mL) and linear ranges (1–100 or 5–100 μg/mL) with a high correlation coefficients (R2 > 0.9987). The intra‐day, inter‐day and column‐to‐column relative standard deviations (RSDs) in the migration times ranged from 0.44 to 4.87%, and the peak area RSDs ranged from 0.80 to 7.28%. The developed capillary electrochromatography method can be successfully utilized for the determination of sulfonamides in tap water and milk samples.  相似文献

By using ionic liquid as membrane liquid and tri-n-octylphosphine oxide (TOPO) as additive, hollow fiber supported liquid phase microextraction (HF-LPME) was developed for the determination of five sulfonamides in environmental water samples by high-performance liquid chromatography with ultraviolet detection The extraction solvent and the parameters affecting the extraction enrichment factor such as the type and amount of carrier, pH and volume ratio of donor phase and acceptor phase, extraction time, salt-out effect and matrix effect were optimized. Under the optimal extraction conditions (organic liquid membrane phase: [C8MIM][PF6] with 14% TOPO (w/v); donor phase: 4 mL, pH 4.5 KH2PO4 with 2 M Na2SO4; acceptor phase: 25 μL, pH 13 NaOH; extraction time: 8 h), low detection limits (0.1–0.4 μg/L, RSD ≤ 5%) and good linear range (1–2000 ng/mL, R2 ≥ 0.999) were obtained for all the analytes. The presence of humic acid (0–25 mg/L dissolved organic carbon) and bovine serum albumin (0–100 μg/mL) had no significant effect on the extraction efficiency. Good spike recoveries over the range of 82.2–103.2% were obtained when applying the proposed method on five real environmental water samples. These results indicated that this present method was very sensitive and reliable with good repeatabilities and excellent clean-up in water samples. The proposed method confirmed hollow fiber supported ionic liquid membrane based LPME to be robust to monitoring trace levels of sulfadiazine, sulfamerazine, sulfamethazine, sulfadimethoxine and sulfamethoxazole in aqueous samples.  相似文献

Ionic liquid-based homogeneous liquid-liquid microextraction (IL-based HLLME) high-performance liquid chromatography was developed and applied to the extraction, separation and determination of some antibiotics in milk. The proteins and lipids were removed by adding salt and adjusting the pH value. The homogeneous extraction was applied to the improvement of recoveries for IL phase and analytes. The experimental parameters of the IL-based HLLME, including salt concentration in sample solution, pH value of sample solution, volume of [C(6)MIM][BF(4)], amount of ion-pairing agent (NH(4)PF(6)), and extraction time, were evaluated. The limits of detection for enoxacin, pefloxacin, norfloxacin, enrofloxacin, sulfamethoxazole and sulfadimethoxine were 15.8, 7.07, 5.13, 4.00, 7.79 and 8.33 μg L(-1), respectively. When the proposed method was applied to the analysis of milk samples the recoveries of the analytes ranged from 92.5 to 118.6% and relative standard deviations were lower than 7.00%.  相似文献

This paper describes a rapid method for the determination of 14 kinds of sulfonamides (SAs) in wastewater using SPE, and LC-MS/MS with positive ESI (ESI(+)) and selected reaction monitoring (SRM) mode. The SPE was performed on an Oasis hydrophilic-lipophilic-balanced (HLB) cartridge. Chromatographic separation on a C18 column was achieved using a binary eluent containing methanol and water with 0.2% formic acid. Typical recoveries of the analytes ranged from 22.3 to 87.0% at a fortification level of 100 ng/L. The LODs in wastewater except sulfathiazole (3 ng/L) could be detected and quantified at levels as low as 1 ng/L. Finally, the method was applied to water from the municipal outlet and the aquaculture wastewater effluent. Sulfamethazine (SM(2)), sulfamethoxypyridazine (SMP), and sulfamethoxazole (SMZ) were most frequently found in wastewater in a concentration range between 1.2 and 31.7 ng/L.  相似文献

A simple, rapid and environment‐friendly technique of single‐drop liquid‐phase microextraction has been developed for the determination of sulfonamides in environmental water. Several important parameters including stirring rate, extraction solvent, extraction pH, salinity and extraction time were optimized to maximize the extract efficiency. Extraction solvent 1‐octyl‐3‐methylimidazolium hexafluorophosphate [C8MIM][PF6] ionic liquid showed better extraction efficiency than 1‐butyl‐3‐methylimidazolium hexafluorophosphate [C4MIM][PF6] and 1‐octanol. The optimum experimental conditions were: pH, 4.5; sodium chloride content, 36% w/v; extraction time, 20 min. This method provided low detection limits (0.5–1 ng/mL), good repeatability (the RSD ranging from 4.2 to 9.9%, n=5) and wide linear range (1–1500 ng/mL), with determination coefficients (r2) higher than 0.9989 for all the target compounds. Real sample analysis showed relative recoveries between 63.5 and 115.8% for all the target compounds.  相似文献

The sulfonamides are stable chemotherapeutics used against the bacterial disease affecting bees, known as American foulbrood (Bacillus larvae), so their residues could appear in the honey of treated bees. Their presence at a concentration above the limit value is a potential hazard to human health. Brazilian authorities have included in the National regulatory monitoring program, the control of the three most widely used sulfonamides in honey production, i.e., sulfathiazole, sulfamethazine and sulfadimethoxine. A method for the determination of residual sulfonamides in honey, using sulfapyridine as an internal standard has been developed, optimized and validated. Some changes were implemented on current available methodologies for the analysis of sulfonamides in honey in order to adopt such procedures to Brazilian honey samples. Sulfonamides were extracted from honey with dichloromethane after dissolution with 30% sodium chloride, and cleaned up with solid phase extraction on Florisil columns. The eluate was analyzed by high-performance liquid chromatography with ultraviolet detection. The limit of detection was determined at 3 μg kg−1, 4 μg kg−1 and 5 μg kg−1 for sulfathiazole, sulfamethazine and sulfadimethoxine, respectively with average recoveries of 61.0% for sulfathiazole; 94.5% for sulfamethazine and 86.0% for sulfadimethoxine at the 100 μg kg−1 level. As the final step of validation procedure, the analysts were submitted to a blind spiked sample prepared by the quality assurance officer which results were successfully obtained regarding recovery and deviations.  相似文献

A novel restricted access-molecularly imprinted material (RA-MIP) with selectivity for sulfonamides was synthesized using initiator-transfer agent-terminator (iniferter) method, a "living"/controlled radical polymerization technique. The material was prepared by grafting two layers with different functions on the silica support. To perform a "grafting from" polymerization, iniferter was immobilized on the surface of silica. The internal sulfamethazine imprinted polymer and the external poly(glycidyl methacrylate) [poly(GMA)] were then grafted successively. The hydrophilic structures were formed on the external layer of the material by the hydrolysis of the linear poly(GMA) for protein removal. The result has shown that this restricted access-MIP grafted silica (RA-MIP-SG) not only has the selectivity for the template and its analog, but also has the ability of exclusion for bovine serum albumin (BSA). It indicated that the material possesses both properties of molecularly imprinted polymer (MIP) and restricted access material (RAM). Using RA-MIP-SG as pre-column, a column-switching HPLC method was established for the determination of sulfonamides in bovine milk. Direct sample injection was performed in the analysis, which provides a convenient analytical procedure. Good linearity in the range of 2-400 ng mL(-1) (R>0.998) has been obtained for seven sulfonamides in the study. The recoveries of all the analytes at three concentration levels are between 93% and 107% with the RSD<8.0%. The limits of quantification and limits of detection are less than or equal to 2.7 ng mL(-1) and 0.8 ng mL(-1) respectively. It demonstrated this RA-MIP-SG can be applied in sample extraction and clean-up for the determination of sulfonamides in bovine milk by direct injection and column-switching HPLC with good efficiency and reliability.  相似文献

A hybrid stationary phase, XTerra MS C18, has been evaluated for the high-temperature reversed-phase liquid chromatography of selected hydrophobic steroids. The effects on the retention and efficiency at temperatures up to 130°C and eluent compositions from methanol–water mixtures to superheated water were studied. The thermodynamic data of the separations were determined. It was shown that increasing the temperature enabled the percentage of methanol to be reduced. High mobile-phase flow rates could be used, but for these non-polar analytes, the retention times with superheated water as the eluent were still high.  相似文献

Graphene‐Fe3O4 nanoparticles were prepared using one‐step solvothermal method and characterized by X‐ray diffraction, FTIR spectroscopy, scanning electron microscopy, and vibrating sample magnetometry. The results demonstrated that Fe3O4 nanoparticles were homogeneously anchored on graphene nanosheets. The as‐synthesized graphene‐Fe3O4 nanoparticles were employed as sorbent for magnetic solid‐phase extraction of sulfonamides in milk prior to capillary electrophoresis analysis. The optimal capillary electrophoresis conditions were as follows: 60 mmol/L Na2HPO4 containing 2 mmol/L ethylenediaminetetraacetic acid disodium salt and 24% v/v methanol as running buffer, separation voltage of 14 kV, and detection wavelength of 270 nm. The parameters affecting extraction efficiency including desorption solution, the amount of graphene‐Fe3O4 nanoparticles, extraction time, and sample pH were investigated in detail. Under the optimal conditions, good linearity (5–200 μg/L) with correlation coefficients ≥0.9910 was obtained. The limits of detection were 0.89–2.31 μg/L. The relative standard deviations for intraday and interday analyses were 4.9–8.5 and 4.0–9.0%, respectively. The proposed method was successfully applied to the analysis of sulfonamides in milk samples with recoveries ranging from 62.7 to 104.8% and relative standard deviations less than 10.2%.  相似文献

Matrix solid‐phase dispersion coupled with homogeneous ionic liquid microextraction was developed and applied to the extraction of some sulfonamides, including sulfamerazine, sulfamethazine, sulfathiazole, sulfachloropyridazine, sulfadoxine, sulfisoxazole, and sulfaphenazole, in animal tissues. High‐performance liquid chromatography was applied to the separation and determination of the target analytes. The solid sample was directly treated by matrix solid‐phase dispersion and the eluate obtained was treated by homogeneous ionic liquid microextraction. The ionic liquid was used as the extraction solvent in this method, which may result in the improvement of the recoveries of the target analytes. To avoid using organic solvent and reduce environmental pollution, water was used as the elution solvent of matrix solid‐phase dispersion. The effects of the experimental parameters on recoveries, including the type and volume of ionic liquid, type of dispersant, ratio of sample to dispersant, pH value of elution solvent, volume of elution solvent, amount of salt in eluate, amount of ion‐pairing agent (NH4PF6), and centrifuging time, were evaluated. When the present method was applied to the analysis of animal tissues, the recoveries of the analytes ranged from 85.4 to 118.0%, and the relative standard deviations were lower than 9.30%. The detection limits for the analytes were 4.3–13.4 μg/kg.  相似文献

建立了一种高效、低成本的固相萃取-高效液相色谱(SPE-HPLC)同时测定鸡粪中6种常见抗生素(2种四环素类、2种喹诺酮类和2种磺胺类)的分析方法。样品经EDTA-McIlvaine缓冲液和有机混合提取液(甲醇-乙腈-丙酮,2:2:1,v/v/v)提取,过HLB固相萃取柱净化,甲醇-二氯甲烷(7:3,v/v)洗脱,高效液相色谱-二极管阵列检测器测定,检测波长λ=270 nm,柱温32℃,流动相为乙腈-0.7%(v/v)磷酸水溶液。该方法在0.5~100 mg/L质量浓度范围内的标准曲线相关系数r2在0.9999~1之间,样品加标回收率在70.0%~116.3%之间,相对标准偏差为1.2%~16.6%。方法检出限为1.3~6.7 μg/kg,定量限为3.5~9.2 μg/kg。应用该方法对辽宁省抚顺市某养鸡场当天的鸡粪进行检测,诺氟沙星和恩诺沙星(喹诺酮类)含量为未检出~9.23 mg/kg和1.57~7.69 mg/kg,磺胺二甲嘧啶(磺胺类)含量为2.02~13.05 mg/kg,磺胺甲恶唑、土霉素和四环素未测出。  相似文献

采用Chirex(S)-LEU(S)-NEA、ChiralcelOD-H和ChiralpakAD-H手性色谱柱直接拆分了2′-羟基-1,1′-联萘-2-苯甲酸酯(HBNB)、1,1′-联萘-2,2′-二苯甲酸酯(BNDB)和2′-甲氧基-1,1′-联萘-2-苯甲酸酯(MBNB)对映体。分别考察了流动相组成、柱温和化合物结构对手性分离的影响。结果表明:3对联萘二酚苯甲酸酯对映体在ChiralpakAD-H柱上的拆分效果最好。当采用正己烷/异丙醇(40/60,v/v)为流动相时,HBNB、BNDB和MBNB对映体的分离因子(α)和分离度(Rs)分别为1.76、1.74、1.40和6.47、7.81、4.75。对比联萘二酚(BN)的分离,从联萘分子中2-位取代基、对映体出峰顺序和热力学参数等方面探讨了相关手性分离机理。  相似文献

A simple, convenient, and economic self-assembly miniaturized syringe assisted extraction (mini-SAE) using poly (hydroxyethyl methacrylate) polymer (PHEMA) as sorbent coupled with liquid chromatography was proposed for rapid screening of sulfadiazine (SD) and sulfamonomethoxine (SMM) in milk. The water-compatible PHEMA was synthesized by reversible addition-fragmentation chain transfer precipitation polymerization using trithiocarbonate as chain transfer agent and methanol–water system as reaction medium. The obtained PHEMA sorbent showed good affinity to sulfonamides and was successfully applied as a special sorbent for a mini-SAE device for simultaneous extraction and isolation of SD and SMM in milk samples. Under the optimum condition, good linearity was obtained in a range of 7.0–700 ng g−1 (r ≥ 0.9995) and the average recoveries of SD and SMM at three spiked levels were ranged from 85.6 to 100.3% with the relative standard deviations (RSD) ≤6.5%. The presented PHEMA-mini-SAE protocol could be potentially applied as an alternative tool for analyzing the residues of SAs in complicated biological samples.  相似文献

A simple and rapid HPLC method has been developed for simultaneous determination of the four resveratrol forms (aglycon and glycosidic) in a Grenache wine from Chateauneuf du Pape (Vaucluse). These analyses were achieved by using two commercial monolith HPLC columns and diode array detection. The method provided reliable separations at low pressure with a short analysis time. The limit of detection (LD) and limit of quantification (LQ) were calculated for each standard. The molecules were separated and quantified in a single run without any purification of the sample.  相似文献

A HPLC method with diode-array detection, at 265 nm, was developed and validated for the determination of ten sulfonamides (SAs): sulfadiazine (SDZ), sulfathiazine (STZ), sulfamethoxine (SMTH), sulfamethizole (SMZ), sulfamethoxypyridazine (SMPZ), sulfamonomethoxine (SMMX), sulfamethoxazole (SMXZ), sulfisoxazole (SIX), sulfadimethoxine (SDMX), and sulfaquinoxaline (SQX) in milk. A mixture of ethyl acetate, n-hexane, and isopropanol was used for the extraction of target analytes from milk. The mobile phase, a mixture of 0.1% v/v formic acid, CH(3) CN, and CH(3) OH was delivered to the analytical column under a gradient program. The procedure was validated according to the European Union regulation 2002/657/EC in terms of selectivity, stability, decision limit, detection capability, accuracy, and precision. Mean recoveries of sulfonamides from milk samples spiked at three concentration levels (0.5×MRL, 1×MRL, and 1.5×MRL) (MRL, maximum residue level) were 93.9-115.9% for SDZ, 97.8-102.9% for STZ, 94.6-107.0% for SMTH, 98.3-111.5% for SMZ, 95.3-108.4% for SMPZ, 97.9-106.0% for SMMX, 97.6-111.3% for SMXZ, 94.3-104.6% for SIX, 96.4-109.1% for SDMX, and 98.2-111.2% for SQX. All RSD values were lower than 8.8%. The decision limits CCa calculated by spiking 20 blank milk samples at MRL (100 μg/kg) ranged from 101.61 to 106.84 μg/kg, whereas the detection capability CCb ranged from 105.64 to 119.01 μg/kg.  相似文献

A new chromatographic stationary phase obtained by coating a reversed phase amide column with phosphatidylcholine based liposomes solution to yield a phospholipid modified column (PLM). The modification is achieved by the dynamic coating method which recycles the coating solution through the column in a closed loop for a period of 24 h. The chromatographic properties of the new column have changed significantly as compared to the original amide column due to the phospholipid coating. A good correlation was observed between n-octanol/water logP values and the logarithm of the retention factor obtained on the PLM column for a large number of solutes. In addition the PLM column was characterized using the linear solvation energy relationship (LSER). The values of the LSER system constants for the PLM column were calculated and were found to be very close to those of the n-octanol/water extraction system thus suggesting that the PLM column can be used for the estimation of n-octanol/water partition coefficient and serve as a possible alternative to the shake-flask method for lipophilicity determination. In addition, the results suggest that the PLM column can provide an alternative to other phospholipid-based column such as the IAM and the DPC columns.  相似文献

In this paper, a methodology to evaluate the probability of false non-compliance and false compliance for screening methods, which give first or second-order multivariate signals is proposed. For this task 120 samples of 6 different kinds of milk have been measured by excitation-emission fluorescence. The samples have been spiked with different amounts of three sulfonamides (sulfadiazine, sulfamerazine and sulfamethazine). These substances have been classified in group B1 (veterinary medicines and contaminants) of annex I of Directive 96/23/EC. The European Union (Commission Regulation EC no. 281/96) has set the maximum residue level (MRL) of total sulfonamides at 100 μg kg−1 in muscle, liver, kidney and milk.The work shows that excitation-emission fluorescence together with the partial least squares class modeling (PLS-CM) procedure may be a suitable and cheap screening method for the total amount of sulfonamides in milk. Three models, PLS-CM, have been built, for the emission and excitation spectra (first-order signals) and for the excitation-emission matrices (second-order signals). In all the cases it reaches probabilities of false compliance below 5% as required by Decision 2002/657/EC.With the same flourescence signals, the total quantity of sulfonamide was calibrated using 2-PLS, 3-PLS and PARAFAC regressions. Using this quantitative approach, the capability of detection, CCβ, around the MRL has been estimated between 114.3 and 115.1 μg kg−1 for a probability of false non-compliance and false compliance equal to 5%.  相似文献
