本文研究了三种羧酸取代的四苯基卟啉衍生物在空气/Cd^2^+水溶液界面上所形成的单层膜及LB膜。这三种卟啉衍生物中, 一种没有脂链, 另外两种具有不同长度的脂链。由π-A等温线得到的平均表观分子面积相差很大。紫外-可见光谱表明, LB膜中卟啉的Soret吸收带相对于溶液的吸收均红移, 但红移程度不同。LB膜的偏振紫外-可见光谱表明, LB膜中三种卟啉衍生物的卟啉环具有基本一致的取向。运用亚相降低法得到了三种卟啉衍生物单层LB膜, 其紫外-可见光谱与用垂直提拉法得到的LB膜的紫外-可见光谱具有一致的特征。这些结果表明: 卟啉衍生物有无取代链及取代链长的不同对平均表观分子面积的大小和膜中环间的距离有影响, 但对环的取向没有影响。环的取向由环本身及环上的亲水取代基来确定。气/液界面上三种卟啉衍生物的单层膜中环也具有一致的取向, 且与LB膜中环的取向相差不大。提拉不会对膜中环的取向及膜的结构造成大的改变。  相似文献

Multilayers consisting of negatively charged phospholipid DMPA and myelin basic protein (MBP) were assembled by Langmuir-Blodgett deposition of floating Langmuir monolayers from the air/water interface to solid substrates. Protein/lipid samples were obtained by binding MBP from the aqueous subphase to the phospholipid monolayers before deposition. The vertical organization of these model membranes (i.e., with organization perpendicular to the substrate surface) was investigated in detail by neutron reflectivity measurements, and the internal distribution of water molecules was determined from the change of contrast after in-situ H2O/D2O exchange. The multilayers were well ordered, with repeating lipid bilayers as fundamental structural unit. MBP was inserted in between adjacent lipid headgroups, such as in the natural myelin membrane. Water molecules in the multilayers were present mainly in the lipid headgroup and protein slab. On exposition of the pure lipid multilayers to a dry atmosphere, a reduction of the bilayer spacing was determined, whereas the global lamellar order was not affected. In contrast, drying of the protein/lipid multilayers induced degradation of the laminar order. The data demonstrate that ordered Langmuir-Blodgett multilayers are versatile model systems for studying how competing interactions between lipid, protein, water, and ions affect the global organization of such multilamellar lipid/protein assemblies. Here, the water molecules were found to be a necessary mediator to maintain the laminar order in a multilayer from DMPA and myelin basic protein.  相似文献

The two-dimensional self-assembly at the air/water (A/W) interface of two dendrimer-like copolymers based on polystyrene and poly(tert-butyl acrylate) (PS-b-PtBA) or poly(acrylic acid) (PS-b-PAA) was investigated through surface pressure measurements (isotherms, isochores, and compression-expansion hysteresis experiments) and atomic force microscopy (AFM) imaging. The two dendrimer-like block copolymers have an 8-arm PS core (Mn = 10 000 g/mol, approximately 12 styrene repeat units per arm) with a 16-arm PtBA (Mn = 230 000 g/mol, approximately 112 tert-butyl acrylate repeat units per arm) or PAA (Mn = 129 000 g/mol, approximately 112 acrylic acid repeat units per arm) corona. The PS-b-PtBA sample forms stable Langmuir monolayers and aggregates into circular surface micelles up to a plateau observed in the corresponding isotherm around 24 mN/m. Beyond this threshold, the monolayers collapse above the interface, resulting in the formation of large and irregular desorbed aggregates. The PS-b-PAA sample has ionizable carboxylic acid groups, and its A/W interfacial self-assembly was therefore investigated for various subphase pH values. Under basic conditions (pH = 11), the carboxylic acid groups are deprotonated, and the PS-b-PAA sample is therefore highly water-soluble and does not form stable monolayers, instead irreversibly dissolving in the aqueous subphase. Under acidic conditions (pH = 2.5), the PS-b-PAA sample is less water-soluble and becomes surface-active. The pseudoplateau observed in the isotherm around 5 mN/m corresponds to a pancake-to-brush transition with the PAA chains dissolving in the water subphase and stretching underneath the anchoring PS cores. AFM imaging revealed the presence of circular surface micelles for low surface pressures, whereas the biphasic nature of the pseudoplateau region was confirmed with the gradual aggregation of the micellar PS cores above the PAA chains. The aggregation numbers for both samples were estimated around 3-5 dendrimer-like copolymers per circular surface micelle. These rather low values confirmed the tremendous influence of molecular architecture on the two-dimensional self-assembly of block copolymers.  相似文献

The interactions of mixed monolayers of two lipids, zwitterionic 1,2-dipalmitoyl-phosphatidylcholine (DPPC) and positively charged 1,2-dipalmitoyl-3-trimethylammonium-propane (DPTAP), with phytohormone indolilo-3-acetic acid (IAA) and selenate anions in the aqueous subphase were studied. For this purpose, isotherms of the surface pressure versus the mean molecular area were recorded. Domain formation was investigated by using Brewster angle microscopy (BAM). The method of grazing incidence X-ray diffraction (GIXD) was also applied for the characterization of the organization of lipid molecules in condensed monolayers. It was found that selenate ions contribute to monolayer condensation by neutralizing the positive net charge of mixed monolayers whereas IAA molecules penetrated the lipid monolayer, causing its expansion/fluidization. When both solutes were introduced into the subphase, a competition between them for interaction with the positively charged lipids in the monolayer was observed.  相似文献

Membrane lipids are active contributors to cell function as key mediators in signaling pathways controlling cell functions including inflammation, apoptosis, migration, and proliferation. Recent work on multimolecular lipid structures suggests a critical role for lipid organization in regulating the function of both lipids and proteins. Of particular interest in this context are the polyphosphoinositides (PPI's), especially phosphatidylinositol (4,5) bisphosphate (PIP 2). The cellular functions of PIP 2 are numerous but the organization of PIP 2 in the inner leaflet of the plasma membrane, as well as the factors controlling targeting of PIP 2 to specific proteins, remains poorly understood. To analyze the organization of PIP 2 in a simplified planar system, we used Langmuir monolayers to study the effects of subphase conditions on monolayers of purified naturally derived PIP 2 and other anionic or zwitterionic phospholipids. We report a significant molecular area expanding effect of subphase monovalent salts on PIP 2 at biologically relevant surface densities. This effect is shown to be specific to PIP 2 and independent of subphase pH. Chaotropic agents (e.g., salts, trehalose, urea, temperature) that disrupt water structure and the ability of water to mediate intermolecular hydrogen bonding also specifically expanded PIP 2 monolayers. These results suggest a combination of water-mediated hydrogen bonding and headgroup repulsion in determining the organization of PIP 2, and may contribute to an explanation for the unique functionality of PIP 2 compared to other anionic phospholipids.  相似文献

We report X-ray reflectivity (XRR) and grazing incidence X-ray diffraction (GIXD) measurements of archaeal bipolar tetraether lipid monolayers at the air-water interface. Specifically, Langmuir films made of the polar lipid fraction E (PLFE) isolated from the thermoacidophilic archaeon Sulfolobus acidocaldarius grown at three different temperatures, i.e., 68, 76, and 81 °C, were examined. The dependence of the structure and packing properties of PLFE monolayers on surface pressure were analyzed in a temperature range between 10 and 50 °C at different pH values. Additionally, the interaction of PLFE monolayers (using lipids derived from cells grown at 76 °C) with the ion channel peptide gramicidin was investigated as a function of surface pressure. A total monolayer thickness of approximately 30 ? was found for all monolayers, hinting at a U-shaped conformation of the molecules with both head groups in contact with the interface. The monolayer thickness increased with rising film pressure and decreased with increasing temperature. At 10 and 20 °C, large, highly crystalline domains were observed by GIXD, whereas at higher temperatures no distinct crystallinity could be observed. For lipids derived from cells grown at higher temperatures, a slightly more rigid structure in the lipid dibiphytanyl chains was observed. A change in the pH of the subphase had an influence only on the structure of the lipid head groups. The addition of gramicidin to an PLFE monolayer led to a more disordered state as observed by XRR. In GIXD measurements, no major changes in lateral organization could be observed, except for a decrease of the size of crystalline domains, indicating that gramicidin resides mainly in the disordered areas of the monolayer and causes local membrane perturbation, only.  相似文献

The peptide corresponding to the sequence (279-298) of the Hepatitis G virus (HGV/GBV-C) E2 protein was synthesized, and surface activity measurements, pi-A compression isotherms, and penetration of E2(279-298) into phospholipid monolayers spread at the air-water interface were carried out on water and phosphate buffer subphases. The results obtained indicated that the pure E2(279-298) Langmuir monolayer exhibited a looser packing on saline-buffered than on pure water subphase and suggest that the increase in subphase ionic strength stabilizes the peptide monolayer. To better understand the topography of the monolayer, Brewster angle microscopy (BAM) images of pure peptide monolayers were obtained. Penetration of the peptide into the pure lipid monolayers of dipalmitoylphosphatidylcholine (DPPC) and dimyristoylphosphatidylcholine (DMPC) and into mixtures of dimyristoylphosphatidylcholine/dimyristoylphosphatidylglycerol (DMPC/DMPG) at various initial surface pressures was investigated to determine the ability of these lipid monolayers to host the peptide. The higher penetration of peptide into phospholipids is attained when the monolayers are in the liquid expanded state, and the greater interaction is observed with DMPC. Furthermore, the penetration of the peptide dissolved in the subphase into these various lipid monolayers was investigated to understand the interactions between the peptide and the lipid at the air-water interface. The results obtained showed that the lipid acyl chain length is an important parameter to be taken into consideration in the study of peptide-lipid interactions.  相似文献

Factor X is a blood clotting protein that associates at membrane surfaces to become activated during the coagulation cascade. A molecular level understanding of the protein-membrane phospholipid interactions has not been reached, although it is thought that the protein binds to phospholipids in the presence of calcium through a bridge with the Gla (gamma-carboxyglutamic acid) domain on the protein. In this work, phospholipid Langmuir monolayers have been utilized as model membranes to study factor X association with phospholipid membrane components. Surface pressure measurements indicate that subphase addition of sodium, magnesium, and calcium ions enhances protein penetration of the lipid monolayer, with the largest association found with calcium ions in the subphase. Fluorescence microscopy images collected after protein penetration of lipid monolayers indicate monolayer condensation in the presence of sodium and magnesium ions. Aggregation of lipid domains is induced when calcium is in the subphase, indicating binding-induced flocculation of surface lipid aggregates. Calcium binding to factor X likely causes a conformational change which allows protein-membrane interaction via hydrophobic association with lipid molecules.  相似文献

The association of neuropeptide Y (NPY) at the air/water interface and with phospholipid monolayers on water as subphase has been investigated using external infrared reflection absorption spectroscopy (IRRAS). Studies of the conformation and orientation of NPY suggest that it adopts an alpha-helical structure and is oriented parallel to the air/water interface in neat peptide monolayers. Both secondary structure and orientation are preserved in mixed lipid/NPY monolayers. Comparison of NPY associated with zwitterionic DPPC and with anionic DMPS suggests that electrostatic attraction plays a major role for peptide binding to the membrane surface.  相似文献

The first combination of scanning electrochemical microscopy (SECM) with a Langmuir trough for liquid/liquid interfaces is described. The technique has been examined and demonstrated through investigations of the effect of monolayers of l-α-phosphatidylcholine, distearoyl (DSPC) on the kinetics of oxygen transfer from decane to water. The stability of monolayers, formed in this way, on the timescale of SECM measurements has been identified as a function of compression speed and subphase composition. Monolayers were stable over a wide range of pressures and molecular areas, but at high compression a decrease in surface pressure with time was observed. This effect was attributed to desorption of the lipid from the interface. In this situation, it was possible to perform SECM measurements (tip-interface approach curves) rapidly under surface pressure control, without causing significant disturbance to the monolayer. DSPC had no detectable effect on the oxygen transfer kinetics when the monolayer was in the liquid-expanded phase, but in the liquid-condensed phase a significant decrease in the rate of oxygen transfer was observed.  相似文献

This work investigates the process of incorporation of a glycosylphosphatidyl inositol (GPI)-anchored alkaline phosphatase into Langmuir monolayers of dimyristoyl phosphatidic acid (DMPA). Three different methods of protein incorporation were assayed. When the protein solution was injected below the air–water interface after formation of the lipid monolayer a micro-heterogeneous distribution of alkaline phosphatase throughout the interface was observed. Adsorption kinetics studied by fluorescence microscopy, associated with surface pressure measurements, led to the proposition of a model in which the protein penetration is modulated by the surface packing of the monolayer and intermolecular interactions occurring between the phospholipid and the protein. At initial surface pressures higher than 20 mN m−1, the protein is quickly adsorbed on the interface and the lateral diffusion drives the alkyl chains to turn towards the air phase while the polypeptide moiety faces the aqueous subphase.  相似文献

We present the results of a fluorescence microscopy study of the interaction of annexin A1 with dipalmitoylphosphatidylcholine (DPPC) monolayers as a function of the lipid monolayer phase and the pH of the aqueous subphase. We show that annexin A1-DPPC interaction depends strongly on the domain structure of the DPPC monolayer and only weakly on the subphase pH. Annexin A1 is found to be line active, with preferential adsorption at phase boundaries. Also, annexin A1 is found to form networks in the presence of a domain structure in the monolayer. Our results point toward an important contribution of the unique N-terminal domain to the organization of the protein at the interface.  相似文献

The synthesis, physicochemical characterization, and interaction with membrane model systems of a peptide derived from the PA22-2 region of laminin are described. Surface activity studies indicate that this peptide is able to spread at the air-water interface being the maximal spreading pressure 20 mN/m at subphase concentrations around 10 micro M. Besides, these peptide molecules are also able to form stable monolayers. Physicochemical studies concerning the interaction of this peptide with lipids, organized in mono and bilayers, were carried out using Langmuir balance experiments and polarization fluorescence techniques. The peptide penetrates better in monolayers of DPPC than in those of PC and forms condensed mixed monolayers with DPPC. Energies of mixing are small thus indicating that deviations from ideality were almost negligible. Interactions with bilayers were studied through microviscosity changes (DPH and TMA-DPH probes), membrane permeability alterations (CF, NBD-PE/dithionite), and fusion promotion (NBD-PE/Rh-PE, resonance energy transfer). Results indicate that this sequence interacts very softly with bilayers without promoting changes in their organization. These data as well as the lack of interaction with erythrocytes suggest that coating liposomes with this peptide through chemical amide bonds can render stable inmunoliposomes for further biological applications.  相似文献

Great interest has been devoted to the gemini amphiphiles because of their unique properties. In this article, we report some interesting properties of the interfacial films formed by a series of newly designed gemini amphiphiles containing the Schiff base moiety. This novel series of gemini amphiphiles with their Schiff base headgroups linked by a hydrophobic alkyl spacer (BisSBC18Cn, n = 2, 4, 6, 8, 10) could be spread to form stable monolayers and coordinated with Cu(Ac)(2) in situ in the monolayer. The alkyl spacer in the amphiphiles has a great effect on the regulation of the properties of the Langmuir monolayers. A maximum limiting molecular area was observed for the monolayers of the gemini amphiphile with the spacer length of hexa- or octamethylene groups. Both the monolayers on water and on the aqueous Cu(Ac)(2) subphase were transferred onto solid substrates, and different morphologies were observed for films with different spacers. Nanonail and tapelike morphologies were observed for amphiphile films with shorter spacers (n = 2 and 4) on the water surface. Wormlike morphologies were observed for gemini films with longer spacers of C(8) and C(10) when coordinated with Cu(Ac)(2). An interdigitated layer structure was supposed to form in the multilayer films transferred from water or the aqueous Cu(Ac)(2) subphase.  相似文献

The spreading behavior and supramolecular assemblies of some arylbenzimidazoles with 2-substituted aromatic groups such as phenyl, naphthyl, anthryl and pyrenyl on water surface and the subphase containing AgNO3 were investigated. It was observed that although these compounds lack long alkyl chains, they showed surface activity when spread from chloroform solution on water surface and formed the supramolecular assemblies. When AgNO3 was present in the subphase, a coordination between the imidazole group of the compounds and Ag(I) occurred in situ in the spreading film, which was verified by the surface pressure/area (pi-A) isotherms and UV/Vis absorption spectra. Both the spreading films from water and the aqueous AgNO3 subphase were transferred onto solid substrates and their surface morphologies as well as properties were characterized by AFM, UV/Vis absorption and CD spectra. Various surface morphologies such as nanoparticles, block domains and nanoutensils were observed depending on the substituted aromatic groups. Interestingly, although all of these compounds were achiral, supramolecular chirality was obtained for some of the arylbenzimidazole films assembled from either the water surface or the subphase containing AgNO3. It was revealed that chiral assemblies could be obtained from water surface for the benzimidazoles which have pyrenyl or alpha-naphthyl groups. For benzimidazole derivative with anthryl group, chiral assemblies could be obtained when spreading on the aqueous AgNO3 subphase. For the benzimidazoles with phenyl or beta-naphthyl groups, no chirality was obtained. It was suggested that both the overcrowded stacking of the aromatic groups and the cooperative arrangement of the molecules on water surface or aqueous AgNO3 subphase play a crucial role in forming the chiral supramolecular assemblies.  相似文献

The surface activity of the poly–[block (ethylene oxide)]–poly [block (propylene oxide)]–poly [block (ethylene oxide)] copolymers (EO)x–(PO)y–(EO)x adsorbed together with dihexadecyl phosphoric acid (DHP), a synthetic phospholipid, is analyzed from their surface pressure and surface potential isotherms. The block copolymers of (EO)x–(PO)y–(EO)x with variable molecular weight (1100–14 000) were dissolved in the subphase for DHP monolayers. The concentration of the copolymers within the aqueous subphase were selected to render an initial surface tension of 60 mN/m. The simultaneous adsorption of the copolymer and DHP is attested by the observation of a liquid expanded state at large areas, absent for pure DHP monolayers. Above some critical surface pressure all copolymers cited above are expelled from the interface. The surface potential isotherms, which give information on the component of the molecular dipole moment normal to the plane of the monolayer, are interpreted in terms of changes in the copolymer conformation as well as in terms of the copolymer desorption from the air–liquid interface. For an equal hydrophobic/hydrophilic ratio, the size of the chains or molecular weight is decisive in the mechanism of the copolymer expulsion from the air–liquid interface.  相似文献

Griseofulvin (GF) is an oral antibiotic for widely occurring superficial mycosis in man and animals caused by dermaphyte fungi; it is also used in agriculture as a fungicide. The mechanism of the biological activity of GF is poorly understood. Here, the interactions of griseofulvin with lipid membranes were studied using 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC), 1,2-dilauroyl-sn-glycero-3-phosphocholine (DLPC), and 1,2-myristoyl-sn-glycero-3-phosphoethanolamine (DMPE) monolayers spread at the air/water interface. Surface pressure (Pi), electric surface potential (Delta V), grazing incidence X-ray diffraction (GIXD), and Brewster angle microscopy (BAM) were used for studying pure phospholipid monolayers spread on GF aqueous solutions, as well as mixed phospholipid/GF monolayers spread on pure water subphase. Moreover, phospholipase A2 (PLA2) activity toward DLPC monolayers and molecular modeling of the GF surface and lipophilic properties were used to get more insight into the mechanisms of GF-membrane interactions. The results obtained show that GF has a meaningful impact on the film properties; we propose that nonpolar interactions are by and large responsible for GF retention in the monolayers. The modification of membrane properties can be detected using both physicochemical and enzymatic methods. The results obtained may be relevant for elaborating GF preparations with increased bioavailability.  相似文献

Sum-frequency spectroscopy (SFS) in the CH and OH stretching regions was employed to obtain structural information about Langmuir monolayers on the H(2)O subphase of the model lipid dioctadecyldimethylammonium bromide (DOMA) and of the neutral surfactant methyl stearate (SME) and their mixtures and about the interfacial water structure underneath the films. These results were compared with the sum-frequency spectra of the interface between Langmuir monolayers of stearic acid and stearic acid-DOMA monolayers and water to prove that the uncompensated headgroup charge of DOMA at the interface is the reason for structuring of interfacial water close to the studied monomolecular films. Sum-frequency spectra on D(2)O subphase were also studied to account for the interference between the CH and OH spectral signatures because of the coherent nature of the SFS signals. Interfacial water structure proved to be a determining factor in the behavior of the mixed lipid monolayers. A mixing induced amplification in the surface potential DeltaV observed in our previous work was explained with total increase of the dipole moment for the mixed films, bigger than the arithmetic average for DOMA and SME monolayers alone. The increase is due to the better packing of the molecules in the mixed films and to the decrease in the interfacial water dipole moment arising from a more disordered water structure underneath the mixed monolayers.  相似文献

Porphyrin nanofiber patterning was generated by air/water interfacial assembly. The air/water interfacial aggregation behavior of two prophyrins, both of which contain two hydrophobic alkyl chains and two carboxylic acid substituent groups at different positions, was investigated using UV spectra, FT-IR spectra, and AFM measurements on the corresponding transferred films. The porphyrin nanofiber patterning can only be produced on ionic liquid (IL) doped water subphases by the assembly of the building blocks with two carboxylic acids located at the para-position (TPPA2b-A). The results suggest that the bulky cations of ionic liquids (ILs) can interact with the carboxylate of porphyrin electrostatically. The appropriate molecular geometries, ionic liquid (IL) doped water subphases, and relatively high surface pressures help the TPPA2b-A to form nanofiber patterns.  相似文献

An important contribution to the surface potential of lipid bilayers and monolayers comes from the intrinsic dipole moment of the lipid molecules. A theoretical model of the monolayer which involves a smeared dipole sheet approximation is introduced. This model is used to explore the nature and origins of the surface potential. In addition, the potential associated with phosphatidyl choline/cholesterol monolayers compressed on a Langmuir-Blodgett trough was measured with a non-contacting electrostatic voltmeter. A trough infusion configuration was fabricated to perform dynamic subphase experiments with compressed films in place. The potential/time response of monolayers to selective bimolecular systems such as antibody-antigen and concanavalin A-saccharide pairs was examined. These reactions induce spontaneous transients in dipole potential of magnitude 20–80 mV and duration of less than 1 s. The potential transients are attributed to local perturbation of lipid orientation and introduction of protein dipole fields caused by the formation of aggregates at the monolayer/water interface.  相似文献
