Acetaminophen is a well-known drug commonly used to provide pain relief, but it can also lead to acute liver failure at high concentrations. Therefore, there is considerable interest in monitoring its concentrations. Sensitive and selective acetaminophen electrochemical sensors were designed by cycling a glassy carbon electrode (GCE) to high potentials in the presence of β-CD in a phosphate electrolyte, or by simply activating the GCE electrode in the phosphate solution. Using cyclic voltammetry, adsorption-like voltammograms were recorded. The acetaminophen oxidation product, N-acetyl benzoquinone imine, was protected from hydrolysis, and this was attributed to the adsorption of acetaminophen at the modified GCE. The rate constants for the oxidation of acetaminophen were estimated as 4.3 × 10–3 cm2 s–1 and 3.4 × 10–3 cm2 s–1 for the β-CD-modified and -activated electrodes, respectively. Using differential pulse voltammetry, the limit of detection was calculated as 9.7 × 10–8 M with a linear concentration range extending from 0.1 to 80 μM. Furthermore, good selectivity was achieved in the presence of caffeine, ascorbic acid and aspirin, enabling the determination of acetaminophen in a commercial tablet. Similar electrochemical data were obtained for both the β-CD-modified and activated GCE surfaces, suggesting that the enhanced detection of acetaminophen is connected mainly to the activation and oxidation of the GCE. Using SEM, EDX and FTIR, no evidence was obtained to indicate that the β-CD was electropolymerised at the GCE.

A carbon paste electrode based on γ-cyclodextrin–carbon nanotube composite (γ-CD–CNT–CME) was developed for the determination of propranolol hydrochloride (PRO). The electrochemical behaviour of PRO was investigated employing cyclic voltammetry, electrochemical impedance spectroscopy and differential pulse adsorptive stripping voltammetry (DPAdSV). Surface morphology of the electrode has been studied by means of scanning electron microscopy. The results revealed that the oxidation of PRO is facilitated at γ-CD–CNT–CME. Under the optimized conditions in Britton–Robinson buffer pH 1.5, the peak currents were found to vary linearly with their concentrations in the range of 1.42 × 10?7 to 4.76 × 10?5 M. A detection limit (S/N = 3) of 4.01 × 10?8 M was obtained for PRO by means of DPAdSV. The proposed method was employed for the determination of PRO in pharmaceutical formulations, urine and blood serum samples.  相似文献

We describe a method for detecting DNA methylation. It is based on direct oxidation of DNA bases at a glassy carbon electrode (GCE) modified with film of a multiwalled carbon nanotube-β-cyclodextrin composite. This nano-structured film causes a strong enhancement on the oxidation current of DNA bases due to its large effective surface area and extraordinary electronic properties. Well-defined peaks were obtained as a result of electro-oxidation of guanine (at 0.67 V), adenine (at 0.92 V), thymine (at 1.11 V), cytosine (at 1.26 V), and 5-methylcytosine (at 1.13 V; all data vs. saturated calomel electrode (SCE)). The potential difference between 5-methylcytosine and cytosine (130 mV) is large enough to enable reliable simultaneous determination and analysis. The interference by thymine can be eliminated by following the principle of complementary pairing between purine and pyrimidine bases in DNA. The modified electrode was successfully applied to the evaluation of 5-methylcytosine in a fish sperm DNA, the methylation level of cytosine was found to be 7.47 %, and the analysis process took less than 1 h.  相似文献

We report on a method for electrochemical enantioselective recognition of tryptophan (Trp) enantiomers. It is based on competitive host-guest interaction between a deoxy-(2-aminoethylamino)-β-cyclodextrin (CD) bound to graphene nanosheets and the Cu(II) complexes of the Trp enantiomers via a ligand exchange mechanism. Chiral recognition was investigated via cyclic voltammetry and electrochemical impedance spectroscopy. The results reveal that the CD bound to graphene displays a stronger interaction with the Cu(II) complex of L-Trp than to that of D-Trp. The method was applied to the determination of the ratio of Trp enantiomers in mixtures.

The electrochemical oxidation of guanosine-5??-monophosphate (GMP) was studied with a glassy carbon electrode modified with a composite made from graphene and multi-walled carbon nanotubes. GMP undergoes an irreversible oxidation process at an oxidation peak potential of 987?mV in phosphate buffer solution. Compared to other electrodes, the oxidation peak current of GMP with this electrode was significantly increased, and the corresponding oxidation peak potential negatively shifted, thereby indicating that the modified material exhibited electrochemical catalytic activity towards GMP. Chronocoulometry demonstrates that the material also effectively increases the surface area of the electrode and increases the amount of GMP adsorbed. Under the optimum conditions, the oxidation current is proportional to the GMP concentration in the range from 0.1 to 59.7???M with a correlation coefficient of 0.9991. The detection limit is 0.025???M (at S/N?=?3).

Calf thymus DNA was electrochemically oxidized at a multi-walled carbon nanotube modified electrode. The potentials for DNA oxidation at pH 7.0 were 0.71 and 0.81 V versus SCE, corresponding to the oxidation of guanine and adenine residues, respectively. A simple, sensitive, and reliable method based on a combination of multi-walled carbon nanotubes with incorporated β-cyclodextrin (β-CD-MWNTs) and a polyaniline (PANI) film-modified glassy-carbon (GC) electrode has been successfully developed for determination of dopamine (DA) in the presence of ascorbic acid (AA). The PANI film had good anti-interference properties and long-term stability, because of the permselective and protective properties of the conducting redox polymer film. The acid-treated MWNTs with carboxylic acid functional groups promoted the electron-transfer reaction of DA and inhibited the voltammetric response of AA. Sensitive detection of DA was further improved by the preconcentration effect of formation of a supramolecular complex between β-CD and DA. The analytical response of the β-CD-MWNTs/PANI film to the electrochemical behavior of DA was, therefore, better than that of a MWNTs/PANI film, a PANI film, or a bare glassy-carbon (GC) electrode. Under the conditions chosen a linear calibration plot was obtained in the range 1.0 × 10−7–1.0 × 10−3 mol L−1 and the detection limit was 1.2 × 10−8 mol L−1. Interference from AA was effectively eliminated and the sensitivity, selectivity, stability, and reproducibility of the electrodes was excellent for determination of DA.  相似文献

A novel β-cyclodextrin (β-CD) derivative containing mono-phenylamino (MPA-β-CD) was newly synthesized by classical Mitsunobu reaction in good yield, and its structure has been confirmed by 1H NMR, 13C NMR and electrospray ionization mass spectra. The compound MPA-β-CD was immobilized onto glassy carbon electrode (GCE) by diazotization, and with this modified electrode the binding behavior of MPA-β-CD for ferrocene (Fc) was investigated qualitatively, and the comparison of differential pulse voltammetry before and after immersion in ferrocene solution indicated that the MPA-β-CD immobilized GCE exhibited the molecular recognition behavior between β-CD and ferrocene.  相似文献

In this work, we investigate the electrochemical activity of dopamine (DA) and uric acid (UA) using both a bare and a modified carbon paste electrode as the working electrode, with a platinum wire as the counter electrode and a silver/silver chloride (Ag/AgCl) as the reference electrode. The modified carbon paste electrode consists of multi-walled carbon nanotubes (>95%) treated with α-cyclodextrine, resulting in an electrode that exhibits a significant catalytic effect toward the electro-chemical oxidation of DA in a 0.2-M Britton–Robinson buffer solution (pH 5.0). The peak current increases linearly with the DA concentration within the molar concentration ranges of 2.0 × 10−6 to 5.0 × 10−5 M and 5.0 × 10−5 to 1.9 × 10−4 M. The detection limit (signal to noise >3) for DA was found to be 1.34 × 10−7 M, respectively. In this work, voltammetric methods such as cyclic voltammetry, chronoamperometry, chronocuolometry, differential pulse and square wave voltammetry, and linear sweep and hydrodynamic voltammetry were used. Cyclic voltammetry was used to investigate the redox properties of the modified electrode at various scan rates. The diffusion coefficient (D, cm2 s−1 = 3.05 × 10−5) and the kinetic parameters such as the electron transfer coefficient (α = 0.51) and the rate constant (k, cm3 mol−1 s−1 = 1.8 × 103) for DA were determined using electrochemical approaches. By using differential pulse voltammetry for simultaneous measurements, we obtained two peaks for DA and UA in the same solution, with the peak separation approximately 136 mV. The average recovery was measured at 102.45% for DA injection.  相似文献

A glassy carbon electrode (GCE) was modified with gold nanoparticles and a film of a composite made from poly(arginine) and multi-walled carbon nanotubes. Antibody against ractopamine was immobilized on the surface of the modified GCE which then was blocked with bovine serum albumin. The assembly of the immunosensor was followed by electrochemical impedance spectroscopy. Results demonstrated that the semicircle diameter increases, indicating that the film formed on the surface hinders electron transfer due to formation of the antibody-antigen complex on the modified electrode. Under optimal conditions, the peak current obtained by differential pulse voltammetry decreases linearly with increasing ractopamine concentrations in the 0.1 nmol?L?1 to 1 μmol?L?1 concentration range. The lower detection limit is 0.1 nmol?L?1. The sensor displays good stability and reproducibility. The method was applied to the analysis of spiked swine feed samples and gave satisfactory results. Gold nanoparticles-poly(luminol) (Plu-AuNPs) hybrid film and multi-walled carbon nanotubes with incorporated β-cyclodextrin modified glassy carbon electrode (β-CD-MWCNTs/Plu-AuNPs/GCE) was successfully prepared for simultaneous determination of dopamine (DA) and uric acid (UA). The surface of the modified electrode has been characterized by X-ray photo-electron spectroscopy (XPS), energy dispersive X-ray spectroscopy (EDS), field-emission scanning electron microscope (SEM) and transmission electron microscope (TEM). Cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS) and differential pulse voltammetry (DPV) have been used to investigate the β-CD-MWCNTs/Plu-AuNPs composite film. Gold nanoparticles anchored into poly(luminol) film exhibited catalytic activity for DA. MWCNTs with incorporated β-CD can greatly promote the direct electron transfer. In 0.10 M phosphate buffer solution (PBS, pH 7.0), the DPV response of the β-CD-MWCNTs/Plu-AuNPs/GCE sensor to DA is about 8-fold as compared with the Plu-AuNPs/GCE sensor, and the detection limit for DA is about one order of magnitude lower than the Plu-AuNPs/GCE sensor. The steady-state current response increases linearly with DA concentration from 1.0 × 10−6 to 5.6 × 10−5 M with a low detection limit (S/N = 3) of 1.9 × 10−7 M. Moreover, the interferences of ascorbic acid (AA) and uric acid (UA) are effectively diminished. The applicability of the prepared electrode has been demonstrated by measuring DA contents in dopamine hydrochloride injection.  相似文献

A novel determination method of electroinactive molecules by means of electrochemical technique is presented. A new self-assembled monolayer containing cyclodextrin (CD) is prepared with mono(6-o-p-tolylsulfonyl)-β-cyclodextrin. Although this derivatization process leads to a β-CD coverage of 10% of a full monolayer, this layer shows an effective host-guest response to ferrocene. The interfacial ferrocene complexation gives a response similar to that expected for a Langmuir adsorption isotherm yielding a stability constant of 4.2 ×104 mol-1· L and a maximum ferrocene coverage of 8.6×10-12 mol/cm2. The redox peak currents of the surface-confined ferrocene decrease upon addition of competing β-CD guest species to the solution, such as m-toluic acid (mTA) and sodium dodecyl sulfonate (SDS). This principle has been used for the determination of the electroinactive molecules, mTA and SDS in the concentration ranges of 0.8-2.7 μmol/L and 5-100 nmol/L, respectively.  相似文献

An electrochemical sensor for fluoroquinolones (FQs) based on polymerization of β-cyclodextrin (β-CD) and l-arginine (l-arg) modified carbon paste electrode (CPE) (P-β-CD-l-arg/CPE) was built for the first time. Synergistic effect of l-arg and β-CD was used to construct this sensor for quantification of these important antibiotics. Scanning electron microscope (SEM) image shows that polymer of β-CD and l-arg has been successfully modified on electrode. Electrochemical impedance spectroscopy (EIS) and cyclic voltammograms (CV) further indicate that polymer of β-CD and l-arg efficiently decreased the charge transfer resistance value of electrode and improved the electron transfer kinetic between analyte and electrode. Under the optimized conditions, this modified electrode was utilized to determine the concentrations of ciprofloxacin, ofloxacin, norfloxacin and gatifloxacin. The differential pulse voltammogram (DPV) exhibits the oxidation peak currents were linearly proportional to their concentration in the range of 0.05–100 μM for ciprofloxacin, 0.1–100 μM for ofloxacin, 0.1–40 μM for norfloxacin and 0.06–100 μM for gatifloxacin, respectively. This method was also successfully used to detect the concentrations of each drug in pharmaceutical formulations and human serum samples. In addition, this proposed fluoroquinolones sensor exhibited good reproducibility, long-term stability and fast current response.  相似文献

We describe a molecularly imprinted electrochemical sensor for selective and sensitive determination of β2-agonists. It is making use of a combination of single-wall carbon nanotubes (SWNTs) with a molecularly imprinted sol–gel. The SWNTs were introduced in order to enhance electron transport and sensitivity. The imprinted sol–gel film with its specific binding sites acts as a selective recognition element and as a preconcentrator for β2-agonists. The morphology of the imprinted film was characterized by scanning electron microscopy. The optimized sensor displays high sensitivity and excellent selectivity for the β2-agonists as shown for their determination in human serum samples.

A sensitive amperometric sensor has been constructed for the determination of hydrogen peroxide (HP). It is based on a glassy carbon electrode modified with a composite made from thionin, EDTA, multiwalled carbon nanotubes, and chitosan. Thionin was covalently immobilized on the surface of the electrode. The sensor exhibits a powerful electrocatalytic activity for the reduction of HP. The amperometric signal is proportional to the concentration of HP in the range from 0.2 μM to 85.0 μM, with a detection limit of 0.065 μM. The sensor displays excellent selectivity, good reproducibility and long-term stability.  相似文献

A novel and sensitive biosensor was developed for the determination of nitrite. Firstly, multi-walled carbon nanotubes–poly(amidoamine)–chitosan (MWNT–PAMAM–Chit) nanocomposite along with the incorporation of DNA was used to modify the glassy carbon electrode. Then the immobilization of Cyt c was accomplished using electrochemical deposition method by consecutive cyclic voltammetry (CV) scanning in a neutral Cyt c solution. CV behaviors of the modified electrodes showed that the MWNT–PAMAM–Chit nanocomposite is a good platform for the immobilization of DNA and Cyt c in order, at the same time, an excellent promoter for the electron transfer between Cyt c and the electrode. At high potential, the immobilized Cyt c could be further oxidized into highly reactive Cyt c π-cation by two-step electrochemical oxidation, which could oxidize NO2 into NO3 in the solution. Therefore, a nitrite biosensor based on the biocatalytic oxidation of the immobilized Cyt c was fabricated, which showed a fast response to nitrite (less than 5 s). The linear range of 0.2–80 μM and a detection limit of 0.03 μM was obtained. Finally, the application in food analysis using sausage as testing samples was also investigated.  相似文献
