In the field desorption mass spectrometry of oligosaccharides, traces of contaminating salts in polar solvents decrease the detection sensitivity, and in some cases no ion could be detected in the molecular range. The present work shows that the addition of alkaline salts (NaI or KI) greatly increases the sensitivity, and cationized molecular ions are obtained. The influence of the molar ratio sugar/salt on the nature and relative intensity of the desorbed species has been studied on mixtures of α-D -trehalose and NaI. At the low salt ratio (100:1), high molecular weight clusters [X M + Na]+ are preferentially formed. At the high salt ratio (1:10), mixed clusters [M + NaI + Na]+, doubly charged ions [M + 2Na]2+ and monomeric cationized ions [M + Na]+ are observed along with salt clusters [NaI + Na]+ and [2NaI + Na]+. In the range of molar ratios sugar/salt of 10:1 to 1:1, the field desorption mass spectra exhibit a cationized ion [M + Na]+, which contributes more than 80% of Σ100. This cationization technique has been applied to the field desorption mass spectrometry of several oligosaccharides. In all cases, the salt effect causes the replacement of the [M + H]+ ion by a [M + C]+ ion. (Note: the term [M + C]+ and similar ones mean an association between the whole molecule studied (M) and an alkaline cation [C]+ ([K)]+, [Na]+, [Li]+).) Thus, di, tri- and tetrasaccharides exhibit intense [M + Na]+ ions in the presence of NaI and only a few fragment peaks are observed in their field desorption mass spectra. This technique has been applied successfully to the detection of the hexasaccharide ajugose in a natural sample of pentasaccharide, and has also allowed the unambiguous determination of the composition of a mixture of partially methylated trehaloses. The salt effects are dicussed in terms of selective adsorption on the emitter, pre-existing soluble complexes sugar-salt, interactions between these species in the electric field and dissociative desorption of ionic complexes.  相似文献

The use of 5-aminosalicylic acid (5-ASA) as a new matrix for in-source decay (ISD) of peptides including mono- and di-phosphorylated peptides in matrix-assisted laser desorption/ionization (MALDI) mass spectrometry (MS) is described. The use of 5-ASA in MALDI-ISD has been evaluated from several standpoints: hydrogen-donating ability, the outstanding sharpness of molecular and fragment ion peaks, and the presence of interference peaks such as metastable peaks and multiply charged ions. The hydrogen-donating ability of several matrices such as α-cyano-4-hydroxycinnamic acid (CHCA), 2,5-dihydroxybenzoic acid (2,5-DHB), 1,5-diaminonaphthalene (1,5-DAN), sinapinic acid (SA), and 5-ASA was evaluated by using the peak abundance of a reduction product [M + 2H + H]+ to that of non-reduced protonated molecule [M + H]+ of the cyclic peptide vasopressin which contains a disulfide bond (S-S). The order of hydrogendonating ability was 1,5-DAN > 5-ASA > 2,5-DHB > SA = CHCA. The chemicals 1,5-DAN and 5-ASA in particular can be classified as reductive matrices. 5-ASA gave peaks with higher sharpness for protonated molecules and fragment ions than other matrices and did not give any interference peaks such as multiply-protonated ions and metastable ions in the ISD mass spectra of the peptides used. Particularly, 1,5-DAN and 5-ASA gave very little metastable peaks. This indicates that 1,5-DAN and 5-ASA are more “cool” than other matrices. The 1,5-DAN and 5-ASA can therefore be termed “reductive cool” matrix. Further, it was confirmed that ISD phenomena such as N-Cα bond cleavage and reduction of S-S bond is a single event in the ion source. The characteristic fragmentations, which form a− and (a + 2)-series ions, [M + H − 15]+, [M + H − 28]+, and [M + H − 44]+ ions in the MALDI-ISD are described.  相似文献

The dinuclear copper complex (α-cyano-4-hydroxycinnamic acid (CHCA) copper salt (CHCA)4Cu2), synthesized by reacting CHCA with copper oxide (CuO), yields increased abundances of [M + xCu − (x−1)H]+ (x = 1–6) ions when used as a matrix for matrix-assisted laser desorption ionization (355 nm Nd:YAG laser). The yield of [M + xCu − (x−1)H]+ (x = 1∼6) ion is much greater than that obtained by mixing peptides with copper salts or directly depositing peptides onto oxidized copper surfaces. The increased ion yields for [M + xCu − (x−1)H]+ facilitate studies of biologically important copper binding peptides. For example, using this matrix we have investigated site-specific copper binding of several peptides using fragmentation chemistry of [M + Cu]+ and [M + 2Cu − H]+ ions. The fragmentation studies reveal interesting insight on Cu binding preferences for basic amino acids. Most notable is the fact that the binding of a single Cu+ ion and two Cu+ ions are quite different, and these differences are explained in terms of intramolecular interactions of the peptide-Cu ionic complex.  相似文献

Mixtures of unsubstituted 15-crown-5 and its analogues containing ortho- and para-methoxyphenoxymethyl substituents with sodium salts were investigated by matrix assisted laser desorption/ionization (MALDI) mass spectrometry. Peaks of cationized molecules [M+Na]+ and cluster ions [2M+2Na+An]+, where M is the crown ether molecule and An is monobasic acid anion, were observed in the mass spectra. It was shown that an increase of the shielding degree of the sodium cation in complexes with crown ethers, i.e., the lariat effect, led to a significant decrease in the intensity of peaks of the cluster ions.  相似文献

The collision-induced decomposition (CID) mass spectra of the protonated and cationized molecules of a number of carbohydrate antibiotics of RMM ranging from 700 to 1500 were studied by means of a four-sector mass spectrometer with a floated collision cell. Helium and argon were used as collision gases. This work illustrates that cationized rather than protonated carbohydrate antibiotics give an increased yield of high-mass ions of diagnostic value. Further, when helium is replaced by argon as collision gas, differences in the CID spectra of MH+ ions become apparent only for molecules of RMM > 1400 whereas for [M + Na]+ ions differences are observed for molecules of RMM as low as 1000. These results have been attributed to the deposition of more internal energy in the precursor ion when argon is used, resulting in increased fragmentation.  相似文献

Red pigment-concentrating hormone (RPCH), an octapeptide found in crustaceans and insects with the sequence pGlu-Leu-Asn-Phe-Ser-Pro-Gly-Trp-NH2, is an N- and C-terminally blocked uncharged peptide. These structural features are shared with many members of the larger adipokinetic hormone (AKH)/RPCH peptide family in insects. We have applied vacuum UV matrix-assisted laser desorption/ionization (MALDI)-Fourier transform ion cyclotron mass spectrometry (FTMS) to the direct analysis of crustacean sinus gland tissues, using 2,5-dihydroxybenzoic acid (DHB) as the MALDI matrix, and have found that RPCH is detected in the cationized, [M + Na]+, form under conditions where other peptides in the direct tissue spectra are protonated without accompanying [M + Na]+ or [M + K]+ satellite peaks. The [M + H]+ ion for RPCH is not detected in tissue samples or for an RPCH standard, even when care is taken to eliminate metal ions. This behavior is not unprecedented; however, both direct tissue spectra and SORI-CID spectra provide no clues to suggest that the ionizing agent is a metal cation. In this communication, we characterize the MALDI-FTMS ionization and SORI-CID mass spectra of the [M + Na]+ and [M + K]+ ions from RPCH, and report on the detection of this neuropeptide in sinus gland tissues from the lobster Homarus americanus and the kelp crab Pugettia producta. We describe two strategies, an on-probe extraction procedure and a salt-doping approach, that can be applied to previously analyzed MALDI tissue samples to enhance and unmask sodiated peptides that may otherwise be mistaken for novel neuropeptides.  相似文献

A good understanding of gas‐phase fragmentation chemistry of peptides is important for accurate protein identification. Additional product ions obtained by sodiated peptides can provide useful sequence information supplementary to protonated peptides and improve protein identification. In this work, we first demonstrate that the sodiated a3 ions are abundant in the tandem mass spectra of sodium‐cationized peptides although observations of a3 ions have rarely been reported in protonated peptides. Quantum chemical calculations combined with tandem mass spectrometry are used to investigate this phenomenon by using a model tetrapeptide GGAG. Our results reveal that the most stable [a3 + Na ? H]+ ion is present as a bidentate linear structure in which the sodium cation coordinates to the two backbone carbonyl oxygen atoms. Due to structural inflexibility, further fragmentation of the [a3 + Na ? H]+ ion needs to overcome several relatively high energetic barriers to form [b2 + Na ? H]+ ion with a diketopiperazine structure. As a result, low abundance of [b2 + Na ? H]+ ion is detected at relatively high collision energy. In addition, our computational data also indicate that the common oxazolone pathway to generate [b2 + Na ? H]+ from the [a3 + Na ? H]+ ion is unlikely. Silver ion complexes of peptides [M + (Ag)n]+, M = angiotensin I or substance P where n = 1–8 and 17–23 for angiotensin I and n = 1–5 for substance P, are identified and characterized using matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOFMS). The Ag+ coordination number exceeds the number of available amino acid residues in angiotensin I whereas the number of observed complexes in substance P is less than the number of amino acid residues in it. The larger coordination number of angiotensin I with Ag+ indicates the simultaneous binding of several Ag+ ions to the amino acid residue present in it. The lower number of observed complexes in substance P suggests the binding of two or more residues to one Ag+ ion. The presence of trifluoroacetic acid in the peptide samples reduces the Ag+ coordination ability in both the peptides which indicates that the basic residues in it are already protonated and do not participate in the Ag+‐binding process. The Ag+ ion also forms a complex with the α‐cyano‐4‐hydroxycinnamic acid (CHCA) matrix and is observed in the MALDI mass spectra and the formation of [CHCA + Ag]+, [CHCA + AgNO3]+ and [(CHCA)2 + Ag]+ ions is due to the high binding affinity of Ag+ to the CN group of CHCA. The positive-ion fast atom bombardment mass spectra of permethylated aldo- and pseudoaldobiouronic acids can be used to distinguish these classes of compounds. The collisional-induced dissociation spectra of the [M + H]+ ions show fragment ions resulting from glycosidic bond cleavage and successive losses of methanol molecules. These spectra together with those of the [M + H ? MeOH]+ oxonium ions allow the identification of the type of interglycosidic linkage. Collisional activation of the relatively stable [M + Na]+ ions show many fragmentations which are common to alkali cationized permethylated saccharides. Moreover, fragment ions resulting from two-bond ring cleavage processes yield additional information with respect to the linkage between the hexose and uronic acid units.  相似文献

Combining desorption ionization with tandem mass spectrometry overcomes the disadvantage of limited fragmentation accompanying desorption and permits mixtures of closely related substances to be investigated directly. These features of the combination are illustrated by completing the structure-proof of a minor component of an ornithine-containing lipid mixture isolated from Thiobacillus thiooxidans. The minor component is a homolog of the major constituent and differs from the principal component owing to the presence of a double bond in lieu of a cyclopropyl ring in one of the constituent fatty acids. Another feature of the combined method is the potentially complementary nature of collision-activated dissociation spectra of protonated and cationized biomolecules. This is illustrated by the differences in the collision-activated dissociations of the [M + Na]+ of sucrose, desorbed by field desorption, and [M + H]+, desorbed by fast atom bombardment. A third illustration is the application of field desorption and tandem mass spectrometry to an organometallic compound. The combined approach allows the ligands to be identified and the relative ligand binding energies to be approximated.  相似文献

Geometrically isomeric dicarboxylic acids, such as maleic and fumaric acid and their methyl homologues, and the isomeric phthalic acids, have been investigated using fast atom bombardment, field ionization and field desorption mass spectrometry. The most intense peak in the positive ion fast atom bombardment spectra corresponds with the [M + H]+ ion. This ion, when derived from the E -acids, tragments either by successive loss of water and carbon monoxide or by elimination of carbon dioxide. In the case of the Z -acids only elimination of water from the [M + H]+ ions is observed to occur to a significant extent. The same is true for the [M + H]+ ions of the isomeric phthalic acids, that is the [M + H] ions derived from iso- and terephthalic acid exhibit more fragmentation than those of phthalic acid. All these acids undergo much less fragmentation upon field ionization, where not only abundant [M + H]+ ions, but also abundant [M] ions, are observed. Upon field desorption only the [M + H]+ and [M + Na]+ ions are observed under the measuring conditions. Negative ion fast atom bombardment spectra of the acids mentioned have also been recorded. In addition to the most abundant [M? H]? ions relatively intense peaks are observed, which correspond with the [M]?˙ ions. The fragmentations observed for these ions appear to be quite different from those reported in an earlier electron impact study and in a recent atmospheric pressure ionization investigation.  相似文献

Experimental data on correlations between the directions of the incident primary ion and the ejected protonated [M + H]+ and alkali metal adduct [M + Cs]+ molecules for three peptide samples with an incident beam of 72.3 MeV 127I14+ ions are reported. Measurements were carried out in a linear time-of-flight mass spectrometer by monitoring the secondary ion yield as a function of electrostatic deflection in a direction perpendicular to the spectrometer axis. No difference was observed in the direction in which [M + H]+ and [M + Cs]+ ionic species are preferentially desorbed. The results obtained suggest that alkali metal cation attachment to biomolecules in plasma desorption mass spectrometry is realized in a close spatial location and time interval with protonation. Formation of ion-molecule complexes occurs at an early stage of the desorption event and precedes their ejection into the gas phase.  相似文献

The gas-phase H/D exchange of bradykinin [M + H]+, [M + Na]+, [M + 2H]2+, and [M + H + Na]2+ ions; des-Arg1-bradykinin, des-Arg9-bradykinin, and bradykinin fragment 2-7 [M + H]+ ions; and O-methylbradykinin [M + H]+ and [M + 2H]2+ ions with D2O have been examined by electrospray ionization Fourier transform ion cyclotron resonance mass spectrometry at 9.4 T. The different peptides vary widely in exchange rate and extent of deuterium incorporation. H/D exchange was slowest and deuterium incorporation was least for bradykinin [M + H]+, [M + H + Na]2+ and bradykinin methyl ester [M + 2H]2+ ions. In contrast, H/D exchange and extent of deuteration are higher for des-Arg1-bradykinin, des-Arg9-bradykinin, and bradykinin fragment 2-7 [M + H]+ ions; and highest for bradykinin [M + Na]+ and [M + 2H]2+, and O-methylbradykinin [M + H]+. Because the most likely site of protonation is the guanidino group of arginine, the above reactivity pattern strongly supports a zwitterion form for protonated gas-phase bradykinin.  相似文献

The nature and location of a variety of modifications of fatty acids are determined by collisional activation (CA) of [M + 2Li ? H]+ ions. The sample molecules are cationized in situ on the probe tip, desorbed by fast atom bombardment and, upon CA, undergo charge-remote decompositions. This approach is a direct, totally instrumental method for structure elucidation. Advantages of CA of [M + 2Li ? H]+ ions are that fatty acids with substituents in close proximity to the carboxylate terminus and modified short-chain acids are readily determined: decompositions of carboxylate anions of these fatty acids result in collision-activated dissociation (CAD) spectra that give incomplete structural information. However, the CAD spectra of some [M ? H]? ions, such as those from epoxy acids, are simpler to interpret than those of the [M + 2Li ? H]+ ions. Thus, CA of fatty acid [M + 2Li ? H]+ ions is a complementary approach to CA of [M ? H]? ions for determining the fatty acid structures investigated here. The use of this approach for analyzing complex mixtures of modified fatty acids is also evaluated.  相似文献

The chemical ionization mass spectra of different dicarboxylic acids, including saturated and unsaturated aliphatic, aromatic, hydroxyl and amino-substituted dicarboxylic acids, have been studied using pure methanol as the reagent gas. Biomolecular monoesterification and diesterification product ions [M+15]+ and [M+29]+, and adduct ion [M+33]+, were observed, in addition to the protonated molecule [MH]+ and unimolecular water elimination product ions. The formation of a protonated molecule with bridged intramolecular hydrogen bond, and its effect on the esterification of dicarboxylic acids is discussed. Geometric isomers, such as maleic and fumaric acid, and ortho and meta isomers of phthalic acids can be distinguished from each other by methanol chemical ionization mass spectra. When ethanol was used as the reagent gas, similar mass spectra of some dicarboxylic acids were obtained.  相似文献

The reactions of dimethyl ether ions with neutral amino alcohols were examined in both a quadrupole ion trap mass spectrometer and a triple quadrupole mass spectrometer. These ion-molecule reactions produced two types of ions: the protonated species [M+l]+ and a more complex product at [M+13]+. The abundance of the [M+13]+ ions relative to that of the [M+1]+ ions decreases with increasing formal interfunctional distance. Multistage collision-activated dissociation techniques were used to characterize the [M+13]+ product ions, their reactivities, and the mechanisms for their formation and dissociation. In addition, molecular semiempirical calculation methods were used to probe the thermochemistry of these reactions. Reaction at the amino alcohol nitrogen site is favored, and the resulting [M+13]+ addition products may cyclize for additional stabilization. Comparisons were made among the behavior of related compounds, such as alcohols, diols, amines, and diamines. The alcohols reacted only to form the protonated species, but the diols, amines, and diamines all formed significant amounts of [M+13]+ ions or related dissociation products.  相似文献

The product ion mass spectra of protonated and cationated peptides of relative molecular mass (RMM) 555–574 Da have been obtained by surface-induced dissociation of MH+ and [M + Cat] ions in a four-sector tandem mass spectrometer equipped with a specially designed collision cell. A linked scan of the electric and magnetic sector field strengths of the second mass spectrometer was used to transmit the fragment ions arising from collisions with a stainless steel surface. The resulting mass spectra contained broad metastable ion peaks produced by the dissociation of MH+ and [M + Cat]+ ions before the second magnetic sector, in the fourth field-free region of the instrument.  相似文献

2,3‐Dimethyl‐2,3‐dinitrobutane (DMNB) is an explosive taggant added to plastic explosives during manufacture making them more susceptible to vapour‐phase detection systems. In this study, the formation and detection of gas‐phase [M+H]+, [M+Li]+, [M+NH4]+ and [M+Na]+ adducts of DMNB was achieved using electrospray ionisation on a triple quadrupole mass spectrometer. The [M+H]+ ion abundance was found to have a strong dependence on ion source temperature, decreasing markedly at source temperatures above 50°C. In contrast, the [M+Na]+ ion demonstrated increasing ion abundance at source temperatures up to 105°C. The relative susceptibility of DMNB adduct ions toward dissociation was investigated by collision‐induced dissociation. Probable structures of product ions and mechanisms for unimolecular dissociation have been inferred based on fragmentation patterns from tandem mass (MS/MS) spectra of source‐formed ions of normal and isotopically labelled DMNB, and quantum chemical calculations. Both thermal and collisional activation studies suggest that the [M+Na]+ adduct ions are significantly more stable toward dissociation than their protonated analogues and, as a consequence, the former provide attractive targets for detection by contemporary rapid screening methods such as desorption electrospray ionisation mass spectrometry. N-Acetylcysteine and nine N-acetylcysteine conjugates of synthetic origin were characterized by positive- and negative-ion plasma desorption mass Spectrometry. For sample preparation the electrospray technique and the nitrocellulose spin deposition technique were applied. The fragmentation of these compounds, which are best seen as S-substituted desaminoglycylcysteine dipeptides, shows a similar behaviour to that of linear peptides. In the positive-ion mass spectra intense protonated molecular ion peaks are observed. In addition, several sequence-specific fragment ions (A+, B+, [Y + 2H]+, Z+), immonium ions (I+) and a diagnostic fragment ion for mercap-turic acids (RM+) are detected. The negative-ion mass spectra exhibit deprotonated molecular ions and in contrast only one fragment ion corresponding to side-chain specific cleavage ([RXS]?) representing the xenobiotic moiety. In the case of a low alkali metal concentration on the target, cluster molecular ions of the [nM + H]+ or [nM - H]? ion type (n = 1-3) are observed. The analysis of an equimolar mixture of eight N-acetylcysteine conjugates shows different quasi-molecular ion yields for the positive- and negative-ion spectra.  相似文献

Analysis of the isobutane chemical ionization mass spectra of hexenols, cyclohexenols and various syn/anti pairs of bicyclic and tricyclic homoallylic alcohols shows that: (i) the spectra of the allylic alcohols are dominated by [M + H – H2O]+ and [M + C4H9–H2O]+ ions and contain traces of [M + H]+ ions; (ii) [M + H]+ ions are prominent in the spectra of acyclic and certain cyclic homoallylic alcohols; and (iii) [M + H]+ ions dominate the spectra of other acyclic unsaturated alcohols. The [M + H]+ ions may result from either: (a) protonation of the hydroxyl group, followed by a very rapid intramolecular proton transfer from the protonated hydroxyl group to the carbon–carbon double bond or internal solvation of the protonated hydroxyl group by the carbon–carbon double bond; and/or (b) direct protonation of the carbon–carbon double bond with significant internal solvation of the resulting carbocation by the hydroxyl group, which may lead to carbon–oxygen bond formation to give a protonated cyclic ether. The consequences of placing various geometric constraints on the possible intramolecular interactions between the hydroxyl group and the carbon–carbon double bond in unsaturated alcohols are explored.  相似文献
