合成了未见文献报道的水溶性的3'-磺酸钠-4',7-二羧甲氧基异黄酮(L1)和3'-磺酸钠-4'-羟基-7-羧甲氧基异黄酮(L2), 采用IR, UV, 1H NMR和元素分析对其结构进行了表征, 利用荧光光谱法研究了它们和母体大豆甙元(D)对羟基自由基的清除活性, 用紫外光谱法研究了其对超氧阴离子自由基和1,1-二苯基-2-苦肼基自由基(1,1-diphenyl-2-picrylhydrazyl free radical, 简称DPPH)的清除活性; 并采用量子化学AM1方法在全几何构型优化的基础上进行了电荷布居分析, 计算了它们抽氢反应的生成热(∆Hf), 从而从理论上探讨了目标化合物清除羟基自由基的活性. 实验结果表明本文合成的两种水溶性化合物清除超氧阴离子自由基和DPPH自由基的活性要优于母体大豆甙元, 对于目标化合物清除羟基自由基的活性, 实验和理论结果都显示其清除活性要优于大豆甙元.  相似文献

A new route for the synthesis of substituted 8‐methyl‐6‐phenyl‐5,6‐dihydro‐4H‐1,3,2‐benzodioxaphosphocine‐2‐oxide derivatives has been developed by using cinnamic acid and p‐cresol via condensation, reduction, and followed by phosphorylation steps. The title compounds were characterized by IR, 1H, 13C, 31P, and mass spectral studies and elemental analysis. The title compounds have been investigated for their antioxidant activity with respect to their IC50 values using 2,2‐diphenyl‐1‐picrylhydrazyl, NO radical scavenging activities, and reducing power assay. The results obtained from the aforementioned methods revealed that 2‐phenylamino derivatives have shown greater free radical scavenging activity when compared with those of the phenoxy derivatives and is attributed to the presence of secondary amino group, which is able to produce free radicals easily.  相似文献

采用自由基比色法考察苯并呋喃酮衍生物结构对其DPPH捕获能力的影响。结果发现,母体苯环5,7位上甲基和叔丁基的改变以及取代苯环上强供电子取代基的存在对苯并呋喃酮衍生物的DPPH捕获能力没有明显影响。取代苯环2’位存在明显的位阻作用,对苯并呋喃酮衍生物的DPPH捕获能力具有明显的抑制作用:并且当此位为不含活泼氢的氢键受体取代基时,苯并呋喃酮衍生物的DPPH捕获能力会由于取代基与3位活泼氢的氢键作用而被进一步削弱;但此位为含有活泼氢的氢键受体取代基时,此取代基的氢将会由于3位活泼氢与取代基的氢键作用而活化,从而使其位阻作用得到一定程度的抑制。  相似文献

The effect of grape seed extract (GSE) on the lipid peroxidation of rat heart mitochondria was studied using the spin trapping agent α-(4-pyridyl-1-oxide)-N-t-butylnitrone (4-POBN) with electron spin resonance (ESR) spectrometer. Two fatty acid spin labels, 5-doxyl-stearic acid (5-DOXYL) and 16-doxyl-stearic acid (16-DOXYL) were used to study the effect of GSE on mitochondrial membrane fluidity. The results showed that the GSE could scavenge the lipid free radicals generated from the mitochondria of rat heart, and protect the mitochondrial membranes damaged by lipid peroxidation. Both the scavenging effect and the protective effect are better in water phase than in lipid phase. And the data indicate that perhaps triners and other polymers have better scavenging effect on lipid free radicals than monomers.  相似文献

A novel screening method was developed for the detection and identification of radical scavenging natural antioxidants based on a free radical reaction combined with liquid chromatography with tandem mass spectrometry. Functionalized graphene quantum dots were prepared for loading free radicals in the complex screening system. The detection was performed with and without a preliminary exposure of the samples to specific free radicals on the functionalized graphene quantum dots, which can facilitate charge transfer between free radicals and antioxidants. The difference in chromatographic peak areas was used to identify potential antioxidants. This is a novel approach to simultaneously evaluate the antioxidant power of a component versus a free radical, and to identify it in a vegetal matrix. The structures of the antioxidants in the samples were identified using tandem mass spectrometry and comparison with standards. Fourteen compounds were found to possess potential antioxidant activity, and their free radical scavenging capacities were investigated. The order of scavenging capacity of 14 compounds was compared according to their free radical scavenging rate. 4′,5,6,7‐Tetrahydroxyflavone (radical scavenging rate: 0.05253 mL mg?1 s?1) showed the strongest capability for scavenging free radicals.  相似文献

Uralenol and neouralenol are two typical licorice root extracts that presents multiple reactive hydroxyl groups, which are considered as good free radical scavengers. A theoretical study on the primary antioxidant activity of uralenol and neouralenol toward hydroxyl and hydroperoxyl radicals has been carried out using the density functional theory (DFT). A total of 10 reaction pathways of uralenol and neouralenol scavenging two radicals in gas phase and in water phase have been tracked. Neouralenol was found to be a better hydroxyl and hydroperoxyl scavenger than uralenol. In vivo, the more reactive sites in uralenol are U5 and U’1, respectively, for scavenging ·OH and ·OOH; and the more reactive sites in neouralenol are N4 and N’5 for scavenging ·OH and ·OOH, respectively.  相似文献

Robinia pseudoacacia flowers have attracted much attention because of numerous bioactivities. In this study, its extract showed the potential scavenging ability for 2,2′-azinobis-(3-ethylbenzthiazoline-6-sulphonate) and 1,1-diphenyl-2-picrylhydrazyl free radicals. Under the guidance of antioxidant activity, the antioxidant extract was enriched by liquid-liquid extraction. The partition coefficients of the two main components in antioxidant extracts differed greatly, so in this study, elution-extrusion counter-current chromatography with the solvent system of n-hexane-ethyl acetate-methanol-water (2.5:5:2.5:5, v/v) was used to enhance the separation efficiency, and the two main components were successfully obtained. Among them, kaempferol showed strong antioxidant activity, which can be responsible for the activity of the extract. In order to deeply understand the antioxidant mechanism of kaempferol, the thermodynamics, frontier molecular orbital, and kinetics of scavenging free radicals were investigated by density functional theory. The results showed that 4′-OH in kaempferol was the most active group, which can scavenge free radicals by hydrogen atom transfer in non-polar solvents and activate 3-OH to generate double hydrogen atom transfer in the gas phase. But in polar solvents, it was more inclined to clear radicals through single electron transfer and proton transfer. The kinetic result showed that kaempferol needed 9.17 kcal/mol of activation energy to scavenge free radicals.  相似文献

Cigarette smoke contains free radicals that are considered to be a major group of carcinogens. In the present study, we used plant antioxidants to treat cigarette filters for scavenging gas phase free radicals in cigarette smoke. The scavenging activities of four antioxidants against gas phase free radicals were analyzed using an improved spin trap protocol. The scavenging rates ranked as lycopene > grape seed extract > pycnogenol > tomato powder. A scavenging rate of 62% (p < 0.001) was obtained from the lycopene treatment. The use of plant antioxidants as potential scavengers for reducing free-radicals in cigarette smoke is discussed.  相似文献

A crude polysaccharide was extracted from Physalis alkekengi L. fruit. HPLC was used for the component analysis of the polysaccharide. The results indicate that Physalis alkekengi L. polysaccharide(PAP) was composed of rhamnose, xylose, arabinose, galactose, and glucose. Free radicals scavenging activity of PAP was studied through 3 free radicals scavenging tests. PAP exhibited high scavenging effects on OH and DPPH radicals, and both the scavenging rates were about 80%. The scavenging rate of O2^- radical was about 22%.  相似文献

自由基清除是富勒烯及其衍生物的一种重要特性. 首次制备了1种二加成亚甲基富勒烯[60]膦酸酯衍生物(bis-methanophosphonate [60]fullerene, BMPF)的纳米颗粒水悬液(n-BMPF), 并采用邻苯三酚自氧化法结合分光光度法, 测定了n-BMPF对超氧阴离子自由基的清除作用. 结果显示, 制备的n-BMPF溶液较稳定, 适合生物学实验. 同时, 它表现出很强的清除超氧阴离子自由基的能力, 并具有浓度依赖性. 当n-BMPF的终浓度为12 μmol/L时, 其对超氧阴离子自由基的清除率达到91.96%. n-BMPF的活性远大于已报道具有自由基清除作用的水溶性富勒烯衍生物富勒醇. 这些数据表明n-BMPF可能作为一种新型高效的自由基清除剂, 在生物医学领域具有潜在的应用价值.  相似文献

The experimental and theoretical study for evaluation of scavenging activity of edaravone (S1) and related derivatives, such as antipyrine (S2), dipyrone (S3), and phenylbutazone (S4), was carried out against DPPH and ABTS radicals. Structure–activity relationship study was performed using quantum chemical calculations at the DFT/B3LYP level of theory along with the 6-31G* basis sets. S1 and S4 are more effective scavengers against DPPH and ABTS. We observed little effects of S2 and S3 at several concentrations against these two free radicals. The calculations of HOMO, ionization potential, and bond dissociation energy confirmed that a hydrogen transfer is more preferential than an electron transfer. The radical stability of these compounds is related with spin densities. In accordance with experimental and theoretical results, edaravone is more active than phenylbutazone as scavenging drug.  相似文献

利用产多糖菌Enterobacter cloacae Z0206(E.cloacaeZ0206)的深层发酵法制备了E.cloacae Z0206细菌富硒多糖;测定了其还原能力和清除1,1-diphenyl-2-picrylhydrazyl(DPPH)自由基、超氧阴离子及羟自由基的能力.结果表明,通过深层富硒发酵、醇沉离心等制备富硒多糖SEPS的产量为9.28g/L,富硒量为2.314mg/g;E.cloacae Z0206富硒多糖对DPPH自由基和羟自由基具有较好的清除作用,在浓度为5g/L时对DPPH自由基和羟自由基的清除率分别为80.35%和84.26%,并具有较强的还原能力,但其对超氧阴离子自由基的清除能力较差.  相似文献

The purpose of this study was to examine the free radical scavenging and antioxidant activities of ellagic acid (EA) and ellagic acid peracetate (EAPA) by measuring their reactions with the radicals, 2,2-diphenyl-1-picrylhydrazyl and galvinoxyl using EPR spectroscopy. We have also evaluated the influence of EA and EAPA on the ROS production in L-6 myoblasts and rat liver microsomal lipid peroxidation catalyzed by NADPH. The results obtained clearly indicated that EA has tremendous ability to scavenge free radicals, even at concentration of 1 µM. Interestingly even in the absence of esterase, EAPA, the acetylated product of EA, was also found to be a good scavenger but at a relatively slower rate. Kinetic studies revealed that both EA and EAPA have ability to scavenge free radicals at the concentrations of 1 µM over extended periods of time. In cellular systems, EA and EAPA were found to have similar potentials for the inhibition of ROS production in L-6 myoblasts and NADPH-dependent catalyzed microsomal lipid peroxidation.  相似文献

利用邻菲罗啉化学发光法研究了中草药有效成分氯原酸、连翘酯甙等肉桂酸类抗氧化剂对HO·的清除作用。量子化学计算[3-21G、B3LYP/6-31G(d)]对实验结果给予了合理的解释。 计算结果显示抗氧化剂形成半醌式自由基前后的能量之差(ΔE)能较好地衡量其活性的高低;抗氧化剂半醌式自由基与邻位取代基形成分子内氢键或通过共振形成邻苯醌可使内能降低,半醌式自由基更稳定;脱氢酚羟基的对位含有给电子取代基团时,可使半醌式自由基易形成并提高其稳定性。  相似文献

Phenolic Schiff bases are known for their diverse biological activities and ability to scavenge free radicals. To elucidate (1) the structure–antioxidant activity relationship of a series of thirty synthetic derivatives of 2-methoxybezohydrazide phenolic Schiff bases and (2) to determine the major mechanism involved in free radical scavenging, we used density functional theory calculations (B3P86/6-31+(d,p)) within polarizable continuum model. The results showed the importance of the bond dissociation enthalpies (BDEs) related to the first and second (BDEd) hydrogen atom transfer (intrinsic parameters) for rationalizing the antioxidant activity. In addition to the number of OH groups, the presence of a bromine substituent plays an interesting role in modulating the antioxidant activity. Theoretical thermodynamic and kinetic studies demonstrated that the free radical scavenging by these Schiff bases mainly proceeds through proton-coupled electron transfer rather than sequential proton loss electron transfer, the latter mechanism being only feasible at relatively high pH.  相似文献

A novel free radical reaction combined with liquid chromatography electrospray ionization tandem mass spectrometry (FRR-LC–PDA-ESI/APCI-MS/MS) screening method was developed for the detection and identification of radical-scavenging natural antioxidants. Functionalized graphene was prepared by chemical method for loading free radicals (superoxide radical, peroxyl radical and PAHs free radical). Separation was performed with and without a preliminary exposure of the sample to specific free radicals on the functionalized graphene, which can facilitate reaction kinetics (charge transfers) between free radicals and potential antioxidants. The difference in chromatographic peak areas is used to identify potential antioxidants. The structure of the antioxidants in one sample (Swertia chirayita) is identified using MS/MS and comparison with standards. Thirteen compounds were found to possess potential antioxidant activity, and their free radical-scavenging capacities were investigated. The thirteen compounds were identified as 1,3,5-trihydroxyxanthone-8-O-β-d-glucopyranoside (PD1), norswertianin (PD2), 1,3,5,8-tetrahydroxyxanthone (PD3), 3, 3′, 4′, 5, 8-penta hydroxyflavone-6-β-d-glucopyranosiduronic acid-6′-pentopyranose-7-O-glucopyranoside (PD4), 1,5,8-trihydroxy-3-methoxyxanthone (PD5), swertiamarin (PS1), 2-C-β-d-glucopyranosyl-1,3,7-trihydroxylxanthone (PS2), 1,3,7-trihydroxylxanthone-8-O-β-d-glucopyranoside (PL1), 1,3,8-trihydroxyl xanthone-5-O-β-d-glucopyranoside (PL2), 1,3,7-trihydroxy-8-methoxyxanthone (PL3), 1,2,3-trihydroxy-7,8-dimethoxyxanthone (PL4), 1,8-dihydroxy-2,6-dimethoxy xanthone (PL5) and 1,3,5,8-tetramethoxydecussatin (PL6). The reactivity and SC50 values of those compounds were investigated, respectively. PD4 showed the strongest capability for scavenging PAHs free radical; PL4 showed prominent scavenging capacities in the lipid peroxidation processes; it was found that all components in S. chirayita exhibited weak reactivity in the superoxide radical scavenging capacity. The use of the free radical reaction screening method based on LC–PDA-ESI/APCI-MS/MS would provide a new approach for rapid detection and identification of radical-scavenging natural antioxidants from complex matrices.  相似文献

The radical scavenging activity of the stable derivatives, which are O-substituted at the C-2 position of ascorbic acid (AA), against 1,1-diphenyl-2-picrylhydrazyl (DPPH) was evaluated in buffer under different pH conditions, and compared with those of AA and alpha-tocopherol. AA was shown to have 50% radical scavenging ability (EC50) at a concentration of 2.2 x 10(-5) M against 0.1 mM DPPH in 60% ethanol. Ascorbyl 6-palmitate, a lipophilic AA derivative which has a free endiol group and is therefore unstable, also showed potent radical scavenging activity with EC50 of 2.9 x 10(-5) M. A typical lipophilic antioxidant, alpha-tocopherol gave a similar EC50 value as that of AA. In contrast, ascorbyl 2,6-dipalmitate, AA 2-phosphate and AA 2-sulfate exhibited negligible scavenging activity. On the other hand, 2-O-alpha-D-glucopyranosyl-L-ascorbic acid (AA-2G) and a series of 6-O-acyl-2-O-alpha-D-glucopyranosyl-L-ascorbic acids (6-Acyl-AA-2G) themselves exhibited the radical scavenging activity of EC50: 6.1 x 10(-5) M and 4.4 x 10(-5)-5.9 x 10(-5) M, respectively, although their activities were lower than that of AA. Among 6-Acyl-AA-2G derivatives, the EC50 values tended to decrease with increasing length of their acyl carbon group. Increasing pH of the buffer resulted in decrease in the scavenging activity of all compounds tested as expected. We speculate that the difference in the radical scavenging activity of derivatives O-substituted at the C-2 position of AA may be ascribed to the linkage type of the substituent group to the endiol-lactone resonance system and the degree of dissociation of the C-3 proton.  相似文献

Eight novel silybin analogues (7a-h) were synthesized and their antioxidant properties including the capability of scavenging superoxide anion free radicals and the inhibitory effect on DPPH free radicals were determined.
