In this paper, a simple and green modification method is developed for biomolecules analysis on poly(dimethylsiloxane) (PDMS) microchip with successful depression of nonspecific biomolecules adsorption. O-[(N-succinimdyl)succiny]-o'-methyl-poly(ethylene glycol) was explored to form hydrophilic surface via in-situ grafting onto pre-coated chitosan (Chit) from aqueous solution in the PDMS microchannel. The polysaccharide chains backbone of Chit was strongly attracted onto the surface of PDMS via hydrophobic interaction combined with hydrogen bonding in an alkaline medium. The methyl-poly(ethylene glycol) (mPEG) could produce hydrophilic domains on the mPEG/aqueous interface, which generated brush-like coating in this way and revealed perfect resistance to nonspecific adsorption of biomolecules. This strategy could greatly improve separation efficiency and reproducibility of biomolecules. Amino acids and proteins could be efficiently separated and successfully detected on the coated microchip coupled with end-channel amperometric detection at a copper electrode. In addition, it offered an effective means for preparing biocompatible and hydrophilic surface on microfluidic devices, which may have potential use in the biological analysis.  相似文献

The adsorption properties of a graft copolymer of poly(ethylene glycol) (PEG) with a polycationic backbone, namely, poly( l-lysine)- graft-poly(ethylene glycol) (PLL- g-PEG), onto nonpolar, hydrophobic PDMS surfaces from aqueous solution and the lubrication properties of the self-mated sliding contacts of PDMS surfaces modified with PLL- g-PEG have been investigated. Whereas PLL- g-PEG is spontaneously attracted to negatively charged surfaces as a result of the polycationic PLL backbone, the collective interaction of (CH 2) 4 hydrocarbon moieties on the lysine units in the PLL backbone with nonpolar, hydrophobic surfaces also enables the adsorption of PLL- g-PEG onto hydrophobic surfaces such as PDMS. The adsorption and lubrication properties of PLL- g-PEG have been investigated by varying the aqueous solution parameters, such as pH (2, 7, and 12) and KCl concentration (0, 0.01, 0.1, and 1 M) as well as the length of the PLL backbone of the copolymer (20 vs 375 kDa). In the absence of tribological stress, the adsorption of PLL- g-PEG onto PDMS surfaces was mainly governed by the KCl concentration, whereas the role of pH or the molecular weight of the copolymer was of relatively minor importance; for all pH values, the adsorbed mass decreased with increasing KCl concentration. Under tribological stress, however, a clear dependence of the lubrication properties of PLL- g-PEG on all of the studied parameters, including pH, KCl concentration, and backbone molecular weight, was observed. The adsorption strength of PLL- g-PEG on PDMS surfaces, rather than the adsorbed mass itself, appeared to be the most critical parameter in determining the lubrication properties.  相似文献

The electrostatic adsorption onto charged surfaces of comb copolymers comprising a polyelectrolyte backbone and pendent PEG side chains, such as poly(l-lysine)-g-poly(ethylene glycol) (PLL-g-PEG), has in previous studies provided protein-repellent thin coatings, particularly on metal oxide surfaces. A drawback of this approach is, however, the instability of such adsorbed layers under extreme pH values or high ionic strength. We have overcome this limitation in the present study by covalently immobilizing PLL-g-PEG copolymers onto aldehyde plasma-modified substrates. Silicon wafers, optical waveguide chips, and perfluorinated ethylene-co-propylene (FEP) polymer substrates were first coated with a thin plasma polymer layer using a propionaldehyde plasma, followed by covalent immobilization of PLL-g-PEG via reductive amination between amine groups of the PLL backbone with aldehyde groups on the plasma-deposited interlayer. The stability in high salt media and the protein resistance of different molecular architectures of immobilized PLL-g-PEG layers were quantitatively investigated by XPS, an optical waveguide technique (OWLS), and ToF-SIMS. The adsorption of bovine serum albumin was found to be below the detection limit (<2 ng/cm(2)), as for electrostatically adsorbed PLL-g-PEG layers. However, after 24 h of exposure of covalently immobilized layers of PLL-g-PEG to high ionic strength buffer (2400 mM NaCl), no significant change in the protein resistance was observed, whereas under the same conditions electrostatically adsorbed PLL-g-PEG coatings lost their protein resistance. Moreover, covalent immobilization via an aldehyde plasma interlayer enabled the application of PLL-g-PEG layers onto substrates such as FEP onto which electrostatic binding is not possible. These findings create a generic platform for the covalent immobilization of PLL-g-PEG onto a wide variety of substrates.  相似文献

This paper reports a simple microwave plasma patterning of poly(dimethylsiloxane) (PDMS) surfaces, which is accomplished by allowing selective surface areas to microwave plasma exposure in the presence of gaseous monomer. When maleic anhydride is used for microwave plasma reaction in the presence of physical barrier on the PDMS substrate, the resulting patterned surfaces with chemically bonded maleic anhydride and carboxylic acid groups are generated. In this particular study we attached amoxicillin via ammonolysis under weak base conditions in the presence of a catalyst as well as poly(ethyleneglycol) (PEG). A combination of internal reflection IR imaging (IRIRI) and atomic force microscopy (AFM) revealed that amoxicillin and PEG can be readily reacted on the microwave plasma patterned PDMS surfaces. Surface areas directly exposed to microwave plasmons exhibit the highest reactivity due to higher content of functional groups. These studies also show that molecular weight of PEG has also significant effect on kinetics of surface reactions.  相似文献

We report about the surface modification of polystyrene (PSt) with photoreactive alpha-4-azidobenzoyl-omega-methoxy poly(ethylene glycol)s (ABMPEG) of three different molecular weights (MWs of approximately 2, approximately 5, and approximately 10 kg/mol) and with two poly(ethylene glycol)/poly(propylene glycol) triblock copolymers (PEG-PPG-PEG) of about identical PEG/PPG ratio (80/20, w/w) and MW(PEG) of approximately 3 and approximately 6 kg/mol, all via adsorption from aqueous solutions. For ABMPEGs, an additional UV irradiation was used for photografting to the PSt. Contact angle (CA) and atomic force microscopy data revealed pronounced differences of the hydrophilicity/hydrophobicity and topography of the surfaces as a function of PEG type and concentration used for the modification. In all cases, an incomplete coverage of the PSt was observed even after modification at the highest solution concentrations (10 g/L). However, clear differences were seen between PEG-PPG-PEGs and ABMPEGs; only for the latter was a nanoscale-ordered interphase structure with an influence of MW(PEG) on the PEG density observed; after modification at the same solution concentrations, the density was significantly higher for lower MW(PEG). The adsorption of three proteins, myoglobin (Mgb), bovine serum albumin (BSA), and fibrinogen to the various surfaces was analyzed by surface plasmon resonance. Pronounced differences between the two PEG types with respect to the reduction of protein adsorption were found. At high, but still incomplete, surface coverage and similar CA, the shielding of ABMPEG layers toward the adsorption of Mgb and BSA was much more efficient; e.g., the adsorbed Mgb mass relative to that of unmodified PSt was reduced to 10% for ABMPEG 2 kg/mol while for both PEG-PPG-PEGs the Mgb mass was still around 100%. In addition, for the ABMPEG layers an effect of MW(PEG) on adsorbed protein mass-decrease with decreasing MW-could be confirmed; and the highest Mgb/BSA selectivities were also observed. A "two-dimensional molecular sieving", based on PEG molecules having a nanoscale order at the hydrophobic substrate polymer surface has been proposed, and the main prerequisites were the use of PEG conjugates which are suitable for an "end-on" grafting (e.g., ABMPEGs), the use of suitable (not too high) concentrations for the surface modification via adsorption/self-assembly, optionally the photografting on the substrate (possible only for ABMPEG), and presumably, a washing step to remove the excess of unbound PEGs. The results of this study also strongly support the hypothesis that the biocompatibility of hydrophobic materials can be very much improved by PEG modifications at surface coverages that are incomplete but have an ordered layer structure controlled by the size and steric interactions of surface-bound PEGs.  相似文献

Inclusion of a polymer cushion between a lipid bilayer membrane and a solid surface has been suggested as a means to provide a soft, deformable layer that will allow for transmembrane protein insertion and mobility. In this study, mobile, tethered lipid bilayers were formed on a poly(ethylene glycol) (PEG) support via a two-step adsorption process. The PEG films were prepared by coadsorbing a heterofunctional, telechelic PEG lipopolymer (1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-poly(ethylene glycol)-2000-N-[3-(2-(pyridyldithio)propionate]) (DSPE-PEG-PDP) and a nonlipid functionalized PEG-PDP from an ethanol/water mixture, as described in a previous paper (Munro, J. C.; Frank, C. W. Langmuir 2004, 20, 3339-3349). Then a two-step lipid adsorption strategy was used. First, lipids were adsorbed onto the PEG support from a hexane solution. Second, vesicles were adsorbed and fused on the surface to create a bilayer in an aqueous environment. Fluorescence recovery after photobleaching experiments show that this process results in mobile bilayers with diffusion coefficients on the order of 2 microm2/s. The mobility of the bilayers is decreased slightly by increasing the density of tethered lipids. The formation of bilayers, and not multilayer structures, is also confirmed by surface plasmon resonance, which was used to determine in situ film thickness, and by fluorimetry, which was used to determine quantitatively the fluorescence intensity for each 18 by 18 mm sample. Unfortunately, fluorescence microscopy also shows that there are large defects on the samples, which limits the utility of this system.  相似文献

A new environmentally friendly method is developed for preventing nonspecific biomolecules from adsorption on poly(dimethylsiloxane) (PDMS) surface via in situ covalent modification. o-[(N-Succinimdyl)succiny]-o'-methyl-poly(ethylene glycol) (NSS-mPEG) was covalently grafted onto PDMS microchannel surface that was pretreated by air-plasma and silanized with 3-aminopropyl-triethoxysilanes (APTES). The modification processes were carried out in aqueous solution without any organic solvent. The mPEG side chains displayed extended structure and created a nonionic hydrophilic polymer brushes layer on PDMS surface, which can effectively prevent the adsorption of biomolecules. The developed method had improved reproducibility of separation and stability of electroosmotic flow (EOF), enhanced hydrophilicity of surface and peak resolution, and decreased adsorption of biomolecules. EOF in the modified microchannel was strongly suppressed, compared with those in the native and silanized PDMS microchips. Seven amino acids have been efficiently separated and successfully detected on the coated PDMS microchip coupled with end-channel amperometric detection. Relative standard deviations (RSDs) of their migration time for run-to-run, day-to-day and chip-to-chip, were all below 2.3%. Moreover, the covalent-modified PDMS channels displayed long-term stability for 4 weeks. This novel coating strategy showed promising application in biomolecules separation.  相似文献

Arrays of releasable micropallets with surrounding walls of poly(ethylene glycol) (PEG) were fabricated for the patterning and sorting of adherent cells. PEG walls were fabricated between the SU-8 pallets using a simple, mask-free strategy. By utilizing the difference in UV-transmittance of glass and SU-8, PEG monomer was selectively photopolymerized in the space surrounding the pallets. Since the PEG walls are composed of a cross-linked structure, the stability of the walls is independent of the pallet array geometry and the properties of the overlying solution. Even though surrounded with PEG walls, the individual pallets were detached from the array by the mechanical force generated by a focused laser pulse, with a release threshold of 6 microJ. Since the PEG hydrogels are repellent to protein adsorption and cell attachment, the walls localized cell growth to the pallet top surface. Cells grown in the microwells formed by the PEG walls were released by detaching the underlying pallet. The released cells/pallets were collected, cultured and clonally expanded. The micropallet arrays with PEG walls provide a platform for performing single cell analysis and sorting on chip.  相似文献

In this study, we applied photo-induced graft polymerization to micropatterned surface modification of polydimethylsiloxane (PDMS) with poly(ethylene glycol). Two types of monomers, polyethylene glycol monoacrylate (PEGMA) and polyethylene glycol diacrylate (PEGDA), were tested for surface modification of PDMS. Changes in the surface hydrophilicity and surface element composition were characterized by contact angle measurement and electron spectroscopy for chemical analysis. The PEGMA-grafted PDMS surfaces gradually lost their hydrophilicity within two weeks. In contrast, the PEGDA-grafted PDMS surface maintained stable hydrophilic characteristics for more than two months. Micropatterned protein adsorption and micropatterned cell adhesion were successfully demonstrated using PEGDA-micropatterned PDMS surfaces, which were prepared by photo-induced graft polymerization using photomasks. The PEGDA-grafted PDMS exhibited useful characteristics for microfluidic devices (e.g. hydrophilicity, low protein adsorption, and low cell attachment). The technique presented in this study will be useful for surface modification of various research tools and devices.  相似文献

Block copolymers composed of hydrophilic poly(ethylene glycol) (PEG) and hydrophobic biodegradable polyesters have been reported as thermogelling polymers, because they feature temperature-dependent sol-to-gel or gel-to-sol transitions in aqueous solutions. In this study, a series of thermogelling poly(ethylene glycol methyl ether)-block–poly(cyclohexylenedimethylene adipate)-block–poly(ethylene glycol methyl ether) triblock copolymers and PEG-block–poly(cyclohexylenedimethylene adipate) multiblock copolymers was synthesized by reacting hydroxyl-terminated poly(cyclohexylenedimethylene adipate) (PCA) with poly(ethylene glycol methyl ether) and PEG, respectively, using 1,6-diisocyanatohexane as the coupling agent. Two hydroxyl-terminated PCAs, i.e., poly(1,4-cyclohexylenedimethylene adipate) and poly(1,3/1,4-cyclohexylenedimethylene adipate), were synthesized by the condensation reaction of adipic acid (AA) with 1,4-cyclohexanedimethanol (CHDM) and 1,3/1,4-CHDM, respectively, and used as the hydrophobic polyester blocks of these thermogelling copolymers to compare the effect of crystallinity on the sol-to-gel transition behavior.The polymers were characterized using proton nuclear magnetic resonance, Fourier transform infrared spectroscopy, gel permeation chromatography, differential scanning calorimetry, solubility testing, and rheological analysis. Experimental results revealed that the structure of the PCA block (crystalline vs. amorphous), the molecular weights of the hydrophobic PCA and hydrophilic PEG blocks, and the type of thermogelling polymer (triblock vs. multiblock) influenced the solubility, polymer micelle packing characteristics, maximum storage modulus, and sol-to-gel temperature of the polymers. Among all the samples at 40 wt.% aqueous solutions, triblock copolymer TB3 showed sol-to-gel temperature at 22 °C, and had the highest maximum storage modulus about 170 Pa.  相似文献

The surface of AISI 316 grade stainless steel (SS) was modified with a layer of poly(ethylene glycol) (PEG) (molecular weight 5000) with the aim of preventing protein adsorption and bacterial adhesion. Model SS substrates were first modified to introduce a very high density of reactive amine groups by the adsorption of branched poly(ethylenimine) (PEI) from water. Methoxy-terminated aldehyde-poly(ethylene glycol) (M-PEG-CHO) was then grafted onto the PEI layers using reductive amination at the lower critical solution temperature (LCST) of the PEG in order to optimize the graft density of the linear PEG chains. The chemical composition and uniformity of the surfaces were determined using X-ray photoelectron spectroscopy (XPS) and time-of-flight static secondary ion mass spectrometry (ToF-SSIMS) in the imaging mode. The effects of PEI concentration and different substrate pre-cleaning methods on the structure and stability of the final PEG layer was examined. Piranha solution proved to be the most effective method for removing adventitious hydrocarbon contamination, compared to cleaning with ultrasonication in organic solvents, and was the SS substrate that produced the most stable and thickest PEI layer. The surface density of PEI was shown to increase with increasing PEI concentration (up to 30 mg/ml), as determined from XPS measurements, and subsequently produced the PEG layer with the highest density of attached chains. In model experiments using β-lactoglobulin no protein adsorption was detected on the optimized PEG surface as determined by XPS and ToF-SSIMS analysis. However, neither the adhesion of a Gram-negative (Pseudomonas sp.) nor a Gram-positive (Listeria monocytogenes) bacterium was affected by the coating as equal numbers adhered to all surfaces tested. Our results show that preventing protein adsorption is not a prerequisite stopping bacterial adhesion, and that other mechanisms most likely play a role.  相似文献

Tensile drawing of PET via the mechanism of solvent crazing in adsorption-active media containing poly(ethylene glycol) with M < 1 × 106 is accompanied by their penetration into the porous structure of the matrix polymer. In this case, the amount of PEG in PET exceeds its concentration calculated on the assumption that the porous structure is filled with the polymer solution. In order to achieve a simple covalent hydrophilic polymer coating on poly(dimethylsiloxane) (PDMS) microfluidic chip, epoxy modified hydrophilic polymers were synthesized in aqueous solution with a persulfate radical initiation system, and crosslinked onto PDMS pretreated by oxygen plasma and silanized with 3-aminopropyl-triethoxysilanes (APTES). Glycidyl methacrylate (GMA) was copolymerized with acrylamide (poly(AAM-co-GMA)) or dimethylacrylamide (poly(DAM-co-GMA)), and graft polymerized with polyvinylpyrrolidone (PVP-g-GMA) or polyvinylalcohol (PVA-g-GMA). The epoxy groups in the polymers were determined by UV spectra after derivation with benzylamine. Reflection absorption infrared spectroscopy (RAIRS) confirmed covalent grafting of GMA-modified polymers onto PDMS surface. Electroosmotic flow (EOF) in the polymer grafted microchannel was strongly suppressed within the range pH 3-11. Surface adsorption of lysozyme and bovine serum albumin (BSA) was reduced to less than 10% relative to that on the native PDMS surface. On the GMA-modified polymer coated PDMS microchip, basic proteins, peptides, and sodium dodecyl sulfate (SDS) denatured proteins were separated successfully.  相似文献

We report a poly(ethylene glycol)‐poly(L ‐alanine)‐azobenzene‐poly(L ‐alanine)‐poly(ethylene glycol) (PEG‐PA‐Z‐PA‐PEG) as a temperature and light sensitive polymer. The poly(ethylene glycol)‐poly(L ‐alanine) diblock copolymers with a flexible‐rigid block structure were coupled by an azobenzene group that undergoes a reversible configurational change between “trans” and “cis” upon exposure to UV and vis light. The single azobenzene molecule embedded in the middle of a block copolymer with a flexible (shell)‐rigid (core) structure significantly affected molecular assembly, micelle size, polypeptide secondary structure, and sol‐to‐gel transition temperature of the polymer aqueous solution, depending on its exposure to UV or vis light. © 2012 Wiley Periodicals, Inc. Uniformly sized microspheres tangled with cup-stacked carbon nanofibers (CSCNFs) were directly synthesized by the pyrolysis of poly(ethylene glycol) (PEG) with a nickel catalyst. A PEG/Ni membrane was prepared on a silicon wafer surface by heating it to 750 °C at a heating rate of 15 °C min(-1). The wafer was heated to a temperature of 400 °C and was held at that temperature for 1 h before raising the temperature to 750 °C for 10 min to form the CSCNF microspheres. The final CSCNF microspheres and the intermediates were evaluated using scanning electron microscopy, transmission electron microscopy, X-ray diffractometry, and Raman spectroscopy to elucidate the growth mechanism. Furthermore, the CSCNF microspheres were successfully dispersed and maintained their spherical shape in an aqueous solution containing 0.5% Nafion. The CSCNF microspheres have the potential to work as a sophisticated carrier with high adsorption and fast electron-transfer exchange properties based on the graphene edges of the nanofiber surface.  相似文献

The fabrication of hydrogel microstructures based upon poly(ethylene glycol) diacrylates, dimethacrylates, and tetraacrylates patterned photolithographically on silicon or glass substrates is described. A silicon/silicon dioxide surface was treated with 3-(trichlorosilyl)propyl methacrylate to form a self-assembled monolayer (SAM) with pendant acrylate groups. The SAM presence on the surface was verified using ellipsometry and time-of-flight secondary ion mass spectrometry. A solution containing an acrylated or methacrylated poly(ethylene glycol) derivative and a photoinitiator (2,2-dimethoxy-2-phenylacetophenone) was spin-coated onto the treated substrate, exposed to 365 nm ultraviolet light through a photomask, and developed with either toluene, water, or supercritical CO2. As a result of this process, three-dimensional, cross-linked PEG hydrogel microstructures were immobilized on the surface. Diameters of cylindrical array members were varied from 600 to 7 micrometers by the use of different photomasks, while height varied from 3 to 12 micrometers, depending on the molecular weight of the PEG macromer. In the case of 7 micrometers diameter elements, as many as 400 elements were reproducibly generated in a 1 mm2 square pattern. The resultant hydrogel patterns were hydrated for as long as 3 weeks without delamination from the substrate. In addition, micropatterning of different molecular weights of PEG was demonstrated. Arrays of hydrogel disks containing an immobilized protein conjugated to a pH sensitive fluorophore were also prepared. The pH sensitivity of the gel-immobilized dye was similar to that in an aqueous buffer, and no leaching of the dye-labeled protein from the hydrogel microstructure was observed over a 1 week period. Changes in fluorescence were also observed for immobilized fluorophore labeled acetylcholine esterase upon the addition of acetyl acholine.  相似文献

We studied the dynamic surface tension of aqueous solutions of Triton X-100 and Triton X-405 by the maximum bubble pressure and the inclined plate methods in the lifetime range from 0.001 s up to 10 s. It is established that in the region of large and ultimately small surface pressure and time the adsorption follows diffusion kinetics, but in the region of intermediate values of lifetime and surface pressure both the surfactants decrease the surface tension faster than predicted by the existing diffusion theory. We offer a model that provides for the ability of poly(ethylene glycol) chains to adsorb on the water-air interface and to change the area that a molecule occupies on the surface. For this model we achieve full coincidence of the measured values and the values calculated according to the diffusion theory of the dynamic surface tension.

It is ascertained that the Triton X-405 molecule can exist in the surface layer in different states: with the poly(ethylene glycol) chain fully expanded or with it partially or fully submerged in solution. The first state is most probable at surface pressures less than 5 mN m−1, and the second is probable at a pressure of about 8–10 mN m−1. At pressures larger than 15–20 mN m−1, the poly(ethylene glycol) chain is fully submerged in the solution. The Triton X-100 molecule can also expand its poly(ethylene glycol) chain at low pressures and fully submerges it in the solution at higher values of the surface pressure.  相似文献

A poly(dimethylsiloxane) (PDMS) microfluidic chip surface was modified by multilayer-adsorbed and heat-immobilized poly(vinyl alcohol) (PVA) after oxygen plasma treatment. The reflection absorption infrared spectrum (RAIRS) showed that 88% hydrolyzed PVA adsorbed more strongly than 100% hydrolyzed one on the oxygen plasma-pretreated PDMS surface, and they all had little adsorption on original PDMS surface. Repeating the coating procedure three times was found to produce the most robust and effective coating. PVA coating converted the original PDMS surface from a hydrophobic one into a hydrophilic surface, and suppressed electroosmotic flow (EOF) in the range of pH 3-11. More than 1,000,000 plates/m and baseline resolution were obtained for separation of fluorescently labeled basic proteins (lysozyme, ribonuclease B). Fluorescently labeled acidic proteins (bovine serum albumin, beta-lactoglobulin) and fragments of dsDNA phiX174 RF/HaeIII were also separated satisfactorily in the three-layer 88% PVA-coated PDMS microchip. Good separation of basic proteins was obtained for about 70 consecutive runs.  相似文献

Poly(ethylene oxide) (PEO) and low molecular weight poly(ethylene glycol) (PEG) were covalently immobilized on silicon wafers and gold films by way of the CH insertion reaction of perfluorophenyl azides (PFPAs) by either photolysis or thermolysis. The immobilization does not require chemical derivatization of PEO or PEG, and polymers of different molecular weights were successfully attached to the substrate to give uniform films. Microarrays were also generated by printing polymer solutions on PFPA-functionalized wafer or Au slides followed by light activation. For low molecular weight PEG, the immobilization was highly dependent on the quality of the film deposited on the substrate. While the spin-coated and printed PEG showed poor immobilization efficiency, thermal treatment of the PEG melt on PFPA-functionalized surfaces resulted in excellent film quality, giving, for example, a grafting density of 9.2×10(-4)?(-2) and an average distance between grafted chains of 33? for PEG 20,000. The anti-fouling property of the films was evaluated by fluorescence microscopy and surface plasmon resonance imaging (SPRi). Low protein adsorption was observed on thermally-immobilized PEG whereas the photoimmobilized PEG showed increased protein adsorption. In addition, protein arrays were created using polystyrene (PS) and PEG based on the differential protein adsorption of the two polymers.  相似文献
