Abstract

Chiral oxazolidin‐2‐ones containing isopropylidene and cyclohexylidene functionalities, readily available from D‐mannitol, have been demonstrated to undergo highly diastereoselective β‐lactam synthesis via the Staudinger reaction with Mukaiyama reagent. Stereoselectivity for cis‐β‐lactam was the result of the [2?+?2] cycloaddition reaction of ketene to trans imines, and cyclohexylidene showed better yield and stereoselectivity than the isopropylidene auxiliary.  相似文献

The cycloaddition of ketenes and imines (Staudinger cycloaddition) is a general method for the synthesis of various β‐lactams. However, reactions of imines and ketenes with electron‐withdrawing substituents produce α,β‐unsaturated alkenamides, ring‐opening products of the intermediates generated from imines and the ketenes, even as sole products, besides the desired β‐lactams. The mechanism of the formation of α,β‐unsaturated alkenamides was investigated. The results indicate that the α,β‐unsaturated alkenamides are generated via a base‐induced C?C bond isomerization followed by electrocyclic ring opening of the formed azacyclobutenes (=1,2‐dihydroazetes; cf. An efficient and simple procedure for the synthesis of ketene imines via a one‐pot three‐component reaction of cyclohexyl isocyanide, diethyl acetylenedicarboxylate, and 1,4‐dihydropyridines under ultrasonic irradiation is described. The remarkable advantages are the simplicity of the experimental procedures, high yields, and short reaction times.  相似文献

An atroposelective synthesis of a new class of 3,3’‐bisindoles bearing axial and central chirality has been established via catalytic asymmetric addition reactions using isatin‐derived imines as electrophiles (23 examples, up to 80% yield, > 95 : 5 dr, 98 : 2 er). This approach takes advantage of chiral phosphoric acid‐catalyzed dynamic kinetic resolution of 2‐substituted 3,3’‐bisindoles via nucleophilic addition of such substrates with isatin‐derived imines. In this approach, isatin‐derived imines acted as a class of competent electrophiles due to their high reactivity and bulky size, which provided an easy access to axially chiral 3,3'‐bisindoles incorporated with a biologically important chiral 3‐aminooxindole unit. This approach has greatly expanded the generality and applicability of the strategy of dynamic kinetic resolution for the synthesis of enantioenriched 3,3’‐bisindole derivatives bearing both axial and central chirality.  相似文献

A series of novel 1,2,4‐oxadiazoline derivatives containing 2‐(1,2,4‐triazol‐1‐yl)quinoline were synthesized by the reaction of imines with benzohydroximinoyl chlorides in the presence of Et3N via 1,3‐diplolar cycloaddition reaction. The structures of the target compounds were confirmed by IR, 1H NMR, MS, elemental, and X‐ray crystallographic analysis.  相似文献

We report an efficient synthetic strategy to diverse hydantoins and thiohydantoins involving a three‐component reaction with the aid of deep eutectic solvent. Here, N,N′‐dimethyl urea and N,N′‐dimethyl thiourea play a dual role as reactant and reaction medium along with l ‐(+)‐tartaric acid. The three‐component reaction provides an easy access to 5‐amino‐1,3‐dialkyl‐substituted hydantoins and thiohydantoins in good yields.  相似文献

An effective and pragmatic strategy for the synthesis of structurally diverse indolo[2,3‐c]isoquinolin‐5‐ones has been developed via a Rh(III)‐catalyzed C?H activation and [4+2] annulation reaction of N‐methoxybenzamides and 3‐diazoindolin‐2‐imines. The reaction involves the efficient formation of two new (one C?C and one C?N) bonds under operationally simple conditions and has the benefits of a broad substrate scope.  相似文献

A highly stereoselective aza‐Henry reaction of α‐aryl nitromethanes with aromatic N‐Boc imines was established by using C1‐symmetric chiral ammonium betaine as a bifunctional organic base catalyst. Various substituted aryl groups for both imines and nitromethanes were tolerated in the reaction, and a series of precursors for the synthesis of unsymmetrical anti‐1,2‐diaryl ethylenediamines was provided.  相似文献

In the presence of 10 mol% lanthanide amide [(Me3Si)2N]3Ln(µ‐Cl)Li(THF)3, the aza‐Henry reaction of N‐tosyl imines with nitroalkanes (1:5 molar ratio) could be performed in good yields. The lanthanide amide‐catalyzed aza‐Henry reaction has the features of mild reaction conditions, tolerance of a variety of aromatic aldehyde‐derived imines and nitroalkanes, short time and good chemical yields. A catalytic mechanism for the reaction was also proposed.  相似文献

An anti‐selective Mannich reaction of aldehydes with N‐sulfonyl imines has been developed by using a 4‐hydroxypyrrolidine in combination with an external Brønsted acid. The catalyst design is based on three elements: the α‐substituent of the pyrrolidine, the 4‐hydroxy group, and the Brønsted acid, the combination of which is essential for high chemical and stereochemical efficiency. The reaction works with aromatic aldehyde‐derived imines, which have rarely been employed in previously reported enamine‐based anti‐Mannich reactions. Additionally, both N‐tosyl and N‐nosyl imines can be successfully used and the Mannich adducts can be easily reduced or oxidized, and after N‐deprotection the corresponding β‐amino acids and β‐amino alcohols can be obtained with good yields. The results also show that this ternary catalytic system may be practical in other enamine‐based reactions.  相似文献

A simple and convenient method for the direct, aminocatalytic, and highly enantioselective Mannich reactions of aldehydes with in situ generated N‐carbamoyl imines has been developed. Both α‐imino esters and aromatic imines serve as suitable electrophilic components. Moreover, the judicious selection of commercially available secondary amine catalysts allows selective access to the desired stereoisomer of the Ntert‐butoxycarbonyl (Boc) or N‐carbobenzyloxy (Cbz) Mannich adducts, with high control over the syn or anti relative configuration and almost perfect enantioselectivity. Besides the possibility to fully control the stereochemistry of the Mannich reaction, the main advantage of this method lies in the operational simplicity; the highly reactive N‐carbamate‐protected imines are generated in situ from stable and easily handled α‐amido sulfones.  相似文献

Unsymmetric cyclic ketenes were generated from N‐acyl‐1,3‐thiazolidine‐2‐carboxylic acids 1a – c by means of Mukaiyama's reagent, and then reacted with imines 2a – c to the new, isomeric spiro‐β‐lactams 3 and 4 via [2+2] cycloaddition (Staudinger ketene–imine reaction; Scheme 1). The reactions were stereoselective (Table 1) and mainly afforded the spiro‐β‐lactams with a relative trans configuration. The spiro‐β‐lactams could be transformed into the corresponding monocyclic β‐lactams by means of thiazolidine ring opening or into substituted thiazolidines via hydrolysis of the β‐lactam ring.  相似文献

A multicomponent double Mannich reaction of amines, aldehydes and ketones was efficiently catalyzed by molecular iodine, producing a series of 4‐piperidones in a stereoselective way. A variety of amines, aldehydes and ketones were tolerated in this tandem process, including those with labile functinal groups. Further investigation of the reaction between alkyl‐imines and ketones showed that imines from amines and ketones were formed in situ and isomerized to enamine in the presence of molecular iodine to accelerate the corresponding Mannich addition.  相似文献

In this review, the recent developments in catalytic asymmetric inverse‐electron‐demand hetero‐Diels−Alder reaction, which is recognized as one of the most powerful routes to construct highly functionalized and enantioenriched six‐membered heterocycles, are described. The article is organized on the basis of different kinds of electron‐deficient heterodienes, including α,β‐unsaturated ketones/aldehydes, o‐benzoquinones, α,β‐unsaturated imines, N‐aryl imines, o‐benzoqinone imides, and other aza‐olefins.  相似文献

The reaction of N,N′‐diarylselenoureas 16 with phenacyl bromide in EtOH under reflux, followed by treatment with NH3, gave N,3‐diaryl‐4‐phenyl‐1,3‐selenazol‐2(3H)‐imines 13 in high yields (Scheme 2). A reaction mechanism via formation of the corresponding Se‐(benzoylmethyl)isoselenoureas 18 and subsequent cyclocondensation is proposed (Scheme 3). The N,N′‐diarylselenoureas 16 were conveniently prepared by the reaction of aryl isoselenocyanates 15 with 4‐substituted anilines. The structures of 13a and 13c were established by X‐ray crystallography.  相似文献

A novel efficient method for the synthesis of earlier unknown 2-substituted analogs of ornithine and homolysine from substituted 5-aminopentyl- and 3-midopropylhydantoins (prepared from cyclic imines and amino and amido ketones) was developed. Primary and secondary amines can be rapidly and quantitatively oxidized to the corresponding imines by singlet oxygen. This reactive form of oxygen was produced using a variable‐temperature continuous‐flow LED‐photoreactor with a catalytic amount of tetraphenylporphyrin as the sensitizer. α‐Aminonitriles were obtained in good to excellent yields when trimethylsilyl cyanide served as an in situ imine trap. At 25°C, primary amines were found to undergo oxidative coupling prior to cyanide addition and yielded secondary α‐aminonitriles. Primary α‐aminonitriles were synthesized from the corresponding primary amines for the first time, by an oxidative Strecker reaction at –50 °C. This atom‐economic and protecting‐group‐free pathway provides a route to racemic amino acids, which was exemplified by the synthesis of tert‐leucine hydrochloride from neopentylamine.  相似文献

The photochemical reaction between three A5‐steroids (1–3) and a series of substituted 1,4‐benzoquinones and their mechanistic study were reported. The reaction in nitrogen atmosphere led to the formation of three products including the steroid‐quinone coupling compound (A), 7‐hydroxy derivatives of Δ5‐steroids (B) and substituted 1, 4‐hydroquinone (C). Both chemical and spectrometric evidences such as UV‐Visible spectra, ESR, chemically induced dynamic nuclear polarization (CIDNP) and cyclic voltammetry (CV) verified that the title reaction underwent a predominant photoinduced electron transfer pathway via the triplet quinone.  相似文献
