A simple, rapid and sensitive method for the determination of five estrogens, estrone, 17beta-estradiol, estriol, ethynyl estradiol, and diethylstilbestrol, was developed using a fully automated method consisting of in-tube solid-phase microextraction (SPME) coupled with liquid chromatography-tandem mass spectrometry (LC/MS/MS). These estrogens were separated within 8 min by HPLC using an XDB-C8 column and 0.01% ammonia/acetonitrile (60/40, v/v) at a flow rate of 0.2 mL/min. Electrospray ionization conditions in the negative ion mode were optimized for MS/MS detection of the estrogens. The optimum in-tube SPME conditions were 20 draw/eject cycles of 40 microL of sample using a Supel-Q PLOT capillary column as an extraction device. The extracted compounds were easily desorbed from the capillary by passage of the mobile phase, and no carryover was observed. Using the in-tube SPME LC/MS/MS method, good linearity of the calibration curve (r > or = 0.9996) was obtained in the concentration range from 10 to 200 pg/mL for all compounds examined. The limits of detection (S/N= 3) of the five estrogens examined ranged from 2.7 to 11.7 pg/mL. The in-tube SPME method showed 34-90-fold higher sensitivity than the direct injection method (5 microL injection). This method was applied successfully to the analysis of environmental water samples without any other pretreatment and interference peaks. Several surface water and wastewater samples were collected from the area around Asahi River, and estriol was detected at 35.7 pg/mL in the effluent of a sewage treatment plant. The recoveries of estrogens spiked into river waters were above 86%, except for estriol, and the relative standard deviations were below 0.9-8.8%.  相似文献

We developed a sensitive and useful method for the determination of five fluoroquinolones (FQs), enoxacin, ofloxacin, ciprofloxacin, norfloxacin, and lomefloxacin in environmental waters, using a fully automated method consisting of in-tube solid-phase microextraction (SPME) coupled with liquid chromatography-tandem mass spectrometry (LC/MS/MS). These compounds were analysed within 7 min by high-performance liquid chromatography (HPLC) using a CAPCELL PAK C8 column and aqueous ammonium formate (pH 3.0, 5 mM)/acetonitrile (85/15, v/v) at a flow rate of 0.2 mL/min. Electrospray ionization conditions in the positive ion mode were optimized for MS/MS detection. In order to optimize the extraction of FQs, several in-tube SPME parameters were examined. The optimum in-tube SPME conditions were 20 draw/eject cycles of 40 μL of sample at a flow-rate of 150 μL/min, using a Carboxen 1010 PLOT capillary column as an extraction device. The extracted compounds were easily desorbed from the capillary by passage of the mobile phase. Using the in-tube SPME LC/MS/MS method, good linearity of the calibration curve (r ≥ 0.997) was obtained in the concentration range from 0.1 to 10 ng/mL for all compounds examined. The limits of detection (S/N = 3) of the five FQs ranged from 7 to 29 pg/mL. The in-tube SPME method showed 60-94-fold higher sensitivity than the direct injection method (5 μL injection). This method was applied successfully to the analysis of environmental water samples without any other pretreatment and interference peaks. Several surface waters and wastewaters were collected from the area around Asahi River, and ofloxacin was detected in wastewater samples of a sewage treatment plant and other two hospitals at 17.5-186.2 pg/mL. The recoveries of FQs spiked into river water were above 81% for a 0.1 or 0.2 ng/mL spiking concentration, and the relative standard deviations were below 1.9-8.6%.  相似文献

The process of surfactant-assisted dispersive liquid-liquid microextraction (SA-DLLME) followed by high-performance liquid chromatography-UV detection was successfully applied for the extraction and determination of selected cannabinoids (cannabidiol, Δ(9)-tetrahydrocannabinol, and cannabinol) in urine samples. The effective parameters on the extraction efficiency were studied and optimized utilizing two different optimization methods: one variable at a time (OVAT) and face center design (FCD). Under the optimum conditions (extraction solvent and its volume, toluene, 85 μL; disperser agent and its concentration, 1.0 mL of ultra-pure water containing 0.5 mmol/L tetradecyl tremethyl ammonium bromide (TTAB); sample pH, 2.0 and salt concentration, 11% w/v NaCl), the limits of detection of the method were in the range of 0.1-0.5 μg/L and the repeatability and reproducibility of the proposed method, expressed as relative deviation, varied between 4.1 and 8.5% and 6.7 and 11.6%, respectively. Linearity was found to be in the range of 1.0-200 μg/L and under the optimum conditions, the preconcentration factors (PFs) were between 190 and 292. This proposed method was successfully applied in the analysis of three male advocate urine samples and good recoveries were obtained.  相似文献

An optimised method using stir bar sorptive extraction (SBSE) and a thermal desorption‐GC‐electron capture detector (GC‐ECD) for the determination of short‐chain chlorinated paraffins from water samples was developed. Recoveries near to 100% were obtained by using 20 mm×0.5 mm (length×film thickness) PDMS commercial stir bars from 200 mL spiked water samples and 20% methanol addition with an extraction period of 24 h. Method sensitivity, linearity and precision were evaluated for surface water and wastewater spiked samples. A LOD of 0.03 and 0.04 μg/L was calculated for surface and wastewater, respectively. The precision of the method given as an RSD was below 20% for both matrices. The developed method was applied for the analysis of two real samples from a contaminated river and a wastewater treatment plant. Results were in accordance with those obtained using a previously developed method based on solid phase microextraction (SPME).  相似文献

This paper presents the development of a procedure, which enables the analysis of nine pharmaceutical drugs in wastewater using gas chromatography‐mass spectrometry (GC‐MS) associated with solid‐phase microextraction (SPME) for the sample preparation. Experimental design was applied to optimize the in situ derivatization and the SPME extraction conditions. Ethyl chloroformate (ECF) was employed as derivatizing agent and polydimethylsiloxane‐divinylbenzene (PDMS‐DVB) as the SPME fiber coating. A fractional factorial design was used to evaluate the main factors for the in situ derivatization and SPME extraction. Thereafter, a Doehlert matrix design was applied to find out the best experimental conditions. The method presented a linear range from 0.5 to 10 μg/L, and the intraday and interday precision were lower than 16%. Applicability of the method was verified from real influent and effluent samples of a wastewater treatment plant, as well as from samples of an industry wastewater and a river.  相似文献

A method was developed for viable and rapid determination of seven polychlorinated biphenyls (PCBs) in water samples with vortex-assisted liquid-liquid microextraction (VALLME) using gas chromatography-mass spectrometry (GC-MS). At first, the most suitable extraction solvent and extraction solvent volume were determined. Later, the parameters affecting the extraction efficiency such as vortex extraction time, rotational speed of the vortex, and ionic strength of the sample were optimized by using a 2(3) factorial experimental design. The optimized extraction conditions for 5 mL water sample were as follows: extractant solvent 200 μL of chloroform; vortex extraction time of 2 min at 3000 rpm; centrifugation 5 min at 4000 rpm, and no ionic strength. Under the optimum condition, limits of detection (LOD) ranged from 0.36 to 0.73 ng/L. Mean recoveries of PCBs from fortified water samples are 96% for three different fortification levels and RSDs of the recoveries are below 5%. The developed procedure was successfully applied to the determination of PCBs in real water and wastewater samples such as tap, well, surface, bottled waters, and municipal, treated municipal, and industrial wastewaters. The performance of the proposed method was compared with traditional liquid-liquid extraction (LLE) of real water samples and the results show that efficiency of proposed method is comparable to the LLE. However, the proposed method offers several advantages, i.e. reducing sample requirement for measurement of target compounds, less solvent consumption, and reducing the costs associated with solvent purchase and waste disposal. It is also viable, rapid, and easy to use for the analyses of PCBs in water samples by using GC-MS.  相似文献

The feasibility of solid-phase microextraction (SPME) for the determination of several organophosphorus flame retardants and plastizicers in water samples by gas chromatography-nitrogen phosphorous detection (GC-NPD) is evaluated. These compounds have a wide range of polarities and volatilities and require a thorough optimisation of the different SPME parameters. Considering also possible contamination and carryover sources, the best compromise microextraction conditions were found to be direct extraction of 22 ml samples, containing 300 mg/ml of NaCl, with a PDMS-DVB coated fibre at room temperature. Although equilibrium was not achieved, an extraction time of 40 min allowed obtaining a good sensitivity (quantification limits between 0.010 and 0.025 ng/ml), comparable to that achieved by solid-phase extraction (SPE) of 1l samples, producing both similar values of precision and accuracy. Furthermore, the SPME method has shown to be free of matrix effects, avoiding the need of employing the standard addition procedure for quantification, and was suitable for the determination of eight of the nine considered compounds. Only tris-(2-ethylhexyl)-phosphate was neither determinable by SPME nor by SPE. Finally, the application of the developed methodology to the analysis of wastewater samples, showed that important concentrations of these compounds (up to 10 ng/ml) have been detected in treated sewage water, being discharged into the aquatic environment.  相似文献

A sensitive and solvent-free procedure for the determination of non-steroidal acidic anti-inflammatory drugs in water samples was optimized using solid-phase microextraction (SPME) followed by on-fiber silylation of the acidic compounds and gas chromatography-mass spectrometry (GC-MS) determination. Microextraction was carried out directly over the filtered water samples using a polyacrylate fiber. Derivatization was performed placing the SPME fiber, loaded with the extracted analytes, in the headspace of a vial containing 50 microl of N-methyl-N-(tert-butyldimethylsilyl)-trifluoroacetamide (MTBSTFA). Derivatives were desorbed for 3 min in the GC injector. Influence of several parameters in the efficiency of microextraction (volume of sample, time, pH, type of fiber coating, etc.) and derivatization steps (time, temperature and volume of MTBSTFA) was systematically investigated. In the optimal conditions an excellent linearity over three orders of magnitude and quantification limits at the ng/l level (from 12 to 40 ng/l) were achieved. The proposed method was applied to the determination of acidic compounds in sewage water and results compared to those obtained using solid-phase extraction (SPE) followed by the derivatization of the compounds in the organic extract of the solid-phase extraction cartridge.  相似文献

A solid-phase micro-extraction (SPME) method using an SPME fiber device and graphite furnace (GF) for extracting Se compounds was proposed. Various factors affecting the derivatization and extraction of Se(IV) by SPME-GF were evaluated, including the effect of acid (type and concentration), the concentration of the derivatizing agent, the derivatization temperature, the extraction and derivatization times and the extraction temperature. After optimizing these conditions, the quantification of Se(IV) was performed by Gas Chromatography-Mass Spectrometry (GC-MS). The limit of detection was 0.37 μg L(-1) for Se(IV). The method was successfully applied to the total Se determination in certified reference materials (BCR-414 and SRM 1643e). A recovery of 97% was obtained for water (SRM 1643e). After microwave oven decomposition and the reduction of selenium using a mixture of 2 mol L(-1) HCl and 1% (w/v) KBr, a recovery of 101% and a relative standard deviation of 3.5% were attained for plankton (BCR-414). The SPME-GF method combined with GC-MS was also applied to the determination of the total selenium in a drug sample (selenium chelate).  相似文献

Volatile organic compounds (VOCs) are toxic compounds in the air, water and land. In the proposed method, ultrasound-assisted emulsification microextraction (USAEME) combined with gas chromatography-mass spectrometry (GC-MS) has been developed for the extraction and determination of eight VOCs in water samples. The influence of each experimental parameter of this method (the type of extraction solvent, volume of extraction solvent, salt addition, sonication time and extraction temperature) was optimized. The procedure for USAEME was as follows: 15 μL of 1-bromooctane was used as the extraction solvent; 10 mL sample solution in a centrifuge tube with a cover was then placed in an ultrasonic water bath for 3 min. After centrifugation, 2 μL of the settled 1-bromooctane extract was injected into the GC-MS for further analysis. The optimized results indicated that the linear range is 0.1-100.0 μg/L and the limits of detection (LODs) are 0.033-0.092 μg/L for the eight analytes. The relative standard deviations (RSD), enrichment factors (EFs) and relative recoveries (RR) of the method when used on lake water samples were 2.8-9.5, 96-284 and 83-110%. The performance of the proposed method was gauged by analyzing samples of tap water, lake water and river water samples.  相似文献

This paper proposes a new analytical procedure based on the headspace solid‐phase microextraction (HS‐SPME) technique and gas chromatography‐selected ion monitoring‐mass spectrometry (GC‐SIM‐MS) for the determination of 16 phenols extracted from leather samples. The optimized conditions for the HS‐SPME were obtained through two experimental designs – a two‐level fractional factorial design followed by a central composite design – using the commercial SPME fiber polyacrylate 85 μm (PA). The best extraction conditions were as follows: 200 μL of derivatizing agent (acetic anhydride), 20 mL of saturated aqueous NaCl solution and extraction time and temperature of 50 min and 75°C, respectively. All optimized conditions were obtained with fixed leather sample mass (250 mg), vial volume (40 mL) and phosphate buffer pH (12) and concentration (50 mmol/L). Detection limits ranging from 0.03 to 0.20 ng/g, and relative standard deviation (RSD) lower than 10.23% (n=6) for a concentration of 800 ng/g (chlorophenols) and 1325 ng/g (2‐phenylphenol) in the splitless mode were obtained. The recovery was studied at three concentration levels by adding different amounts of phenols to the leather sample and excellent recoveries ranging from 90.0 to 107.2% were obtained. The validated method was shown to be suitable for the quantification of phenols in leather samples, as it is simple, relatively fast and sensitive.  相似文献

Graphene is a novel and interesting carbon material that could be used for the separation and purification of some chemical compounds. In this investigation, graphene was used as a novel fiber‐coating material for the solid‐phase microextraction (SPME) of four triazine herbicides (atrazine, prometon, ametryn and prometryn) in water samples. The main parameters that affect the extraction and desorption efficiencies, such as the extraction time, stirring rate, salt addition, desorption solvent and desorption time, were investigated and optimized. The optimized SPME by graphene‐coated fiber coupled with high‐performance liquid chromatography‐diode array detection (HPLC‐DAD) was successfully applied for the determination of the four triazine herbicides in water samples. The linearity of the method was in the range from 0.5 to 200 ng/mL, with the correlation coefficients (r) ranging from 0.9989 to 0.9998. The limits of detection of the method were 0.05‐0.2 ng/mL. The relative standard deviations varied from 3.5 to 4.9% (n=5). The recoveries of the triazine herbicides from water samples at spiking levels of 20.0 and 50.0 ng/mL were in the range between 86.0 and 94.6%. Compared with two commercial fibers (CW/TPR, 50 μm; PDMS/DVB, 60 μm), the graphene‐coated fiber showed higher extraction efficiency.  相似文献

A sensitive procedure for the determination of three UV filters: ethylhexyl salicylate (EHS), 3,3,5-trimethylcyclohexyl salicylate (Homosalate, HMS), 2-hydroxy-4-methoxybenzophenone (BP-3) and two related hydroxylated benzophenones (2,4-dihydroxybenzophenone, BP-1 and 2,2′-dihydroxy-4-methoxybenzophenone, BP-8) in water samples is presented. Analytes were first concentrated on the coating of a solid-phase microextraction (SPME) fibre, on-fibre silylated and then determined using gas chromatography combined with tandem mass spectrometry (GC-MS/MS). Factors affecting the performance of extraction and derivatization steps are thoroughly evaluated and their effects on the yield of the sample preparation discussed. Under final working conditions, a PDMS-DVB coated SPME fibre was exposed directly to 10 mL of water, adjusted at pH 3, for 30 min. After that, the fibre was placed in the headspace (HS) of a 1.5 mL vial containing 20 μL of N-methyl-N-(trimethylsilyl)-trifluoroacetamide (MSTFA). On-fibre silylation of hydroxyl groups contained in the structure of target compounds was performed at 45 °C for 10 min. The whole sample preparation process was completed in 40 min, providing limits of quantification from 0.5 to 10 ng L−1 and acceptable precision (RSDs under 13%) for samples spiked at different concentrations. All compounds could be accurately determined in river and treated wastewater (relative recoveries from 89 to 115%) using standards in ultrapure water, whereas standard addition is recommended to quantify their levels in untreated wastewater. Analysis of wastewater revealed the systematic presence of BP-3 and BP-1 in raw samples with maximum concentrations close to 500 and 250 ng L−1, respectively.  相似文献

A solid-phase microextraction (SPME) method for the determination of triclosan, methyl triclosan, 2,4-dichlorophenol and 2,3,4-trichlorophenol (considered as possible triclosan metabolites) in water samples was optimised. Analytes were first concentrated on a SPME fibre, directly exposed to the sample, and then triclosan and the two chlorinated phenols on-fibre silylated using N-methyl-N-(tert.-butyldimethylsilyl)-trifluoroacetamide (MTBSTFA). Methyl triclosan remained unaffected during the derivatization step. Compounds were determined using gas chromatography in combination with mass spectrometry (GC-MS). Influence of different factors on the efficiency of extraction and derivatization steps was systematically investigated. Using a polyacrylate (PA) fibre quantification limits below 10 ng/l, and acceptable relative standard deviations, were obtained for all compounds after an extraction time of 30 min. On-fibre silylation was carried out in only 10 min. Moreover, the efficiency of the procedure was scarcely affected by the type of water sample. The method was applied to several samples of treated and raw wastewater, triclosan was found in all samples, at concentrations from 120 to 14,000 ng/l, and 2,4-dichlorophenol in most of them, at levels up to 2222 ng/l.  相似文献

A solid‐phase extraction (SPE) method was developed to extract 14 pesticides simultaneously from environment samples using cigarette filter as the sorbent before gas chromatography‐mass spectrometry (GC‐MS) analysis. Parameters influencing the extraction efficiency, such as the sample loading flow rate, eluent and elution volume, were optimized. The optimum sample loading rate was 3 mL/min, and the retained compounds were eluted with 6 mL of eluent at 1 mL/min under vacuum. Good linearity was obtained for all the 14 pesticides (r2>0.99) from 0.1 to 20 μg/L for water and from 2 to 400 μg/kg for soil samples. The detection limits (signal‐to‐noise=3) of the proposed method ranged from 0.01 to 0.20 μg/L for water samples and from 0.42 to 6.95 μg/kg for soil samples. The developed method was successfully applied for determination of the analytes in real environmental samples, and the mean recoveries ranged from 76.4 to 103.7% for water samples and from 79.9 to 105.3% for soil samples with the precisions (relative standard deviation) between 2.0 and 13.6%.  相似文献

A sensitive and efficient solid-phase microextraction (SPME) method for the determination of organophosphorous (OPPs) and pyrethroid pesticides (Pyrs) in aquaculture-seawater samples by using GC with MS/MS (GC-MS/MS) was developed. Dichlorvos and chlorpyrifos (OPPs); permethrin, alpha-cypermethrin and deltamethrin (Pyrs) were selected according to their use as chemotherapeutants in the aquaculture industry. Different parameters affecting extraction efficiency such as fibre coating, agitation, pH and extraction time profiles were investigated. An experimental central composite design (alpha = 1) and desirability functions were used for the simultaneous optimization of extraction temperature and sample volume. Finally, a method based on direct SPME in 40 min at 75 degrees C using 100-microm-thick poly(dimethyl)siloxane (PDMS) fibre and 20 mL of sample volume is proposed. The method was validated, exhibiting good linearity, precision and accuracy parameters with picogram per millilitre LODs. The proposed methodology was applied to determine the ultratrace levels of OPPs and Pyrs in aquaculture-seawater samples by the standard addition approach, which proved to be reliable and sensitive, in addition to requiring only small amounts of sample.  相似文献

A solid-phase microextraction (SPME)-gas chromatography (GC)-mass spectrometry (MS) analytical method for the simultaneous separation and determination of 16 polycyclic aromatic hydrocarbons (PAHs) from aqueous samples has been developed, based on the sorption of target analytes on a selectively sorptive fibre and subsequent desorption of analytes directly into GC-MS. The influence of various parameters on PAH extraction efficiency by SPME was thoroughly studied. Results show that the fibre exposure time and the use of agitation during exposure are critical in enhancing SPME performance. The presence of colloidal organic matter (as simulated by humic acid) in water samples is shown to significantly reduce the extraction efficiency, suggesting that SPME primarily extracts the truly dissolved compounds. This offers the significant advantage of allowing the differentiation between freely available dissolved compounds and those associated with humic material and potentially biologically unavailable. The method showed good linearity up to 10 μg/l. The reproducibility of the measurements expressed as relative standard deviation (R.S.D.) was generally <20%. The method developed was then applied to extract PAHs from sediment porewater samples collected from the Mersey Estuary, UK. Total PAH concentrations in porewater were found to vary between 95 and 742 ng/l with two to four ring PAHs predominating. Results suggest that SPME has the potential to accurately determine the dissolved concentrations of PAHs in sediment porewater.  相似文献
