In an extensive search for bioactive compounds from plant sources, the quantitative and qualitative characterisation of the compounds present in Cynoglossum cheirifolium extracts was studied. Total phenolic and flavonoid contents were determined by spectrophotometric techniques. In vitro antioxidant and radical scavenging profiling was determined through DPPH? scavenging activity and Ferric reducing antioxidant power (FRAP). Our study showed that leaves produce more phenolic compounds, followed by flowering aerial part. The butanolic fraction obtained from leaves extract exhibited the highest total phenolics (78.65 ± 3.58 mg GAE/g DW) and flavonoids (22.15 ± 4.66 mg CE/g DW). In contrast, this fraction displayed the highest DPPH? scavenging ability with IC50 values of 0.06 ± 0.003 mg/mL. The RP-HPLC-PDA analysis of phenolic compounds from leaves of C. cheirifolium lets to identify: rosmarinic acid, ferulic acid, caffeic acid, p-coumaric acid, syringic acid, sinapic acid and rutin. The obtained results indicate that this plant represent a valuable source of natural antioxidants.  相似文献

Presented work summarizes the data about polyphenolic profiles in various plant parts (leaves, shoots, berries) of sea buckthorn (Hippophaë rhamnoides L.) during the annual growth cycle. A reversed-phase high performance liquid chromatography method (RP-HPLC) coupled with diode-array detection (DAD) was optimized for determination of catechin, epicatechin, gallic acid, p-coumaric acid, caffeic acid, ferulic acid, rutin (quercetin 3-rutinoside) and quercitrin (quercetin 3-rhamnoside). The content of these polyphenolic compounds was monitored in extracts of sea buckthorn plant samples from April to October. The total antioxidant activity was determined using scavenging of 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonate) cation radical (ABTS·+) and 1,1-diphenyl-2-picrylhydrazyl radical (DPPH·). The total content of polyphenols was estimated by conventional spectrophotometric method using Folin-Ciocalteu reagent. The monitoring of temporal changes of selected polyphenolic compounds by RP-HPLC showed that catechin, epicatechin and gallic acid were the most abundant analytes in annual green shoots and leaves, and their content varied significantly during the studied period.   相似文献

A method for the determination of rutin was developed from in an O/W emulsion with decyl oleate and Ceteareth-20 containing Ginkgo biloba L., Hedera helix L., and Thymus vulgaris L. extracts. Total quantification of rutin was performed by determination in the aqueous and oily phase. Rutin was extracted from the oily phase by ultrasonication for 30 min at 37°C in methanol and resolved by liquid chromatography on a column containing a Gemini 3 µ C18 110A stationary phase.

Chromatography was carried out using gradient elution with the mobile phase composed of water:formic acid and water:acetonitrile:formic acid, which was varied from 100:0.1 to 50:50:0.1 (v/v/v) over 80 min and delivered at a flow rate of 0.3 ml/min. The analytes were quantified using a UV-VIS absorption spectrometer at a wavelength of 254 nm and a single quadrupole mass spectrometer employing an ESI interface operated in the positive ion mode with single ion monitoring at m/z = 609.14.

Finally, a simple and rapid method for the extraction and determination of rutin in hydrophilic and lipophilic phases of an O/W emulsion with good precision and acceptable recovery was developed. This method might be of special importance for the analysis of rutin and other flavonoids in O/W or W/O emulsion matrices.  相似文献

The chromatographic conditions for the separation of a complex set of flavonoids (aglycones and glycosides) by micellar liquid chromatography with spectrophotometric detection were optimized. A good separation for all analytes was obtained and satisfactory peak shapes were achieved by isocratic elution with Ultrasphere ODS column (250 mm × 4.6 mm, 5 μm). The optimal mobile phase range for flavonoids separation is: SDS concentration between 0.014 and 0.018 mol L?1 and 1-propanol volume fraction between 2.2 and 4.5% (v/v) in a diluted (1:5) phosphate buffer solution pH 6.86. The flavonoids (robinin, rutin, hyperoside, quercitrin, liquroside, luteolin-7O-glucoside, apigenin-7O-glucoside, isosalipurposide, myricetin, fisetin, luteolin, apigenin, quercetin and caempferol) were successfully separated within 40 min with isocratic elution. The developed method is an alternative to reversed-phase LC in the assay of flavonoids in plants, plant extracts and plant extract containing drugs.  相似文献

A simple and fast method was developed for simultaneous quantitative determination of three biologically active phenolic compounds i.e. quercetin, rutin and coumaric acid in flowers of Rhododendron arboreum using high-performance thin-layer chromatography (HPTLC). The separation was performed on TLC aluminium plates precoated with silica gel RP-18 F254S. Good separation was achieved in the mobile phase of methanol-water-formic acid (40:57:3, v/v/v) and densitometric determination of these compounds was carried out at 280 nm in reflectance/absorbance mode. The linear regression data for the calibration plots showed a good linear relationship with r = 0.9971, 0.9953 and 0.9960 for quercetin, rutin and coumaric acid, respectively. Accuracy of the method was checked by recovery study conducted at two different levels with the average recovery of 99.90%, 99.02% and 99.16% for quercetin, rutin and coumaric acid, respectively. The present method is being reported first time and may be used for routine quality control of the flowers of R. arboreum.  相似文献

Following optimization of extraction, separation, and analytical conditions, a simple, rapid, and sensitive HPLC-UV method was developed for the simultaneous determination of seven major bioactive components in Sambucus chinensis Lindl, including chlorogenic acid, caffeic acid, p-coumaric acid, ferulic acid, kaempferol-3-O-β-D-galactopyranoside, kaempferol-3-O-β-D-glucopyran-oside, and kaempferol-3-O-(6-actyl)-β-D-galacto-pyranoside. The good chromatographic separation was performed on a Gemini C18 reversed-phase analytical column (250 mm × 4.6 mm, 5 μm) by gradient elution with acetonitrile and formate aqueous buffer (containing 0.8% formic acid, V/V) at a flow rate of 1.0 mL/min. The detection wavelength was set at 326 nm. The intra-day and inter-day precisions were evaluated with the R.S.D. values less than 4.0%. The mean recoveries of the seven compounds were in the range of 92.4%–104.8%. The method was successfully applied to determine the seven bioactive compounds in six different origins of Sambucus chinensis Lindl samples, and there was a significant variation in the contents of the seven compounds among the six samples. Therefore, this method provided a new basis of overall assessment for routine use in the quality control of Sambucus chinensis Lindl.  相似文献

An HPLC procedure is proposed for the determination of 12 phenolic compounds in plant tissues by reversed-phase HPLC with gradient elution and UV detection. The influence of pH, composition of the mobile phase, concentration of the organic modifier, and temperature on the separation of gallic, protocatechuic, trans-ferulic, trans-caffeic acid, rutin, quercetin, dihydroquercetin, and (_)-epicatechin for 30 min is studied. The lower limit of quantification of phenolic compounds is 1–2.5 μg/L. The procedure was applied to the determination of phenolic compounds in aqueous extracts of Hypericum perforatum; its sample was found to contain protocatechuic acid, (_)-epicatechin, and also rutin.  相似文献

A simple, accurate and reproducible capillary electrophoresis method with UV detection has been developed for the simultaneous determination of four iridoid glycosides, 6-O-methyl-catalpol, aucubin, harpagide, and harpagoside, in Scrophularia ningpoensis (Xuan-shen). The running buffer was 100 mM borate (pH 9.3) containing 20% methanol. Applied voltage was 20 kV and temperature was 25 °C. Diphylloside A was used as an internal standard (IS) and detection was at 200 nm. The effects on separation of buffer pH, buffer concentration, and organic modifiers were investigated. The extracts of S. ningpoensis were well separated within 45 min.

A reverse phase liquid chromatography method with diode array detection was developed to evaluate the quality of Cirsium setosum through establishing chromatographic fingerprint and simultaneous determination of six phenolic compounds, namely chlorogenic acid, caffeic acid, rutin, linarin, luteolin and apigenin. The chromatographic separation was performed on an Agilent SB-C18 column (250 × 4.6 mm, 5.0 μm) with a gradient elution program using a mixture of acetonitrile and 0.5% aqueous acetic acid (v/v) as mobile phase within 25 min at 326 nm wavelength. The correlation coefficients of similarity were determined from the LC fingerprints, and they shared a close similarity. The LC with electrospray ionization mass spectrometry experiment was performed to further confirm the identity of phenolic compounds. The six phenolic compounds showed good regression (R 2 > 0.9995) within test ranges and the recovery of the method was in the range of 95.8–102.8%. In addition, the content of those six phenolic compounds in C. setosum growing in different locations of China was determined to establish the effectiveness of the method. The results indicated that the developed method by having a combination of chromatographic fingerprint and quantification analysis could be readily utilized as a quality control method for C. setosum and its related traditional Chinese medicinal preparations.  相似文献

The objective of this study was to set up a method to detect five compounds in fresh smashed apples by HPLC/DAD simultaneously. Different methods have been tested to control browning and ascorbic acid with ultrasonication was adopted. Methanol–water–acetic acid (30:69:1, v/v) containing 2.0 g of ascorbic acid L?1 was chosen as the extract solvent. The method effectively simplified the sample treatment compared with the traditional ways. And primarily, the results were used to identify between different varieties. The chromatographic separation was performed on an Atlantis C18 (250 mm × 4.5 mm, particle size 5 μm) with a gradient elution program using a mixture of acetonitrile and 2% aqueous acetic acid (v/v) as mobile phase within 20 min at 270 nm wavelength. The variation of the content of five compounds was gallic acid (ND ~1.81 μg g?1), protocatechuic acid (ND ~1.79 μg g?1), chlorogenic acid (13.81–189.4 μg g?1), caffeic acid (6.82–45.02 μg g?1) and rutin (0.96–18.55 μg g?1). The results could successfully be used to discriminate between different apple varieties (Gala, Fuji, Delicious, 8th Apple US, Golden Apple, Green Apple and Red Rose); chlorogenic acid and rutin being the polyphenols that contribute most to the differentiation.  相似文献

A micellar electrokinetic capillary chromatography (MEKC) method, using UV detection, was developed for the determination of polyphenols in Toona sinensis (Meliaceae); the procedure involved precipitation of polyphenols from the leaves of T. sinensis using methanol. The structures can be established with fifteen compounds including methyl gallate, gallic acid, kaempferol, quercitin, quercitrin, rutin, kaempferol‐glucoside, catechin, epicatechin, stearic acid, palmitic acid, β‐sitosterol, stigmasterol, β‐sitosteryl‐glucoside, and stigmasteryl‐glucoside by spectroscopic analysis. However, there has been no investigation to quantitate the polyphenols that form T. sinensis. Thus, seven polyphenols of T. sinensis with UV absorbance, catechin (C), epicatechin (EC), methyl gallate (MG), rutin (R), gallic acid (G), quercitrin (Q), and kaempferol (K) were separated within 10 min with a 40 cm uncoated fused‐silica column, with the RSD < 3% (migration times), voltage at 15 kV using this method. On‐column detection was carried out at 254 nm. The detection limit of this method for all analytes ranged from 19.5 to 0.02 μM (RSD < 3.1%). The method provided a rapid and sensitive identification of polyphenols of interest in T. sinensis and is suitable for biological activity studies.  相似文献

Acetone extract of faba bean (Vicia faba L.) was found to be highest total phenol and flavonoid content among all extracts. Antioxidant activity for inhibition percentage (free radical scavenging activity) had 86.47% for acetone extract, and 97.36% for ascorbic acid respectively. IC50 value of ascorbic acid and acetone extact were found to be 9 μg/mL ± 0.20 and 30 μg/mL ± 0.21. Faba bean seeds had catechin, epicatechin, gallic acid and ellagic acid which on molecular docking study revealed that it binds effectively with xanthine oxidase by binding energy of –7.78, –6.11, –6.39, –5.78 kcal/mol respectively compared to allopurinol drug having binding energy of –4.94 kcal/mol. Gallic acid, ellagic acid, catechin, epicatechin (polyphenols) and allopurinol bind other than catalytic residues (Glu-1261) of xanthine oxidase. In vitro and in silico analysis recommended that mode of enzyme inhibition was mixed type.  相似文献

UHPLC-MS/MS method using BEH C18 analytical column was developed for the separation and quantitation of 12 phenolic compounds of Chamomile (Matricaria recutita L.). The separation was accomplished using gradient elution with mobile phase consisting of methanol and formic acid 0.1%. ESI in both positive and negative ion mode was optimized with the aim to reach high sensitivity and selectivity for quantitation using SRM experiment. ESI in negative ion mode was found to be more convenient for quantitative analysis of all phenolics except of chlorogenic acid and kaempherol, which demonstrated better results of linearity, accuracy and precision in ESI positive ion mode. The results of method validation confirmed, that developed UHPLC-MS/MS method was convenient and reliable for the determination of phenolic compounds in Chamomile extracts with linearity >0.9982, accuracy within 76.7-126.7% and precision within 2.2-12.7% at three spiked concentration levels. Method sensitivity expressed as LOQ was typically 5-20 nmol/l.Extracts of Chamomile flowers and Chamomile tea were subjected to UHPLC-MS/MS analysis. The most abundant phenolic compounds in both Chamomile flowers and Chamomile tea extracts were chlorogenic acid, umbelliferone, apigenin and apigenin-7-glucoside. In Chamomile tea extracts there was greater abundance of flavonoid glycosides such as rutin or quercitrin, while the aglycone apigenin and its glycoside were present in lower amount.  相似文献

Parthenium hysterophorus L., is an obnoxious weed known for its environmental health hazards and medicinal uses. These characteristics are due to presence of sesquiterpene lactones and organic acids; therefore a rapid and sensitive analytical procedure using HPLC-PDA-MS-MS was developed and optimized for separation, identification, and quantification of parthenin and six organic acids. Separation and characterization of compounds was achieved on a RP-C18 column with 1% acetic acid in water (A) and acetonitrile (B) as a mobile phase at a flow rate of 0.6 mL min?1 and by matching their UV and mass spectra with reference compounds. Six organic acids (ferulic acid, 0.1 mg g?1 to coumaric acid, 13.6 mg g?1) and parthenin (27.4 mg g?1) were characterized within 26 minutes of chromatographic separation in plant extract. The calibration curves are linear with correlation coefficients from 0.985 to 0.998, limit of detection and quantification ranged between 1.0 µg mL?1 (anisic acid) to 2.2 µg mL?1 (parthenin) and 2.5 µg mL?1 (coumaric acid) to 5.2 µ g mL?1 (parthenin) and recovery ranged between 90.9% to 97.3%. To the best of our knowledge this is the first report for the simultaneous separation of parthenin and organic acids. The method is applicable for screening of commercial crops, medicinal plants, and their products which might be mixed with P. hysterophorus during harvesting period.  相似文献

Abstract

The concentration and antioxidant activity of the known phenolics macluraxanthone, rutin, chlorogenic acid, and psoralen present in M. pomifera, F. carica, and F. elastica extracts were determined to evaluate the plant extracts potency as new antioxidant suppliers.

The quantification of the compounds was obtained by HPLC-DAD. The antioxidant activity of plant extracts and compounds was evaluated using DPPH and ABTS methods. The results seem to indicate that these extracts can be promising as new antioxidant suppliers, particularly F. carica and F. elastica leaves because only about 30% and about 50%, respectively, of the total activity was due to the phenolics determined.  相似文献

In this study the antioxidant and cytotoxicity activity of the Adonidia merrillii fruits were investigated using different solvent polarities (methanol, ethyl acetate and water). The results showed that the total phenolic and flavonoid contents of the methanolic extract was higher compare with other extract with respective values of 17.80 ± 0.45 mg gallic acid equivalents/g dry weight (DW) and 5.43 ± 0.33 mg rutin equivalents/g DW. Beside that The RP-HPLC analyses indicated the presence of gallic acid, pyrogallol, caffeic acid, vanillic acid, syringic acid, naringin and rutin. In the DPPH, NO2 and ABTS scavenging assays, the methanolic extract exhibited higher antioxidant activity as compared to the ethyl acetate and water extracts. The extracts exhibited moderate to weak cytotoxic activity in the assays using human hepatocytes (Chang liver cells) and NIH/3T3 (fibroblasts cell) cell lines. The findings showed the Adonidia merrillii fruit extracts to possess considerable antioxidant and cytotoxicity properties. The fruit, therefore, is a potential candidate for further work to discover antioxidant and cytotoxic drugs from natural sources.  相似文献

The antioxidant activity of buckwheat and barley extracts by reaction with 1,1-diphenyl-2-picrylhydrazyl and the total of phenolic compounds have been determined using the Folin-Ciocalteu reagent. It has been found that water-ethanol extracts of buckwheat are characterized by higher antioxidant activity (6.2 ± 0.5 μM-eq. of Trolox/g) and concentration of phenolic compounds (4.41 ± 0.07 mg-eq. of rutin/g) compared to barley extracts (4.2 ± 0.3 μM-eq. of Trolox/g and 2.4 ± 0.1 mg-eq. of rutin/g, respectively). A series of phenolic compounds have been identified by HPLC with UV detection and mass spectrometric detection with electrospray ionization. The main phenolic compounds-antioxidants in buckwheat extracts are rutin, catechin and epicatechin, 1-O-caffeoyl-O-rutinoside (m/z 487), and epicatechin-O-3,4-dimethylgallate (m/z 469), and in the barley extract, catechin, prodelphinidin B3 (m/z 593), procyanidin B3 (m/z 577), and procyanidin C2 (m/z 865).  相似文献

Micro two-dimensional separations were performed on polar bonded stationary phases of the type cyanopropyl-silica using non-aqueous eluents (polar modifier dissolved in n-heptane) as the first direction eluents and aqueous eluents (organic modifier—MeOH dissolved in water) as the second direction eluents. The chromatographic process was performed in micro scale using 5 × 5 cm plates, small volumes of eluents and 10 μL of plant extracts to obtain satisfying separation. Plates developed in horizontal chambers were dried and observed in UV light (254 nm and 366 m) photographed by digital camera and derivatized by DPPH to detect antioxidants (free radical scavengers) or derivatized by Naturstoff reagent to detect phenolic compounds (characteristic luminescence of some phenolic compounds). The above experiments give the possibility to construct fingerprints for investigated Polygonum hydropiper, Betula verrucosa and Pulmonaria officinalis extracts. It can be used in quality control of the plant material and its antioxidative activity. Novelty of the paper is the micro-scale of the separation by two-dimensional thin layer chromatography mode. For the first time two-dimensional separation of plant extracts on 5 × 5 cm plates in two directions is performed.  相似文献

A rapid, simple and reliable analytical procedure has been developed for the determination of polyphenols, including chlorogenic acid, rutin and scopoletin, in tobacco samples. After extraction, the polyphenols were analyzed by micellar liquid chromatography coupled with UV detection at 340 nm without sample preparation step. The compositions of the micellar mobile phase for the separation of these polyphenols were selected adequately by a chemometric method. The optimal resolution and satisfactory peak shapes were achieved with 0.022 mol L?1 sodium dodecyl sulfate and 0.45% (volume fraction) 1-propanol solution at pH 5.0 using Eclipse XDB-C18 (250 mm × 3.0 mm, 5 μm) column. The limits of detection for chlorogenic acid, rutin and scopoletin were 0.15, 0.29 and 0.22 μg mL?1, respectively. This validated method was successfully applied to qualitatively and quantitatively analyze the polyphenols in tobacco in less than 10 min.  相似文献

The antimicrobial activity of acetone, methanol and aqueous extracts of Asphodeline anatolica E. Tuzlaci leaves was evaluated against American type culture collection, food and clinical isolates (Listeria monocytogenes and Staphylococcus aureus including methicillin-resistant strains-MRSA). Biofilm formation, toxicity and characterisation of the polyphenolic content were analysed. The acetone extract demonstrated a higher antibacterial activity against S. aureus including MRSA strains, L. monocytogenes and Pseudomonas aeruginosa than against other extracts. No effect was observed in biofilm formation. The extracts resulted non-toxic against Artemia salina Leach. The phytochemical screening of extracts indicated that they mainly contained six polyphenols identified as catechin 3-O-gallate, protocatechuic acid, diosmin, rutin, cirsimaritin and kaempferol glucoside. This study is the first report on antimicrobial activity and phenolic content of A. anatolica and contributes to enrich the literature data on the biological properties of this plant. A. anatolica leaves have a potential as source of natural antimicrobial compounds.  相似文献
