The lipophylic extracts of three Viscum album subspecies growing on different host plants in Turkey were comparatively analyzed for derived methyl esters of their fatty acids by capillary gas chromatography-mass spectrometry (GC-MS). The sample of V. album ssp. album growing on apricot trees was remarkably rich in palmitic acid (11.47%). Arachidic acid was found in only ssp. austriacum and ssp. abietis samples. The unsaturated fatty acids, mainly oleic and linoleic acids, were identified in nine lipophylic extracts obtained from V. album samples belonging to ssp. album. The amounts of linoleic and oleic acid were the highest in the sample of ssp. album growing on apricot trees (12.18 and 9.19%, respectively). The fruit of Canarium album, also called Chinese olive, is a popular food and traditional Chinese herb. The ethyl acetate extract of Canarium album fruits was found to exhibit an inhibitory effect on six-helix bundle formation of the human immune deficiency virus (HIV) glycoprotein transmembrane subunit gp41 in our previous study. In this paper, a preparative separation of anti-HIV components from an ethyl acetate extract of Canarium album fruits was performed by high-speed counter-current chromatography, using a solvent system of n-hexane–ethyl acetate–ethanol–0.5% acetic acid aqueous (0.5:9.5:2:8, v/v/v/v). Among the six fractions obtained from a single separation process, three exhibited over 90% purity as determined by HPLC, two compounds were identified as 5-hydroxymethylfurfural and 3,4,5-trihydroxybenzoic acid, and four fractions displayed significant inhibitory effects on HIV-1 IIIB infection. These findings suggest that the high-speed counter-current chromatography is an effective and reliable technique for separating bioactive components from medicinal herbs. Further identification of the active compounds in Canarium album fruits and studying their mechanism of action are warranted.  相似文献

Abstract

Chemical characteristics of novel seed oils, yet not investigated, from three endemic Arecaceae (palm) species from Reunion Island are described. Fatty acid profiles are performed using two-dimensional gas chromatography-mass spectrometry. Carotenoid contents are determined by high performance liquid chromatography-mass spectrometry. The results of the investigations emphasize the particular composition of the unconventional red seed oil from Hyophorbe indica. Characteristic features of this oil reveal a high degree of unsaturation (50% of polyunsaturated fatty acids, with a high content (17%) of omega-3), which is possibly a unique fatty acid composition in the Arecaceae family. The two other palm oils from Dictyosperma album and Latania lontaroides contain high level of saturated fatty acids very similar to that of the edible palm oil. H. indica oil is also very rich in valuable carotenoids; in particular, lutein, β-carotene and lycopene are detected in a high content (respectively 45, 23 and 35?mg.kg?1 in oil).  相似文献

The content of plant secondary metabolites is not stable, and factors such as the region/location effect and seasonal variations have an impact on their chemical composition, especially in parasitic plants. Research in this area is an important step in the development of quality parameter standards of medicinal plants and their finished products. The effects of the time and place of harvest and the host tree species on the chemical composition and antioxidant activity of mistletoe extracts were investigated. Statistical tools were used to evaluate the results of the spectrophotometric and LC-ESI-MS/MS studies of the phenolic composition and antioxidant activity. The investigations indicate that the qualitative and quantitative composition, influencing the biological activity of mistletoe extracts, largely depends on the origin of the plant. The mistletoe extracts exhibited a rich phenol profile and high antioxidant activity. The chemometric analysis indicated that mistletoe collected from conifers (Viscum abietis and Viscum austriacum) had the most advantageous chemical composition and antioxidant activity. Moreover, the chemical profile and biological activity of the plant material were closely related to the climatic conditions and location of the harvested plant. Higher levels of phenolic compounds and high antioxidant activity were found in extracts obtained from plant material collected in cold weather with the presence of snow and less sunshine (autumn–winter period).  相似文献

A new β-tetralonyl glucoside, methylberchemiaside (1), along with five known compounds (2–6) were isolated from a fungus Colletotrichum sp. GDMU-1 derived from the leaves of Santalum album. Their structures were determined by detailed analysis of spectroscopic data. All compounds were tested for the inhibitory effects on the nitric oxide (NO) production in lipopolysaccharide (LPS)-treated RAW264.7 cells. Among them, compounds 4 and 5 showed moderate anti-inflammatory activity with IC50 value of 30.4 and 8.9 μM, respectively.  相似文献

Sandalwood oils are highly desired but expensive, and hence many counterfeit oils are sold in high street shops. The study aimed to determine the content of oils sold under the name sandalwood oil and then compare their chromatographic profile and α- and β santalol content with the requirements of ISO 3518:2002. Gas chromatography with mass spectrometry analysis found that none of the six tested “sandalwood” oils met the ISO standard, especially in terms of α-santalol content. Only one sample was found to contain both α- and β-santalol, characteristic of Santalum album. In three samples, valerianol, elemol, eudesmol isomers, and caryophyllene dominated, indicating the presence of Amyris balsamifera oil. Another two oil samples were found to be synthetic mixtures: benzyl benzoate predominating in one, and synthetic alcohols, such as javanol, polysantol and ebanol, in the other. The product label only gave correct information in three cases: one sample containing Santalum album oil and two samples containing Amyris balsamifera oil. The synthetic samples described as 100% natural essential oil from sandalwood are particularly dangerous and misleading to the consumer. Moreover, the toxicological properties of javanol, polysantol and ebanol, for example, are unknown.  相似文献

The bioassay-oriented fractionation of mistletoe crude extracts (MCEE) using 75% ethanol and culture products of mistletoe transformed by Rhodobacter sphaeroides, a photosynthetic bacterium (PSBT), revealed that the high cytotoxic activities were due to the petroleum ether extracts (PEs) and the acid-precipitated proteins from the aqueous extracts (AQs) of MCEE and PSBT. The isolated triterpenes may account for the activities of the PEs of MCEE and PSBT, respectively. Extraction of MCEE using petroleum ether led to the isolation of 3-epi-betulinic acid (1), betulonic acid (2), oleanolic acid (3), and β-amyrin acetate (4), while petroleum ether extraction of PSBT led to the isolation of 1,3,4,betulinic acid (5), erythrodiol (6), and (3β)-olean-12-ene-3,23-diol (7). The PE of PSBT exerted higher cytotoxicity than the PE of MCEE, which was due to the different triterpene contents of these two extracts. The cytotoxic activities of all compounds were tested, and the results revealed that compounds 1, 2, 3, 5, 6, and 7 contributed significantly to the cytotoxicities of both PEs. The AQ of the PSBT exerted almost the same cytotoxic activity and lower toxicity compared to the AQ of the MCEE. These findings indicate that mistletoe products biotransformed by R. sphaeroides could be used to treat cancers, since they have lower toxicities and higher antitumor activities compared to standard treatments.  相似文献

The aim of this study was to conduct a histochemical analysis to localize lipids, terpenes, essential oil, and iridoids in the trichomes of the L. album subsp. album corolla. Morphometric examinations of individual trichome types were performed. Light and scanning electron microscopy techniques were used to show the micromorphology and localization of lipophilic compounds and iridoids in secretory trichomes with the use of histochemical tests. Additionally, the content of essential oil and its components were determined using gas chromatography-mass spectrometry (GC-MS). Qualitative analyses of triterpenes carried out using high-performance thin-layer chromatography (HPTLC) coupled with densitometric detection, and the iridoid content expressed as aucubin was examined with spectrophotometric techniques. We showed the presence of iridoids and different lipophilic compounds in papillae and glandular and non-glandular trichomes. On average, the flowers of L. album subsp. album yielded 0.04 mL/kg of essential oil, which was dominated by aldehydes, sesquiterpenes, and alkanes. The extract of the L. album subsp. album corolla contained 1.5 × 10−3 ± 4.3 × 10−4 mg/mL of iridoid aucubin and three triterpenes: oleanolic acid, β-amyrin, and β-amyrin acetate. Aucubin and β-amyrin acetate were detected for the first time. We suggest the use of L. album subsp. album flowers as supplements in human nutrition.  相似文献

In this study, vic‐dioxime ligand, (1E,2E)‐2‐(hydroxyimino)‐N′‐[(1E)‐2‐oxo‐2‐phenylethylidene]ethanehydroximohydrazide (LH2), and its Cu (II) and Ni (II) transition metal complexes were synthesized and characterized using analytical and spectroscopic techniques. Furthermore, in vitro cytotoxic and apoptotic effects of this vic‐dioxime ligand and its Cu (II) and Ni (II) complexes on Caco‐2 heterogeneous human epithelial colorectal adenocarcinoma cells were evaluated. The effect of the vic‐dioxime ligand and its Ni (II) and Cu (II) complexes in combination with Campto on the cells was also investigated. The cytotoxicity test was carried using the MTT assay, and the apoptotic effect was tested by DNA diffusion assay. Campto was used as a standard anti‐cancer drug, Caco‐2 cancer cells treated with dimethylsulfoxide acted as solvent control, and human peripheral lymphocytes were used as control. The ligand and its complexes exhibit concentration‐dependent cytotoxic and apoptotic behavior. The ligand induces the weakest cytotoxic and apoptotic effects on both Caco‐2 cancer cells and lymphocytes. The Ni (II) complex of ligand induces high cytotoxic and apoptotic effects on both Caco‐2 cancer cells and lymphocytes. The Cu (II) complex of ligand has high cytotoxic and apoptotic effects on Caco‐2, but weak cytotoxic and apoptotic effects on lymphocytes. The cytotoxic and apoptotic effects of the ligand and its Ni (II) and Cu (II) complexes were found to be concentration dependent, i.e. the higher the concentration is the more cytotoxic it will be. The present findings suggest that Cu (II) complex has the potential to act as a promising anti‐cancer compound against Caco‐2 colon cancer cells.  相似文献

A new HPLC-DAD method was developed and validated for simultaneous determination of six main phenylethanoid glycosides (calceolarioside D, neocalceolarioside D, verbascoside, isoverbascoside, leucoseptoside A and martynoside) in the aerial parts of four Scutellaria L. taxa from flora of Turkey. All standard compounds showed a good linearity (R 2 > 0.999) in a relatively wide concentration range (1–120 μg/mL). The LOD of the compounds was in the range of 0.104–1.295 μg/mL and the LOQ was in the range of 0.450–2.536 μg/mL. The recoveries of the selected compounds were calculated in the range of 97.46–117.85%. The amounts of the phenylethanoid glycosides showed variation in the extracts. The developed method was found to be accurate, precise and reproducible, and successfully applied to identify and quantify the phenylethanoid glycoside composition of Scutellaria species.  相似文献

Calotropis procera (family: Asclepiadaceae) contains cardiac glycosides which are cytotoxic to cancer cells. The extracts of C. procera have been reported to be cytotoxic to many cancer cell lines and this is the first report against the human skin melanoma cells (SK-MEL-2). The SK-MEL-2 cells treated with C. procera methanolic extract (CPME) were analysed for growth inhibition and apoptosis. The exposure of phosphatidylserine in apoptotic SK-MEL-2 was analysed by using the Annexin-V FITC flow cytometry method. In CPME-treated SK-MEL-2 cells, 19.6% of apoptotic and 58.3% dead cells were observed. The 15.97% and 15.85% of early apoptotic cells were found at 20 μg/mL of the ouabain and paclitaxel, respectively. Active caspases, nuclear degradation confirmed apoptotic SK-MEL-2 cells in time- and dose-dependent manner. The cell cycle analysis shows that CPME treated cells halt at G2/M phase. Significant cytotoxic activity of CPME against SK-MEL-2 may be attributed to its high cardenolide content.  相似文献

The cellulose system of the viscous fibrous cellulosic polysaccharide (viscan) in the viscin tissue of the European mistletoe, Viscum album L., was analyzed by chemical and physicochemical techniques including sugar analysis, optical and transmission electron microscopy, X-ray and electron diffraction together with solid state CP/MAS 13C-NMR spectroscopy. The results confirmed that in the elongated thin viscin cells, the cellulose microfibrils (having a diameter of around 3 nm) were tightly coiled with their axes perpendicular to the long axis of the cell. Upon stretching these cells became deformed by more than a hundred fold. In such a deformation, the cellulose microfibrils became unwound to be perfectly aligned along the stretching direction. Based on solid-state CP/MAS 13C-NMR spectroscopic analysis of the viscin tissue, it was found that its cellulose consisted of I and I polymorphs in the ratio 1:1.  相似文献

A method to identify anticancer compounds in plants was proposed based on the hypothesis that these compounds are primarily present in plants to provide them with an ecological advantage over neighboring plants and other competitors. According to this view, identifying plants that contain compounds that inhibit or interfere with the development of other plant species may facilitate the discovery of novel anticancer agents. The method was developed and tested using Magnolia grandiflora, Gynoxys verrucosa, Picradeniopsis oppositifolia, and Hedyosmum racemosum, which are plant species known to possess compounds with cytotoxic activities. Plant extracts were screened for growth inhibitory activity, and then a thin-layer chromatography bioautography assay was conducted. This located the major antileukemic compounds 1, 2, 4, and 5 in the extracts. Once the active compounds were located, they were extracted and purified, and their structures were determined. The growth inhibitory activity of the purified compounds showed a significant correlation with their antileukemic activity. The proposed approach is rapid, inexpensive, and can easily be implemented in areas of the world with high biodiversity but with less access to advanced facilities and biological assays.  相似文献

Natural and synthetic β-lactam derivatives constitute an interesting class of compounds due to their diverse biological activity. Mostly used as antibiotics, they were also found to have antitubercular, anticancer and antidiabetic activities, among others. In this investigation, six new 3,3-dichloro-β-lactams prepared in a previous work were evaluated for their hemolytic and cytotoxic properties. The results showed that the proposed compounds have non-hemolytic properties and exhibited an interesting cytotoxic activity toward squamous cell carcinoma (A431 cell line), which was highly dependent on the structure and concentration of these β-lactams. Among the molecules tested, 2b was the most cytotoxic, with the lowest IC50 values (30–47 µg/mL) and a promising selectivity against the tumor cells compared with non-tumoral cells.  相似文献

Chenopodium album is a weedy annual plant in the genus Chenopodium. C. album pollen represents a predominant allergen source in Iran. The main C. album pollen allergens have been described as Che a 1, Che a 2, and Che a 3. The aim of this work was to clone the Che a 1 in Escherichia coli to establish a system for overproduction of the recombinant Che a 1 (rChe a 1). In order to clone this allergen, the pollens were subjected to RNA extraction. A full-length fragment encoding Che a 1 was prepared by polymerase chain reaction amplification of the first-strand cDNA synthesized from extracted RNA. Cloning was carried out by inserting the cDNA into the pET21b (+) vector, thereafter the construct was transformed into E. coli Top10 cells and expression of the protein was induced by IPTG. The rChe a 1 was purified using histidine tag in recombinant protein by means of Ni–NTA affinity chromatography. IgE immunoblotting, ELISA, and inhibition ELISA were done to evaluate IgE binding of the purified protein. In conclusion, the cDNA for the major allergen of the C. album pollen, Che a 1, was successfully cloned and rChe a 1 was purified. Inhibition assays demonstrated allergic subjects sera reacted with rChe a 1 similar to natural Che a 1 in crude extract of C. album pollen. This study is the first report of using the E. coli as a prokaryotic system for Che a 1 cloning and production of rChe a 1.  相似文献

The total content of polyphenols and flavonoids determined in the same plant and their corresponding antioxidant activities may vary widely, depending on the extraction conditions applied. This study was conducted to optimise the extraction conditions of phenolics and flavonoids from the mistletoe plant. Various extraction methods, i.e. ultrasound-assisted extraction technology, maceration, maceration with stirring, accelerated solvent extraction (ASE), and extraction under reflux were evaluated for their percentage extraction of polyphenols (TPC) and flavonoids (TFC) from Viscum album subsp. abietis. In addition, the anti-radical activity of extracts was analysed using the 2,2-diphenyl-1-picrylhydrazyl method. The effects of temperature, solvent type, and concentration on the phenolic extraction efficiency and antioxidant activity were studied using chemometric and statistical methods. The results showed that the extracts of V. album subsp. abietis contained large amounts of polyphenols and flavonoids (up to 57.673 mg g?1 and 9.955 mg g?1 of dry extract, respectively) and exhibited potent antioxidant activity, hence representing promising sources of powerful antioxidants. Due to its high extraction efficiency and considerable saving of time and solvent, ASE was more effective than the other extraction techniques. Extracts prepared with water-polar solvent mixtures displayed the highest TPC, TFC, and antioxidant activity, while organic polar solvents were the least efficient extractants.  相似文献

Summary. 3,4-Dihydroisoquinoline-Reissert-compounds were alkylated to 1-benzyl- and 1-picolyl-derivatives, which in turn could selectively be hydrolized yielding various carboxylic acids, among others certain amino acids related to 3,4-deoxynorlaudanosoline carboxylic acid (DNLCA). These on treating with ethanolic KOH underwent cyclization to dibenzoquinolizine- and isoquinonaphthyridine-13a-carboxylic acids. Alternatively this cyclization also could be achieved by a more convenient one-pot procedure starting from the same dihydro-Reissert-compounds. Aim of this study was to identify pentacyclic triterpenoids betulinic acid (BA), oleanolic acid (OA) and ursolic acid (UA) from five Swertia species endemic to Western Ghats, which are used as substitutes as well as adulterants to Swertia chirayita. Our results indicate that the concentration of active compound vary largely among and within the species from different localities. OA was found in a higher amount in all species compared to BA and UA. From the results, it was clear that BA, OA and UA are present in the endemic species collected from Western Ghats, thus advocating the use of these species as alternate sources to S. chirayita. This in due course may release pressure of exploitation from natural resources of S. chirayita and help to bring it out from an endangered category from conservation point of view.  相似文献

Seed oils of 15Sideritisspecies collected from different regions in Turkey(S. athoa, S. brevidens, S. caesarea, S. condensata, S. congesta, S. dichotoma, S. erythrantha var. cedretorum, S. germanicopolitana ssp. germanicopolitana, S. hololeuca, S. lanata, S. libanotica ssp. violascens, S. lycia, S. niveotomentosa, S. perfoliata, S. phrygia, S. pisidica)were obtained by a Soxhlet apparatus using hexane. The oil yields were found to be between 5.6-36.3%. Fatty acids in the oils were converted to methyl esters and their compositions were determined by GC/MS. The main fatty acid components of the oils from all the species are linoleic (45.4-64.0%), oleic (12.3-26.5%), 6-octadecynoic (4.5-26.8%), palmitic (0.3-9.4%), and linolenic (0.8-2.0%) acids.  相似文献

Plinia cauliflora is a plant species with edible fruits but its chemical composition is not totally known yet although former studies showed its potential as antifungal agent. This work aimed the chemical analysis of the leaves, the activity against fungi species and evaluated cytotoxicity. Extract was obtained with 70% ethanol. An ethyl acetate fraction was obtained and glycosylated quercetin and myricetin were isolated. Samples were tested against Candida species, dermatophytes and entomopathogenic fungi. Cytotoxicity was evaluated against fibroblast cells. Extract showed good activity against C. albicans (minimum inhibitory concentration at 156 μg/mL), C. parapsilosis (78 μg/mL), C. krusei (19 μg/mL), Trichophyton rubrum (78 μg/mL) and Microsporum canis (156 μg/mL). Isolated compounds were more active against C. krusei and T. rubrum. The extract, which was the more active sample, demonstrated low cytotoxic effect and encourage more studies against rising non-albicans species and dermatophyte T. rubrum.  相似文献
