In vitro anti-tumour and anti-radical activities of the acetone extract of the freshwater sponge Ochridaspongia rotunda were the subject of this study. The extract was found to be highly cytotoxic to human lung tumour cell line A-549 reaching IC50 value of 5.01 ± 0.21 μg/mL. Indeed, it displayed only 2-fold less anti-tumour activity than doxorubicin (IC50 value 2.42 ± 0.13 μg/mL) used as a positive control. The same extract was also found to be almost 37-fold more selective against A-549 vs. MRC-5 (normal) lung cells, in difference to weak selectivity of doxorubicin (less than 3-fold). Its profound anti-DPPH radical activity comparable to that of quercetin (IC50 values 3.68 ± 0.19 and 3.14 ± 0.09 μg/mL, respectively) coupled with no signs of genotoxicity in the comet assay (MRC-5 cell line, vs. doxorubicin) has actually implicated the importance of this animal bioresource in searching for pharmaceutically useful bioactive compounds of natural origin.  相似文献

The dark reduction kinetics of micromolar concentrations of Fe(III) in aqueous solution were studied in the presence of millimolar concentrations of ferrozine (FZ) over the pH range 4.0–7.0. A pseudo-first-order kinetics model was used to describe Fe(III) reduction at pH 4.0 and 5.0, and the reduction rate decreased with increasing pH or initial Fe(III) concentration. A more molecular-based kinetics model was developed to describe Fe(III) reduction at pH 6.0 and 7.0. From this model, the intrinsic rate constants (k1) of Fe(III) reduction by FZ in the dark were obtained as 0.133 ± 0.004 M?1 s?1 at pH 6.0 and 0.101 ± 0.009 M?1 s?1 at pH 7.0. It was also found in this model that a higher pH, a higher concentration of Fe(III), a lower concentration of FZ and less incubation time led to a lower fraction of Fe(III) reduction by FZ in the dark.  相似文献

Our quest to isolate and characterize natural products with in vitro solid tumor selectivity is driven by access to repositories of Indo-Pacific sponge extracts. In this project an extract of a species of Haplosclerida sponge obtained from the US NCI Natural Products Repository displayed, by in vitro disk diffusion assay (DDA) and IC50 determinations, selective cytotoxicity with modest potency to a human pancreatic cancer cell line (PANC-1) relative to the human lymphoblast leukemia cell line (CCRF-CEM). Two brominated indoles, the known 6-bromo conicamin (1) and the new derivative, 6-Br-8-keto-conicamin A (2), were identified and 2 (IC50 1.5 μM for the natural product vs 4.1 μM for the synthetic material) was determined to be responsible for the cytotoxic activity of the extract against the PANC-1 tumor cell line. The new natural product and ten additional analogs were prepared for further SAR testing.  相似文献

In the last years, several studies have recently evaluated the beneficial effects of red orange juice (Citrus sinensis (L.) Osbeck) and its active components in weight management and obesity. Moro orange is a cultivar of red orange, particularly rich in active compounds such as anthocyanins, hydroxycinnamic acids, flavone glycosides and ascorbic acid, which displays anti-obesity effects in in vitro and in vivo studies. In this clinical study, the effect of a Moro juice extract (Morosil®, 400 mg/die) supplementation was evaluated in overweight healthy human volunteers for 12 weeks. Results showed that Moro juice extract intake was able to induce a significant reduction in body mass index (BMI) after 4 weeks of treatment (p < 0.05). Moreover, in subjects treated with Moro extract, body weight, BMI, waist and hip circumference were significantly different from the placebo group (p < 0.05). In conclusion, it could be suggested that the active compounds contained in Moro juice have a synergistic effect on fat accumulation in humans and Moro juice extract can be used in weight management and in the prevention of human obesity.  相似文献

The antioxidant, antiproliferative and cytotoxic activities against different human cancer cells were investigated in local and recently introduced plants of Mentha sp., Rosmarinus officinalis L. (ROL) and Origanum majorana L. (OML). ROL exhibited the highest antioxidant activities (IC50 8.4 ± 0.2 μg/mL) followed by OML and mint species such as Mentha suaveolens ‘apple mint’ and Mentha longifolia L. exhibiting moderate antioxidant activities. HPLC analysis of leaf extract revealed that rosmarinic acid is the main component followed by caffeic acid. Herbal leaf extracts varied in their proliferation inhibition and cytotoxicity against HeLa, MCF-7 and Jurkat cancer cells in a dose-dependent matter. The highest antiproliferative inhibition and cytotoxic activity were detected in ROL and OML followed by mint. Local herbs might have a potential role as anticancer natural medicines in addition to their high antioxidant activities due to the presence of different phenolics in their aqueous tea extracts.  相似文献

The dispersion and agglomeration behavior of flaky graphite particles in an aqueous solution was studied. The small particles of flaky graphite naturally formed a large and compact agglomeration in an aqueous solution at pH 7.0 and 9.0, respectively. The theoretical calculation result based on typical DLVO theory was opposite the phenomenon observed by optical microscope, indicating that the interaction force between the flaky graphite particles exceeded the van der Waals and electrostatic double-layer forces. When the extended DLVO theory was used, the total interaction energy (UT = UA + UR + UHI) was negative over most of the distance range, indicating that the agglomeration of the small particles was attributed to the attraction. This result indicated that hydrophobic interactions dominated the dispersion and agglomeration behavior of the hydrophobic flaky graphite particles.  相似文献

Acacia ligulata A.Cunn. ex Benth. (Fabaceae: Mimosoideae) is a native Australian plant used traditionally by Australian Aboriginal groups. This study was undertaken to investigate the bioactivity of A. ligulata extracts and to evaluate their chemical composition. Potential antibacterial, cytotoxic and enzyme inhibitory effects relevant to traditional medicinal and food uses of the species were examined and LC-MS/MS was performed to investigate the chemical composition. Antibacterial activity was observed for bark and leaf extracts with an MIC for the bark extract of 62.5 μg/mL against Streptococcus pyogenes. Pod extracts showed cytotoxic effects against cancer cells, with the highest activity against melanoma SK-MEL28 cells with IC50 values between 40.8 and 80.6 μg/mL. Further, the leaf and pod extracts also inhibited α-amylase EC-3.2.1.1 and α-glucosidase EC-3.2.1.20 with IC50 values between 9.7–34.8 and 12.6–64.3 μg/mL, respectively. The LC-MS/MS profiling indicated that several different saponins were present in the active extracts.  相似文献

The aim of this work is the phytochemical study of the butanolic extract of the aerial parts of Veronica rosea. Four compounds 14 have been isolated using different chromatographic methods. The structures of these compounds were determined by NMR spectral analysis and mass spectroscopy. The adsorption and anticorrosion effects of this extract were investigated towards the corrosion of copper in 1 M HNO3 aqueous by the weight loss technique and potentiodynamic polarization. The results showed that the butanolic extract is a good inhibitor and the inhibition efficiency increases with increasing of concentration of the inhibitor. The adsorption of this extract on the copper specimen surface was spontaneous and obeyed the Langmuir’s adsorption isotherm. Large value of adsorption equilibrium Constant (K ads = 35 L g?1) was obtained. The polarization experiments confirmed the data obtained by gravimetric weight-loss. Tafel plot of polarization curves indicates that the extract acts as a mixed type inhibitor.  相似文献

Brassica oleracea L. var. capitata f. rubra (Red Cabbage) dye is composed mainly of natural pigment called anthocyanins used as a natural colourant. Wool and silk fibres were dyed with the aqueous extract obtained from red cabbage. The dyeing process was investigated and the combined effects of dyeing conditions on the colour yield parameter (K/S) were studied. Resulted fastness to wash, rubbing and light of the dyed fabrics were evaluated. Chemical Oxygen Demand (COD) and Biological Oxygen Demand (BOD5) of the residual effluent were measured. Best dyeing conditions were found to be: 50 g/100 mL, pH 2, 60 min and 100 °C, respectively, for the red cabbage weight, pH, dyeing duration and temperature. Good fastnesses properties were found in both cases: for wool and silk fabrics. It was found also that the calculated biodegradability ratio (COD/BOD5) of the residual bath of dyeing wool and silk with red cabbage extract are lower than 1.5 which means that these baths are biodegradable.  相似文献

This study was carried out to determine the cytotoxic effect of seven plant extracts and the isolated compounds – syringin and 4-methoxycinnamyl alcohol – on cancerous and non-cancerous cells. The ethanol extract of Foeniculum vulgare was found to exhibit the most significant toxicity with an IC50 value of 19.97 μg/mL on HeLa cells. Bioassay-guided fractionation led to the isolation of two compounds, syringin (1) and 4-methoxycinnamyl alcohol (2). Both compounds showed toxicity against MCF-7, HeLa and DU145 cancer cell line. The results showed that compound 2 showed high toxicity against all the cancer cell lines with IC50 values of 14.24, 7.82 and 22.10 μg/mL, respectively. 4-Methoxycinnamyl alcohol also showed no apoptotic effect in cell cycle analysis after 48 h at a concentration of 10 μg/mL. However, DNA fragmentation study revealed that necrosis took place at a concentration of 10 μg/mL after 48 h exposure.  相似文献

A preliminary study on Echinops giganteus (Asteraceae) showed that the methanolic extract has interesting cytotoxicities against a panel of cancer cell lines. From this extract, a lignan, a flavonoid and a polyacetylenic thiophene identified were three times less cytotoxic than the extract. In the search of the metabolites responsible for the bioactivity, a new harvested E. giganteus was subjected to a phytochemical study using chromatographic methods. In the course of the work, two new compounds: a brominated oleanolide (1) and a tetrahydrofurano-ceramide (2) were obtained along with β-amyrin acetate (3), 2-(penta-1,3-diynyl)-5-(4-hydroxybut-1-ynyl)-thiophene (4), 2-(penta-1,3-diynyl)-5-(3,4-dihydroxybut-1-ynyl)-thiophene (5) and 4-hydroxy-2,6-di-(3′,4′-dimethoxyphenyl)-3,7-dioxabicyclo-(3.3.0)octane (6). Their structures were determined on the basis of NMR spectroscopy and mass spectrometry data in conjunction with those reported in the literature. The cytotoxicity of 1, 2 and 5 was evaluated by employing resazurin assay against a panel of cancer cell lines with IC50 values in range 6.12 ± 0.46–46.96 ± 3.61 μM.  相似文献

A sensitive and rapid liquid chromatographic method was successfully developed and validated for the determination of sibutramine hydrochloride in bulk and capsules. Sibutramine in the presence of its degradation products was analyzed using UV detection at 225 nm. Chromatography was performed on a reversed-phase C8 (150 × 4.0 mm I.D., 5 μm) analytical column under isocratic conditions. The mobile phase was composed of acetonitrile:water (aqueous phase containing 0.3% triethylamine and pH adjusted to 7.0) (75:25, v/v) at a flow-rate of 1.1 mL min−1. No chromatographic interference was found during the analysis. Light was the stress condition which most contributed to sibutramine degradation. The method showed a linear response (r > 0.999) from 30 to 90 μg mL−1. The mean recovery for capsules was 101.2%. Inter-day assays showed relative standard deviations of 0.42 and 1.62% for bulk and capsules, respectively. The developed method is able to separate sibutramine from its major degradation products and it may be used in the quality control of this active pharmaceutical ingredient in both bulk and capsules.

Ferula gummosa Boiss. has medicinal applications in treating a wide range of diseases including cancer. The objective of this study was to evaluate the antiproliferative activities of the seed and gum extracts of F. gummosa as well as to study the effect of the potent extract on the induction of apoptosis and cell cycle arrest. Our results demonstrated that the ethanolic extract had the lowest IC50 value at 72 h (0.001 ± 1.2 mg/mL) in BHY cells. Moreover, flowcytometry and annexin-V analysis revealed that the ethanolic extract induced apoptosis and cell-cycle arrest in BHY cells at G1/S phase. In addition, colorimetric methods exhibited the highest amount of total phenolics and flavonoids in the aqueous and gum extracts (0.12 ± 0.037, 0.01 ± 2.51 mg/g of dry powder). Generally, the results obtained indicate that F. gummosa ethanol extract may contain effective compounds which can be used as a chemotherapeutic agent.  相似文献

A new tailor-made colorimetric chemosensor 1, containing pyridine and benzothiazole moieties connected through an azo (–N = N–) linkage has been synthesised. In 9:1 (v/v) aqueous THF (pH 7.0 HEPES buffer), it showed a conspicuous naked-eye colour change upon binding to Ni2+ (colourless to light green) and AcO (colourless to orange) resulting in their ratiometric sensing. The cation and anion recognition property of the chemosensor 1 was monitored by UV–vis spectral analysis and 1H NMR titrations.  相似文献

The crude extracts of tubers of Aconitum spicatum (Bruhl) Stapf were investigated for in vitro antileishmanial activity against Leishmania major. The dichloromethane extract at pH 2.5 showed antileishmanial activity with IC50 value of 27.10 ± 0.0 μg/mL. Chromatographic purification of the dichloromethane extract led to isolation of three C-19 norditerpenoid alkaloids indaconitine (1), chasmaconitine (2) and ludaconitine (3). Compounds 3 and 2 showed antileishmanial activity with IC50 = 36.10 ± 3.4 and 56.30 ± 2.1 μg/mL, respectively. Compound 1 was less effective (IC50 > 100 μg/mL). The cytotoxicity of compounds 1, 2 and 3 studied against MCF7, HeLa and PC3 cancer cell lines and 3T3 normal fibroblast cell line did not show cytotoxicity at 30 μM.  相似文献

Nature is a generous source of compounds with the potential for prevention of infections. Antimicrobial screening of aqueous extract from bark of wild Himalayan cherry (Prunus cerasoides) was carried out against various pathogenic microorganisms with inhibition zone ranging from 19 to 24 mm. An optimization strategy, which included classical method and statistical method (RSM), was applied to optimize the effect of process variables. Fifteen percent plant material extracted at 40 °C for 60 min and at its natural pH (4.5) exhibited best antimicrobial activity with an average zone of inhibition ranging from 19 to 29 mm. Statistical optimization using RSM further enhanced the activity by 1.09–1.24 folds. Minimum inhibitory concentration of the aqueous extract against different microorganism ranged from 1 to 10 mg/ml. The aqueous extract was found to be reasonably thermostable at boiling temperature for 1 h. Viable cell count (VCC) studies of the extract showed it to be bactericidal in nature. Further, the aqueous extract was found to be neither cytotoxic nor mutagenic, when evaluated by MTT assay and Ames mutagenicity test. The results suggest that the aqueous extract of P. cerasoides could be a potential source to obtain new antimicrobials and effective herbal medicines to combat the problem of ever emerging microbial resistance.  相似文献

Ipomoea carnea Jacq. is an important folklore medicinal plant, assessed for its underexplored biological potential. Antioxidant, cytotoxic, antiproliferative and polyphenolic profile of whole plant was evaluated using various techniques. Maximum extract recovery (29% w/w), phenolic [13.54 ± 0.27 μg GAE/mg dry weight (DW)] and flavonoid (2.11 ± 0.10 μg QE /mg DW) content were recorded in methanol-distilled water (1:1) flower extract. HPLC-DAD analysis quantified substantial amount of six different polyphenols ranging from 0.081 to 37.95 μg/mg extract. Maximum total antioxidant and reducing potential were documented in methanol-distilled water and acetone-distilled water flower extracts (42.62 ± 0.47 and 24.38 ± 0.39 μg AAE/mg DW) respectively. Ethanol-chloroform root extract manifested highest free radical scavenging (IC50 of 61.22 μg/mL) while 94.64% of the extracts showed cytotoxicity against brine shrimps. Ethanol leaf extract exhibited remarkable activity against THP-1 cell line (IC50 = 8 ± 0.05 μg/mL) and protein kinases (31 mm phenotype bald zone).  相似文献

Five polar steroid derivatives, including one new glycosylated polyhydroxysteroid namely polyacanthoside A (1), were isolated from the water-soluble materials from the MeOH extract of the Vietnamese starfish Astropecten polyacanthus using various chromatographic separations. The structure elucidation was confirmed by spectroscopic experiments such as HR-ESI-MS, 1D and 2D NMR. Among the isolated compounds, (20R,24S)-3β,6α,8,15β,24-pentahydroxy-5α-cholestane (3) showed significant cytotoxic effect against five human cancer cell lines as HepG2, KB, LNCaP, MCF7 and SK-Mel2 with the IC50 values from 18.03 ± 2.63 to 21.59 ± 3.23 μM.  相似文献

A new stereoisomer Meso-araguspongine C together with nine reported macrocyclic bis-quinolizidine alkaloids araguspongines A, C, E, L, N?P, petrosin, and petrosin A were isolated from marine sponge Xestospongia muta. Stereochemistry of meso-araguspongine C (2) and araguspongines N-P (3?5) were established by their NMR data and conformational analyses. Both araguspongine C (1) and meso-araguspongine C (2) exhibited great cytotoxic activity towards HepG-2, HL-60, LU-1, MCF-7, and SK-Mel-2 human cancer cells (IC50 in the range of 0.43–1.02 μM). At a concentration of 20 μM, isolated compounds (1?10) also showed modest inhibitory effects (from 7.6 to 40.8%) on the NO production in LPS activated RAW264.7 macrophages.  相似文献

A nanoscale catalyst Fe0(FeNPs) supported on the natrolite zeolite nanoparticles (NANPs) is successfully synthesized and characterized by FT-IR, X-ray diffraction (XRD) and scanning electron microscopy (SEM) and thermogravimetric-differential thermal analysis (TG-DTA). The photodegradation of methyl orange (MO) is studied in aqueous suspension containing the catalyst under UV irradiation and H2O2. The effect of various reaction parameters such as initial dye concentration, irradiation time, pH, H2O2 concentration and catalyst dosage on the decolorization of methyl orange is investigated. The degradation study reveals that the reactivity of the catalysts is in order of: photo-NANPs–FeNPs–H2O2 > photo-NANPs–H2O2 > photo-NANPs–FeNPs > photo-H2O2 > NANPs–FeNPs–H2O2. The results show that methyl orange can be effectively decolorized by NANPs–FeNPs via the pseudo-first-order kinetic model.  相似文献
